[Selective photodynamic destruction of leukemic cells].

BACKGROUND: Residual leukemic cells if present in autologous bone marrow grafts or CD34+ concentrates obtained from peripheral blood may increase the risk of relapse after autotransplantation. We are presenting the employment of a new method which was introduced into the photodynamic therapy, namely enhancement of synthesis of the photosensitizing compound, protoporphyrin IX, in cancer cells, following application of its metabolic precursor, 5-aminolevulinic acid, for the specific destruction of leukemic cells. METHODS AND RESULTS: By determining cell viability using tetrazolium salt reductio

Mechanism of iron potentiation of hepatic uroporphyria: studies in cultured chick embryo liver cells.

Effects of iron were studied in cultured chick embryo liver cells to help elucidate the effect of hepatic iron in the human disease porphyria cutanea tarda and in toxic porphyria caused by chemicals. These cultures have proven useful because (a) phenobarbital and phenobarbital-like drugs induce a common form(s) of cytochrome P-450 (P-450-phenobarbital) in these cultures; (b) 20-methylcholanthrene and certain other polycyclic hydrocarbons induce a different form(s) (P-450-methylchol-anthrene), and (c) uroporphyria can be produced rapidly by exposure to suitable chemicals. In these cultures, tre

α-Lipoic Acid Inhibits IL-8 Expression by Activating Nrf2 Signaling in Helicobacter pylori-infected Gastric Epithelial Cells.

Helicobacter pylori (H. pylori) causes gastritis and gastric cancers. Oxidative stress is involved in the pathological mechanism of H. pylori-induced gastritis and gastric cancer induction. Therefore, reducing oxidative stress may be beneficial for preventing the development of H. pylori-associated gastric diseases. Nuclear factor erythroid-2-related factor 2 (Nrf2) is a crucial regulator for the expression of antioxidant enzyme heme oxygenase-1 (HO-1), which protects cells from oxidative injury. α-Lipoic acid (α-LA), a naturally occurring dithiol, shows antioxidant and anti-inflammatory effec

Metabolomic analysis of CSF indicates brain metabolic impairment precedes hematological indices of anemia in the iron-deficient infant monkey.

OBJECTIVES: Iron deficiency (ID) anemia leads to long-term neurodevelopmental deficits by altering iron-dependent brain metabolism. The objective of the study was to determine if ID induces metabolomic abnormalities in the cerebrospinal fluid (CSF) in the pre-anemic stage and to ascertain the aspects of abnormal brain metabolism affected. METHODS: Standard hematological parameters [hemoglobin (Hgb), mean corpuscular volume (MCV), transferrin (Tf) saturation, and zinc protoporphyrin/heme (ZnPP/H)] were compared at 2, 4, 6, 8, and 12 months in iron-sufficient (IS; n = 7) and iron-deficient (ID;

Iron monitoring and supplementation: how do we achieve the best results?

A number of factors have been shown to limit the response to recombinant human erythropoietin (r-HuEPO). One major factor appears to be an inadequate iron supply to the bone marrow. Erythropoiesis is dependent upon a continuous supply of iron to the bone marrow. The rate at which iron can be drawn from existing stores may easily limit the rate of delivery for haemoglobin synthesis. This results in 'functional iron deficiency' which is distinct from 'absolute iron deficiency' caused by depletion of iron stores. At present there are three main parameters available to clinicians wishing to monito

Targeting Oxidative Stress and Neuroinflammation: Analgesic Effects of 4-(phenylsulfanyl)butan-2-one on Chronic Constriction Injury-Induced Peripheral Neuropathic Pain in Rats.

Neuropathic pain arises from complex pathological processes involving oxidative stress and neuroinflammation. Excessive production of reactive oxygen species (ROS) contributes critically to the initiation and persistence of neuropathic pain following nerve injury and inflammatory insults, and antioxidant-based interventions have shown consistent efficacy across multiple preclinical models. This study investigated the analgesic effects of 4-(phenylsulfanyl)butan-2-one (4-PSB-2), a compound with known anti-inflammatory properties, using the chronic constriction injury (CCI) rat model. Identifying biomarkers of the serum proteins and metabolites associated with severe preeclampsia.

BACKGROUND: Preeclampsia is a severe pregnancy-related disorder characterized by hypertension and proteinuria, and affects approximately 2–8% of pregnancies worldwide. It poses substantial risks to both maternal and fetal health, including preterm birth, intrauterine growth restriction, and life-threatening complications, such as eclampsia and organ failure. However, the mechanisms underlying preeclampsia, especially those involving blood-associated proteins, are poorly understood. This study aimed to identify critical blood proteins linked to severe preeclampsia and explore their potential me

Chitosan oligosaccharide-coated carrier-free evodiamine-curcumin nanoassemblies enhance oral delivery and anti-atherosclerotic efficacy.

Oxidative stress and dysregulated vascular smooth muscle cell (VSMC) proliferation drive atherosclerotic (AS) progression and plaque development. Natural phytochemicals, including evodiamine (Evo) and curcumin (Cur), show anti-atherosclerotic potential, but poor aqueous solubility and insufficient intestinal absorption limit their oral use. Here, Evo and Cur were co-assembled into carrier-free Evo-Cur nanoparticles (EC NPs) and coated with chitosan oligosaccharide (COS) to obtain COS-coated EC nanoparticles (COS@EC NPs). Spectroscopic analyses and molecular dynamics simulations indicated that

Heme promotes venetoclax resistance in multiple myeloma through MEK-ERK signaling and purine biosynthesis.

We previously demonstrated that reduced intrinsic electron transport chain (ETC) activity predicts and promotes sensitivity to the B-cell lymphoma 2 (BCL-2) antagonist, venetoclax (Ven), in multiple myeloma (MM). Heme, an iron-containing prosthetic group and metabolite, is fundamental to maintaining ETC activity. Interrogation of the cyclin D1 group 2 subgroup of MM from the Relating Clinical Outcomes in MM to Personal Assessment of Genetic Profile (CoMMpass) trial (NCT01454297), which can be used as a proxy for Ven-sensitive MM (VS MM), shows reduced expression of the conserved heme biosynthe

A microneedle platform Co-encapsulating chondral organoids and PpIX for spatiotemporally orchestrated tumor ablation and osteochondral regeneration.

Tumor-derived osteochondral defects (OCDs) pose significant clinical challenges, necessitating integrated strategies for local tumor ablation while preserving the regenerative niche and enabling coordinated cartilage and subchondral bone repair. Herein, we introduce a spatiotemporal microneedle (MN) platform that synergistically combines protoporphyrin IX (PpIX)-mediated sonodynamic therapy (SDT) for precise tumor eradication with chondral organoid (CO)-driven regeneration, leveraging differential mechanical cues for lineage-specific guidance. COs were engineered from bone marrow stem cells (B

Heme oxygenase-1 determines the cell fate of ferroptotic death of alveolar macrophages in COPD.

BACKGROUND: Despite an increasing understanding of chronic obstructive pulmonary disease (COPD) pathogenesis, the mechanisms of diverse cell populations in the human lung remain unknown. Using single-cell RNA sequencing (scRNA-Seq), we can reveal changes within individual cell populations in COPD that are important for disease pathogenesis and characteristics. METHODS: We performed scRNA-Seq on lung tissue obtained from donors with non-COPD and mild-to-moderate COPD to identify disease-related genes within different cell types. We testified the findings using qRT-PCR, immunohistochemistry, im

Body composition, mitochondrial oxygen metabolism and metabolome of patients with obesity before and after bariatric surgery (COMMITMENT): protocol for a monocentric prospective cohort study.

INTRODUCTION: Obesity, defined as a body mass index ≥30 kg/m2, is one of the most prevalent health conditions worldwide. It is part of the metabolic syndrome, which encompasses arterial hypertension, dyslipoproteinaemia and diabetes. Obesity is viewed as a systemic disease with pathophysiological mechanisms on the molecular level. Dysfunction of the mitochondrion and systemic low-grade inflammation are among the proposed causes for the metabolic changes. In severe cases of obesity, laparoscopic sleeve gastrectomy, a bariatric operation, can achieve the desired weight loss and has been associat

Mechanisms of isoniazid and rifampicin-induced liver injury and the effects of natural medicinal ingredients: A review.

Isoniazid (INH) and rifampicin (RFP) are the first-line medications for tuberculosis treatment, and liver injury is the major adverse effect. Natural medicinal ingredients provide distinct benefits in alleviating patients' symptoms, lowering the liver injury risk, delaying disease progression, and strengthening the body's ability to heal. This paper summarises the recent research on the mechanisms of INH and RFP-induced liver injury and the effects of natural medicinal ingredients. It is believed that INH-induced liver injury may be attributed to oxidative stress, mitochondrial dysfunction, dr

HMOX1 is partly responsible for phenotypic and functional abnormalities in mesenchymal stem cells/stromal cells from placenta of preeclampsia (PE) patients.

BACKGROUND: Preeclampsia is a common obstetric syndrome affecting women in their first pregnancy and characterized by hypertension and proteinuria, which appears after 20 weeks of gestation. It is characterized by high blood pressure and occasional damage to another organ system most often the liver and kidneys. Currently, the etiology and pathogenesis of this syndrome are not fully understood. Since mesenchymal stem cells/stromal cells (MSCs) are intimately associated with endothelial cells that line vessel walls in the decidua they may play some role in the pathogenesis of this syndrome. The Interplay between Molten Globules and Heme Disassociation Defines Human Hemoglobin Disassembly.

Hemoglobin functions as a tetrameric oxygen transport protein, with each subunit containing a heme cofactor. Its denaturation, either in vivo or in vitro, involves autoxidation to methemoglobin, followed by cofactor loss and globin unfolding. We have proposed a global disassembly scheme for human methemoglobin, linking hemin (ferric protoporphyrin IX) disassociation and apoprotein unfolding pathways. The model is based on the evaluation of circular dichroism and visible absorbance measurements of guanidine-hydrochloride-induced disassembly of methemoglobin and previous measurements of apohemog
