The "skin window" test with phytohemagglutinin and pyrogenal reveals the multiple character of disturbances in the cooperative interaction of dermal T-lymphocytes and macrophages in patients with clinical forms of erysipelas, which indicates the necessity of differentiated immunological correction.

BACKGROUND: Anaplastic thyroid cancer (ATC) is characterized by high invasiveness and rapid progression and has a poor prognosis. The aim of this study was to investigate the role of ketone body metabolism in ATC and provide a novel approach for ATC treatment.
METHODS: Human ATC cell lines, including 8505C and CAL-62, were used as the research objects. Cell Counting Kit-8 and colony formation assays appraised cell proliferation. Flow cytometry was performed to evaluate the cell cycle and apoptosis. Wound healing and transwell assays verified the migration and invasion of cells. Furthermore, t

Progressive multifocal leukoencephalopathy (PML) is a rare, opportunistic infection of the central nervous system, caused by reactivation of the ubiquitous JC virus. PML is a devastating disease that is frequently fatal, and although survival rates have improved, patients who survive PML often experience considerable neurological deficits. PML was associated with a variety of immunosuppressive therapies in the past decade, but attribution of causality is difficult owing to the presence of confounding factors and to an inadequate understanding of the underlying pathogenesis of this disease. Thi

Asthmatic triad (AT) is a clinical syndrome incorporating bronchial asthma (BA), recurrent nasal and sinus polyps, intolerance of aspirin, derivatives of pirasolone and other nonsteroid antiinflammatory drugs. Specific diagnosis of aspirin and analgin intolerance in patients with AT was made using the test of natural leukocyte emigration inhibition in vivo in the oral cavity. The test was performed in 22 patients with AT and 13 patients with BA with pollenosis without aspirin and analgin intolerance. In all the patients the test provided positive results, in control patients these were negativ

Acute rheumatic fever (ARF) has the characteristics of an autoimmune disease, triggered by cross-reactive antigens shared by the group A streptococcus and a variety of tissues including the heart, endothelium, and basal ganglia. Using two parameters of cellular reactivity, migration inhibition and blastogenic transformation, ARF patients from Trinidad show significant lymphocyte reactivity to streptococcal antigens, particularly those from an ARF associated streptococcal strain. This reactivity, studied over a 2-year period, peaked at 1 to 6 months after the acute onset and remained significan

Production of leucocyte migration inhibition factor by peripheral blood leucocytes in response to challenge with gluten fractions has been proposed as a reliable in vitro test for the diagnosis of coeliac disease. We have performed the leucocyte migration inhibition test with two different gluten fractions, GFIII and B2, in untreated and treated coeliac patients, patients with other intestinal diseases (abnormal controls) and healthy controls, and evaluated the sensitivity, specificity and positive and negative predictability of the test for the diagnosis of coeliac disease. Using GFIII as ant

Cellular immune reactions against normal biliary tract antigens have been investigated in 21 patients with primary biliary cirrhosis (PBC) and 18 with chronic active hepatitis (CAH) using the leucocyte migration inhibition test with partially purified antigens from normal human gall-bladder bile. Four antigen fractions, containing (either separately or together) three previously-described biliary antigens, were employed: (1) Antigen I; (2) Canalicular antigen; (3) Canalicular and Ductular antigens; (4) All three antigens. Seventeen (81%) of the PBC patients showed migration inhibition with all

The development of cellular immunity was investigated in mice infected experimentally with tick-borne encephalitis virus strains (TBE) isolated from patients. In the presence of strain variants of the response, two main trends in the dynamics of cellular immunity induced by different virus strains were established: a rapid increase in the level of sensitization of lymphoid cells in the first days after virus inoculation followed in the stage of paralysis by the desensitization phenomenon (the dynamics of the waning type), and a gradual increase in the level of cellular response in the course o

A Listeria monocytogenes infection in guinea pigs was used to study the interrelationship between antigen-induced macrophage migration inhibition, delayed-type hypersensitivity, and acquired cellular resistance. Early after infection (at 2 and 7 days), very significant enhancement of macrophage migration was observed. Migration inhibition was detected beginning on day 14 and was uniformly observed only on day 21 of the infection, after which a shift again to enhancement was seen. The early detection (by day 2) of migration enhancement suggested that this assay may be more sensitive than assess

The existence of CMI to tumor-associated antigens present in 3 M KCl extracts of breast carcinomas was demonstrated in a group of breast cancer patients by the leukocyte migration inhibition (LMI) assay. When crude KCl extracts were tested, 3 of 5 breast cancer patients gave a positive response to autologous tumor extracts. Eleven of 20 gave a positive response to allogeneic extracts as compared to 3 of 22 controls (including 6 patients with benign breast disease, 7 with non-mammary cancers and 9 normal controls). Extracts of fibrocystic disease tissue gave positive LMI tests in 2 of 5 breast

Previous studies using synthetic immunogenic molecules containing two haptenic peptides (ther NH2-terminal heptapeptide and the COOH-terminal pentapeptide of oxidized ferredoxin (O-Fd) from C. pasteurianum) have shown that both peptides individually are capable of initiating lymphocyte transformation and inhibiting migration in populations of lymphocytes from O-Fd-sensitized guinea pigs. While migration inhibition could readily be stimulated by single haptenic peptides, lymphocyte treasformation appeared to be more easily induced by molecules containing two or more haptenic peptides (these cou

Guinea-pigs made tolerant to dinitrochlorobenzene (by prior intracardiac injection of dinitrobenzene sulphonate) failed to give positive skin reactions after contact sensitization, but nevertheless had peritoneal cells which reacted with the hapten in macrophage migration inhibition (MMI) tests. This reactivity was blocked in vitro by the addition of serum from tolerant animals but not by serum from hypersensitive animals. Cells from hypersensitive guinea-pigs were anomalous, in that their reaction with hapten in MMI was not blocked by tolerant serum. Hypersensitive serum, though not active by

Cryptotanshinone, a natural compound isolated from the roots of Salvia miltiorrhiza Bge (Danshen), has been found to induce cancer cells apoptosis and impair cell migration and invasion in various malignancies, but its antiproliferation and chemosensitization effects of Cryptotanshinone on tongue squamous cell carcinoma(TSCC)still remain fully elucidated. In this study, the effects of Cryptotanshinone on the proliferation, apoptosis and cell cycle of human TSCC cells, including CAL 27 and SCC 9 cells, were measured. The results demonstrated that Cryptotanshinone dose-dependently inhibited cell

The human placenta is an invasive structure in which highly proliferative, migratory, and invasive extravillous trophoblast (EVT) cells migrate and invade the uterus and its vasculature. Using in vitro propagated normal first-trimester EVT cells and immortalized EVT cells, which share all of the phenotypic and functional characteristics of the normal EVT cells, it has been shown that migration/invasion of human EVT cells is stringently regulated by many growth factors, their binding proteins, extracellular matrix (ECM) components, and some adhesion molecules in an autocrine/paracrine manner at
