Alterations in Retinal Signaling Across Age and Sex in 3xTg Alzheimer's Disease Mice.

BACKGROUND: Visual disturbances often precede cognitive dysfunction in patients with Alzheimer's disease (AD) and may coincide with early accumulation of amyloid-β (Aβ) protein in the retina. These findings have inspired critical research on in vivo ophthalmic Aβ imaging for disease biomarker detection but have not fully answered mechanistic questions on how retinal pathology affects visual signaling between the eye and brain. OBJECTIVE: The goal of this study was to provide a functional and structural assessment of eye-brain communication between retinal ganglion cells (RGCs) and their prima

AMPK modulation ameliorates dominant disease phenotypes of CTRP5 variant in retinal degeneration.

Late-onset retinal degeneration (L-ORD) is an autosomal dominant disorder caused by a missense substitution in CTRP5. Distinctive clinical features include sub-retinal pigment epithelium (RPE) deposits, choroidal neovascularization, and RPE atrophy. In induced pluripotent stem cells-derived RPE from L-ORD patients (L-ORD-iRPE), we show that the dominant pathogenic CTRP5 variant leads to reduced CTRP5 secretion. In silico modeling suggests lower binding of mutant CTRP5 to adiponectin receptor 1 (ADIPOR1). Downstream of ADIPOR1 sustained activation of AMPK renders it insensitive to changes in AM

Diagnosability of Synthetic Retinal Fundus Images for Plus Disease Detection in Retinopathy of Prematurity.

Advances in generative adversarial networks have allowed for engineering of highly-realistic images. Many studies have applied these techniques to medical images. However, evaluation of generated medical images often relies upon image quality and reconstruction metrics, and subjective evaluation by laypersons. This is acceptable for generation of images depicting everyday objects, but not for medical images, where there may be subtle features experts rely upon for diagnosis. We implemented the pix2pix generative adversarial network for retinal fundus image generation, and evaluated the ability

Retinal microvascular signs and risk of diabetic kidney disease in asian and white populations.

The objective was to examine prospectively the association between retinal microvascular signs and development of diabetic kidney disease (DKD) in Asian and White populations. We analysed two population-based cohorts, composing of 1,221 Asians (SEED) and 703 White (WESDR) adults with diabetes. Retinal microvascular signs at baseline included vascular caliber (arteriolar-CRAE, and venular-CRVE) and diabetic retinopathy (DR). Incident cases of DKD were identified after ~ 6-year. Incident cases were defined based on eGFR in SEED and proteinuria or history of renal dialysis in WESDR. The incidence

[Retinal vascular network: Changes with aging and systemic vascular disease (cardiac and cerebral)].

For over 10 years, the description of the retinal microvascular network has benefited from the development of new imaging techniques. Automated retinal image analysis software, as well as OCT angiography (OCT-A), are able to highlight subtle, early changes in the retinal vascular network thanks to a large amount of microvascular quantitative data. The challenge of current research is to demonstrate the association between these microvascular changes, the systemic vascular aging process, and cerebrovascular and cardiovascular disease. Indeed, a pathophysiological continuum exists between retina

Retinal changes in Alzheimer's disease- integrated prospects of imaging, functional and molecular advances.

Alzheimer's Disease (AD) is a devastating neurodegenerative disorder of the brain, clinically characterised by cognitive deficits that gradually worsen over time. There is, at present, no established cure, or disease-modifying treatments for AD. As life expectancy increases globally, the number of individuals suffering from the disease is projected to increase substantially. Cumulative evidence indicates that AD neuropathological process is initiated several years, if not decades, before clinical signs are evident in patients, and diagnosis made. While several imaging, cognitive, CSF and blood

In Vivo Assessment of Retinal Biomarkers by Hyperspectral Imaging: Early Detection of Alzheimer's Disease.

A noninvasive and cost-effective means to detect preclinical Alzheimer's disease (AD) and monitor disease progression would be invaluable. The retina is a developmental extension of the brain and has been viewed as a window to evaluate AD-related pathology. Cross-sectional studies have shown structural changes in the retina of AD patients that include thinning of the retinal nerve-fiber layer and changes in retinal vasculature. However, such changes do not manifest in early stages of the disease nor are they specific biomarkers for AD. Described herein is the utilization of our retinal hypersp

Determination of Mitochondrial Oxygen Consumption in the Retina Ex Vivo: Applications for Retinal Disease.

Mitochondrial dysfunction, and consequently altered aerobic energy metabolism, is associated with numerous retinal diseases, including photoreceptor degeneration and diabetic retinopathy. Here, we describe a detailed protocol to directly measure oxygen consumption in the intact retina ex vivo using microplate-based fluorescence technology. We have used this method to assess preferred energy substrate for retinal tissue and suggested its application for investigating mechanisms of retinal disease.
