A novel mesenchymal stem cell-targeting dual-miRNA delivery system based on aptamer-functionalized tetrahedral framework nucleic acids: Application to endogenous regeneration of articular cartilage.

Articular cartilage injury (ACI) remains one of the key challenges in regenerative medicine, as current treatment strategies do not result in ideal regeneration of hyaline-like cartilage. Enhancing endogenous repair via microRNAs (miRNAs) shows promise as a regenerative therapy. miRNA-140 and miRNA-455 are two key and promising candidates for regulating the chondrogenic differentiation of mesenchymal stem cells (MSCs). In this study, we innovatively synthesized a multifunctional tetrahedral framework in which a nucleic acid (tFNA)-based targeting miRNA codelivery system, named A-T-M, was used.

Highly-sensitive detection of Salmonella typhi in clinical blood samples by magnetic nanoparticle-based enrichment and in-situ measurement of isothermal amplification of nucleic acids.

Enteric fever continues to be a major cause of mortality and morbidity globally, particularly in poor resource settings. Lack of rapid diagnostic assays is a major driving factor for the empirical treatment of enteric fever. In this work, a rapid and sensitive method 'Miod' 'has been developed. Miod includes a magnetic nanoparticle-based enrichment of target bacterial cells, followed by cell lysis and loop-mediated isothermal amplification (LAMP) of nucleic acids for signal augmentation along with concurrent measurement of signal via an in-situ optical detection system. To identify positive/ne…

Scaffold RNA Reengineering for a Leakage-Suppressed CRISPR/Cas12a Cascade System Toward Precise Nucleic Acid Diagnostics.

Amplification-free CRISPR detection has attracted significant attention for its capacity to circumvent the intricacies of nucleic acid amplification. Yet, the persistent challenge of high background signals remains a critical bottleneck, motivating intensive efforts toward background suppression strategies. Here, we present a 3'-end DNA extension of scaffold RNA (3'D-SR) conformational switch to drive the cascade-activatable and leakage-suppressed Cas12a system, a mechanism we denote as CALCas. By engineering the crRNA of Cas12a system and introducing steric hindrance as an activation barrier,

Integrating Peroxidase-like Supramolecular Crystal with Nucleic Acid Amplification Cascades for Ultrasensitive Electrochemical Detection of Mucin 1.

Mucin 1 (MUC1) is an important biomarker for epithelial-derived malignancies, yet its ultratrace abundance in early tumorigenesis demands analytical methods with exceptional sensitivity. Herein, we report an ultrasensitive electrochemical aptasensing platform that integrates a biomimetic supramolecular crystal with target recycling amplification (TRA) and catalytic hairpin assembly (CHA). The supramolecular crystal (Fe-TCPP-SC) is rationally engineered by direct self-assembly of Fe(III) meso-tetra(4-carboxyphenyl)porphyrin (Fe-TCPP-(COOH)4) via hydrogen bonding, eliminating inactive metal node

A PfAgo-assisted artificial nucleic acid circuit for extraction-free and ultrasensitive detection of Staphylococcus aureus in dairy products.

Staphylococcus aureus is a major foodborne pathogen associated with dairy products; however, rapid and reliable detection remains challenging due to complex food matrices. Herein, we developed a one-step isothermal biosensing strategy based on an artificial nucleic acid circuit-assisted Argonaute system (PfAgo-ANCA) for rapid and sensitive detection of S. aureus. By targeting the species-specific nuc gene, the platform utilizes the high-temperature endonuclease activity of Pyrococcus furiosus Argonaute (PfAgo). The strategy integrates programmable PfAgo-mediated target recognition with downstr

Electrokinetic Nucleic Acid Amplification (E-NAAMP) Using Paper-PDMS Microfluidics and High-Frequency Joule Heating.

We present a novel paper-based nucleic acid amplification (NAA) technique using electrokinetic nucleic acid amplification (E-NAAMP). In E-NAAMP, a high radio frequency (RF) potential is applied across a conductive aqueous sample to induce an Ohmic current and drive the sample temperature to increase by Joule heating. Using this RF approach, we investigate the ability to induce E-NAAMP in pressurized paper-based microfluidic channels. We use the microfluidic pressure-in-paper (µPiP) method to encapsulate synthetic and natural fiber-based paper channels between thin sheets of polydimethylsiloxan

Peroxyoxalate chemiexcitation enables optical-excitation-free duplex end-point readout of fluorogenic nucleic acid assays.

Nucleic acid amplification tests (NAATs), including PCR, are considered the gold standard for pathogen detection. Amplification is commonly detected by fluorescence from probes or intercalating dyes, with internally-quenched probes most widely used for enhanced specificity. However, these methods require elaborate instrumentation for fluorescence detection, limiting their usability at point-of-care. We found that the chemical excitation of fluorophores, as used in consumer glow sticks, can be used to detect PCR probe hydrolysis with minimal instrumentation using a smartphone or simple digital

Engineering of a wireless single capacitive discharge conductivity-clamped gene electrotransfer probe for naked nucleic acid therapeutics.

Pulsed electric field gene electrotransfer addresses the need for safe, directed "vector-free" delivery of DNA/mRNA therapeutics in deep tissue targets. We report the development of a disposable needle-type "electro-lens" probe for precision delivery of DNA/mRNA to tissues via translocation of nucleic acids into cells using focused single-pulse electric fields. We developed and manufactured a novel ganged linear bionic electrode array enhanced by incorporating integrated microfluidics to deliver naked DNA expression cassettes in a low-conductivity carrier ("conductivity-clamping," CC). This sy…

Locked nucleic acid probes enable enzyme-free photoelectrochemical detection of double-stranded DNA cancer biomarkers.

The accurate detection of double-stranded DNA (dsDNA) cancer biomarkers remains a critical challenge in biosensing technologies, as native strand pairing hampers probe hybridization. While photoelectrochemical (PEC) biosensors hold promise for rapid and cost-effective nucleic acid diagnostics, their application has largely been confined to synthetic single-stranded DNA (ssDNA), limiting their clinical applicability. Here, we introduce an enzyme-free PEC biosensing strategy that directly detects DNA targets within their native double-stranded form by integrating locked nucleic acid (LNA) probes

Advances in Detecting Viable/Dead Foodborne Microorganisms Using Diverse Functional Nucleic Acid-Based Molecular Recognition.

Accurately detecting viable foodborne pathogenic bacteria is essential for food safety risk assessments and public health interventions. Traditional plate counting is time-consuming and operationally cumbersome. Immunological assays are unable to distinguish viable from dead cells, whereas conventional nucleic acid amplification is often affected by residual DNA originating from dead bacteria. These limitations render conventional approaches inadequate for rapid and precise field detection. Functional nucleic acids (FNAs) offer a promising alternative for viability detection because of their h…

A two-dimensional nanosheet-enhanced fluorescent biosensor with nucleic acid amplification for breast cancer-related miRNA detection.

Breast cancer remains a major global health burden with high incidence and mortality, necessitating advanced diagnostic solutions. While microRNAs (miRNAs) have emerged as promising biomarkers, the clinical translation faces a formidable obstacle: the lack of reliable detection techniques that are convenient, ultrasensitive, and highly specific enough for accurate diagnosis. To tackle these challenges, we presented an innovative strategy combined with catalyzed hairpin assembly (CHA) and a two-dimensional nanosheet for miRNA biomarker to enable precise breast cancer diagnosis (CHA-[Cu(tz)] sys

Magnetically Driven Dual-miRNA Framework Nucleic Acid Biosensing Platform for Precise Classification of Breast Cancer Subtypes.

The significant variation in treatment strategies among breast cancer subtypes establishes precise early subtyping as a critical prerequisite for effective therapy. While molecular profiling offers accurate classification, developing a rapid and reliable detection method remains challenging. Hence, we constructed an integrated platform by integrating a DNA tetrahedral probe (DTP) and surface antiadhesive magnetic micro/nanorobots (MNRs), enabling rapid contact between the MNR-DTP system and target analytes. The probe enhances fluorescence via target-triggered strand displacement and CHA-mediat

A highly sensitive detection platform for scrub typhus using multi-fluorescence nucleic acid probes and STORM imaging.

BACKGROUND: Nucleic acid testing (NAT) is essential for pathogen identification, disease diagnosis, and biomedical research. Traditional amplification-based methods, while widely used, often face challenges such as long processing times and susceptibility to false positives. Additionally, single-molecule detection approaches are limited by weak signal intensities and rapid photobleaching. To address these limitations, we present an amplification-free biosensing platform for the highly sensitive and specific single-molecule detection of Orientia tsutsugamushi DNA. RESULTS: This platform employ

Dual-Action Nucleic Acid Nanoparticles: Innate Immune Activation and Silver Nanocluster-Mediated Targeting of Intracellular Bacteria.

Intracellular bacterial infections, such as those caused by S. aureus, are particularly challenging to treat because pathogens reside within host cells, evading immune defenses and limiting antibiotic efficacy, subsequently promoting drug resistance. To address this, we developed immunostimulatory nucleic acid nanoparticles (NANPs) functionalized with antibacterial silver nanoclusters to enable a dual mechanism of antibacterial action. These NANPs are engineered to activate RIG-I-mediated innate immune responses, which are often bypassed by intracellular bacteria to avoid host clearance. The e
