The effects of local micro injections of opiates and enkephalins into the forebrain on the electrocorticogram of the rat.

The effects of various opiate compounds have been studied on the electrocorticogram (ECoG) of the rat following local injection into various brain areas. Injections of all compounds studied into both the caudate-putamen and the basal forebrain, in particular the olfactory tubercles, induced changes in the ECoG. Injections of saline vehicle into these areas were ineffective as were injections of morphine into the corpus callosum. Potency was etorphine greater than morphine = codeine greater than thebaine. Naloxone alone was inactive following injection but if combined with morphine markedly att

Novel Cyclic Endomorphin Analogues with Multiple Modifications and Oligoarginine Vector Exhibit Potent Antinociception with Reduced Opioid-like Side Effects.

Endomorphins (EMs) are potent pharmaceuticals for the treatment of pain. Herein, we investigated several novel EM analogues with multiple modifications and oligoarginine conjugation. Our results showed that analogues 1-6 behaved as potent μ-opioid agonists and enhanced stability and lipophilicity. Analogues 5 and 6 administered centrally and peripherally induced significant and prolonged antinociceptive effects in acute pain. Both analogues also produced long-acting antiallodynic effects against neuropathic and inflammatory pain. Furthermore, they showed a reduced acute antinociceptive toleran

Green Light Antinociceptive and Reversal of Thermal and Mechanical Hypersensitivity Effects Rely on Endogenous Opioid System Stimulation.

Benefits of phototherapy were characterized in multiple diseases including depression, circadian rhythm disruptions, and neurodegeneration. Studies on migraine and fibromyalgia patients revealed that green light-emitting diodes (GLED) exposure provides a pragmatic and safe therapy to manage chronic pain. In rodents, GLED reversed hypersensitivity related to neuropathic pain. However, little is known about the underlying mechanisms of GLED efficacy. Here, we sought to understand how green light modulates the endogenous opioid system. We first characterized how exposure to GLED stimulates releas

Endometriosis: what a pain it is.

The contrasting findings obtained in the studies that have attempted to correlate the stage of endometriosis with severity of pain symptoms suggest that some specific characteristics of the lesions are more implicated in the genesis of pain than disease extension per se. Thus, fresh, metabolically active, intraperitoneal implants may cause functional pain symptoms such as dysmenorrhea, whereas infiltrating, nodular and fibrotic lesions are responsible for organic-type pain such as deep dyspareunia. Women with symptomatic endometriosis seem to have reduced peripheral beta-endorphin production i

Interactions between the blood-brain barrier and endogenous peptides: emerging clinical implications.

The effects of peptides on brain function suggest therapeutic and pathologic roles for these substances. Many peptides cross the blood-brain barrier (BBB) by transmembrane diffusion as a function of their lipid solubilities. Other peptides, such as the enkephalins, Tyr-MIF-1, vasopressin-related peptides, and peptide T-like peptides, are transported by carrier-mediated systems. Passage is influenced by aging, stress, lighting, drugs, amino acids, and neurotoxins. Disruption of the BBB results in complex changes in the blood and CSF levels of peptides. Peptides influence the passage of glucose,

Sleep disorders in Tourette's syndrome.

Sleep disorders have been reported in approximately 80% of Tourette's syndrome (TS) patients. Sleep studies in TS patients have demonstrated a 30% reduction in delta-sleep (slow-wave sleep) in nontreated subjects, decreased percentage of REM-sleep and the presence of tics during sleep. A subgroup of young TS patients was reported to have an increased percentage of delta sleep. Although these findings were initially thought to result from deranged dopaminergic and serotoninergic functions in TS, we suggest that abnormalities of hypothalamic-mediated control mechanisms involving the intrinsic op

The endogenous opioid system in Gilles de la Tourette syndrome.

The various neurotransmitter systems postulated to be involved in the pathogenesis of Gilles de la Tourette syndrome(GTS) are described with special reference to the endogenous opioid system(EOS). Malfunction of the opioid system is proposed as the underlying disturbance in this disease causing secondary dysfunction of the other systems. Furthermore, the various symptoms of the illness are examined also in terms of dysfunction of the endogenous opioid system.

Suppression of renin release by antagonism of endogenous opiates in the dog.

The influence of blockade of endogenous opioids on the release of renin due to partial renal arterial constriction was determined acutely and chronically in unilaterally nephrectomized dogs. In acute preparations changes in plasma renin activity, arterial blood pressure, and heart rate were determined after 15 min of 60% renal arterial constriction before and after administration of either a saline vehicle, the opiate antagonist naloxone (0.05 mg/kg), or morphine (2 mg/kg). Acute antagonism of endogenous opiates abolished the increase in plasma renin activity and mean arterial pressure associa

Effects of centrally administered naloxone on phrenic nerve activity.

The present study examined the effect on respiration of naloxone administered centrally. Experiments were performed on anesthetized, paralyzed cats whose vagi and carotid sinus nerves had been cut. The animals' end-tidal pCO2 and body temperature were kept constant with servocontrollers. Naloxone administered into the third cerebral ventricle in doses of 40 micrograms/kg and 400 micrograms/kg resulted in significant (P less than 0.05) increases in phrenic neural activity. The response to the 400 micrograms/kg dose was significantly greater (P less than 0.05) than the response to the 40 microgr

Amphetamine-induced analgesia does not involve brain opioids.

The intrinsic analgesic properties of amphetamine were studied in rats. Subcutaneous injection of amphetamine exerted an additive effect on morphine-induced analgesia in the hot-plate test. Amphetamine itself showed intrinsic analgesic activity in a dose-dependent manner. Administration of naloxone failed to affect the analgesia induced by amphetamine. However, injection of haloperidol totally suppressed the amphetamine-induced change in pain response latency. Both naloxone and haloperidol failed to affect the pain threshold when injected alone, but inhibited morphine-induced analgesia. It is

The effect of spontaneous seizures on pentylenetetrazole and maximum electroshock induced seizures in the Mongolian gerbil.

The seizure sensitive Mongolian gerbil, a genetic model of epilepsy, has been used to investigate the effects of prior spontaneous seizures on the threshold for pentylenetetrazole (PTZ) and maximum electroshock (MES) induced convulsions. In animals which had spontaneous epileptic seizures prior to testing the threshold for PTZ induced convulsions was raised compared to non-seized animals. This inhibitory effect of a spontaneous seizure was not observed for MES convulsions. Pretreatment with naltrexone, an opioid antagonist, attenuated the post-ictal inhibitory effect on PTZ induced convulsions

Elevated opioid activity in sera of chronic schizophrenics.

The effect of native serum samples from schizophrenic and control patients on the electrically induced contractions of the isolated mouse vas deferens (MVD) preparations was investigated. It was demonstrated that only the samples of schizophrenic origin elicited a naloxone dependent inhibition on the contractions of the MVD preparations, while sera from healthy individuals and those from non schizophrenic but mentally ill patients proved to be ineffective in this respect. By using ultrafiltration and gel chromatographic techniques, four fractions disclosing MVD related biological activity coul

A new Met-enkephalin analogue suppresses plasma vasopressin in man.

Metkephamid acetate, a newly synthetized analogue of methionine enkephalin, suppressed plasma vasopressin levels from 2.4 pg/ml to 0.8 pg/ml after intramuscular administration to 4 normal human volunteers. This suppression occurred in the absence of significant changes in either plasma osmolality or blood pressure. Thus, metkephamid acetate, like many other opiates which bind to the mu subtype of opiate receptors, has a direct suppressive effect on plasma vasopressin levels.

Role of Physical Activity in Women's Mid-life: Neuroscience, Longevity, and Disease Prevention.

Physical activity (PA) is vital for managing menopausal symptoms and reducing noncommunicable diseases in middle-aged women. Despite the known benefits, many women do not meet the recommended activity levels and view household tasks as sufficient owing to factors such as time constraints, lack of motivation, or physical limitations. This article explores the significance, forms, and health impacts of PA in menopausal women. Regular PA, including aerobic exercise, resistance training, and yoga, offers several health benefits. Exercise enhances adult neurogenesis, brain structure, learning, memo

A Mouse Model of Sporadic Alzheimer's Disease with Elements of Major Depression.

After olfactory bulbectomy, animals are often used as a model of major depression or sporadic Alzheimer's disease and, hence, the status of this model is still disputable. To elucidate the nature of alterations in the expression of the genome after the operation, we analyzed transcriptomes of the cortex, hippocampus, and cerebellum of the olfactory bulbectomized (OBX) mice. Analysis of the functional significance of genes in the brain of OBX mice indicates that the balance of the GABA/glutamatergic systems is disturbed with hyperactivation of the latter in the hippocampus, leading to the devel

Rett syndrome: the state of clinical and basic research, and future perspectives.

To clarify the pathophysiology of brain and spinal cord impairment in Rett syndrome (RTT), we report on the current status of research on Rett syndrome and review the abnormalities reported in neurotransmitters, neuromodulators and other biological markers in patients with RTT. We have previously investigated the levels of various factors in the blood, plasma, and cerebrospinal fluid (CSF) of RTT patients, including biogenic amines, lactate, melatonin, pyruvate and other citric acid cycle intermediates, substance P, β-endorphin and other neuropeptides, and a neuromodulator of β-phenylethylamin
