Targeting oncogenic TβRI signaling inhibits androgen-independent prostate cancer growth and metastasis.

Metastatic castration-resistant prostate cancer (mCRPC) remains the primary cause of prostate cancer-related mortality. Despite the availability of treatments, the molecular mechanisms underlying tumor invasion and metastasis are not fully understood, highlighting the need for novel therapeutic strategies. In this study, we developed fully human monoclonal antibodies (mAbs) that prevent the proteolytic cleavage of the transforming growth factor-beta (TGFβ) type I receptor (TβRI) by steric hindrance. This cleavage, mediated by the metalloprotease ADAM17 (a disintegrin and metalloprotease domain

Oncogenic VEGFR2 rewires the extracellular matrix to impair endothelial mechanics and destabilize tumor vasculature.

Solid tumors are highly dynamic environments, in which endothelial cells (ECs) interface with blood, cancer cells, and the extracellular matrix (ECM), thereby regulating tumor behavior and metastatic dissemination. We recently showed that aberrant Vascular Endothelial Growth Factor Receptor 2 (VEGFR2) activation promotes tumor progression through ECM remodeling. Here we investigated whether VEGFR2-driven ECM remodeling affects EC behavior and vessel stability. Using decellularized ECM from Sk-Mel-31 melanoma cells expressing either VEGFR2WT or the aberrant VEGFR2R1032Q mutant, we found that VE

Tumoral Metabolism at the Intersection of Oncogene Signaling, Epigenetics and Immunology: Emerging Therapeutic Strategies in Cancer.

Metabolic reprogramming is a unifying characteristic of cancer and involves orchestrated changes in glucose, amino acid, lipid, and mitochondrial metabolism that go beyond the well-known Warburg effect. Evidence is accumulating that these metabolic states are actively remodeled by oncogene signaling and tumor suppressor loss, allowing cancer cells to sustain anabolic growth, redox homeostasis, and therapeutic stress. This review provides an overview of new findings on tumor metabolism, mechanisms, and the molecular networks governing this reprogramming. We discuss how the major oncogenic pathw

The Oncogenic and Tumor-Suppressive Roles of SNHG18: A Double-Edged Long Noncoding RNA in Cancer.

Numerous recent studies have shown that long noncoding RNAs (lncRNAs) play a crucial role in the development of diverse cancers. Small nucleolar RNA host gene 18 (SNHG18) is a cancer-related lncRNA in the SNHG family, whose expression is dysregulated in various tumors and acts as either an oncogenic or tumor-suppressive factor. Several studies have demonstrated that SNHG18 exhibits dual effects in various cancers. The role of this molecule is context-dependent, acting as a tumor suppressor in certain cancers while exhibiting oncogenic properties in others. The expression of SNHG18 is cancer-ty

KIT mutation and AHN-associated oncogenic profiles in systemic mastocytosis with an associated hematological neoplasm.

In 5% to 15% of patients with systemic mastocytosis (SM), SM coexists with an associated hematological neoplasm (AHN). Most patients with SM-AHN harbor a KIT mutation, mainly KIT p.D816V, together with other AHN-related genetic alterations; however, limited data exist about the frequency and clinical impact of the coexistence of both types of genetic alterations in the same vs distinct bone marrow (BM) cell compartments. Here, we analyzed 79 patients with SM-AHN classified into patients who (1) displayed distinct unrelated genetic alterations in BM mast cells (MC) and AHN cells (21/79 [27%]);

Slap restricts oncogenic Src-family kinase signaling to maintain colonic epithelial homeostasis.

Src-family kinases (SFKs) regulate proliferation in colonic epithelial cells (CECs), but the mechanisms that restrain their activity remain poorly defined. We identify Src-like adaptor protein (SLAP), a negative regulator of receptor tyrosine kinase signaling, as a key suppressor of SFK activity in the colon. Constitutive and inducible epithelial-specific Slap deletion using a villin-CreERT2 model increases CEC proliferation and accelerates tumorigenesis in the azoxymethane/dextran sodium sulfate model. Slap deficiency also enhances SFK-dependent expansion of normal and tumor-derived colonic o

Inverted logic of ecDNA localization converts nuclear periphery into oncogenic transcription hubs in cancer.

Extrachromosomal DNA (ecDNA) is a major driver of cancer pathogenesis, however, its spatial organization in the nucleus is not currently understood. Here we show that ecDNAs, in contrast to chromosomal DNA, preferentially localize to the nuclear periphery yet resist peripheral gene silencing. Single-molecule multi-omic sequencing and imaging-based CRISPR screening demonstrate enhanced ecDNA hub formation, H3K27ac and H3K9me3 bivalency, late DNA replication, long-range CpG hypomethylation, and increased transcriptional output when ecDNAs localize to the nuclear periphery. EcDNA anchors to the n

Neuraminidase-Associated Vaginal Dysbiosis Correlates with HPV Infection Across Oncogenic Risk Subtypes.

BACKGROUND: Human papillomavirus (HPV) infection is a major cause of cervical cancer, and increasing evidence suggests that cervicovaginal microbial dysbiosis may influence HPV susceptibility. This study evaluated the association between microbial enzymatic markers and HPV infection across different risk subtypes. METHODS: A cross-sectional analysis was conducted in 43,426 women undergoing HPV screening. HPV status and subtype classification (high-, possible-, and low-risk) were determined using standardized genotyping. Clinical and laboratory parameters including age, white blood cells (WBC)

CSTF2-mediated 3'UTR shortening drives oncogenic activation of GPC3 in hepatocellular carcinoma.

Alternative polyadenylation (APA) generates mRNA isoforms with distinct 3' untranslated regions (3'UTRs), thereby influencing transcript stability and translation. In cancer, 3'UTR shortening can activate oncogenes by escaping microRNA (miRNA)-mediated repression, but its role in hepatocellular carcinoma (HCC) remains poorly defined. Here, we profiled mRNA length alterations in multistage human HCC transcriptome datasets and investigated their functional consequences. Approximately 77% of mRNAs with altered length exhibited 3'UTR shortening. Glypican-3 (GPC3) was the most prominently upregulat…

Does the ERBB/EGF Signaling Network Serve as a Nexus for Oncogenic Signals in Uveal Melanoma?

Uveal melanomas (UMs) metastasize at a high frequency, and metastatic UMs are associated with very poor clinical outcomes. Consequently, there is considerable interest in the genetics and biochemistry of UM and in how these insights may lead to novel and effective approaches to treating it. This work reviews the biology and pharmacotherapy of UM. It also summarizes signaling by the network comprising ERBB receptor tyrosine kinases and EGF family peptide growth factors, including mechanisms of crosstalk with G protein-coupled receptors. Next, this work discusses signaling changes that appear to

Epitranscriptomic Regulation in Oncogenic Viruses: Emerging Roles of RNA Modifications in Viral Persistence and Tumourigenesis.

Oncogenic viral pathogens, such as human papillomavirus (HPV), Epstein-Barr virus (EBV), hepatitis B and C viruses (HBV, HCV), and Kaposi's sarcoma-associated herpesvirus (KSHV), are involved in a significant proportion of human cancers worldwide. While genetic and epigenetic mechanisms underlying viral oncogenesis have been thoroughly investigated, emerging evidence underscores a critical role for epitranscriptomic regulation in virus-host interactions and their clinical consequences. RNA modifications such as N6-methyladenosine (m6A), 5-methylcytosine (m5C), and pseudouridine (Ψ) dynamically…

Functional characterization of FAM120A as a novel effector in the progranulin/EphA2 oncogenic axis in bladder cancer.

BACKGROUND: Bladder cancer (BC) is one of the most deadly diseases in the USA, with 84,530 new cases and 17,870 estimated deaths in 2026. The growth factor progranulin is involved in several human pathologies, including frontotemporal dementia (FTD), immune response, and cancer. We showed that in BC, progranulin and its signaling receptor, EphA2, drive tumor cell motility, invasion, and in vivo tumor formation, making it a critical pathway in tumor establishment. However, the molecular mechanisms of progranulin/EphA2 action are still poorly defined. METHODS: Progranulin-dependent EphA2 intera

Fusion-driven oncogenic programs shape the immune landscape in translocation renal cell carcinoma.

Renal cell carcinomas (RCC) comprise multiple molecularly distinct cancers but most are treated empirically with therapies designed for clear cell RCC (ccRCC), the most common subtype, due to incomplete understanding of subtype-specific biology. We analyzed single-cell transcriptomes and chromatin accessibility profiles from translocation renal cell carcinoma (tRCC), an aggressive RCC defined by oncogenic TFE3 gene fusions. We show that, despite arising from a proximal tubule cell of origin similar to ccRCC, tRCCs display distinct oncogenic programs and an immunosuppressive tumor microenvironm

NSD Histone Methyltransferases in Solid Tumors: Biological Functions, Oncogenic Mechanisms and Therapeutic Targeting.

Epigenetic dysregulation is a defining feature of solid tumors. Among epigenetic regulators, the nuclear receptor-binding SET domain (NSD) family of histone methyltransferases, comprising NSD1, NSD2, and NSD3, have emerged as critical mediators of oncogenic chromatin remodeling and transcriptional regulation. These enzymes primarily catalyze histone H3 lysine 36 (H3K36) methylation, thereby regulating chromatin accessibility, transcriptional programs, DNA damage repair, and genome stability. Aberrant expression, mutation, amplification, and chromosomal rearrangement of NSD family members have

STAG2 mutations in the normal colon induce upregulation of oncogenic pathways in neighbouring wildtype cells.

While driver mutations in the normal colon have been described, characterizing the role and function of these driver mutations in relation to colorectal oncogenesis remains incomplete. Here, we investigated the role of STAG2 mutants in the normal colon using patient-derived wildtype organoids. Using CRISPR-Cas9 gene editing, we generated STAG2 mutants, and co-cultured these mutants with wildtype organoids, mimicking the presence of such STAG2 mutants in the normal colon. We sought to determine the transcriptional impact of co-culture using scRNAseq. Surprisingly, we uncovered a possible cell-c…

Oncogenic activation of EVC/EVC2 defines a therapeutically targetable subset of acute myeloid leukemia.

Acute myeloid leukemia (AML) is an aggressive hematologic malignancy characterized by high incidence, poor prognosis, and marked genetic and clinical heterogeneity. This heterogeneity poses a significant challenge to effective treatment and underscores the urgent need for tailored therapeutic strategies. Here, we identified the EvC ciliary complex subunits EVC and EVC2 as aberrantly overexpressed in a subset of AMLs, particularly those harboring ASXL1 mutations or the t(8;21) translocation, with elevated expression correlating with poor patient prognosis. Functional studies demonstrated that E

Hedgehog signaling drives glial cell plasticity and oncogenic reprogramming in gastroenteropancreatic neuroendocrine neoplasms.

UNLABELLED: Disruption of the Men1 locus in epithelial and endocrine tissues fails to generate the full spectrum of gastroenteropancreatic neuroendocrine tumors (GEP-NETs), raising the possibility of a potential stromal source for these cancers. Neural crest-derived glial cells were previously implicated in neuroendocrine tumors arising in the pituitary and pancreas, yet these studies lacked a clear mechanism for these events. Here, we investigated the hypothesis that Men1 -driven Hedgehog (HH) signaling redirects the glial cell fate to give rise to neuroendocrine tumors in the gastrointestina
