Clusters of Drug-Resistant Mycobacterium tuberculosis Detected by Whole-Genome Sequence Analysis of Nationwide Sample, Thailand, 2014-2017.

Multidrug-resistant tuberculosis (MDR TB), pre-extensively drug-resistant tuberculosis (pre-XDR TB), and extensively drug-resistant tuberculosis (XDR TB) complicate disease control. We analyzed whole-genome sequence data for 579 phenotypically drug-resistant M. tuberculosis isolates (28% of available MDR/pre-XDR and all culturable XDR TB isolates collected in Thailand during 2014-2017). Most isolates were from lineage 2 (n = 482; 83.2%). Cluster analysis revealed that 281/579 isolates (48.5%) formed 89 clusters, including 205 MDR TB, 46 pre-XDR TB, 19 XDR TB, and 11 poly-drug-resistant TB isol…

Analysis of brain region-specific co-expression networks reveals clustering of established and novel genes associated with Alzheimer disease.

BACKGROUND: Identifying and understanding the functional role of genetic risk factors for Alzheimer disease (AD) has been complicated by the variability of genetic influences across brain regions and confounding with age-related neurodegeneration. METHODS: A gene co-expression network was constructed using data obtained from the Allen Brain Atlas for multiple brain regions (cerebral cortex, cerebellum, and brain stem) in six individuals. Gene network analyses were seeded with 52 reproducible (i.e., established) AD (RAD) genes. Genome-wide association study summary data were integrated with th

Cluster analysis to estimate the risk of preeclampsia in the high-risk Prediction and Prevention of Preeclampsia and Intrauterine Growth Restriction (PREDO) study.

OBJECTIVES: Preeclampsia is divided into early-onset (delivery before 34 weeks of gestation) and late-onset (delivery at or after 34 weeks) subtypes, which may rise from different etiopathogenic backgrounds. Early-onset disease is associated with placental dysfunction. Late-onset disease develops predominantly due to metabolic disturbances, obesity, diabetes, lipid dysfunction, and inflammation, which affect endothelial function. Our aim was to use cluster analysis to investigate clinical factors predicting the onset and severity of preeclampsia in a cohort of women with known clinical risk fa…

A Delphi exercise and cluster analysis to aid in the development of potential classification criteria for systemic sclerosis using SSc experts and databases.

OBJECTIVES: Since the 1980 ACR classification criteria for systemic sclerosis (SSc) do not identify 20% with SSc, revised criteria are necessary. METHODS: Suggested new criteria from the literature were sent in random order to 96 SSc experts. A 3-round Delphi Consensus eliminated criteria. Then cluster analysis reduced items. The Canadian Scleroderma Research Group (CSRG) database was used to determine the prevalence of each item. RESULTS: Seventy-one of 96 (71%) completed all 3 rounds; 47 items were expanded to 76 in round 2. Thirty items had at least 50% consensus and 18 had >75% agreement…

Clustering-Based Accelerometer Measures of Physical Activity Patterns in Children With Overweight or Obesity: Baseline Cross-Sectional Methodological Analysis.

BACKGROUND: Accelerometers produce high-resolution physical activity data, but commonly used summary measures often reduce these data to total volume, intensity, or variability and may not retain interpretable temporal structure across the day. Cluster-based summaries may provide a way to characterize daily physical activity profiles while preserving information about when activity occurs. OBJECTIVE: This study evaluated whether cluster-derived accelerometer summary measures could represent daily physical activity patterns and explain variation in pediatric cardiometabolic outcomes comparably

Radiomics-Based AI for the Diagnosis and Prognosis of Vessels Encapsulating Tumor Clusters in Hepatocellular Carcinoma: Systematic Review and Meta-Analysis.

BACKGROUND: Vessels encapsulating tumor clusters (VETCs), a CD34-positive vascular pattern in hepatocellular carcinoma (HCC), are linked to aggressive biology, early recurrence, and poor survival. Because pathologic VETC assessment requires postoperative immunohistochemistry and may be affected by sampling, preoperative noninvasive prediction remains clinically important. Radiomics-based artificial intelligence (AI) applied to routine contrast-enhanced imaging may provide a surrogate marker, but evidence across has not been comprehensively appraised. OBJECTIVE: This study aimed to evaluate th

Fluorescence Imaging Analysis of PTK7 Clustering in Situ via a Programmable DNA Network for Apoptosis Induction and Drug Resistance Reversal.

Inducing cell surface receptor clustering to mediate downstream signaling and reverse drug resistance has become a core cancer therapeutic strategy. However, controllable in situ formation of receptor superclusters remains a key bottleneck. To address these issues, a programmable DNA self-assembly strategy was established by integrating three units of rolling circle amplification (RCA) vector unit, two long DNA strands loaded with aptamer recognition units and the hairpin assembly unit. Fluorescence imaging analysis demonstrated that targeting binding to protein tyrosine kinase 7 (PTK7) can tr

Integrated transcriptomic and proteomic analysis reveals isolation and culture associated molecular changes in neonatal porcine pancreatic cell clusters.

BACKGROUND: While porcine Neonatal Pancreatic Cell Clusters (NPCCs) were potential alternative source of islets, their immaturity and heterogeneity make them inefficiently ameliorate hyperglycemia in rodents. Our previous work showed that NPCCs regress to a transitional-like state immediately after isolation and undergo dynamic transcriptional changes during short-term in vitro culture. The present study aimed to further delineate the molecular cues underlying these changes during porcine pancreatic tissue isolation and cultivation. METHOD: Transcriptomic and proteomic analysis were performed

Clustering analysis of volatile organic compound biomarkers with tobacco exposure and the association with cardiovascular health outcomes in an observation study cohort.

INTRODUCTION: Volatile organic compounds (VOCs) are toxic compounds found in tobacco smoke. Despite research on cigarette generated single VOCs, scant evidence exists on the mixtures of VOCs associated with different tobacco products. We aimed to explore whether distinct VOC exposure profiles exist among users of combustible cigarettes, e-cigarettes, and non-users, and to assess their associations with cardiovascular (CV) health markers. METHODS: Participants who self-reported use of e-cigarettes, cigarettes, or no tobacco (n=348; mean age 26 ± 7 years) enrolled in The Cardiovascular Injury d

Gender-biased clustering of attitudes towards physical intimate partner violence: A social network analysis in south-central Ethiopia.

Changing social norms, shared beliefs about what is acceptable, is a key focus of global health campaigns aimed at ending intimate partner violence against women (IPVAW). In Ethiopia, it is estimated that one in four women have been assaulted by a male partner, and over half the population hold attitudes supportive of this form of violence. To date, efforts to change people's attitudes towards IPVAW have been hindered by uncertainty over how social norms are acquired or learned. Here, we consider whether people's acceptance of IPVAW is maintained through social influence or "contagion," using
