Oral Salvia officinalis phenolics abolish mortality and mitigate inflammation in acute TNBS murine colitis.

BACKGROUND: This study is the first to evaluate a phenolic extract of Salvia officinalis in the TNBS murine model of colitis. Inflammatory bowel disease (IBD) involves transmural oxidative and nitrosative stress that up-regulates inducible nitric-oxide synthase (iNOS) and cyclo‑oxygenase-2 (COX-2). Although phenolic constituents of S. officinalis are anti-inflammatory in vitro, their efficacy has never been tested in the trinitrobenzene-sulphonic acid (TNBS) model of colitis. METHODS: Aqueous-ethanolic sage leaves were extracted and standardized [(total phenolics = 6208 ± 200 mg gallic acid e

Rosmarinic acid mitigates intestinal inflammation and oxidative stress in bullfrogs (Lithobates catesbeiana) fed high soybean meal diets.

High proportions of soybean meal in aquafeed have been confirmed to induce various intestinal pathologies. This study aims to investigate the regulatory effects of rosmarinic acid (RA), an antioxidant with anti-inflammatory and antimicrobial properties, when added to high soybean meal feeds in different doses, (0, 0.5, 1, and 4 g/kg). During the 56-day feeding trial, results indicated that, compared to the control group without RA (0 g/kg), the 1 g/kg and 4 g/kg RA groups increased bullfrog survival rates and total weight gain while reducing feed coefficient. Additionally, these doses markedly

Rosmarinic acid protects mice from imiquimod induced psoriasis-like skin lesions by inhibiting the IL-23/Th17 axis via regulating Jak2/Stat3 signaling pathway.

IL-23/Th17 (IL-17) axis plays a critical role in psoriasis. Rosmarinic acid (RA) was proved the inhibitory effect of T cell infiltration in the skin. However, whether and how RA has beneficial effects on psoriasis did not really know yet. So lipopolysaccharide (LPS)-induced abnormal proliferation Hacat cell line and Imiquimod (IMQ)-induced psoriasis-like mouse dermatitis were used to assess the pharmacological effects and mechanisms of RA by Psoriasis Area Severity Index (PASI) score, histopathology, flow cytometry, reverse transcription-polymerase chain reaction (RT-PCR) and western blotting.

The TRPV1 channel regulates glucose metabolism.

Endocannabinoids (ECs) mediate effects via cannabinoid receptor types 1 and 2 (CB1 and 2) and transient receptor potential channel-vanilloid subfamily member 1 (TRPV1) channels. In high-fat diet (HFD)-induced obese mice overactivity of the EC system and inhibition of CB1 increase skeletal muscle glucose uptake. We explored the role of TRPV1. Male TRPV1+/+(WT) and TRPV1-/-(KO)-mice were fed (20 wk) a standard laboratory diet (SLD) or HFD. An intraperitoneal glucose tolerance test was performed. RT-PCR was performed to measure mRNA of genes involved in glucose/lipid metabolism and the EC system

Homocysteine-induced caspase-3 activation by endoplasmic reticulum stress in endothelial progenitor cells from patients with coronary heart disease and healthy donors.

Previous studies have suggested an association of hyperhomocysteinemia-induced vascular pathology with enhanced apoptotic potential of endothelial progenitor cells in patients with coronary heart disease. Our results indicate that 500 µmol/L homocysteine induced endothelial progenitor cell apoptosis and activation of caspase-3, both of which were abolished by 100 µmol/L and 200 µmol/L salubrinal, an agent that prevents endoplasmic reticulum stress-induced apoptosis. The addition of 500 µmol/L homocysteine caused a release of Ca(2+) from intracellular stores, and enhanced phosphor-eukaryotic in

New Cinnamamide and Derivatives as New Larvicides against Aedes aegypti Vector Larvae: Facile Synthesis, In Silico Study, In Vitro Noncytotoxicity, and Nontoxicity against Zebrafish.

Every year, millions of people are infected by arboviruses, such as Dengue, a neglected disease that mainly affects regions with lower socioeconomic development. One of the main strategies to combat these diseases is population control of mosquito vectors, such as Aedes aegypti. Chemical control is an effective approach to reducing the population of these vectors; however, challenges such as toxicity and increasing resistance in populations make it necessary to search for new bioactive and selective substances continuously. In this sense, this study presents the synthesis and evaluation of the

Rosmarinic Acid Ameliorates Type 2 Diabetic Osteoporosis by Reducing NLRP3 Expression and Alleviating Osteoblast Pyroptosis via the FOXO1/TXNIP Signaling Pathway.

Unlike postmenopausal osteoporosis, Type 2 diabetic osteoporosis (T2DOP) occurs in a chronic high-glucose, inflammatory microenvironment, increasing fracture risk. Emerging evidence links osteoblast pyroptosis to T2DOP pathogenesis. While rosmarinic acid (RA) exhibits antidiabetic and anti-inflammatory properties, its role in T2DOP remains unclear. We established a T2DOP mouse model using a high-fat diet and low-dose STZ, confirmed by micro-CT. RA's effects on osteoblast function and mitochondrial homeostasis were evaluated, with network pharmacology and molecular docking identifying potential

Origin of Ca2+ necessary for carbachol-induced contraction in longitudinal muscle of the proximal colon of rats.

The origin of Ca2+ necessary for carbachol (CCh)-induced contraction of longitudinal muscle of the proximal colon of rats was studied. CCh induced contraction of the muscle consisting of two phases, phasic and tonic phases, with a concomitant biphasic increase in [Ca2+]i. After removal of Ca2+ from the bathing solution of the colonic segments, CCh-induced contraction was rapidly inhibited; there was almost complete inhibition 1 min after the removal. Nicardipine, a blocker of voltage-dependent calcium channel, also significantly inhibited CCh-induced contraction. On the other hand, treatment o
