Intergenerational inheritance of paternal metabolic disorder: miR-29a/c-3p/glucose-6-phosphatase 1 axis in glucose intolerance propagation.

The escalating global burden of obesity and diabetes underscores the need to elucidate generational effects of parental metabolic health. This study investigates the generational inheritance of paternal obesity and glucose dysregulation in offspring, along with its underlying epigenetic mechanisms. Four-week-old male C57BL/6 J mice were randomly assigned to either a control diet (CON) or high-glucose/high-fat (HGF) group. After 4 weeks, these males were mated with healthy females to produce F1 male offspring, with the same breeding strategy applied to generate F2 males. Metabolic phenotyping r

The Effect of Kangaroo Care Provided by Fathers in the Neonatal Intensive Care Unit on Paternal Stress and Neonatal Comfort: A Pilot Randomized Controlled Trial.

BACKGROUND: Fathers of infants hospitalized in neonatal intensive care units (NICUs) frequently experience high levels of stress and limited involvement in direct infant care. While kangaroo care is well-established for mothers, evidence regarding father-provided kangaroo care and its effects on both paternal stress and neonatal comfort remains limited. PURPOSE: To examine the effects of kangaroo care provided by fathers on paternal stress levels and neonatal comfort in an NICU setting. METHODS: This pilot randomized controlled trial was conducted from March to December 2024 in a city hospit

The effect of partner-assisted haptonomy on maternal and paternal attachment and marital adjustment in pregnant women following infertility treatment: a randomized controlled trial.

Infertility treatment presents significant psychological challenges for couples, affecting prenatal attachment and marital adjustment. Haptonomy, a touch-based intervention, has been proposed to enhance parent-infant bonding and strengthen partner relationships during pregnancy. However, limited research has explored its impact on couples conceiving through assisted reproductive technologies (ART). This study examined the effects of partner-assisted haptonomy on prenatal attachment and marital adjustment in women who conceived through infertility treatment. A randomized controlled trial was co

Paternal exposure to polystyrene nanoplastics induces inter- and transgenerational bronchopulmonary dysplasia-like damage in male offspring by FtMt hypermethylation-mediated ferroptosis.

Bronchopulmonary dysplasia (BPD) is a major cause of chronic lung disease in both preterm infants and adults, but its etiology remains incompletely understood. In this study, F0 generation mice were exposed to polystyrene nanoplastics (PS-NPs), and F1 to F3 generations were obtained by breeding. Multi-omics sequencing including whole genome methylation sequencing, single cell transcriptome sequencing and transcriptome sequencing was performed on the lungs of offspring. The levels of Fe2+, lipid peroxidation products and key gene expression were determined. Male mice exposed to PS-NPs at enviro

Diagnostic limitations of routine low-coverage CNV-seq in detecting mosaic paternal uniparental disomy in Beckwith-Wiedemann syndrome: a case report.

BACKGROUND: Beckwith-Wiedemann syndrome (BWS) is an overgrowth disorder primarily caused by imprinting defects at 11p15.5. Mosaic paternal uniparental disomy (pUPD) accounts for approximately 20% of cases but is challenging to detect prenatally using routine low-coverage CNV-seq, as this approach has limited sensitivity for copy-number-neutral events such as uniparental disomy, particularly when present in a mosaic state. CASE PRESENTATION: We report a newborn presenting with macroglossia, giant omphalocele, and a birth weight of 4,200 g (>97th percentile). Critically, routine prenatal CNV-se

Association of paternal age with embryological outcomes and β-hCG positivity in women with polycystic ovary syndrome undergoing ICSI: a retrospective cohort study.

BACKGROUND: The independent effect of paternal age on assisted reproductive technology outcomes remains uncertain, particularly when maternal characteristics and conventional semen parameters are considered. This study investigated the association between paternal age and embryological outcomes and β-hCG positivity in women with polycystic ovary syndrome (PCOS) undergoing intracytoplasmic sperm injection (ICSI). METHODS: This single-center retrospective cohort study included 192 ICSI cycles in women with PCOS aged ≤35 years. Male partners were categorized as <35 years (n = 129), 35-39 years (

Prenatal Psychological Adjustment, Relationship Satisfaction, and Parental Reflective Functioning: An Examination of Actor and Partner Effects in the Context of Paternal Moderate to Heavy Drinking.

OBJECTIVE: Parental reflective functioning (PRF) emerges during the prenatal period and helps fathers and mothers prepare for the transition to parenthood. Few studies have considered how PRF could support at-risk fathers and their partners across this transition. In a sample of moderate to heavy drinking fathers, an actor-partner interdependence mediation model (APIMeM) was used to examine concurrent indirect effects between prenatal psychological symptoms and paternal and maternal PRF through interparental relationship satisfaction while accounting for the interdependence among father-mother

Embryonic heat conditioning induces paternal heredity of immunological cross- tolerance: coordinative role of CpG DNA methylation and miR-200a regulation.

BACKGROUND: Enhancing an organism's survival hinges on the development of balanced and adaptable stress response systems. While the initial stress-response set-points in the hypothalamus may be genetically determined, they are further influenced by epigenetic factors during embryonic development. A debate persists regarding the heritability of such behavioral traits. The chick in ovo heat conditioning model offers a unique insight into this fundamental question, where manipulation during embryonic development can induce heat resilience and even cross-tolerance to promote immunological resilien

Paternal adverse childhood experiences and offspring's attentional disengagement from faces at 8 months-Results from the FinnBrain Birth Cohort Study.

Paternal adverse childhood experiences (ACEs) have been recently linked to offspring's brain development. Yet, none of the previous studies in humans have explored the association between paternal ACEs and a child's attentional bias for facial expressions of emotion. Our study fills this gap. Data were collected from 239 fathers (mean age 32.15; SD 5.04) and their children at 8 months of age who were part of the FinnBrain Birth Cohort Study. Paternal ACEs were evaluated using the Trauma and Distress Scale (TADS) in five domains: emotional and physical neglect, emotional and physical abuse, and

Paternal heroin self-administration in rats increases drug-seeking behavior in male offspring via miR-19b downregulation in the nucleus accumbens.

Accumulating evidence indicates that drug addiction may lead to adaptive behavioral changes in offspring, potentially due to epigenetic modifications in parental germline. However, the underlying mechanisms remain inadequately understood. In this study, we show that paternal heroin self-administration (SA) increased heroin-seeking behavior in the F1 generation, when compared with offspring sired by yoke-infused control males, indicating cross-generational impact of paternal voluntary heroin seeking behavior. Notably, the increase of heroin seeking behavior in offspring was replicated by zygoti

Associations of Paternal Seminal Plasma Metals with their Spouses' Unexplained Recurrent Spontaneous Abortion (URSA) Risk and the Potential Mediating Role of Oxidative Stress.

Unexplained recurrent spontaneous abortion (URSA) is a prevalent reproductive issue but its etiology remains obscure. Male exposure to environmental chemicals is suggested to elevate URSA risk in female partners. Herein, a case-control design set out to investigate associations between metal levels in human seminal plasma with URSA risk, plus to determine evidence of mediating effects by oxidative stress. Levels of 15 metal elements and oxidative stress marker malondialdehyde (MDA) in seminal plasma were measured in 125 male spouses of URSA cases compared to 108 male partners of women with suc

Determining the effects of paternal obesity on sperm chromatin at histone H3 lysine 4 tri-methylation in relation to the placental transcriptome and cellular composition.

Paternal obesity has been implicated in adult-onset metabolic disease in offspring. However, the molecular mechanisms driving these paternal effects and the developmental processes involved remain poorly understood. One underexplored possibility is the role of paternally induced effects on placenta development and function. To address this, we investigated paternal high-fat diet-induced obesity in relation to sperm histone H3 lysine 4 tri-methylation signatures, the placenta transcriptome, and cellular composition. C57BL6/J male mice were fed either a control or high-fat diet for 10 weeks begi

A Small De Novo CNV Deletion of the Paternal Copy of FOXF1, Leaving lncRNA FENDRR Intact, Provides Insight into Their Bidirectional Promoter Region.

Pathogenic single-nucleotide variants (SNVs) and copy-number variant (CNV) deletions involving the FOXF1 transcription factor gene or CNV deletions of its distant lung-specific enhancer are responsible for alveolar capillary dysplasia with misalignment of pulmonary veins (ACDMPV), a rarely diagnosed lethal lung developmental disorder in neonates. In contrast to SNVs within FOXF1 and CNV deletions involving only the FOXF1 enhancer, larger-sized deletions involving FOXF1 and the adjacent, oppositely oriented lncRNA gene FENDRR have additionally been associated with hypoplastic left heart syndrom

Paternal exposure to exercise and/or caffeine and alcohol modify offspring behavioral and pathophysiological recovery from repetitive mild traumatic brain injury in adolescence.

Only recently has the scope of parental research expanded to include the paternal sphere with epidemiological studies implicating stress, nutrition and alcohol consumption in the neurobiological and behavioral characteristics of offspring. This study was designed to determine if paternal exposure to caffeine, alcohol and exercise prior to conception would improve or exacerbate offspring recovery from adolescent repetitive mild traumatic brain injury (RmTBI). Sires received 7 weeks of standard drinking water, or caffeine and ethanol and were housed in regular cages or cages with running wheels,

Two's Company, Three's a Crowd? Maternal and Paternal Talk About Their Infant Differs in Associations With Wellbeing, Couple Relationship Quality, and Caregiving Sensitivity.

Problems of depression and anxiety are common in early parenthood and adversely affect parenting quality (1). Rumination is closely linked to poor wellbeing (2), suggesting that self-focus may be one mediator of the association between wellbeing and caregiving [e.g., (3)]. Framed within an international study of first-time mothers and fathers (4), the current study included 396 British mothers and fathers (in 198 heterosexual cohabiting couple relationships) of first-born 4-month-old infants. Parents reported on their symptoms of depression, anxiety and satisfaction in their couple relationshi

Maternal and paternal ages at conception of index child and risk of childhood acute leukaemia: A multicentre case-control study in Greater Mexico City.

BACKGROUND: The parental age at conception has been reported to be a risk factor for childhood acute leukaemia (AL); however, the relationship is controversial. The aim of the present study was to investigate the association between parental age at conception and the risk of AL in Mexican children, a population with a high incidence of the disease and a high prevalence of pregnancies in adolescents and young adults. METHODS: A multicentre case-control study was conducted. Incident AL cases younger than 17 years of age diagnosed between 2010 and 2015 were included. Controls were matched with c

Warburg Micro Syndrome 1 due to Segmental Paternal Uniparental Isodisomy of Chromosome 2 Detected by Whole-Exome Sequencing and Homozygosity Mapping.

Warburg micro syndrome (WARBM) is a rare autosomal recessive disorder characterized by microcephaly, cortical dysplasia, intellectual disability, ocular abnormalities, spastic diplegia, and microgenitalia. WARBM has 4 subtypes arising from pathogenic variants in 4 genes (RAB18, RAB3GAP1, RAB3GAP2, and TBC1D20). Here, we report on a patient with a homozygous pathogenic c.665delC (p.Pro222HisfsTer30) variant in the RAB3GAP1 gene identified by whole-exome sequencing (WES) analyses. Only his father was a heterozygous carrier, and homozygosity mapping analysis of the WES data revealed large loss-of

Paternal exposure to environmental 17-alpha-ethinylestradiol concentrations modifies testicular transcription, affecting the sperm transcript content and the offspring performance in zebrafish.

The synthetic estrogen 17-α-ethinylestradiol (EE2), a major constituent in contraceptive pills, is an endocrine disrupting chemical (EDC) present in the aquatic environment at concentrations of ng/L. Developmental exposure to these low concentrations in fish can induce several disorders. Zebrafish (Danio rerio) is a perfect organism for monitoring the effects of environmental contaminants. Our hypothesis is that changes promoted by EE2 in the germ line of male adults could be transmitted to the unexposed progeny. We exposed male zebrafish to 2.5, 5 and 10ng/L of EE2 during spermatogenesis and
