A monoclonal antibody specific to Müller cells and selective synaptic sites in the retina.

We have produced a monoclonal antibody which stains the Müller cells in the region of the photoreceptors, nerve terminals surrounding the horizontal cells, and nerve terminals in the inner plexiform layer in carp, goldfish, and white perch retinas. Electron microscopy showed that the staining in the outer and inner retina was confined to Müller cells and presynaptic terminals, respectively. In the teleost brain, the antibody stained only the optic tectum and efferent fibers from the stratum album centrale. Biochemical characterization by immunoblotting showed that this monoclonal antibody reco…

Characterization of galactose-containing glycoconjugates in the human retina: a lectin histochemical study.

Seven specimens of morphologically normal formalin-fixed and paraffin-embedded human retina were studied using a panel of fourteen biotinylated lectins, all of which react with glycoconjugates containing galactose and N-acetylgalactosamine residues. Agaricus bisporus (ABA), Bauhinia purpurea (BPA), Phaseolus vulgaris (PHA-E), peanut (PNA) and Ricinus communis (RCA-I) agglutinins labeled photoreceptor cells prior to enzymatic predigestion. BPA and PNA bound specifically to cones. The plexiform layers reacted with ABA, BPA and PHA-E, while only ABA and PHA-E labeled the nuclear layers. After pre…

Microglial cell responses in the rabbit retina following transection of the optic nerve.

The response of retinal microglial cells, which accompanies retrograde degeneration of ganglion cell axons and perikarya (induced by transection of the optic nerve), was studied in whole-mounted rabbit retinae labeled enzyme-histochemically for nucleoside diphosphatase (NDPase), which is a microglial cell marker. A few days after transection, the number of microglial cells/mm2, as well as their staining intensity, began to increase in the inner plexiform layer. The mosaic-like distribution of the star-shaped microglial cells present in the inner plexiform layer of a normal rabbit retina was pr…

Actions of excitatory amino acids on brisk ganglion cells in the cat retina.

1. Retinal ganglion cell activity was recorded extracellularly in the intact cat eye. We examined the effects of iontophoretically applied glutamate (GLU), aspartate (ASP), and the specific agonists kainate (KA), quisqualate (QQ), (RS)-alpha-amino-3-hydroxy-5-methyl-4-isoxazole-propionic acid (AMPA), and N-methyl-D-aspartate (NMDA) on the spontaneous and light-driven activity of ganglion cells. 2. ASP and GLU increased the spontaneous as well as the light-driven activity of all brisk cell types. The effects of the two drugs were very similar. The activity of most cells remained at a constant i…

[Post-natal maturation of the retina in the albino rat. I. The pigment epithelium].

The Authors studied the postnatal development of the retinal pigment epithelium in the albino rat, in order to elucidate its morphological and functional evolution, correlated to the numerous functional roles played in Vertebrates (Scheme 1). At birth, epithelial cells show few cytoplasmic organules and the apical surface provided of small depressions. From the third to the fifth postnatal day the first apical microfolds surround the depressions. From the seventh to the ninth day inner segments develop, whilst the apical surface of the epithelial cells is covered by many finger-like microfolds…

Immunocytochemical localization of vitamin A in the retina and pineal organ of the frog, Rana esculenta.

Vitamin A immunoreactive sites were studied in the retina and pineal organ of the frog, Rana esculenta, by the peroxidase antiperoxidase, avidin-biotinperoxidase and immunogold methods. In dark-adapted material, strong immunoreaction was found in the outer and inner segments of the photoreceptor cells of both retina and pineal organ, as well as in the pigment epithelium, retinal Müller cells and pineal ependymal cells. In light-adapted retina, cones and green (blue-sensitive) rods were immunopositive. At the electron microscopic level, immunogold particles were found on the membranes of the ph…

Neovascular glaucoma and the occurrence of twin vessels in congenital arteriovenous communications of the retina.

Two cases with unilateral congenital arteriovenous communications of the retina are described. One of them developed a neovascular glaucoma. Both cases had twin vessels of the retina. The possible significance of twin vessels in these cases is discussed.

The structural basis of the inner blood-retina barrier in the eye of Macaca mulatta.

This is a morphological analysis of the inner blood-retina barrier in various segments of the retinal vasculature in the eye of Macaque monkeys. The primary aims of this study are to identify the components of the walls of the arteries, central capillaries, peripheral capillaries and veins of the retina using light and electron microscopy, and to compare and contrast junctional morphology as revealed by thin section electron microscopy and freeze-fracture. The walls of these vascular segments are composed of continuous endothelium, muscle cells or pericytes, and connective tissue. Endothelial …

A chick neural retina adhesion and survival molecule is a retinol-binding protein.

A 20,000-D protein called purpurin has recently been isolated from the growth-conditioned medium of cultured embryonic chick neural retina cells (Schubert, D., and M. LaCorbiere, 1985, J. Cell Biol., 101:1071-1077). Purpurin is a constituent of adherons and promotes cell-adheron adhesion by interacting with a cell surface heparan sulfate proteoglycan. It also prolongs the survival of cultured neural retina cells. This paper shows that purpurin is a secretory protein that has sequence homology with a human protein synthesized in the liver that transports retinol in the blood, the serum retinol-…

An ultrastructural study of the retina in the Jansky-Bielschowsky type of neuronal ceroid-lipofuscinosis.

A 7 1/2-year-old boy died of the Jansky-Bielschowsky type of neuronal ceroidlipofuscinosis. Although an electroretinogram showed reduced activity, pigmentary retinal atrophy was not demonstrable by ophthalmoscopy two years before death. At autopsy, the retina had severe loss of photoreceptor cells, pigment displacement, gliosis, and uniformly structured lipopigment bodies with an internal structure of curvilinear profiles in ganglionic and Müller cells, in pigment epithelia, in the remaining photoreceptors, and in the elements of the inner nuclear layer. These findings conformed to those in th…
