Inflammatory mediator stimulation of astrocytes and meningeal fibroblasts induces neuronal degeneration via the nitridergic pathway.

The role of inflammatory cytokines in the pathogenesis of neurological disorders is not entirely clear. The neurotoxic effects of cytokines, and perhaps indirectly bacterial endotoxins, could be mediated by the stimulation of immunocompetent cells in the brain to produce toxic concentrations of nitric oxide (NO) and reactive nitrogen oxides. NO is a short-lived, diffusible molecule that has a variety of biological activities including vasorelaxation, neurotransmission, and cytotoxicity. Both constitutive and inducible NO synthase has been described in astrocytes in vitro. Here we demonstrate t…

Interleukin-6 production by brain tissue and cultured astrocytes infected with Theiler's murine encephalomyelitis virus.

Theiler's murine encephalomyelitis virus (TMEV) is known to interact with cells of the central nervous system (CNS). Here we report that, interestingly, it is a potent inductor of interleukin-6 (IL-6) in the CNS of infected animals and in pure cultures of astrocytes. Maximal IL-6 gene transcription in glial cells, as detected by bioassay and ELISA, was observed at 6 and 24 h after infection. Astrocytes from both SJL/J and Balb/c (strains of mice susceptible and resistant, respectively, to TMEV-induced demyelination) produced similar amounts of IL-6, measured in tissue culture supernatants. The…

Degeneration of astrocytes in feline retinopathy of prematurity causes failure of the blood-retinal barrier.

This study addresses the role of astrocytes in the genesis of retinopathy of prematurity, examined in the feline model of this condition. Evidence is presented that the hypoxia of retinopathy of prematurity, in addition to inducing vasoproliferation, damages the retina directly. Retinal neurons survive the hypoxia, but the astrocytes, which are involved in the formation of the glia limitans of the retinal vessels, degenerate. Astrocytes subsequently recolonize the retina after a delay that matches the period of leakiness of the proliferative vasculature (described in the companion article). Gi…

Increased susceptibility of human fetal astrocytes to human T-lymphotropic virus type I in culture.

Human T-lymphotropic virus type I (HTLV-I) has been considered as an agent responsible for tropical spastic paraparesis and HTLV-I associated myelopathy. However, the pathogenesis of the diseases remains unclear. In a previous study we demonstrated that HTLV-I could infect adult human astrocytes and oligodendrocytes in vitro, although the rates of infected cells were low, at a rate of 0.1% and 0.01-0.05% respectively. Since mother-to-child transmission has been proposed as one of the major pathways for the prevalence of HTLV-I endemic, in the present study we investigated the susceptibility of…

Ultracytochemical demonstration of alkaline phosphatase activity in astrocytes and subependymal cells in the rat brain.

Ultrastructural localization of alkaline phosphatase activity was studied in astrocytes of the cerebral cortex and corpus callosum, and in the ependymal and subependymal cells of the lateral ventricle of normal rat brain by means of electron microscopic histochemistry employing the lead citrate method. Astrocytes exhibited alkaline phosphatase activity on the plasma membrane of the endfeet facing the vascular basement membrane and at the gap junctions, but not on other parts of the plasma membrane or in the cytoplasm. Immature astrocytes containing small bundles of filaments, a few glycogen gr…

Glial-Dopamine crosstalk: Astrocytic and microglial gatekeepers of neuroinflammation, plasticity, and motivation.

Dpamine (DA) signaling has long been framed through a neuron-centric lens; yet, mounting evidence reveals that glial cells (astrocytes and microglia) serve as indispensable gatekeepers of dopaminergic tone, synaptic plasticity, and neuroimmune balance. Single-cell, spatial, and optical imaging studies have redefined DA circuits as multicellular ecosystems in which glial receptors, transporters, and gliotransmitters dynamically sculpt neuromodulation and behavior. Astrocytes fine-tune DA clearance, glutamate buffering, and metabolic coupling, while microglia integrate immune and stress cues rec…

Neurotransmitter Receptor Interactions Control Astrocyte Ca²⁺ Signal in the Basolateral Amygdala.

INTRODUCTION: The endocannabinoid (eCB) system plays fundamental roles in both physiological and pathological aspects of brain function. Activation of type 1 cannabinoid (CB1) receptors in astrocytes by eCBs elevates their intracellular calcium through a signaling pathway that involves activation of Gq/11 proteins, phospholipase C, and IP3R2 to mobilize calcium. Hippocampal astrocytes have been proposed to integrate synaptic information through nonlinear interactions between glutamate and acetylcholine in eliciting calcium responses. However, whether similar integrative interactions occur amon…

Astrocytic GABAergic signaling in perioperative neurocognitive disorders: mechanisms and intervention strategies.

Perioperative neurocognitive disorders (PND) are a major burden for older surgical patients, yet little is known about the cellular events that set them in motion, and no therapy currently changes their course. Anesthesia and amnesia after etomidate, sevoflurane, or isoflurane depend mainly on gamma-aminobutyric acid type A receptors (GABA(A)Rs), and an increasing body of work shows that astrocytes also respond to these drugs. This review examines an astrocyte-centered GABAergic cascade that may link anesthetic exposure to PND and grades the strength of the evidence at each node. When anesthet…

Astrocytic NLRP3 cKO Mitigates Cognitive Impairment Induced by Paclitaxel in Mice.

Chemotherapy-induced cognitive impairment (CICI) is a prevalent and debilitating sequela in cancer patients receiving paclitaxel. Our previous study revealed that glial cells, particularly astrocytes, regulate NLRP3-mediated neuroinflammation, which is crucial to the pathophysiology of cognitive deficits. However, the therapeutic potential of targeting astrocytic NLRP3 for CICI remains largely unexplored. This study sought to determine whether astrocyte-specific NLRP3 knockout (KO) could mitigate paclitaxel-induced cognitive impairment. A mouse model of CICI was established via intraperitoneal…

ACSA2 is not astrocyte-specific: implications for cell sorting strategies in the rodent brain.

INTRODUCTION: Astrocyte-specific cell surface antigen-2 (ACSA2) has been established as the gold-standard marker for isolating astrocytes via magnetic-activated cell sorting (MACS) or fluorescence-activated cell sorting (FACS) for downstream transcriptomic studies. In a prior study of the astrocyte response to cortical stroke, we used ACSA2-based cell sorting prior to single cell RNA sequencing (scRNAseq). We found a substantial population of ACSA2+ cells exhibiting robust microglial gene expression signatures, suggesting contamination of purified astrocyte preparations. METHODS: An intraveno…

Astrocyte autophagy-pyroptosis crosstalk in Alzheimer's: insights from knowledge graphs.

Astrocytes play a crucial role in brain health and are implicated in various aspects of Alzheimer’s disease (AD) progression. However, the mechanism underlying astrocyte autophagy and pyroptosis in AD remains unclear. To advance this knowledge, we constructed a knowledge graph of AD and dementia (AdDKG) by integrating literature-derived data with the Systematized Nomenclature of Medicine – Clinical Terms (SNOMED CT). The core knowledge base, addkg, was created through named entity recognition and semantic annotation of 172 283 articles using a concept identifier (CI-er) tool. AdDKG knowledge g…

Light-based modulation of astrocytic calcium for regulation of organelle dynamics and morphogenesis.

Astrocytes are essential for neuronal homeostasis, and synaptic modulation and development. While astrocytes exhibit distinct Ca2+ signaling patterns, decoding their physiological roles remains challenging because conventional approaches generate nonphysiological, spatially imprecise Ca2+ surges that obscure endogenous signals. Here, we developed a tunable light-based tool to modulate astrocytic Ca2+ activity, mimicking localized spikes and global waves elicited by graded glutamate through endogenous mGluR-Gq-IP3R signaling. IP3R triple knockout abolished light-evoked Ca2+ elevations, indicati…

Modeling Astrocyte-Driven Repair of Visuomotor Deficits in Alzheimer's Thalamic Circuitry.

Alzheimer's Disease (AD) frequently manifests in visuomotor impairments, disrupting cholinergic signaling from the brainstem to the Thalamic Reticular Nucleus (TRN). The contribution of this work lies in the development of a biologically informed computational framework to model astrocyte mediated restoration of synaptic transmission, measured by a 'release probability (PR)' in TRN under cholinergic depletion. The model simulates three physiological states-healthy, damaged, and recovered by incorporating dual astrocytic mechanisms i.e., depolarization induced suppression excitation (DSE) and e

Bayesian modelling of amyloid-beta dynamics and astrocyte influence in Alzheimer's disease.

BACKGROUND: Alzheimer's disease (AD) is a complicated neurological condition defined by the deposition of amyloid-beta (Aβ) plaques. Despite extensive research, the dynamics of Aβ growth, particularly the role of astrocytes, remain poorly understood, limiting the development of effective treatments. NEW METHOD: This study addresses this gap by introducing a Bayesian inference framework for modelling Aβ dynamics, incorporating both strong and weak astrocyte effects utilizing Alzheimer's Disease Neuroimaging Initiative (ADNI) clinical data. RESULTS: Through a combination of stochastic growth m…

Novel Roles for the Ectoenzyme CD38 in the Maintenance of Transcriptional and Metabolic Homeostasis in Astrocytes.

CD38 is an ectoenzyme that converts NAD+ to NAM to help maintain bioenergetic homeostasis. CD38 dysregulation and gene variation is reported in neurodegenerative conditions such as Parkinson's disease (PD) and Alzheimer's disease (AD), highlighting the need to better understand CD38 biology within the brain. Here, we demonstrate enrichment of Cd38 in midbrain astrocytes and describe how CD38 deficiency influences brain metabolism, astrocytic gene expression, and bioenergetics. We demonstrate increased NAD content, decreased NAM content, and increased NAD/NAM in the midbrain and striatum of CD3…

Astrocytic Gq pathway activation enhances the efficacy of rehabilitation after stroke.

Stroke is a leading cause of functional impairment. As rehabilitation has a limited effect, it is essential to advance future treatments and enhance the efficacy of rehabilitation. This study focused on the use of astrocytes to augment the efficacy of rehabilitation for strokes. It utilized chemogenetics to specifically activate astrocytes in the peri-infarct region of a mouse photothrombosis model. The mice were subjected to immunohistochemical analysis and behavioral tests to evaluate the effects of astrocytic Gq pathway activation after an induced stroke. The timing of astrocytic activation…

Astrocytic connexins in alcohol use disorder: mechanisms of neuroinflammation and therapeutic potential.

Astrocytes are critical regulators of brain homeostasis and are known to be disrupted during alcohol use disorder (AUD). Here, we describe what is known about astrocytic dysfunction in AUD and the emerging literature supporting a mechanistic link to connexins in the pathophysiology of this disorder. We integrate what is known in the current literature and highlight areas where a lack of evidence leads to inconclusive interpretations. Overall, this is a comprehensive review of astrocytes and connexins in AUD, providing ideas and insights to basic and clinical audiences interested in pursuing fu…

Retinal Astrocytic Hamartoma in a Child With Neurofibromatosis Type 1 Treated With Selumetinib.

Retinal astrocytic hamartoma (RAH) is a rare benign retinal tumor most commonly associated with tuberous sclerosis complex and is typically stable. RAH may also rarely occur in neurofibromatosis type 1 (NF1), an autosomal dominant disorder characterized by skin pigmentation abnormalities and multiple neurofibromas. We present a case of a child with NF1 and symptomatic, inoperable plexiform neurofibromas, for which selumetinib was initiated. Incidentally, the patient also had a progressive RAH involving the optic disc, leading to severe exudative retinal detachment. Following selumetinib therap

Astrocyte-oligodendrocyte crosstalk dependent myelination via secreted protein YKL40.

Although oligodendrocyte differentiation and myelin formation are inherent properties of oligodendrocyte progenitor cells (OPCs) that are guided by intrinsic transcriptional and epigenetic programs, this process in the brain is finely regulated by signals coming from other cells, including astrocytes. Here, we identified the astrocyte secreted protein YKL40 at the center of astrocyte-OPC cross-communication and myelination in the developing brain. We find that YKL40 is expressed by astrocytes within white matter areas in the developing brain, coinciding with the OPC differentiation and myelina…
