Evolution of the frontal aslant tract and implications for primate vocalization and human speech.

Monkeys, chimpanzees and humans share dorsomedial and ventrolateral frontal regions for vocalization, yet only humans have speech. These regions are interconnected by the frontal aslant tract (FAT), a pathway increasingly recognized as central to speech production. Using spherical deconvolution tractography, we compared the FAT across primate species. In humans, the FAT is left-lateralized and enlarged, predominantly in its anterior parts connecting prefrontal areas. This expansion parallels the enlargement of the arcuate fasciculus, which conveys auditory input to the ventrolateral frontal re

Distinct neural geometries for target position and velocity in the primate superior colliculus.

Moving targets pose a fundamental challenge for sensorimotor control: by the time a target's position is transformed into neural signals, the target has already moved. Primates compensate for this delay and accurately intercept objects by accounting for target velocity. However, the neural mechanisms by which sensorimotor structures represent target velocity remain poorly understood. Here we recorded spikes and local field potentials from the superior colliculus (SC) of male macaques viewing or directing saccades to stationary and moving targets and used time-resolved dimensionality reduction …

Surround motion modulates the encoding properties of primate retinal ganglion cells.

Antagonistic interactions between center and surround regions of the receptive field are widely observed across sensory systems. In the early visual system, these interactions contribute to important computations such as edge detection. However, less is known about how center-surround interactions depend on the spatiotemporal properties of the visual input. Here, we show that surround motion strongly modulates the response properties of two understudied primate ganglion cell types. Broad thorny cell responses are strongest when motion in the center and surround is uncorrelated, similar to obje

Challenges and perspectives in xenoliver research: lessons from decedent and primate models.

Liver xenotransplantation is a potential strategy to address the shortage of donor livers, either as a temporary bridge to allotransplantation, bridge to native liver recovery, or destination therapy. Advances in genetic engineering and immunosuppression have enabled liver xenografts to evade hyperacute rejection, yet durable graft survival remains elusive. Profound thrombocytopenia, consumptive coagulopathy, and xenotransplantation-associated thrombotic microangiopathy limit long-term graft survival. Evidence from nonhuman primates, decedents, and recent clinical models suggests that hematolo…

Gene regulatory innovations from transposable elements in primate cerebellum development.

Transposable elements are hypothesized to have driven gene regulatory innovation, yet their contributions to primate brain development at the cell type level remain underexplored. Here, we use single-cell multiomics data from human, macaque, marmoset, and mouse cerebella to show that transposable element contributions to different cell types are shaped by varying degrees of constraints across cell types, as well as the preferential co-option of certain transposable elements in specific cell states. Using a sequence-based deep-learning model that predicts cell-type-specific chromatin accessibil

Fossil evidence favors a role for vision in the modular evolution of the primate neocortex.

Neocortical expansion is a key innovation of crown primates, although fossil evidence for how the neocortex has evolved remains elusive. We studied neocortical evolution using virtual brain endocasts from extant and Eocene-Miocene fossil primates. We found that neocortex size increased in parallel across crown primate clades, with rapid increases in tarsiers and anthropoids. Although the frontal lobe is commonly described as having expanded disproportionately in primates, we show that it evolved gradually, following a common pattern for allometric scaling. By contrast, nonfrontal neocortical r…

Pulvinar subdivisions and connectivity patterns across primate species: a comparative perspective.

With the advances in our ability to perturb brain activity in recent years, new stimulation techniques have become essential tools in human neuroscience. Non-invasive stimulation methods, such as transcranial magnetic stimulation (TMS), as well as deep brain stimulation (DBS) delivered invasively to access deep brain structures, have been applied in both basic and clinical research and in the treatment of neurological conditions including Parkinson's disease, essential tremor and epilepsy. In the context of epilepsy, neuromodulatory interventions have demonstrated encouraging results in reduci

Ground-truth encoding of self-motion in the primate cerebellar nodulus and uvula.

Accurate internal estimates of self-motion and orientation relative to gravity are fundamental for stabilizing gaze, controlling posture, and navigating through dynamic environments. Prevailing theories propose that the cerebellar nodulus and uvula (NU) employ internal models to suppress sensory input arising from predictable, self-generated motion. However, this assumption has never been directly tested. Here, we recorded NU Purkinje cell activity in rhesus monkeys during active and passive head movements. We found neurons responsive to passive translations remained equally sensitive to self-

Yellow fever outbreak temporarily changes dispersal patterns in an endangered primate.

Disease outbreaks can severely affect populations in the wild. However, their consequences on individual social behaviour and population demography are poorly understood. We used a multi-site capture-recapture model to investigate the impacts of a yellow fever outbreak on the endangered golden lion tamarin Leontopithecus rosalia, in the fragmented Atlantic forest, Brazil. Annual adult survival rate severely declined in 2017-2018, coinciding with the outbreak period. Simultaneously, dispersal patterns changed temporarily, with a reduction of settlement time for individuals dispersing within the

Nonhuman primate models of HIV reveal reservoir dynamics and promising interventions.

PURPOSE OF REVIEW: HIV cure research seeks to eradicate the virus from people with HIV (PWH) or provide posttreatment control (PTC) of virus sufficient to block new transmissions and provide better health outcomes. Nonhuman primate (NHP) models of HIV have proven instrumental to our understanding HIV infection. This review highlights the remarkable contributions of NHPs in both corroborating data from PWH and expanding our understanding beyond what is possible in a clinical setting by utilizing novel virus models, immune activating therapies, and direct in vivo intervention to clear the viral

Primate-specific sperm lnc-CLCN7 reveals embryo quality in IVF.

Long non-coding RNAs (lncRNAs) are an essential class of regulatory molecules that participate in diverse biological processes. However, whether sperm-derived lncRNAs from infertile men contribute to impaired embryo development during in vitro fertilization (IVF) remains unclear. In this study, we investigate the lncRNA expression profiles in sperm from asthenozoospermic patients with poor embryo development and explore their potential roles in early embryo development. Microarray analyses identify 993 differentially expressed lncRNAs in sperm samples from these patients, including 626 downreg

Translational research on pandemic virus infection using nonhuman primate models.

After the COVID-19 pandemic, nonhuman primate (NHP) models, which are necessary for the rapid development of vaccines and new medical therapies, have become important in studies on infectious diseases because of their genetic, metabolic, and immunological similarities to humans. Our group has long been using NHP models in studies on infectious diseases including H1N1 influenza pandemic and COVID-19. Despite limitations such as the limited number of animals and the husbandry requirements, NHP models have contributed to the prediction of the pathogenicity of emerging viruses and the evaluation o

Tau and tauopathies across primate species: implications for modeling neurodegenerative disorders.

Tauopathies are neurodegenerative disorders characterized by the abnormal accumulation and aggregation of hyperphosphorylated tau protein. T … orts to unravel the mechanisms leading to tauopathies and identifying therapeutic targets. Nonhuman primates (NHPs) present several advantages for the study of tauopathies, as they have complex neuroanatomy and behavior that resembles human traits, and their tau gene and protein are highly conserved. Moreover, aged NHPs, like humans, can present various ta …

Non-human primate: the new frontier model of female reproductive engineering.

Reproductive engineering encompasses a range of advanced tissue engineering techniques aimed at addressing infertility that is non-curable with current assisted reproductive technology (ART). The use of animal models has been crucial for these advancements, with a notable preference for non-human primates (NHPs) given their genetic, anatomical, and physiological similarities to humans. Therefore, NHPs are invaluable for studying reproductive engineering. Thus, in reproductive studies, NHPs bridge the anatomical and physiological gaps between rodent models and humans. Their shared features with…

A new class of human CpG Island promoters with primate-specific repeats.

A subset of imprinting control regions (ICRs) in the human and mouse possess CpG islands associated with imperfect tandem repeats (TRs) that were shown to be essential for genomic imprinting through genetic studies. To identify whether this feature is also present in non-imprinted CpG island promoters, we performed extensive dot plot analyses and identified 342 (326 autosomal and 16 X-chromosomal) human CpG island gene promoters associated with imperfect TRs of ≥ 400 bp, unit lengths 50-150 bp. Most occur as clusters at the human chromosome ends, distinct from the clusters of imprinted genes,

Host ZAP activity correlates with the levels of CpG suppression in primate lentiviruses.

Zinc-finger antiviral protein (ZAP) is thought to drive the suppression of CpG dinucleotides in many viruses to mimic the composition of the … corresponding host lemur ZAPs. In addition, lemur genes also show lower CpG suppression than other primates. Thus, the antiviral activity of ZAP not only affects suppression of CpG dinucleotides in viral transcripts but possibly also host genomes.

The biological scaling of Alzheimer's disease neuropathological changes across primate species.

BACKGROUND: Studying the spontaneous emergence of biological anomalies within the animal kingdom can provide insights into the causes of diseases. It is often assumed that Alzheimer's Disease (AD), like other neurodegenerative diseases, is specific to humans. However, the age-related occurrence of AD neuropathological changes (ADNC) in non-human primates (NHPs) and their comparison with humans has not been formally studied. Moreover, a conceptual framework for interpreting the spontaneous occurrence of ADNC in NHPs has yet to be established. METHODS: We conducted a systematic review of the av…
