Systems-Level Transcriptomic and Cellular Architecture of the Human Striatum in Schizophrenia and Bipolar Psychosis.

Psychosis involves striatal circuit dysfunction, but the regional molecular and cellular changes remain unclear. We re-analyzed 215 postmortem RNA-seq samples (72 donors: 36 controls, 28 schizophrenia, 8 bipolar disorder with psychosis) from the nucleus accumbens, caudate and putamen, asking whether diagnosis-associated transcriptional changes are region-specific and whether they reflect cellular composition or cell-intrinsic state. Region-specific limma-voom models identified 0, 141 and 2152 schizophrenia-associated genes (FDR < 0.10) in the nucleus accumbens, caudate and putamen; the small b

Emerging novel therapies for the treatment of hereditary haemorrhagic telangiectasia.

Hereditary haemorrhagic telangiectasia is the second most common heritable bleeding disorder worldwide, with a prevalence more than twice that of haemophilia, and is the most morbid inherited bleeding disorder of women. As an autosomal dominant vasculopathy due to dysregulated angiogenic signalling, patients with hereditary haemorrhagic telangiectasia develop telangiectasias and arteriovenous malformations which cause numerous complications. These complications include moderate-to-severe chronic epistaxis, gastrointestinal bleeding, iron deficiency anemia, and requirement for intravenous iron

Development of pharmacological interventions for the treatment of sarcopenia.

Sarcopenia is a multifactorial age-related muscle disorder of which its underlying pathophysiological mechanisms remain incompletely understood. To date, treatment strategies of sarcopenia have been largely confined to lifestyle interventions, underscoring an urgent need for pharmacological options that directly target the biological drivers of muscle deterioration. Despite increasing interest in drug development for sarcopenia, no specific pharmacological agent has yet received regulatory approval. This review provides a comprehensive synthesis of recent advances in investigational pharmacoth

Marked clinical and haemodynamic improvement with sotatercept in severe refractory pulmonary arterial hypertension associated with corrected complex congenital heart disease.

BACKGROUND: Pulmonary arterial hypertension (PAH) is a progressive disease associated with high morbidity despite combination therapy. Sotatercept, a recombinant fusion protein that acts as an activin signaling inhibitor, has shown significant clinical and haemodynamic benefit in phase 3 trials. However, evidence in PAH associated with congenital heart disease (PAH-CHD) is very limited. CASE PRESENTATION: 37-year-old woman with repaired pulmonary atresia with ventricular septal defect and major aortopulmonary collateral arteries, who developed severe PAH years later. Despite treatment with ta

Doxycycline for epistaxis in hereditary hemorrhagic telangiectasia: genotype-stratified outcomes.

Patients with hereditary hemorrhagic telangiectasia (HHT) experience recurrent epistaxis. Doxycycline has been proposed as a possible treatment, although its efficacy remains controversial. Whether the most common genotypes (activin receptor-like kinase 1, endoglin, and SMAD4) contribute to a differential response has not been investigated. In this study, we evaluate the effectiveness of doxycycline among the different HHT genotypes. A retrospective cohort study was conducted at the University of Florida's Hereditary Hemorrhagic Telangiectasia Center. Forty-one adult patients (aged ≥18 years)

FGF Signaling Potentiates Müller Glia for Mammalian Retinal Regeneration.

Müller glia possess latent regenerative potential that could be harnessed to restore retinal neurons lost to injury or disease. Although fibroblast growth factor (FGF) signaling is upregulated following retinal damage, its role in mammalian retinal regeneration remains unclear. Here, we investigated the function of FGF signaling in Müller glial reprogramming using genetic, pharmacological, and single-cell transcriptomic approaches. Activation of FGF signaling alone was insufficient to induce Müller glial proliferation in the mouse retina. However, conditional deletion of FGFR1/2 in Müller glia

Exploring the therapeutic potential of GLP-1 receptor agonists in pulmonary arterial hypertension.

Pulmonary hypertension (PH) is a progressive and multifactorial disease marked by elevated pulmonary arterial pressure and right ventricular (RV) dysfunction. Among its subtypes, pulmonary arterial hypertension (PAH) is characterised by profound vascular remodelling, inflammation and fibrosis. Despite therapeutic advances targeting the endothelin, nitric oxide, prostacyclin, and, more recently, the activin signalling pathway via sotatercept, outcomes in PAH remain suboptimal, particularly in patients with cardiovascular comorbidities. This has prompted growing interest in novel metabolic-based

Independent prognostic value of semaphorin-4D, interleukin-1β and complement activation in newly diagnosed multiple myeloma patients.

Multiple myeloma represents a systemic disease of the bone marrow (BM) niche, in which immune and skeletal pathways are tightly interconnected. However, the independent prognostic significance of bone and immune-related markers in newly diagnosed multiple myeloma (NDMM) remains incompletely understood. Semaphorin (Sema) 4D, activin-A, and periostin ELISA, LEGENDplex™ Human Bone Metabolism Panel and proteomic analysis for novel biomarker identification were conducted in 71 consecutive samples from NDMM patients. In 25 patients, genomic analysis was performed on sorted clonal plasma cells. NDMM

Targeted Therapies in Myelofibrosis: Present Landscape, Ongoing Studies, and Future Perspectives.

Myelofibrosis (MF) is a myeloproliferative neoplasm that is accompanied by driver JAK2, CALR, or MPL mutations in more than 90% of cases, leading to constitutive activation of the JAK-STAT pathway. MF is a multifaceted disease characterized by trilineage myeloid proliferation with prominent megakaryocyte atypia and bone marrow fibrosis, as well as splenomegaly, constitutional symptoms, ineffective erythropoiesis, extramedullary hematopoiesis, and a risk of leukemic progression and shortened survival. Therapy can range from observation alone in lower-risk and asymptomatic patients to allogeneic

The effects of anti-obesity medications on bone metabolism: A critical appraisal.

The development of novel sophisticated medications that induce weight loss has revolutionized the management of people living with obesity (PwO). However, when body weight is reduced, muscle and bone are lost along with fat. In the present review, we quote and discuss existing evidence on the effects of the major anti-obesity medications on bone metabolism. Glucagon-like peptide-1 receptor (GLP-1R) agonists have shown a positive impact in preclinical studies but a neutral or negative, albeit not clinically significant, effect on bone turnover markers and bone mineral density in clinical studie

Bimagrumab: Novel Medical Therapy for Inclusion Body Myositis, Sarcopenia, and Medication-Induced Lean Body Mass Loss.

Bimagrumab is a monoclonal antibody that targets activin type II receptors, blocks myostatin and related proteins: it promotes muscle growth and prevents muscle loss, while causing weight loss. It has been effective in sporadic inclusion body myositis with promising results, a disease that had no previous effective therapy. It has also been studied in sarcopenia, leading to improved muscle mass and reduced fat mass. It has been very effective in obese patients with type 2 diabetes, leading to reduced fat mass, increased lean body mass, and improved insulin sensitivity. Subcutaneous administrat

Non-coding RNAs involved in the molecular pathology of Alzheimer's disease: a systematic review.

UNLABELLED: Alzheimer's disease (AD) is the leading cause of dementia globally, having a pathophysiology that is complex and multifactorial. Recent findings highlight the significant role of non-coding RNAs (ncRNAs), specifically microRNAs (miRNAs), long non-coding RNAs (lncRNAs), circular RNAs (circRNAs), and piwi-interacting RNAs (piRNAs) in the molecular mechanisms underlying AD. These ncRNAs are involved in critical biological processes such as cell proliferation, apoptosis, oxidative stress, amyloid-beta aggregation, tau phosphorylation, neuroinflammation, and autophagy, which are pivotal

Shear Stress and Sub-Femtomolar Levels of Ligand Synergize to Activate ALK1 Signaling in Endothelial Cells.

Endothelial cells (ECs) respond to concurrent stimulation by biochemical factors and wall shear stress (SS) exerted by blood flow. Disruptions in flow-induced responses can result in remodeling issues and cardiovascular diseases, but the detailed mechanisms linking flow-mechanical cues and biochemical signaling remain unclear. Activin receptor-like kinase 1 (ALK1) integrates SS and ALK1-ligand cues in ECs; ALK1 mutations cause hereditary hemorrhagic telangiectasia (HHT), marked by arteriovenous malformation (AVM) development. However, the mechanistic underpinnings of ALK1 signaling modulation

Bidirectional Mendelian Randomization Highlights Causal Relationships Between Circulating INHBC and Multiple Cardiometabolic Diseases and Traits.

Human genetic and transgenic mouse studies have highlighted a potential liver-adipose tissue endocrine axis, involving activin C (Act-C) and/or Act-E and ALK7, influencing fat distribution and systemic metabolism. We investigated the bidirectional effects between circulating INHBC, which homodimerizes into Act-C, and adiposity traits, insulin resistance, inflammation, and cardiometabolic disease risk. Additionally, we examined whether Act-C is an ALK7 ligand in human adipocytes. We used Mendelian randomization and in vitro studies in immortalized human abdominal and gluteal adipocytes. Circula

Momelotinib: Mechanism of action, clinical, and translational science.

Myelofibrosis is a chronic myeloproliferative disorder characterized by bone marrow fibrosis, splenomegaly, anemia, and constitutional symptoms, with a median survival of ≈6 years from diagnosis. While currently approved Janus kinase (JAK) inhibitors (ruxolitinib, fedratinib) improve splenomegaly and symptoms, most can exacerbate myelofibrosis-related anemia, a negative prognostic factor for survival. Momelotinib is a novel JAK1/JAK2/activin A receptor type 1 (ACVR1) inhibitor approved in the US, European Union, and the UK and is the first JAK inhibitor indicated specifically for patients with

HOXA3 functions as the on-off switch to regulate the development of hESC-derived third pharyngeal pouch endoderm through EPHB2-mediated Wnt pathway.

OBJECTIVES: Normal commitment of the endoderm of the third pharyngeal pouch (3PP) is essential for the development and differentiation of the thymus. The aim of this study was to investigate the role of transcription factor HOXA3 in the development and differentiation of 3PP endoderm (3PPE) from human embryonic stem cells (hESCs). METHODS: The 3PPE was differentiated from hESC-derived definitive endoderm (DE) by mimicking developmental queues with Activin A, WNT3A, retinoic acid and BMP4. The function of 3PPE was assessed by further differentiating into functional thymic epithelial cells (TEC

[Research progress on molecular mechanism related to skeletal muscle atrophy].

The maintenance of skeletal muscle quality involves various signal pathways that interact with each other. Under normal physiological conditions, these intersecting signal pathways regulate and coordinate the hypertrophy and atrophy of skeletal muscles, balancing the protein synthesis and degradation of muscle. When the total rate of protein synthesis exceeds that of protein degradation, the muscle gradually becomes enlarged, while when the total rate of protein synthesis is lower than that of protein degradation, the muscle shrinks. Myocyte atrophy mainly involves two protein degradation path
