High-dose alkylating agents with autologous hematopoietic stem cell support and trastuzumab in ERBB2 overexpressing metastatic breast cancer: a feasibility study.

BACKGROUND: ERBB2 overexpression predicts a worse outcome for patients receiving high-dose chemotherapy (HDC). Trastuzumab improves response rate and survival in ERBB2 overexpressing metastatic breast cancer patients (MBC). We investigated the feasibility of combining high-dose alkylating agents with autologous hematopoietic stem cell (AHSC) support and trastuzumab in ERBB2 overexpressing MBC. PATIENTS AND METHODS: Eleven consecutive patients with pre-treated ERBB2 overexpressing MBC were enrolled. HDC regimen consisted of a single course of cyclophosphamide 120 mg/kg + melphalan 140 mg/m2 (C

Conversion of alcohols, thiols, and trimethysilyl ethers to alkyl cyanides using triphenylphosphine/2,3-dichloro-5,6-dicyanobenzoquinone/n-Bu(4)NCN.

Triphenylphosphine and 2,3-dichloro-5,6-dicyanobenzoquinone afford an adduct, which in the presence of n-Bu(4)NCN converts alcohols, thiols, and trimethylsilyl ethers into their corresponding alkyl cyanides in good to excellent yields at room temperature. This method is highly selective for the conversion of 1 degrees alcohols in the presence of 2 degrees and 3 degrees ones, thiols and silyl ethers.

Application of a TCA-precipitation method for the determination of 1-alkyl-sn-glycero-3-phosphate: Acetyl-CoA acetyltransferase in human renal tissue.

The activity of 1-alkyl-sn-glycero-3-phosphate:Acetyl-CoA acetyltransferase, which catalyses the first step of the de novo biosynthesis of PAF, was determined and characterised in cortical and medullary human renal tissues. A novel thin-layer chromatographic system as well as a trichloroacetic acid precipitation method, were utilised in order to determine the enzyme's activity. The acetyltransferase activity was associated with the membranous fractions of the renal tissue, it showed an optimum pH of 8.4 and it had a bell-shaped dependence on BSA concentration. One or more disulphide bonds were

Application of the restricted-access precolumn packing material alkyl-diol silica in a column-switching system for the determination of ketoprofen enantiomers in horse plasma.

The group of LiChrospher ADS (alkyl-diol silica) sorbents that make part of a unique family of restricted-access materials, have been developed as special packings for precolumns used in the LC-integrated sample processing of biofluids. The advantage of these sorbents lies in the direct injection of untreated biological fluids, that is without sample clean-up, the elimination of the protein matrix with a quantitative recovery together with an on-column enrichment. The present method is based on previous work applying UV detection at 260 nm for ketoprofen determinations. Plasma samples introduc

Induction of complete haematological remission after monotherapy with anti-CD20 monoclonal antibody (RITUXIMAB) in a patient with alkylating agent resistant Waldenström's macroglobulinaemia.

Response of Waldenström's macroglobulinaemia to chemotherapy with alkylating agents is usually only transient. We report a case with marked bone marrow involvement and resistance to chemotherapy with alkylating agents. The patient was red cell and platelet transfusion dependent. Three weeks after rituximab-monotherapy, he achieved a complete haematological remission which is continuing 6 months after the end of therapy. Our case demonstrates that treatment with the unconjugated anti-CD20-monoclonal antibody (rituximab) may be a new powerful tool for a better treatment of Waldenström's macroglo

Chromosomal breakage, endomitosis, endoreduplication, and hypersensitivity toward radiomimetric and alkylating agents: a possible new autosomal recessive mutation in a girl with craniosynostosis and microcephaly.

A high frequency of spontaneous chromosomal breakage, endomitosis, endoreduplication and hypersensitivity toward both the alkylating agent Trenimon and the radiomimetric drug bleomycin was observed in phytohemagglutinin-stimulated peripheral lymphocytes from a girl with craniosynostosis, microcephaly, ptosis, bird-like facies, and moderate mental retardation. We also observed abnormal chromosomal spiralization and some aspects of abnormal cellular division. Several fruitless attempts were made to establish a cell line. The parents were consanguineous, supporting the existence of a new, rare, a

[Molluscacide activity of a mixture of 6-n-alkyl salicylic acids (anacardic acid) and 2 of its complexes with copper (II) and lead (II)].

The molluscicide activity of hexanic extract from Anacardium occidentale L. (cashew) nut shell, of copper (II) complex, of lead (II) complex and anacardic acid has been compared in the laboratory in an attempt to obtain better stability than anacardic acid. This was obtained from the hexanic extract of the cashew nut shell by precipitation with lead (II) hydroxide or cupric sulfate plus sodium hydroxide or (II) cupric hydroxide followed by treatment of lead (II) complex with a diluted solution of sulfuric acid. Ten products of the mixture obtained were tested on adults snails of Biomphalaria g

Increased levels of platelet-activating factor (1-O-alkyl-2-acetylglycerophosphocholine) in blood after reversal of renal clip hypertension in the rat.

1. This study analyses whole blood in acutely unclipped one-kidney, one-clip (1K, 1C) hypertensive rats for the presence of platelet-activating factor (PAF), a potent vasodilator and a putative mediator of the rapid blood pressure (BP) fall seen after unclipping. 2. Hypertensive 1K, 1C rats were anaesthetized and a carotid and jugular cannula were inserted for BP measurement and anaesthetic infusion respectively. After a stable level of anaesthesia was attained, the constrictive clip was removed and BP was recorded for 30 min. 3. Blood was drawn from the aorta directly into ice-cold acetone. T

Van der Waals Interactions between Nonpolar Alkyl Chains and Polar ZnO Surfaces in Gas Sensing Dynamics of Aliphatic Carboxylic Acids.

Understanding the adsorption behavior of carboxylic acids on oxide surfaces is important in various chemical processes, including heterogeneous catalysis and chemical sensing. Traditionally, these behaviors have been attributed mainly to interactions between the polar carboxyl group and the polar oxide surface. Here, we demonstrate a vital yet previously underestimated effect of van der Waals interactions between the nonpolar alkyl chains and the polar ZnO surfaces on gas adsorption sensing of aliphatic carboxylic acids. Adsorption behaviors of carboxylic acids with different chain lengths (C3

From Fresh Leaves to Tea Infusion: Investigating the Origins, Key Variables, and Sensory Impact of 3-Alkyl-2-Methoxypyrazines in Green Tea.

This study investigated methoxypyrazines (MPs) in green tea infusions using solid-phase microextraction (SPME) and stable isotope dilution assay (SIDA) to extract and quantitate. Quantitative results showed that MPs (IPMP, SBMP, and IBMP) originated from fresh leaves and decreased gradually during processing. Stems of new shoots had the highest MPs levels, which declined from the first to fourth leaf, and shading increased MPs in older leaves. Quantification of relevant amino acids showed that MPs were strongly correlated with glycine, valine, and isoleucine. In 21 green tea infusions, IBMP ap

Disparity in stabilization effect of polyols on the cold- and heat-induced unfolding of apo-myoglobin: Impact of alkyl chain length and number of hydroxyl groups.

The fundamental challenge in protein-based therapeutics and biotechnological applications is maintaining protein stability and activity. This study investigates the effects of various diols (ethylene glycol, 1,2-butanediol, 1,4-butanediol, 1,2-hexanediol, 1,6-hexanediol) and triols (glycerol, 1,2,6-hexanetriol) on the cold and heat-induced denaturation of apo-myoglobin (apoMb). The polyols destabilize the protein during heat denaturation but stabilize it against cold denaturation, unlike conventional denaturants or stabilizers. These polyol-induced effects are enthalpically driven. Fluorescenc

Gold nanoparticle-decorated reduced graphene oxide as a highly effective catalyst for the selective α,β-dehydrogenation of N-alkyl-4-piperidones.

Gold nanoparticles (AuNPs) supported on reduced graphene oxide (AuNPs/rGO) were demonstrated to be a highly reactive catalyst for the selective α,β-oxidative dehydrogenation (ODH) of N-alkyl-4-piperidones, using N-methyl-, N-ethyl- and N-benzyl-4-piperidone. The substrate N-methyl-4-piperidone represents a pharmaceutically relevant system as its reaction product N-methyl-2,3-dihydropyridin-4(1H)-one is highly valuable (>1000 €·g-1) in contrast to the inexpensive starting material (0.15 €·g-1). Various synthesis methods were employed to prepare AuNPs supported on different carbon materials, inc

Lewis Acid-Catalyzed Switchable Functionalization of C,N-Cyclic Ketimines with Vinyl Azides: Access to Diverse C(2)-Alkylated Pseudoindoxyls.

Herein, a divergent coupling of indole-derived C,N-cyclic ketimines with vinyl azides is described for the synthesis of structurally diverse C(2)-alkylated pseudoindoxyls, especially α-indoxylacetamides, ketones, and acetonitrile derivatives. The Lewis acid-catalyzed divergent Schmidt and interrupted Schmidt-type pathways of the transient iminodiazonium ion intermediate enable vinyl azides to effectively function as surrogates for amides, ketones, and cyanomethanides. Furthermore, α-indoxylacetamides exhibited fluorescence properties, demonstrating an impressive fluorescence quantum yield.

A first-in-human phase 1a study of LP-184, a tumor-site activated novel alkylating agent, in patients with advanced solid tumors.

LP-184 is a novel acylfulvene pro-drug that alkylates DNA after bioactivation by the intracellular oxidoreductase prostaglandin reductase 1 (PTGR1). In preclinical studies, potent growth inhibition has been observed against a wide variety of solid tumor models, particularly those carrying DNA damage repair deficiency. Here, we report results of the first-in-human dose escalation trial of LP-184 in advanced solid tumors. A dose escalation Bayesian optimal interval design was used with a starting dose of 0.01 mg/kg. Eligible patients with advanced refractory solid tumors were enrolled to receive

Investigation of the Effect of Alkyl Chain Length on the Size and Distribution of Thiol-Stabilized Silver Nanoparticles for Proton Exchange Membrane Fuel Cell Applications.

This article reports on how the length of the alkyl chain influences the morphological properties of thiol-stabilized silver nanoparticles (Ag NPs) and their subsequent effects on the performance and durability of proton exchange membrane fuel cells (PEMFCs). We synthesized thiol-stabilized Ag NPs by varying the alkyl chain length: 1-hexane thiol (C6), 1-octanethiol (C8), 1-decanethiol (C10), 1-dodecanethiol (C12), and 1-tetradecanethiol (C14), which we achieved using the two-phase Brust-Schiffrin method. X-ray Diffraction (XRD) patterns confirm the formation of crystalline Ag NPs. A morpholog

Induction of ER stress-mediated apoptosis in breast cancer cell line by the powerful alkylating agent bendamustine and insights into its molecular mechanisms.

Bendamustine, an alkylating agent used in treating hematological cancers like non-Hodgkin's lymphoma (NHL) and chronic lymphocytic leukemia (CLL), has recently garnered attention for its potential in breast cancer therapy. This study explores its anticancer effects and molecular mechanisms in breast cancer cells. MDA-MB-231 cells were exposed to various concentrations of bendamustine (0-50 µM), and cytotoxicity was assessed using Alamar Blue and LDH assays, revealing a dose-dependent reduction in cell viability with an IC50 value of 16.98 μM at 24 h (p < 0.001). Alkyl Chain Engineering for High-Brightness Near-Infrared-II Aggregation-Induced Emission Nanoprobes toward High-Resolution In Vivo Bioimaging.

Fluorescence imaging in the second near-infrared window (NIR-II, 1000-1700 nm) has emerged as a powerful tool for in vivo bioimaging, offering deep-tissue penetration alongside high spatiotemporal resolution and contrast. Owing to structural advantages, aggregation-induced emission (AIE) luminogens are recognized as an ideal platform for constructing advanced NIR-II organic probes. However, their brightness remains suboptimal for practical applications due to intrinsic molecular constraints and environmental quenching effects. Addressing these issues, a high-brightness NIR-II AIE nanoprobe, TT

Low-Frequency Vibrational Density of States of Nanophase-Separated Poly(n‑alkyl methacrylate)s: Confined Phonons and Relationship to Specific Heat.

This study investigates the low-frequency vibrational density of states of nanophase-separated poly-(n-alkyl methacrylate)-s (PnMAs) and its relationship to specific heat. This system undergoes a nanophase separation for n > 1 in alkyl side chain-rich domains and a backbone-rich matrix. Using inelastic neutron scattering, the low-frequency vibrational density of states (Boson peak, BP) of PnMAs with varying alkyl side chain lengths (methyl, butyl, hexyl, and octyl) is measured. The results reveal that the BP shifts to higher frequencies with increasing side chain length reaching a maximum. Thi
