BACKGROUND: Many regions in Africa continue to experience a high malaria burden, highlighting the need for improved intervention tools adapted to local eco-epidemiological contexts. The aims of this study were to develop and evaluate adaptive intervention strategies for malaria vector control tailored to these local conditions.
METHODS: This two-stage, cluster, sequential, multiple-assignment, randomised, open-label trial comprised 84 clusters in Muhoroni and Nyakach, subcounties of Kisumu County, western Kenya. Each cluster included 500-600 residents in an area of approximately 2 km2 and all

BACKGROUND: Insecticide-treated nets (ITNs) represent a key tool in reducing human vector contact for malaria control. However, increasing insecticide resistance of malaria vectors threatens the effectiveness of pyrethroid-only nets in reducing malaria risk. Next-generation nets, such as those with dual-active ingredients, have been recommended for use in areas with high malaria burden and confirmed pyrethroid resistance. Here, we assessed the impact of the distribution of Interceptor® G2 (IG2) ITNs on malaria cases in the Western North Region of Ghana distributed in 2021.
METHODS: We analyse

Increasing insecticide resistance in malaria vectors threatens the efficacy of current control tools, however knowledge of metabolic and cuticular mechanisms is widely lacking in Ghana. We examined the metabolic and cuticular resistance mechanisms in Anopheles gambiae mosquitoes from coastal and sahel zones of Ghana. WHO susceptibility tests and synergist assays were performed on F0 field collected An. gambiae s.l. Gene expression profiles of eight key metabolic and cuticular genes were determined using qRT-PCR. Moderate to high pyrethroid resistance (< 70%) were observed across all the sites.

INTRODUCTION: Malaria is still a major health problem in sub-Saharan Africa, where 98% of global malaria mortality occurs. In addition, the spread of Plasmodium falciparum with partial artemisinin resistance in East Africa and beyond is a great concern. The establishment of more effective vector control, in addition to the current long-lasting insecticide-treated net distribution programme, is an urgent task in these areas. One novel vector control candidate is the pyrethroid-PBO ceiling nets (OlysetPlus ceiling nets) which can overcome the problems of variations in net use behaviours and meta

The mosquito Aedes aegypti is the main vector of the etiological agents of dengue, Zika, and chikungunya representing major public health concern in tropical and subtropical regions. Control of A. aegypti relies mainly on pyrethroid insecticides such as deltamethrin, but increasing resistance has reduced their effectiveness. Here, we identified and characterized two arginine kinase genes (ak1 and ak2) in A. aegypti, a key enzyme involved in regulating energy homeostasis in insects and investigated their contribution to insecticide tolerance. We performed protein phylogenetic analysis and prote

Rhipicephalus microplus is a major ectoparasite of cattle in tropical and subtropical regions and poses a serious threat to animal welfare and productivity. Chemical acaricides remain the main method of control in Brazil. Nevertheless, resistance to these compounds has become increasingly widespread. Minas Gerais is the leading milk-producing state in Brazil, yet data on acaricide resistance in one of its key dairy-producing region, the Triângulo Mineiro, remain scarce. This study evaluated the resistance of R. microplus populations from 15 dairy farms from nine municipalities in the Triângulo

BACKGROUND: Effective vector control is key to malaria prevention. However, this is now compromised by increased insecticide resistance due to continued reliance on insecticide-based control interventions. In Kenya, we have observed heterogenous resistance to pyrethroids and organophosphates in Anopheles arabiensis which is one of the most widespread malaria vectors in the country. We investigated the gene expression profiles of insecticide resistant An. arabiensis populations from Migori and Siaya counties in Western Kenya using RNA-Sequencing. The present cluster-randomised control trial aims to assess the entomological efficacy of pyrethroid-pyriproxyfen and pyrethroid-chlorfenapyr LLINs compared to the standard pyrethroid-only LLINs, in their third year of community usage. Adult mosquito collections were performed every 3 months, in 4 randomly selected houses in each of the 60 trial clusters, using human landing catches. Adult mosquitoes were morphologically identified and Anopheles vectors were molecularly speciated and screened for the presence of the L1014F kdr mutation using PCR. Plasmodium falciparum sporozoite infection was

OBJECTIVE: In recent years, it has been known that the melatonin hormone, secreted from the pineal gland, possesses significant antioxidant activity. This study explores the therapeutic effect of melatonin on the deleterious effects of deltamethrin, a pyrethroid pesticide extensively used worldwide, including in Türkiye, on mouse liver cells.
METHODS: Hepatocytes from Balb/C mice were isolated using a two-stage perfusion method, resulting in over 85% live hepatocytes. The isolated cells were cultured with different doses of deltamethrin (1 and 10 µM) and melatonin (100 µM) for 24 and 48 hours

BACKGROUND: Intense pyrethroid resistance threatens the effectiveness of the primary vector control intervention, insecticide-treated nets (ITNs), in Nigeria, the country with the largest malaria burden globally. In this study, the epidemiological and entomological impact of a new type of ITN (piperonyl-butoxide [PBO] ITNs) distributed in Ebonyi State were evaluated. The epidemiological impact was also compared to the impact of standard pyrethroid-only ITNs in Cross River State.
METHODS: A controlled interrupted time series analysis was conducted on monthly malaria incidence data collected at

West Nile virus (WNV) is the leading mosquito-borne disease causing-pathogen in the United States. Concerningly, there are no prophylactics or drug treatments for WNV and public health programs rely heavily on vector control efforts to lessen disease incidence. Insecticides can be effective in reducing vector numbers if implemented strategically, but can diminish in efficacy and promote insecticide resistance otherwise. Vector control programs which employ mass-fogging applications of insecticides, often conduct these methods during the late-night hours, when diel temperatures are coldest, and

BACKGROUND: Broflanilide is a new insecticide being developed for malaria vector control. As new insecticide chemistries become available, strategies to preserve the susceptibility of local malaria vectors and extend their useful life need to be considered before large scale deployment. This requires the development of appropriate testing procedures and identification of suitable discriminating concentrations for monitoring susceptibility in wild vector populations to facilitate decision making by control programmes.
METHODS: Dose-response WHO bottle bioassays were conducted using the insecti

BACKGROUND: Gains in malaria control are threatened by widespread pyrethroid resistance in malaria vectors across sub-Saharan Africa. New long-lasting insecticidal nets (LLINs) containing two active ingredients (dual active-ingredient LLINs) have been developed to interrupt transmission in areas of pyrethroid resistance. We aimed to evaluate the effectiveness of three dual active-ingredient LLINs compared with standard pyrethroid LLINs against pyrethroid-resistant malaria vectors in rural Tanzania.
METHODS: In this study, we did a secondary analysis of entomological data from a four-group, 3

BACKGROUND: Vector control is still a pivotal method for preventing malaria, and its potency is weakened by the increasing resistance of vectors to chemical insecticides. As the most abundant and vital malaria vector in Southeast Asia, the chemical insecticide resistance status in Anopheles sinensis remains elusive in Laos, which makes it imperative to evaluate the true nature of chemical insecticide resistance-associated genetic mutations in An. sinensis in Laos.
METHODS: Adult An. sinensis were collected from three border regions in Laos. DNA was extracted from individual mosquitoes. PCR am

Despite the developmental toxicity reported in animals, few epidemiologic studies have investigated the potential effects of prenatal exposure to pyrethroid pesticides (PYRs) on fetal growth. A birth cohort study was conducted to examine the association between prenatal exposure to PYRs and birth outcomes, and a nested case-control study was conducted in this cohort to evaluate the effects of PYR on congenital defects. The assessment of PYR exposure was based on self-reported household pesticide use and urinary PYR metabolite levels. We found that pregnant women in this region were ubiquitousl

Pyrethroids are a class of widely-used insecticides that can be transported from terrestrial applications to aquatic systems via runoff and tend to sorb to organic carbon in sediments. Pyrethroid occurrence is detrimental to stream ecosystems due to toxicity to sediment-dwelling invertebrates which are particularly at risk of pyrethroid exposure in urban streams. In this work, 49 streams located in watersheds in the northeastern United States were surveyed for nine current-use pyrethroids using two extraction methods. Total sediment concentrations were determined by exhaustive chemical extract

To reduce morbidity and mortality caused by visceral leishmaniasis (VL), the Brazilian Visceral Leishmaniasis Control and Surveillance Program promotes the diagnosis and treatment of cases, vector control, euthanasia of seropositive dogs, and health education. Nevertheless, the effectiveness of these measures is questionable as they lead to little reduction in the transmission of the disease. Thus, the effectiveness of strategies such as insecticide-impregnated collars, spot-on insecticides, and immunization of dogs should be assessed. Herein, we evaluated the effectiveness of deltamethrin-imp

Synthetic pyrethroids have been widely used in Chile to control the sea lice Caligus rogercresseyi, a major ectoparasite of farmed salmon. Although resistance of C. rogercresseyi to pyrethroids has been reported in Chile, there is no information regarding the geographic extent of this problem. In this study we explored the spatial and temporal variation of C. rogercresseyi's response to pyrethroids in Chile from 2012 to 2013. We modeled lice abundance one week after treatment with a linear mixed-effects regression, and then we performed spatial and spatio-temporal cluster analyses on farm-leve

Pesticides have been used extensively for pest control, resulting in serious pesticide resistance. It is extremely valuable to identify the resistance genes related to pest control. In our previous studies, we reported that Ran is the deltamethrin (DM) resistance-associated gene. To clarify whether Ran is also related to the resistance of other pesticides, we selected four kinds of pesticides, including parathion, DDT, carbaryl and pleocidin, for further study. The results showed that Ran could be up-regulated by all pesticides. We further verified the relationship between Ran and resistance t
