[Relationship of single nucleotide polymorphism, environmental factors and the hereditary susceptibility of hepatocellular carcinoma].

Resulting by the interaction between the hereditary factor and the environmental factor, the occurrence and development of hepatocellular carcinoma (HCC) is a complicated course with multi-genes and multi-factors. As the third genetic marker, the single nucleotide polymorphism reflects the hereditary difference among individuals, decides the susceptibility to disease and becomes an important tool to study the hereditary susceptibility of HCC.

A dual signal ratio electrochemical sensor based on DNA tetrahedrons and bimetallic nanocomposites for AFB1 analysis.

Given the hepatotoxicity and widespread contamination of Aflatoxin B1 (AFB1), developing ultra-sensitive, anti-interference analytical platforms is paramount for public health. Herein, a dual signal ratio electrochemical sensing platform for accurate AFB1 analysis was constructed integrating DNA tetrahedrons (TNDA), a dynamic DNA walker, and nitrogen-doped graphene oxide-supported hollow silver-platinum bimetallic nanospheres (NGR-HP-AgPt). The NGR-HP-AgPt cavity provides a uniform microenvironment, synergistically promoting catalytic effects to significantly accelerate interfacial electron tr

Mycotoxin binder for immune and intestinal histopathology ameliorations against Newcastle disease in vaccinated broilers.

BACKGROUND: In broiler farming, vaccination against Newcastle disease (ND) is essential. Nonetheless, during the post-vaccination phase, production may be negatively impacted by mycotoxin contamination in feed. This study aimed to evaluate the effect of mycotoxin binders on immune and intestinal histopathology ameliorations against ND in vaccinated broilers with aflatoxin B1 (AFB1) and ochratoxin A (OTA) toxication. METHODS: A total of 20 broilers were randomly assigned into 4 groups with 5 replications i.e. (C-) broiler groups with basal feed, (C+) broiler groups with AFB1 and OTA feed conta

Mycotoxin-free Aspergillus oryzae strain lineage for alternative and novel protein production at industrial scale.

Advanced industrial strains of Aspergillus oryzae have been used for decades for the production of recombinant proteins including food and feed enzymes at large scale. The A. oryzae strain lineage evaluated in this review derives from the proprietary Novozymes (now Novonesis) strain collection. A. oryzae wild-type strains have the potential to produce three different mycotoxins (aflatoxins (AFL), cyclopiazonic acid (CPA), and 3-nitropropionic acid (3-NPA)). Here, we review the work originally performed at Novozymes to identify a strain (BECh1) that contained a large chromosomal deletion compri…

The genetic and environmental basis of hepatocellular carcinoma.

Liver cancer is among the most common malignancies, impacting significantly across all societies worldwide. The lethal impact of this prevalent cancer is unlikely to change considerably in the near future due to a limited understanding of disease pathogenesis on the molecular, cellular, and environmental levels and how current knowledge might be converted into new preventive, therapeutic and diagnostic approaches. This article highlights the current challenges and opportunities in this critical area of unmet need.

[Differentially expressed genes in hepatocellular carcinoma of tree shrew induced by different factors].

BACKGROUND & OBJECTIVE: Previous studies on differentially expressed genes in hepatocellular carcinoma (HCC) used to perform with para-cancerous tissues as normal control. However, the para-cancerous tissue of HCC is actually abnormal because they frequently contain hepatitis, cirrhosis, hyperplastic nodules or foci, etc. In order to explore the molecular mechanism and the responsible genes for hepatocarcinogenesis, through applying the HCC model of tree shrew (Tupaia belangeri chinensis), this study was designed to compare gene expression levels between HCC induced by different factors and th

Pathogenesis of hepatocellular carcinoma.

The pathogenesis of HCC is poorly understood at present. There is insufficient understanding to propose a robust general model of hepatic carcinogenesis, partly because pathogenic host and environmental factors show significant regional variation, making such generalization difficult. Figure 4 is a model based on data presented in this article. Multiple risk factors for HCC have been identified, including cirrhosis, male gender, increasing patient age, toxins, chronic viral hepatitis, and other specific liver diseases. The understanding of how the individual risk factors result in genetic chan

Mycotoxins and plant alkaloids in plant-based protein foods and meat alternatives: Occurrence, exposure and risk assessment.

Plant-based protein-rich foods and meat alternatives are increasingly used as substitutes or supplements to animal-based products. Despite their potential nutritional and environmental benefits, data on their contamination with natural toxins remain limited, particularly for plant alkaloids. In this study, 57 mycotoxins, 16 tropane alkaloids and 32 pyrrolizidine alkaloids were analysed in 101 plant-based protein samples, including protein isolates/concentrates, extruded products, veggie burgers and other meat alternatives, using U-HPLC-HRMS/MS. Twenty-five mycotoxins and eight alkaloids were q

Nickel Core-Thin Iridium Shell Nanoparticles as Cost-Effective and Highly Active Peroxidase-Mimicking Nanozymes.

Peroxidase-mimicking nanozymes have emerged as powerful catalytic alternatives to natural enzymes for a wide range of applications. Among them, iridium (Ir)-based nanozymes exhibit exceptional intrinsic peroxidase-like activity. However, the high cost and scarcity of noble-metal Ir severely limit practical and large-scale use. Herein, we address this challenge by engineering a core-shell Ni-Ir nanozyme in which inexpensive nickel (Ni) serves as the core and catalytically active Ir forms a thin surface shell, resulting in a low Ir content of only 4.9 at.% (denoted as Ir4.9%Ni). This architectur

Whole-exome sequencing reveals genomic landscape of intrahepatic cholangiocarcinoma and identifies SAV1 as a potential driver.

Intrahepatic cholangiocarcinoma (ICC) is the second most common primary hepatic malignancy after hepatocellular carcinoma, with poor prognosis and limited treatment options. The genomic features of ICC in Chinese patients remain largely unknown. In this study, we perform deep whole-exome sequencing of 204 Chinese primary ICCs and characterize genomic alterations and clonal evolution, and reveal their associations with patient outcomes. We identify six mutational signatures, including Signatures A and F, which are highly similar to previously described signatures linked to aristolochic acid and

Hepatocyte Thorns, A Novel Drug-Induced Stress Response in Human and Mouse Liver Spheroids.

The in vivo-relevant phenotype of 3D liver spheroids allows for long-term studies of, e.g., novel mechanisms of chronic drug-induced liver toxicity. Using this system, we present a novel drug-induced stress response in human and murine hepatocyte spheroids, wherein long slender filaments form after chronic treatment with four different drugs, of which three are PPARα antagonists. The morphology of the thorns varies between donors and the compounds used. They are mainly composed of diverse protein fibres, which are glycosylated. Their formation is inhibited by treatment with fatty acids or anti

In Vivo and In Vitro Models of Hepatocellular Carcinoma: Current Strategies for Translational Modeling.

Hepatocellular carcinoma (HCC) is the sixth most common cancer worldwide and the third leading cause of cancer-related death globally. HCC is a complex multistep disease and usually emerges in the setting of chronic liver diseases. The molecular pathogenesis of HCC varies according to the etiology, mainly caused by chronic hepatitis B and C virus infections, chronic alcohol consumption, aflatoxin-contaminated food, and non-alcoholic fatty liver disease associated with metabolic syndrome or diabetes mellitus. The establishment of HCC models has become essential for both basic and translational

Environmental and Physiological Barriers to Child Growth and Development.

Aggregated analyses of child growth in low- and middle-income countries (LMICs) reveal a remarkably consistent picture of serious growth failure compared to the WHO reference growth curves. Impoverished diets with low dietary diversity are a key driver of poor growth, but there are important additional environmental factors that limit the uptake and utilization of nutrients. This paper considers such factors. A large proportion of the rapid growth deterioration in later infancy can be ascribed to infections and to wider nonspecific effects of living in an unhygienic environment, including the

Design of a Dual Channel Self-Reference Photoelectrochemical Biosensor.

Photoelectrochemical (PEC) biosensors are usually based on the single photocurrent change caused by biorecognition events between analytes and probes. However, the photocurrent may be influenced by other factors besides target analytes and bring a false result. To improve the accuracy and reliability of PEC detection, here we proposed the design of a dual channel self-reference PEC biosensors. CdTe and CdTe-graphene oxide (GO) were chosen as the two PEC active material and modified onto two adjacent areas on the ITO electrode. Then they were functionalized with Aflatoxin B1 (AFB1) aptamer thro

Somatic mutations in human cancer: applications in molecular epidemiology.

The tumour suppressor protein p53 mediates cell-cycle arrest, DNA repair and apoptosis after activation by multiple forms of cellular stresses. When activated, this "master protein" modulates its response depending on the type and intensity of the stress. The TP53 gene with its nearly 20,000 described mutations is the most mutated gene in cancer. Most mutations are missense and occur at over 200 codons within the central portion of the gene. In several cancers, the distribution of mutation types and sites follow a specific pattern reflecting the effects of environmental mutagens. An example fo

Hepatocellular carcinoma: the high-risk patient.

Hepatocellular carcinoma (HCC) is the most common hepatic malignancy worldwide. The primary risk factor for the development of HCC is cirrhosis. Even patients without cirrhosis who develop HCC are typically found to have some underlying hepatic abnormality, such as steatohepatitis or chronic viral hepatitis. Although cirrhosis of any cause increases the risk of developing HCC, cirrhosis associated with chronic hepatitis B or C virus infection or hemochromatosis carries the greatest risk. Additional factors such as patient age and sex, duration and severity of liver disease, concurrent alcohol

Xeroderma pigmentosum and related disorders: examining the linkage between defective DNA repair and cancer.

Xeroderma pigmentosum, Cockayne syndrome, the xeroderma pigmentosum-Cockayne syndrome complex, and trichothiodystrophy cells have defects in DNA repair and are associated with clinical and cellular hypersensitivity to ultraviolet radiation (UV). Familial dysplastic nevus syndrome cells have UV hypermutability. Although xeroderma pigmentosum and dysplastic nevus syndrome have markedly increased cancer risk. Cockayne syndrome and trichothiodystrophy do not. At the molecular level, these disorders are associated with several different genetic defects as evidenced by the existence of multiple over

The importance of glutathione and glutathione transferase for somatic mutations in Drosophila melanogaster induced in vivo by 1,2-dichloroethane.

Two principal pathways of metabolism of the carcinogenic compound 1,2-dichloroethane (DCE) have been proposed. One is a mixed function oxidase dependent pathway requiring oxygen and NADPH. The other pathway depends on the presence of glutathione (GSH) and glutathione transferase (GST). The aim of this study was to investigate the role of the latter pathway for the in vivo mutagenicity of DCE in the somatic wing spot test in Drosophila melanogaster. DCE caused a dose-dependent increase of wing spots. In order to investigate the role of cellular GSH for the mutagenicity, the level of GSH was dec

Reversion of 6-thioguanine resistant Chinese hamster cell lines: agent specificity and evidence for the repair of promutagenic lesions.

The kinetics and mutagen specificity of reversion of an HGPRT(-)TG(R) line of Chinese hamster cells have been examined in detail by measuring the frequency of HAT(R) colonies. Alkylating agents which produce relatively high levels of O-atom reaction were effective in inducing reversion. MMS, DMS and u.v. were less efficient, and aflatoxin B1, acridine orange and N-acetoxy-AAF were completely ineffective. For agents which were effective, the relationship between HAT(R) colony frequency and dose of mutagen was linear at early expression times (6 h). HAT(R) colony frequency fell subsequently at a
