The efficacy and safety of fluoxetine, a new antidepressant agent, were assessed in a double-blind, parallel, randomized study of 44 outpatients with major depressive disorder. Following a 1-week placebo period, patients were randomly assigned to either fluoxetine or amitriptyline for a period of 5 weeks. The mean maintenance dosages were 55 mg/day for fluoxetine and 159 mg/day for amitriptyline. Both drugs were effective in relieving the symptoms of depression. The most frequently reported side effects were nausea and nervousness for fluoxetine, and dry mouth, dizziness, and drowsiness for am…

A consecutive sample of 298 nonpsychotic psychiatric outpatients was classified according to DSM-III and divided into 4 diagnostic groups: pure major depression, mixed major depression/panic disorder, pure panic disorder and a remaining group of other disorders. The patients' report of childhood relationship to parents and siblings, family atmosphere, their own personality characteristics as children and precipitating events were compared in the various groups. In addition, differences in personality and frequencies of personality disorders were investigated by means of various instruments. Ou

The relationship between childhood trauma with major depressive disorder (MDD) and personality disorders is complex. We explored the differences in the subjective reporting of childhood trauma to determine whether there were differences between those with a diagnosis of personality disorder and those with MDD. Adult patients with depressive symptoms were recruited from three adult psychiatry inpatient wards. Sixty inpatients fulfilled the study criteria and were requested to complete the childhood trauma questionnaire (CTQ). At discharge, diagnosis was determined and was allocated mainly to tw…

The international classification of diseases 11th (ICD-11) revision is due by 2015. The ICD-11 beta draft has recently been released, which includes a prospective change in the content of mood disorders. The ICD-11 may separate the disorders into bipolar and depressive disorders as a consequence of an evaluation for the feasibility of a meta-structure for mental and behavioral disorders. In addition, the bipolar disorders may be divided into type I and II disorders. The depressive disorders may include new diseases, i. e., disruptive mood dysregulation disorder, mixed depressive anxiety, and p…

Bipolar II disorder (recurrent depressive and hypomanic episodes) and related disorders (united in the bipolar spectrum) are understudied, despite a prevalence of about 5% in the community and about 50% in depressed outpatients. The apparent increase in prevalence of the bipolar spectrum is related to several changes in diagnostic criteria, including improved probing for history of hypomania (focused more on overactivity than on mood change), lower minimum duration of hypomania, and inclusion of unipolar depressions with bipolar signs (eg, family history of bipolar disorder, mixed depression).

Menstrual disorders are among the most frequent somatic complications and symptoms of eating disorders. This study was designed to assess the prevalence of eating disorders connected to menstrual disorders of a non-organic origin in women at two gynecological out-patient departments. We performed a survey via questionnaires (ANIS: Anorexia Nervosa Inventory Scale; BCDS: Bulimic Cognitive Distortions Scale; BITE: Bulimia Investigation Test, Edinburgh; EDI: Eating Disorders Inventory; BDI: Beck Depression Inventory). Among 75 women tested we found a prevalence figure of 4% for anorexia nervosa (

BACKGROUND: Depressive disorder is a common mental illness associated with substantial functional impairment. Although pharmacotherapy is widely used, its effectiveness is often limited by adverse effects and poor adherence. Acupuncture has been increasingly applied as a complementary treatment for depression, and its neurobiological characteristics remain unclear.
OBJECTIVE: This randomized, sham-controlled trial aimed to evaluate the clinical efficacy of acupuncture for mild to moderate depressive disorder and to investigate its effects on prefrontal cortical function using functional near-…

BACKGROUND: Adolescent depression and suicide are significant issues, with insufficient solutions available. Concerns about electroconvulsive therapy (ECT)'s cognitive side effects have restricted its use in adolescents with major depressive disorder (MDD). Clarifying ECT's cognitive, antidepressant, and anti-suicidal effects is crucial to inform its clinical application.
METHODS: This prospective observational study enrolled 60 adolescents with DSM-5-diagnosed MDD and scheduled for bifrontal ECT, between October 2023 and March 2025. Cognitive outcomes were assessed using the Cambridge Neurop…

INTRODUCTION: The effects of repetitive transcranial magnetic stimulation (rTMS) on the left dorsolateral prefrontal cortex (DLPFC) in patients with major depressive disorder (MDD) have been proved to have antidepressant effects. However, the absence of biomarkers to assess treatment response remains a challenge. This research aims to explore the relationship between frontal lobe activity, measured using near infrared spectroscopy (NIRS), and changes in symptoms among MDD patients following rTMS treatment.
METHODS: A total of 26 MDD patients underwent 20 sessions of 10 Hz rTMS targeting the …

BACKGROUND: Childhood maltreatment continues to pose a great challenge to psychiatry. Although there is growing evidence demonstrating that childhood maltreatment is an important risk factor for depressive disorders, it remains to be elucidated which specific symptoms occur after exposure to different kinds of childhood maltreatment, and whether certain pathways may account for these associations.
PARTICIPANTS AND SETTINGS: A total of 203 adult patients (18-53 years old) with MDD, diagnosed by Structured Clinical Interview for Diagnostic and Statistical Manual of Mental Disorders, Fourth Edit…

BACKGROUND: There is evidence that symptomatology in patients with major depressive disorder (MDD) changes with age. However, studies comparing depressive symptomatology between different age groups during antidepressant therapy are rare. We compared demographic and clinical characteristics in depressed patients of different age groups at baseline and during treatment.
METHODS: 889 MDD inpatients were divided into four age groups (18-29, 30-39, 40-49, 50-65 yrs.). Demographic and clinical characteristics including depressive symptomatology (assessed by the Inventory of Depressive Symptoms) we…

Background: Recent studies have demonstrated that the assessment of postural performance may be a potentially reliable and objective marker of the psychomotor retardation (PMR) in the major depressive disorder (MDD). One of the important facets of MDD-related PMR is reflected in disrupted central mechanisms of psychomotor control, heavily influenced by compelling maladaptive depressive rumination. In view of this we designed a research paradigm that included sequential execution of simple single-posture task followed by more challenging divided attention posture tasks, involving concurring mot…

INTRODUCTION: Cognitive disturbances in Major Depressive Disorder (MDD) could persist beyond the symptomatic phase of the illness. However, the works addressing this topic did not usually account for the possible impact of medication on the cognitive functions of depressed patients. The present study aims to investigate whether MDD patients in remission treated with selective serotonin reuptake inhibitors (SSRI) or dual serotonergic-noradrenergic reuptake inhibitors (SNRI) show cognitive deficits, to study whether the same patients suffer neuropsychological disturbances when they are unmedicat…

BACKGROUND: Self-stigma toward psychiatric disorders is a major barrier to help-seeking and treatment adherence, significantly impacting patients' quality of life. Insight is one of the key factors influencing internalized stigma. Therefore, this study aims to assess the relationship between self-stigma and insight among patients with Major Depressive Disorder (MDD), Generalized Anxiety Disorder (GAD), and Obsessive-Compulsive Disorder (OCD).
METHODS: A total of 140 participants included individuals diagnosed with MDD, GAD and OCD who had not experienced recurrent episodes over the past six m…

Orexin-A, a neuropeptide involved in regulating physiological processes, has potential implications in psychiatric disorders. Studies examining orexin-A levels in schizophrenia, bipolar disorder, and major depressive disorder have produced conflicting results, highlighting the need for further research. To address these inconsistencies, this meta-analysis aims to synthesize existing data on orexin-A levels across these disorders, clarifying its role and potential as a diagnostic or prognostic biomarker. This systematic review, registered with PROSPERO, investigated plasma orexin-A levels in sc…

The inclusion of polygenic scores (PGS) from genetically correlated traits such as Major Depressive Disorder (MDD) and alcohol use disorder (AUD) may improve the prediction of these outcomes and their comorbidity. Despite the importance of this work, few studies have evaluated the efficacy of this possibility. The current study evaluates the use of MDD and AUD PGS individually and together to improve the prediction of MDD, AUD, and comorbid MDD-AUD using a sample of European, African, or Admixed American Ancestry participants from the National Longitudinal Study of Adolescent to Adult Health (…

The blood-brain barrier (BBB) is a unique neurovascular interface essential for central nervous system homeostasis. Beyond its classical protective role, accumulating evidence points to the BBB as a dynamic structure actively involved in brain function in health and disease. The present review synthesizes clinical, molecular, and neuroimaging evidence implicating BBB dysfunction in major depressive disorder (MDD), bipolar disorder (BD), and schizophrenia. The paper was prepared in accordance with the recommendations of the Scale for the Assessment of Narrative Review Articles and was based on …

Objective: Patients with Major depressive disorder (MDD) and a comorbid Borderline personality disorder (BPD) show a poorer prognosis for MDD compared to patients without BPD. Little is known about the therapeutic processes underlying this prognosis. The goal of the present study was to investigate whether patients with more severe BPD symptoms experience less strengthening in patient-therapist movement synchrony (MS) throughout treatment, and whether less strengthening in MS is associated with less effective sessions. Method: Ninety-five patients participating in a randomized control trial we…
