Langerhans cell sarcoma (LCS), malignant indeterminate dendritic cell tumor (IDCT), and interdigitating dendritic cell sarcoma (IDCS) are rare malignant neoplasms that originate from antigen-presenting conventional dendritic cells (cDCs). These tumors, collectively termed cDC sarcoma in this study, have been reported only rarely in association with myeloid malignancies. We collected five cases of cDC sarcoma coexisting with acute myeloid leukemia (AML) or myelodysplastic neoplasm with increased blasts (MDS-IB), conducted clinicopathologic and genetic analyses, and compared these with four repo

Dendritic cells (DCs) are major contributors to generating an effective immune response due to their ability to present antigens to T cells. Recently, nanoparticles have been widely used in different medical applications, such as drug-delivery systems, to enhance the function of impaired immune cells. Objectives: This research aims to develop an effective antitumor DC-based vaccine by adsorption of chitosan-nanoparticles (CH-NPs) onto DCs. Methods: Undifferentiated mouse bone marrow progenitor cells were differentiated into mature DCs using cytokines and lipopolysaccharides. CH-NPs were prepar

Dendritic cells (DCs) are essential for anti-tumor immunity through recognition of tumor-derived signals and antigens, yet their function is frequently compromised within the tumor microenvironment (TME), enabling immune evasion. Consequently, enhancing DC functionality represents a critical strategy for advancing tumor immunotherapy. The novel engineered biomimetic DC-derived vesicle vaccine, g-diDCVac, integrates the STING agonist diABZI and tumor antigen gp100. Designed for facile modification and enhanced stability, g-diDCVac effectively targets lymph nodes, promotes DC maturation, and eli

Hemorrhagic shock induces immune dysfunction via mesenteric lymph return, leading to systemic inflammatory response and multiple organ dysfunction. Dendritic cells (DCs) play a pivotal role in immune response following hemorrhagic shock. Therefore, identifying regulatory targets within DCs is essential for understanding hemorrhagic shock-induced immune dysfunction. Endoplasmic reticulum (ER) autophagy (ER-phagy), a selective form of autophagy, is critical for DC function. Here, we investigated the role of tumor necrosis factor-α-induced protein-8-like 2 (TIPE2), a protein known to regulate aut

Background: Dendritic cells (DCs) are heterogeneous antigen-presenting cells that bridge innate and adaptive immunity. Recent classifications of hematolymphoid neoplasms highlight the complex origins of DC-related neoplasms. DCs have also been associated with the progression of multiple myeloma (MM). This report presents the case of a patient with MM in whom bone marrow analysis revealed an unusual additional clonal population of immature cells, in addition to plasmacytoid DCs, that later evolved into plasmacytoid dendritic cell proliferation associated with acute myeloid leukemia (pDC-AML). M

Chronic myelomonocytic leukemia (CMML) is associated with high rates of transformation to acute myeloid leukemia (AML) and poor overall survival. Plasmacytoid dendritic cell (pDC) proliferations in bone marrow biopsies have been proposed to be prognostically significant in CMML. In this multi-institutional study, we quantified pDC clusters in bone marrow biopsies of patients with CMML by interferon regulatory factor 8 (IRF8) immunohistochemical staining performed in separate laboratories and demonstrated the association of pDC clusters with overall survival. Two hundred five newly diagnosed CM

Mature plasmacytoid dendritic cell proliferation is a condition associated with myeloid neoplasms, most commonly chronic myelomonocytic leukemia. Plasmacytoid dendritic cells can resemble lymphocytes and histiocytes morphologically and immunophenotypically. Mature plasmacytoid dendritic cell proliferation may therefore go unrecognized if the diagnosis is not suspected and appropriate stains for plasmacytoid dendritic cells are not performed. Herein, we present a case of mature plasmacytoid dendritic cell proliferation masquerading clinically and histologically as histiocytoid Sweet syndrome. T

Inflammation plays a crucial role in the pathogenesis of various diseases, necessitating the development of effective anti-inflammatory therapeutics. Dendritic cells (DCs), as professional antigen-presenting cells, are key regulators of immune responses. In this study, we investigated the immunomodulatory effects of LSI312A, a novel compound derived from medicinal plant analogues, on DC function and inflammatory signaling pathways. LSI312A exhibited no cytotoxicity in DC2.4 cells at concentrations up to 20 μM. LSI312A significantly reduced antigen uptake and impaired the expression of co-stimu

We report the design and synthesis of a biodegradable hyperbranched polyether polyketal polyol, containing acid-labile ketal groups with controlled degradation, high water solubility, excellent hemocompatibility, and cell compatibility. For this, we have developed a new AB2-type acid-labile monomer, 2-((4-(2-(oxiran-2-ylmethoxy) ethoxy)tetrahydro-2H-pyran-4-yl)oxy)ethan-1-ol (OTPE). The OTPE monomer is copolymerized with glycidol through anionic ring-opening multibranching polymerization generating OTPE-incorporated hyperbranched polyglycerol (A-BioHPG) with varying degradable monomer content.

Systemic lupus erythematosus (SLE) is a chronic autoimmune disease characterized by immune dysregulation, leading to tissue inflammation and damage. Tolerogenic dendritic cells (DCs) and regulatory T cells (Tregs) play critical roles in restoring immune balance, and their dysfunction is implicated in SLE pathogenesis. This study investigates the immunomodulatory effects of Lactobacillus rhamnosus and Lactobacillus delbrueckii on DC function and T cell subset differentiation in SLE patients. Peripheral blood mononuclear cells (PBMCs) were isolated from SLE patients and healthy controls. Mixed l

Follicular dendritic cell sarcoma, a rare malignant tumor arising from specialized antigen-presenting cells, typically occurs in lymph nodes but can also develop at extranodal sites, including the liver. We present a case of a woman in her late 20s with systemic lupus erythematosus who presented with fever as the chief complaint. Imaging revealed a hepatic mass, and pathology confirmed the diagnosis of hepatic follicular dendritic cell sarcoma. The patient showed no recurrence 1 year post-radical surgery. We analyzed a total of 43 cases from 32 previous reports and the present case. Among thes

BACKGROUND: Fibrosis remains a major complication in several chronic diseases, including systemic sclerosis (SSc). Plasmacytoid dendritic cells (pDCs) are innate immune cells that play a key role in the development of fibrosis in SSc patients, through still poorly defined mechanisms. Interestingly, endoplasmic reticulum (ER) stress signaling pathways are dysregulated in pDCs from patients with SSc, but their contribution to fibrosis remains unclear. Thus, this study aimed to unravel the mechanisms behind the involvement of pDCs and ER stress in fibrosis.
METHODS: To address this question, we

A healthy 17-year-old boy was diagnosed as having blastic plasmacytoid dendritic cell neoplasm (BPDCN), a rare hematologic malignancy. Vision changes prompted ophthalmology consultation, revealing peripheral field changes, retinal findings, and bilateral optic disc edema. Imaging confirmed bilateral optic neuropathy. This case highlights the potential for pediatric ocular involvement in BPDCN.

Myeloid cells, including dendritic cells (DCs) and macrophages, are essential for establishing central tolerance in the thymus by promoting T cell clonal deletion and regulatory T cell (Treg) generation. Previous studies suggest that the thymic DC pool consists of plasmacytoid DC (pDC), XCR1+ DC1, and SIRPα+ DC2. Yet the precise origin, development, and homeostasis, particularly of DC2, remain unresolved. Using single-cell transcriptomics and lineage-defining mouse models, we identify nine major populations of thymic myeloid cells and describe their lineage identities. What was previously cons

Microfold (M) cells are rare intestinal epithelial cells that reside in the follicle-associated epithelium of Peyer's patches1. M cells transport luminal antigens to submucosal antigen-presenting cells2,3. These insights primarily derive from transmission electron microscopy and studies using genetically modified mice2-4. Here we establish an intestinal organoid model to study human M cells and reconstruct the differentiation trajectory of M cells through transcriptome profiling. The results indicate that as well as facilitating luminal antigen transport, human M cells also directly present an

BACKGROUND: Blastic plasmacytoid dendritic cell neoplasm (BPDCN) and acute myeloid leukemia (AML) show overlapping clinicopathological presentations, which makes it challenging to differentiate on a small skin biopsy. NPM1 mutations are the most common genetic lesions in AML, accounting for one third of cases and cause an aberrant cytoplasmic delocalization of NPM1 mutants, which can be detected by an immunohistochemical stain. Frequency of NPM1 mutations in BPDCN remains controversial. We aimed to investigate NPM1 mutations in cutaneous BPDCN cases and compare them with cutaneous NPM1 positiv

The innate immune system relies on pattern recognition receptors (PRRs) to detect pathogen-associated molecular patterns (PAMPs) and guard proteins to monitor pathogen disruption of host cell processes. How different immune cell types engage PRRs and guard proteins to respond to infection is poorly understood. Here, we show that macrophages and dendritic cells (DCs) distinctly respond to bacterial virulence activities. In macrophages, the bacterial pathogen Legionella pneumophila deploys its Dot/Icm type IV secretion system (T4SS) to deliver effector proteins that facilitate robust intracellul

Cancer stem cells (CSCs) can self-renew and differentiate, contributing to tumor heterogeneity, metastasis, and recurrence. Their resistance to therapies, including immunotherapy, underscores the importance of targeting them for complete remission and relapse prevention. Olfactomedin 4 (OLFM4), a marker associated with various cancers such as colorectal cancer, is expressed on CSCs promoting immune evasion and tumorigenesis. However, its potential as a target for CSC-specific immunotherapy remains underexplored. The primary aim of this study is to evaluate the effectiveness of targeting OLFM4
