Hydrogen peroxide-induced epithelial injury: the protective role of intracellular nonprotein thiols (NPSH).

Injury to the alveolar region is a hallmark of the adult respiratory distress syndrome (ARDS) whereas injury to the epithelium of the conducting airways is a characteristic of asthma. Reactive oxygen species have been implicated as mediators of lung injury in both of these conditions. We have investigated the relationship between intracellular nonprotein thiols (NPSH), and the release of the cytosolic enzyme lactate dehydrogenase (LDH), as an index of cell injury, following treatment of the human alveolar type II-like epithelial cell line (A549 cells) or the human bronchial epithelial cell lin

Cytochrome b558 and hydrogen peroxide production in small intensely fluorescent cells of sympathetic ganglia.

Small intensely fluorescent (SIF) cells are paraganglionic cells derived from sympathicoblasts which may serve as interneurons, endo-/paracrine cells or arterial chemoreceptors within sympathetic ganglia. Like paraganglionic cells of other locations, e.g., carotid body glomus cells, they are responsive to hypoxia. Recent studies on glomus cells and other hypoxia-sensing cells suggested the involvement of a b558-type cytochrome and intracellular generation of H2O2 in the process of oxygen sensing. In the present study, we demonstrate the occurrence of the small subunit of cytochrome b558, p22ph

A critical evaluation of anti-peroxidant effect of intravenous magnesium in acute aluminium phosphide poisoning.

The anti-peroxidant effect of intravenous magnesium was evaluated in 50 patients with acute aluminium phosphide poisoning. The patients were divided into two groups, one who received magnesium sulphate therapy (Group I) and the other who did not (Group II). The clinical and biochemical parameters in both groups were comparable. Finding of increased mean malonyl-di-aldehyde (MDA) levels in group I (3.18 +/- 0.93 micromol/L) and group II (3.15 +/- 0.78 mmol/L) combined with low blood levels of reduced glutathione (18.5 +/- 1.6 mg/dl in group I) and (17.8 +/- 1.4 mg/dl in group II) indicated oxid

[Disturbance of regional blood flow and activity of lipid peroxidation in recurrent ulcer and their drug correction].

Regional blood flow and intensity of free-radical lipid peroxidation were evaluated at rheogastrography and biochemically (quantitation of malonic dialdehyde and antioxidant serum activity), respectively, in 25 patients with recurrent ulcer. Recurrent ulcer is shown to induce disturbances in gastric wall regional hemodynamics due to activated vascular tone, reduced arterial blood inflow and venostasis. Lipid peroxidation became overactive and decreased defence potentialities of gastroduodenal serum. Oleum Hippopheae which is a natural antioxidant, improves regional hemodynamics by relaxation o

Effect of acute Trypanosoma vivax infection on cattle erythrocyte glutathione and susceptibility to in vitro peroxidation.

During acute Trypanosoma vivax infection of calves, produced by intravenous inoculation, the mean packed cell volume and red blood cell counts of the infected animals decreased significantly (P < 0.05) between Days 6 and 13 post-infection (pi). The moderately severe normocytic anaemia started to develop during the first wave of parasitaemia which occurred from Day 2 pi and peaked between Days 4 and 5 pi. The mean erythrocyte glutathione (GSH) concentration of the infected calves decreased significantly (P < 0.05) from 58.4 +/- 11.4 mg 100ml-1 red blood cells (RBC) on Day 0 pi to 44.5 +/- 12.8

Cultured rat mesothelial cells generate hydrogen peroxide: a new player in peritoneal defense?

This study was designed to examine whether rat peritoneal mesothelial cells in culture could generate hydrogen peroxide in different experimental conditions. Mesothelial cells, incubated in M-199, spontaneously released hydrogen peroxide. This process was significantly increased by addition of phorbol myristate acetate, as well as of superoxide dismutase to the medium, whereas it was substantially inhibited by catalase. Exposure of mesothelial cells to modified M-199 medium with 1.5% glucose concentration-lactated peritoneal dialysis solution did not seem to interfere either with the spontaneo

Evaluation of real-time techniques to measure hydrogen peroxide in air at the permissible exposure limit.

The major occupational concern from bio-decontamination of equipment using vapor phase hydrogen peroxide (VHP) generation systems is potential operator exposure outside the protective barrier from possible VHP leaks or accidental releases from the sealed piece of equipment during decontamination. For this reason, different real time monitoring techniques were evaluated to determine their ability to accurately measure VHP at concentrations ranging from 0.5 ppm to 5 ppm. The results of this laboratory evaluation suggest that two of the four methods evaluated (the ion mobility spectrometer [IMS]

The pyridoindole antioxidant stobadine prevents alloxan-induced lipid peroxidation by inhibiting its propagation.

Under in vitro conditions, the pyridoindole stobadine inhibited alloxan-induced lipid peroxidation in a model biological membrane with the efficacy comparable with that of the standard Trolox. Intermediary alloxan radicals and hydroxyl radicals were not directly involved in the process of lipid peroxidation, however, the presence of iron chelate was a necessary prerequisite. Since stobadine did not affect the kinetics of alloxan redox-cycling in the presence of GSH, we suggest that the protective action of stobadine against the alloxan-induced lipid peroxidation was mediated predominantly by i

Leukocyte hydrogen peroxide production in a surgical wound in mice. The effects of an amide local anaesthetic.

The oxygen metabolism of polymorphonuclear leukocytes (PMN) is of importance in local tissue repair processes. Amide local anaesthetics are commonly used to relieve surgical wound pain. The cellular effects of local anaesthetics in vivo is poorly described in the literature. However, interactions between amide local anaesthetics and the oxygen metabolism of leukocytes have been reported. To extend that knowledge, this paper investigates the influence of lidocaine treatment on the production of hydrogen peroxide (H2O2) by leukocyte oxygen metabolism. A soft tissue chamber model in the mouse was

Dietary and pharmacologic regimens to reduce lipid peroxidation in non-insulin-dependent diabetes mellitus.

Persons with diabetes are at increased risk for developing coronary heart disease (CHD): although the standard risk factors are also applicable, there are other major nontraditional risk factors for these persons. For example, oxidation of lipoproteins may play an important role in the development of atherosclerosis. Another risk factor is the presence of small, dense low-density lipoprotein (LDL), which may have enhanced atherogenicity because it enters the arterial wall more readily and is more easily oxidized. Both of these factors may be particularly important in persons with non-insulin-d

Iron storage, lipid peroxidation and glutathione turnover in chronic anti-HCV positive hepatitis.

BACKGROUND/AIMS: Little is known about the pathogenesis of liver damage related to hepatitis C virus. The presence of steatosis or increased ferritin levels, and preliminary data on the relevance of iron as a prognostic factor prompted us to ascertain whether hepatitis C virus-related liver damage might be mediated by iron accumulation. METHODS: We evaluated the degree of hepatic inflammation and steatosis, serum ferritin, transferrin saturation and iron levels, tissue iron concentrations and iron index, liver glutathione and malondialdehyde in 33 males and 20 females with chronic hepatitis C

[Lipid peroxidation and the antioxidant system in chronic cholecystitis in persons located in an area of increased radiation].

The studies covered 35 subjects with chronic cholecystitis who had to work in radionuclide-contaminated territory. There appeared changes in examinees' lipid peroxidation and antioxidant defense systems. These were more pronounced in leukocytes, possibly due to direct action of radiation on cell membrane, and may contribute to disease chronicity and recurrences.

Hydrogen peroxide does not function downstream of salicylic acid in the induction of PR protein expression.

The roles of salicylic acid (SA) and H2O2 in the induction of PR proteins in tobacco have been examined. Studies were conducted on wild-type tobacco and plants engineered to express a bacterial salicylate hydroxylase capable of metabolizing SA to catechol (SH-L plants). Wild-type and PR-1a-GUS-transformed plants express PR-1a following challenge with Pseudomonas syringae pathovar syringae, SA or 2,6-dichloro-isonicotinic acid (INA). In contrast, SH-L plants failed to respond to SA but did express PR-1a following INA treatment. H2O2 and the irreversible catalase inhibitor 3-amino-1,2,4-triazole

Role of lipid peroxidation in H2O2-induced renal epithelial (LLC-PK1) cell injury.

The exact sequence of events or mechanisms by which H2O2 induces renal cell injury remains undetermined. Specifically, whether the attendant lipid peroxidation is a cause or an effect remains unclear. Employing H2O2 and LLC-PK1 cells, we tested the hypothesis that lipid peroxidation is a seminal event and that its inhibition is cytoprotective. In a time course study, lipid peroxidation (thiobarbituric acid reaction) and degradation (release of [3H]arachidonic acid) preceded H2O2-induced cytolysis (51Cr and lactate dehydrogenase release). The role of preceding lipid peroxidation in cytolysis wa

Peroxidative damage to cardiac mitochondria. II. Immunological analysis of modified adenine nucleotide translocase.

We have previously reported that treatment of isolated rat heart mitochondria with the free radical-generating system Cu2+/tert-butylhydroperoxide produces striking changes in the adenine nucleotide translocase (ANT) of the inner membrane. These changes include a small increase in apparent molecular weight as determined by sodium dodecyl sulfate-polyacrylamide gel electrophoresis, followed by its disappearance from the polypeptide profile upon further oxidant treatment (Zwizinski and Schmid (1992) Arch. Biochem. Biophys. 294, 178-183). In order to characterize its peroxidative modification in

Expired breath hydrogen peroxide is a marker of acute airway inflammation in pediatric patients with asthma.

Airway inflammation is important in the development and progression of many pulmonary disorders, including asthma. We hypothesized that the hydrogen peroxide (H2O2) concentration in expired breath may be a marker of airway inflammation. Expired breath condensate was collected by cooling and the H2O2 concentration was measured fluorimetrically. Thirty-five samples were collected from 22 pediatric patients with asthma who were 7 to 18 yr of age and from 11 healthy, nonasthmatic controls. Asthmatic subjects were determined to be well or sick (acute disease of the upper or lower respiratory tract)

Effect of endotoxin on rat serum, lung and liver lipid peroxidation and on tissue metallothionein levels.

The effect of endotoxin on serum and lung and liver lipid peroxidation, as measured by thiobarbituric acid reactants (TBARs), as well as on lung and liver metallothionein (MT) has been studied in the rat. Endotoxin consistently increased serum and liver TBARs in a time-response manner. The increase in the serum preceded that in the liver, with peaks 3-6 h and 24 h after endotoxin administration, respectively. In contrast, lung TBARs levels did not increase regardless of the experimental approaches studied, suggesting that the rat is not a good model for the adult respiratory distress syndrome.

Aging and cancer: plasma antioxidants and lipid peroxidation in young and aged breast cancer patients.

The relationship between aging and cancer is complex because the intrication takes place at the cell, the organism and the environment levels. On the other hand, carcinogenesis is a multi-step process, and different mechanisms may be involved in each step. For example, oxidants and antioxidants may play a different role depending upon the phase considered. Tumors in older patients are generally described as slow growing. The difference in tumor aggressiveness between young and older patients is especially obvious in breast cancer patients. The age specificity of some breast risk factors sugges

The extra-weak chemiluminescence generated during oxidation of some tetracycline antibiotics. II. Peroxidation.

This study was undertaken to establish the mechanism of chemiluminescence during the oxidation reaction of tetracycline antibiotics in the presence of molecular oxygen and H2O2. The spectral distribution of chemiluminescence and fluorescence and the quantum yields of chemiluminescence were measured. The chemiluminescence spectrum measured with cut-off filters revealed one broad blue-green band and bands with maxima at 580, 640 and 700 nm. The bands at 580, 640 and 700 nm were similar to those observed for singlet molecular oxygen. The effect of superoxide radical, hydroxyl radical inhibitors,
