The importance of cyanide toxicity as a component of inhalational injury in patients with burns is increasingly being recognised, and its prompt recognition and management is vital for optimising burns survival. The evidence base for the use of cyanide antidotes is limited by a lack of randomised controlled trials in humans, and in addition consideration must be given to the concomitant pathophysiological processes in patients with burns when interpreting the literature. We present a literature review of the evidence base for cyanide antidotes with interpretation in the context of patients wit

BACKGROUND: Aminoglycoside antibiotics are some of the most commonly used agents for treating gram-negative bacterial infections. They are extremely efficacious but can result in ototoxicity. It has been postulated that the mechanism inducing damage is the formation of oxygen free radicals. Many compounds have been employed in an attempt to reduce aminoglycoside-induced hearing loss. We endeavour to do likewise using sodium thiosulphate. This free radical scavenging agent has a proven ability to minimize cochlear damage owing to the chemotherapeutic agent cisplatin.
OBJECTIVES: This study had

Cyanide is a serious environmental pollutant and a biocontrol metabolite in plant growth-promoting Pseudomonas species. Here we report on the presence of multiple sulfurtransferases in the cyanogenic bacterium Pseudomonas aeruginosa PAO1 and investigate in detail RhdA, a thiosulfate:cyanide sulfurtransferase (rhodanese) which converts cyanide to less toxic thiocyanate. RhdA is a cytoplasmic enzyme acting as the principal rhodanese in P. aeruginosa. The rhdA gene forms a transcriptional unit with the PA4955 and psd genes and is controlled by two promoters located upstream of PA4955 and rhdA. Bo

A case of attempted homicide by cyanide ingestion is reported. The victim, a 19-year-old woman, unknowingly ingested cyanide and presented to the Emergency Department unresponsive, in shock, and in profound metabolic acidosis. The differential diagnosis of this presentation and the patient's successful treatment are reviewed. The important management issues surrounding the treatment of cyanide poisoning are discussed.

In cyanide poisoning, metalloproteins and carbonyl groups containing proteins are the main target molecules of nucleophilic attack by cyanide. To defend against this attack, cyanide is metabolized to less toxic thiocyanate via transsulfuration. This reaction is catalyzed by rhodanese and mercaptopyruvate sulfurtransferase (MST). Rhodanese is a well characterized mitochondrial enzyme. On the other hand, little was known about MST because it was unstable and difficult to purify. We first purified MST to homogeneity and cloned MST cDNA from rat liver to characterize MST. We also found that MST wa

Oligodeoxyribonucleotides cross-linked by reductively activated mitomycin C (MC) were prepared and purified for the first time. The cross-linked products were structurally characterized by nucleoside and MC-nucleoside adduct analysis. Optimal conditions were established for the cross-linking reaction, resulting in high yields, typically in the 20-50% range. Nuclease digests of the cross-linked oligonucleotides yielded the same bifunctional MC-deoxyguanosine adduct as that previously isolated from DNA exposed to MC in vitro and in vivo [Tomasz et al. (1987) Science 235, 1204]. The cross-linked

Hydrogen sulphide (H₂S) is produced endogenously via two enzymes dependent on pyridoxal phosphate (PLP): cystathionine beta-synthase (CBS, EC 4.2.1.22), cystathionase γ-liase (CTH, EC 4.4.1.1), and a third, 3-mercaptopyruvate sulfurtransferase (MPST, EC 2.8.1.2). H₂S strengthens the defence mechanisms of the gastric mucosal barrier, and plays an important role in gastroprotection, including the increased resistance to damage caused by various irritants and non-steroidal anti-inflammatory drugs. The study was conducted to determine the role of H₂S in ulcerated gastric mucosa of rats caused by i

OBJECTIVE: The objective of this study was to describe the efficacy of sodium thiosulphate (STS) in tumoral calcinosis (TC).
METHODS: The methodology involved the reporting of four retrospective case reports of TC complicating end-stage renal disease (ESRD).
RESULTS: We investigated STS treatment in four patients (two men; ages 46-70 years) with TC. ESRD was secondary to nephronophthisis (n = 1), membranoproliferative glomerulonephritis (n = 1), diabetic nephropathy (n = 1), and thrombotic microangiopathy (n = 1). TC developed 3-28 years after dialysis began and resulted in articular pain (n

The rhodanese homology domain is a ubiquitous fold found in several phylogenetically related proteins encoded by eubacterial, archeal, and eukaryotic genomes. Although rhodanese-like proteins share evolutionary relationships, analysis of their sequences highlights that they are so heterogeneous to form the rhodanese superfamily. The variability occurs at different levels including sequence, active site loop length, presence of a critical catalytic Cys residue, and domain arrangement. Even within the same genome, multiple genes encode rhodanese-like proteins presenting with variably arranged rh

Organ transplantation is the treatment of choice for patients with organ failure. However, the long-term success of this complex life-saving procedure is severely challenged by several inherent factors such as ischemia-reperfusion injury (IRI), an unavoidable pathological condition which occurs due to temporary cessation of blood supply to the donor organ during procurement and cold preservation, and subsequent blood restoration upon engraftment. IRI decreases organ graft quality and function and increases the incidence of post-transplant complications. While the transplant community has adopt

PURPOSE: Measuring clinical practice guideline (CPG)-consistent care through electronic health record (EHR) queries provides a scalable and efficient approach to evaluating quality of care. Template care pathways are used to operationalize CPGs. Goals were to (1) identify care pathway actions for supportive care in pediatric oncology that are amenable to EHR-based evaluation, (2) prioritize five care pathway actions for further assessment, (3) develop EHR queries to identify patients who received care pathway-consistent care, and (4) describe the proportion of pediatric patients with cancer wh

Ototoxicity represents a clinically significant complication of anticancer therapy in pediatric patients. Cytotoxic agents used in oncology, particularly platinum-based chemotherapy, may induce damage to the auditory and vestibular systems, resulting in hearing loss, tinnitus, and balance disturbances. Even mild hearing impairment during childhood may negatively affect speech perception, language development, communication abilities, and subsequent educational and psychosocial functioning. This narrative review aims to synthesize current evidence on treatment-related ototoxicity in children, w

INTRODUCTION: Salmonella is an important foodborne pathogen that can induce severe diseases such as gastrointestinal disease and typhoid fever. Accumulating evidence revealed that Salmonella's resistance to antibiotics also seriously affects human health. Pathogenic Salmonella enterica serovar Goldcoast (S. Goldcoast) was first detected in 2010 in China and was predicted to have an increasing tendency.
METHODS: The MacConkey agar, Salmonella Shigella agar, three-sugar iron agar slant, and Gram-stained microscopic examination were used for strain identification. Gram-negative bacteria identifi

INTRODUCTION: chironomid larvae are present in aquatic systems, including freshwater, and often are important hosts for Vibrio cholerae. Chironomid larvae interact with V. cholera and may shield the bacterium from environmental stressors, resulting in its dissemination. This study aimed to determine the role of chironomid larvae in the persistence of V. cholera in the water reservoirs of a cholera-endemic area of Cameroon.
METHODS: chironomid larvae and water samples were collected from scoop holes of the Kaliao and Mizao temporary streams in Maroua during the cholera inter-epidemic (dry) sea

There is growing evidence for the potential of biochars (BCs) in remediating mercury-contaminated paddy soils, but the high doses commonly used in laboratory studies discourage BC application in practice. To address these difficulties, we compared the effects of varying amounts of BCs from different sources on the formation of methylmercury (MeHg) in soil and its accumulation in rice through microcosm and pot experiments. The addition of a wide range of added doses (0.3, 0.6, 1, 2, 4 and 5 %, w/w) of BCs derived from different biomass feedstocks (i.e., corn stalk, wheat straw, bamboo, oak and

Near the end of 2019, SARS-CoV-2, a novel highly contagious coronavirus phylogenetically related to the SARS virus, entered the human population with lethal consequences. This special issue devoted to the resulting disease COVID-19 was not planned but instead the articles accumulated organically as researchers in the cell stress response field noticed similarities among the pathophysiology of COVID-19 infections and the responses that they studied in contexts unrelated to viral infection. We preface the issue with an introductory article which begins with a brief review of the structure and bi
