Revise the revised? New dimensions of the neuroanatomical hypothesis of panic disorder.

In 2000, Gorman et al. published a widely acknowledged revised version of their 1989 neuroanatomical hypothesis of panic disorder (PD). Herein, a 'fear network' was suggested to mediate fear- and anxiety-related responses: panic attacks result from a dysfunctional coordination of 'upstream' (cortical) and 'downstream' (brainstem) sensory information leading to heightened amygdala activity with subsequent behavioral, autonomic and neuroendocrine activation. Given the emergence of novel imaging methods such as fMRI and the publication of numerous neuroimaging studies regarding PD since 2000, a c

Propriospinal pathways in the dorsal horn (laminae I-IV) of the rat lumbar spinal cord.

The spinal dorsal horn is regarded as a unit that executes the function of sensory information processing without any significant communication with other regions of the spinal gray matter. Within the spinal dorsal horn, however, the different rostro-caudal and medio-lateral subdivisions intensively communicate with each other through propriospinal pathways. This review gives an overview about these propriospinal systems, and emphasizes that the medial and lateral parts of the spinal dorsal horn show the following distinct features in their propriospinal interconnectivities: (a) A 100-300μm lo

Cortical area inducing chewing-like rhythmical jaw movements and its connections with thalamic nuclei in guinea pigs.

Repetitive electrical stimulation to the cortical masticatory areas (CMA) evokes rhythmical jaw movements (RJM), whose patterns vary depending on the stimulation site, in various species. In guinea pigs, although alternating bilateral jaw movements are usually seen during natural chewing, it is still unclear which cortical areas are responsible for chewing-like RJM. To address this issue, we first defined the cortical areas inducing chewing-like RJM by intracortical microstimulation. Stimulation of the most lateral area of the CMA, the granular cortex, induced chewing-like RJM, but from the re

Decreased pain inhibition in irritable bowel syndrome depends on altered descending modulation and higher-order brain processes.

Irritable bowel syndrome (IBS) is a functional gastrointestinal disorder involving abdominal pain and bowel dysfunction. IBS pain symptoms have been hypothesized to depend on peripheral and central mechanisms, but the pathophysiology is still unclear. The aim of the present study was to assess the contribution of cerebral and cerebrospinal processes to pain inhibition deficits in IBS. Fourteen female patients with diarrhea-predominant IBS (IBS-D) and 14 healthy female volunteers were recruited. Acute pain and the nociceptive withdrawal reflex (RIII reflex) were evoked by transcutaneous electri

Craving, context and the cortex.

A new study challenges the idea that the ventromedial prefrontal cortex inhibits drug relapse, by selectively inactivating a subpopulation of neurons in this brain area and showing attenuation of context-induced reinstatement of heroin seeking.

The hippocampal system as the cortical resource manager: a model connecting psychology, anatomy and physiology.

A model is described in which the hippocampal system functions as resource manager for the neocortex. This model is developed from an architectural concept for the brain as a whole within which the receptive fields of neocortical columns can gradually expand but with some limited exceptions tend not to contract. The definition process for receptive fields is constrained so that they overlap as little as possible, and change as little as possible, but at least a minimum number of columns detect their fields within every sensory input state. Below this minimum, the receptive fields of some colum

Neural circuits revealed by axon tracing and mapping cadherin expression in the embryonic chicken cerebellum.

Fiber connections of the cerebellar cortex are organized into distinct parasagittal domains. Each domain expresses a unique subset of various genes. Brain structures that are directly connected to the cerebellar cortex, such as the deep cerebellar nuclei and the inferior olivary nucleus, show a similarly differential pattern of connectivity and gene expression. For example, several members of the cadherin family of adhesion molecules are expressed differentially in the subdivisions of the cerebellar system in chicken and mouse. Little is known, however, about how the molecular maps in the diff

The involvement of the human cerebellum in eyeblink conditioning.

Besides its known importance for motor coordination, the cerebellum plays a major role in associative learning. The form of cerebellum-dependent associative learning, which has been examined in greatest detail, is classical conditioning of eyeblink responses. The much advanced knowledge of anatomical correlates, as well as cellular and molecular mechanisms involved in eyeblink conditioning in animal models are of particular importance because there is general acceptance that findings in humans parallel the animal data. The aim of the present review is to give an update of findings in humans. E

The role of the striatum in adaptation learning: a computational model.

To investigate the functional role of the striatum in visuo-motor adaptation, we extend the DIRECT-model for visuo-motor reaching movements formulated by Bullock et al.(J Cogn Neurosci 5:408-435,1993) through two parallel loops, each modeling a distinct contribution of the cortico-cerebellar-thalamo-cortical and the cortico-striato-thalamo-cortical networks to visuo-motor adaptation. Based on evidence of Robertson and Miall(Neuroreport 10(5): 1029-1034, 1999), we implement the function of the cortico-cerebellar-thalamo-cortical loop as a module that gradually adapts to small changes in sensori

Central vestibular disorders.

Dizziness or vertigo is an erroneous perception of selfmotion or object-motion as well as an unpleasant distortion of static gravitational orientation. It is caused by a mismatch between the vestibular, visual, and somatosensory systems. Thanks to their functional overlap, the three systems are able to compensate, in part, for each other's deficiencies. Thus, vertigo is not a well-defined disease entity, but rather a multisensory syndrome that results when there is a pathological dysfunction of any of the stabilizing sensory systems (e.g., central vestibular disorders, peripheral vestibular di

Overactive bladder in children. Part 1: Pathophysiology.

PURPOSE: Detrusor overactivity is the most common voiding dysfunction in children. Detrusor overactivity is also known as overactive bladder syndrome, urge syndrome, hyperactive bladder syndrome, persistent infantile bladder and detrusor hypertonia. It has become apparent that the ideas that had been used to dictate the management of this problem in children were based on the foundation that this is a primary bladder problem and/or a delay in maturation in the nervous system of children. The expectation that children would outgrow the problems led many pediatric urologists and other practition

Controlling inflammation: the cholinergic anti-inflammatory pathway.

Innate immune responses and inflammation are regulated in part by neural mechanisms. In the present paper, we summarize experimental evidence that reveals that innate immunity and inflammation are controlled by the vagus nerve, previously known as a regulator of other vital physiological functions. Activation of vagus nerve cholinergic signalling inhibits TNF (tumour necrosis factor) and other pro-inflammatory cytokine overproduction through 'immune' alpha7 nicotinic receptor-mediated mechanisms. This efferent vagus nerve-based 'cholinergic anti-inflammatory pathway' has been elucidated as a c

The role of the extrapersonal brain systems in religious activity.

The neuropsychology of religious activity in normal and selected clinical populations is reviewed. Religious activity includes beliefs, experiences, and practice. Neuropsychological and functional imaging findings, many of which have derived from studies of experienced meditators, point to a ventral cortical axis for religious behavior, involving primarily the ventromedial temporal and frontal regions. Neuropharmacological studies generally point to dopaminergic activation as the leading neurochemical feature associated with religious activity. The ventral dopaminergic pathways involved in rel

Central and peripheral sensitization in tension-type headache.

Recent studies on tension-type headache indicate that the nociceptive input to the central nervous system may be increased as a result of activation or sensitization of peripheral sensory afferents. Moreover, pain perception studies and pharmacologic studies strongly suggest that the central nervous system is sensitized in patients with chronic tension-type headache. The barrage of nociceptive impulses from the periphery may be responsible for the sensitization of second- and third-order neurons in the central nervous system. In this way, sensitization may play a role in the initiation and mai

Evolutionary mismatch, neural reward circuits, and pathological gambling.

Evolutionary mismatch theory has been applied to disorders of self-regulation such as maladaptive eating patterns and drug abuse. Modern gambling represents a refinement of the elements of risk and chance, which draw upon the faculties of judgment and novelty-seeking. A set of neuroanatomical structures, including prefrontal-subcortical systems and associated limbic structures, have been implicated in the processing of reward and punishment, including gambling-related situations. Neurobiological systems guiding choice and behavior have evolved to maximize chances for survival under hunter-gath

Quantification of gastric mill network effects on a movement related parameter of pyloric network output in the lobster.

It has long been known that gastric mill network activity (cycle period 5-10 s) alters pyloric network output (cycle period approximately 1 s), but these effects have not been quantified. Many pyloric muscles extract gastric mill timed variations in pyloric motor neuron firing, and consequently produce gastric mill timed movements even though no gastric mill neurons innervate them. Determining pyloric behavior therefore requires detailed description of gastric mill effects on pyloric neural output. Pyloric muscle activity correlates well with motor neuron overall spike frequency (OSF, burst sp

Follower neurons in lobster (Panulirus interruptus) pyloric network regulate pacemaker period in complementary ways.

Distributed neural networks (ones characterized by high levels of interconnectivity among network neurons) are not well understood. Increased insight into these systems can be obtained by perturbing network activity so as to study the functions of specific neurons not only in the network's "baseline" activity but across a range of network activities. We applied this technique to study cycle period control in the rhythmic pyloric network of the lobster, Panulirus interruptus. Pyloric rhythmicity is driven by an endogenous oscillator, the Anterior Burster (AB) neuron. Two network neurons feed ba

Processing of tactile and kinesthetic signals from bilateral sides of the body in the postcentral gyrus of awake monkeys.

Earlier studies pointed out that in the primary somatosensory cortex (SI) the receptive fields (RF) of bilateral neurons were related exclusively to the body midline. We recently found a substantial number of neurons with bilateral RFs on hand digits, shoulders/arms or legs/feet in the caudalmost part (areas 2 and 5) of the postcentral gyrus in awake macaque monkeys. The RFs of these neurons were generally of the most complex types found in this region of the cortex, and thus they were considered to be at the highest level along the hierarchical chain of information processing. We conclude tha

Spontaneous confabulation, reality monitoring, and the limbic system--a review.

Patients with anterior limbic damage may present a distinct syndrome, spontaneous confabulation: they fail in common memory tests, act on the basis of previous habits rather than currently relevant memories, produce confabulations composed of elements of past true events, are disorientated, and are absolutely convinced about the veracity of their perceived reality. Spontaneous confabulation is independent of other false memories, such as, provoked confabulations or illusory recognition. Studies showed that spontaneous confabulators fail to suppress (inactivate) evoked memories that do not pert
