Beyond canvas: behavioral onset of rfc1-expansion disease in an Italian family-causal or casual?

INTRODUCTION: Biallelic intronic AAGGG repeat expansion in the replication factor C subunit 1 (RFC1) gene was recently identified in two/third of patients with cerebellar ataxia, sensory neuropathy, and bilateral vestibular areflexia syndrome (CANVAS). The phenotypic spectrum has expanded since (i.e., parkinsonism, motor neuron involvement, cognitive decline); no behavioral symptoms have been reported yet. CASE REPORT: We report an Italian family that met the diagnostic criteria for CANVAS, and RFC1-expansion was detected in five of seven. All the affected members presented behavioral-psychia

Locations of androgen-concentrating cells in the brain of Xenopus laevis: autoradiography with 3H-dihydrotestosterone.

The distribution of hormone-concentrating cells in the brains of South African clawed frogs, Xenopus laevis, was examined autoradiographically after the administration of 3H-dihydrotestosterone. Hormone-accumulating cells were found in cranial nerve nucleus IX-X and adjacent smaller cells, a presumed medullary vestibular nucleus, a presumed sensory nucleus of cranial nerve V, dorsal tegmental area of the medulla, laminar nucleus of the torus semicircularis, ventral thalamus, and anterior pituitary. The pattern of dihydrotestosterone-labelled cells differs from previously reported results follo

Age-related degeneration of the serotoninergic fibers in the zitter rat brain.

The serotonin neuron system was studied using immunohistochemical and neurochemical techniques in zitter mutant rats aged 1-14 months, which are characterized by abnormal metabolism of superoxides. The morphology of the serotoninergic neuron system and the serotonin level in the zitter rat were compared to those of age-matched Sprague-Dawley (SD) rats. Up to age 4 month, the density and distribution of serotoninergic fibers in the zitter rat brain were similar to those of control rats. However, several serotoninergic fibers with abnormal morphology, characterized by swollen varicosities, were

Central projections of lagenar primary neurons in the chick.

Perception of linear acceleration and head position is the function of the utricle and saccule in mammals. Nonmammalian vertebrates possess a third otolith endorgan, the macula lagena. Different functions have been ascribed to the lagena in arboreal birds, including hearing, equilibrium, homing behavior, and magnetoreception. However, no conclusive evidence on the function of the lagena in birds is currently available. The present study is aimed at providing a neuroanatomical substrate for the function of the lagena in the chicken as an example of terrestrial birds. The afferents from the lage

Antisecretory Factor in rat cerebellar granule cells augments inhibitory current via GABAA receptors: a mechanism for vertigo reduction in Menière's disease?

UNLABELLED: In this review we propose a mechanism by which the antisecretory factor AF may decrease vertigo symptoms in Menière’s disease patients. This review is based on our previously published work and suggests a way of interpretation of the results obtained in view of the clinical effect of AF factor against vertigo caused by Menière’s disease. The Antisecretory Factor (AF) is an endogenous protein regulating water and ion transport across cellular membranes. AF was originally discovered and isolated from rat and pig pituitary gland but later it has been isolated from all mammalian and no

High-grade astrocytoma in a sheep: clinical, pathology, and genetic investigations.

Primary neoplastic diseases of the CNS in livestock are rare compared to companion animals. Here we describe the clinical, pathologic, and genetic findings in an adult sheep with a high-grade cerebral astrocytoma. Clinically, apathy, ataxia, bilateral amaurosis, and horizontal resting nystagmus were observed. The signs were suggestive of a space-occupying lesion affecting the vestibular and ascending reticular activating systems. The CSF had high total protein, hypercellularity, and pleocytosis. Macroscopically, a friable, gray-pink, ovoid mass (6 × 3 × 4 cm) was observed within most of the ri

Bio-Inspired Central Pattern Generator for Adaptive Gait Generation and Stability in Humanoid Robots on Sloped Surfaces.

Existing research has preliminarily achieved stable walking in humanoid robots; however, natural human-like leg motion and adaptive capabilities in dynamic environments remain unattained. This paper proposes a bionic central pattern generator (CPG) gait generation method based on Kimura neurons. The method maps the CPG output to the spatial motion patterns of the robot's center of mass (CoM) and foot trajectory, modulated by 22 undetermined parameters. To address the vague physical interpretation of CPG parameters, the strong neuronal coupling, and the difficulty of decoupling, this research s

Velocity storage mechanism drives a cerebellar clock for predictive eye velocity control.

Predictive motor control is ubiquitously employed in animal kingdom to achieve rapid and precise motor action. In most vertebrates large, moving visual scenes induce an optokinetic response (OKR) control of eye movements to stabilize vision. In goldfish, the OKR was found to be predictive after a prolonged exposure to temporally periodic visual motion. A recent study showed the cerebellum necessary to acquire this predictive OKR (pOKR), but it remained unclear as to whether the cerebellum alone was sufficient. Herein we examined different fish species known to share the basic architecture of c

Auditory Detection Thresholds and Cochlear Resistivity Differ Between Pediatric Cochlear Implant Listeners With Enlarged Vestibular Aqueduct and Those With Connexin-26 Mutations.

Purpose The goal of this study was to evaluate differences in the electrode-neuron interface as a function of hearing loss etiology in pediatric cochlear implant (CI) listeners with enlarged vestibular aqueduct (EVA) syndrome and in those with autosomal recessive connexin-26 mutations (DFNB1). Method Fifteen implanted ears (9 participants, 5 ears with EVA, 10 ears with DFNB1) were assessed. Single-channel auditory detection thresholds were measured using broad and spatially focused electrode configurations (steered quadrupolar; focusing coefficients = 0 and 0.9). Cochlear resistivity estimates

Type 1 vanilloid receptor expression by mammalian inner ear ganglion cells.

The type 1 vanilloid receptor (VR1) is a non-specific cation channel activated by capsaicin, lipoxygenase (LOX) products, heat and acid. This study demonstrates VR1 and 5-LOX expression by inner ear ganglion cells. A PCR product (210 bp) was amplified from both oligo(dT)- and random primer-generated cDNAs of rat spiral ganglion cells using VR1 gene-specific primers constructed from the 3' non-homologous region. This PCR product shared 100% sequence homology to a rat VR1 cDNA (GenBank accession no. AF029310) and a rat vanilloid receptor splice variant mRNA (GenBank accession no. AF158248). Froz

Motor neuron pathology in CANVAS due to RFC1 expansions.

CANVAS caused by RFC1 biallelic expansions is a major cause of inherited sensory neuronopathy. Detection of RFC1 expansion is challenging and CANVAS can be associated with atypical features. We clinically and genetically characterized 50 patients, selected based on the presence of sensory neuronopathy confirmed by EMG. We screened RFC1 expansion by PCR, repeat-primed PCR, and Southern blotting of long-range PCR products, a newly developed method. Neuropathological characterization was performed on the brain and spinal cord of one patient. Most patients (88%) carried a biallelic (AAGGG)n expans

Return to Play Following Concussion: Role for Imaging?

This review surveys concussion management, focusing on the use of neuroimaging techniques in return to play (RTP) decisions. Clinical assessments traditionally were the foundation of concussion diagnoses. However, their subjective nature prompted an exploration of neuroimaging modalities to enhance diagnosis and management. Magnetic resonance spectroscopy provides information about metabolic changes and alterations in the absence of structural abnormalities. Diffusion tensor imaging uncovers microstructural changes in white matter. Functional magnetic resonance imaging assesses neuronal activi

Fabry's disease as a differential diagnosis of MS.

Fabry's disease is a genetically inherited error of glycosphingolipid metabolism that results from the defective activity of the lysosomal enzyme alpha-galactosidase A (alpha-GalA). The enzymatic defect, caused by an X-linked recessive genes, leads to progressive deposition of neutral glycosphingolipids (predominantly globotriaosylceramide), with terminal alpha-galactosyl moieties, in most visceral tissues and fluids of the body. Cerebrovascular manifestations result from multifocal small-vessel involvement and may include thromboses, basilar arterial ischaemia and aneurysm, seizures, paroxyst

Mumps labyrinthitis, endolymphatic hydrops and sudden deafness in succession in the same ear.

Acute sensorineural hearing loss, mostly unilateral and reversible, is a well-known complication to mumps. Secondary endolymphatic hydrops, Ménière's syndrome, has rarely been associated with a previous mumps infection. This paper presents the case report of a woman who experienced unilateral hearing loss, vestibular symptoms and a caloric depression on the same ear during mumps. The symptoms and findings were reversible. Twelve years later she developed Ménière's symptoms in the same ear. This continued for 2 years after which she suddenly had a sensorineural hearing loss. This was localized

Evaluation of patients with acute vestibular syndrome.

Acute vestibular syndrome is characterized by a rapid unilateral injury to either peripheral or central vestibular structures. It consists of severe vertigo, nausea and vomiting, spontaneous nystagmus, and postural instability. In many cases, a peripheral etiology is considered although it may be due to an underlying serious central pathology. The present study was designed to investigate the feasibility of differentiating the cause of acute vestibular syndrome in such patients using clinical, audiovestibular and radiologic tools. We performed a case series study of patients complaining of acu
