Comparison of the cell walls of Paracoccidioides loboi and Paracoccidioides brasiliensis by using polysaccharide-binding dyes.

Biopsies from patients with Jorge Lobo's disease (keloid blastomycosis) and paracoccidioidomycosis (South American blastomycosis), caused respectively by Paracoccidioides loboi a non-cultivated fungus and Paracoccidioides brasiliensis, were stained with six different dyes having the property of forming complexes with some polysaccharides and oligosaccharides. The cell-walls of both fungi showed similar staining characteristics, suggesting a similar chemical structure. The role of the fungal cell-walls in evasion mechanisms is discussed.

Talaromyces spp. are tunable producers of light-activated secondary metabolites with anticancer potential.

Photodynamic therapy (PDT) is a promising treatment strategy for resistant or non-surgical tumors. To develop photosensitizers tailored to specific cancer types, new chemical scaffolds are needed. Macrofungi have recently been identified as a potent source of such compounds, particularly dimeric anthraquinones. However, the sources previously described (i.e., dermocyboid Cortinarii) cannot be cultivated in a biotechnologically applicable way. Thus, in this proof-of-concept study, we explored Talaromyces species (Ascomycota) for their potential to produce similar light-activated metabolites. Si

Synthesis of Novel Isoxazoline Derivatives with Selective Inhibition of the Drug Efflux Pump Mdr1 to Reverse Drug Resistance of Candida albicans In Vitro and In Vivo.

The transcriptional upregulation of drug efflux pumps constitutes a primary mechanism of azole resistance acquired by diverse fungal pathogens. This overexpression diminishes intracellular azole accumulation, thereby compromising drug efficacy and contributing to therapeutic failure. The coadministration of azole antifungals with efflux pump inhibitors presents a promising therapeutic strategy to combat resistant fungal infections. In the search for novel antifungal drugs, we obtained 49 isoxazoline derivatives through structural modification of the isoxazolin-3-one skeleton. Among them, 6ao s

Human Pathogenic Paecilomyces from Food.

Paecilomyces spp. and Byssochlamys spp. are heat-resistant fungi important to industry because they can cause food and beverage spoilage, incurring economic loss. The consequences of food or beverage fungal colonization is the loss of nutritional value, structure and taste, and the possibility of producing toxic secondary metabolites that may result in medical problems. Furthermore, these fungi can infect animals and humans and it is unknown if contaminated foods may be fomites. P. variotii is the principal agent of food spoilage or contamination and it is most frequently associated with human

Influence of doping ion, capping agent and pH on the fluorescence properties of zinc sulfide quantum dots: Sensing of Cu2+ and Hg2+ ions and their biocompatibility with cancer and fungal cells.

Herein, a facile one-pot synthetic method was explored for the fabrication of glutathione capped Mn2+ doped‑zinc sulphide quantum dots (GSH-Mn2+-ZnS QDs) for both fluorescent detection of Cu2+ and Hg2+ ions and for fluorescence imaging of two cancer (RIN5F and MDAMB231) and fungal (Rhizopus oryzae) cells. Particularly, doping of Mn2+ into ZnS QDs nanocrystal structure resulted a great improvement in the fluorescence properties of ZnS QDs. The emission peak of undoped ZnS QDs was found at 447 nm, which is due to the large number of surface defects in the ZnS QDs nanostructures. Under identical

Chemical Interaction among Termite-Associated Microbes.

Bacteria and fungi in shared environments compete with one another for common substrates, and this competition typically involves microbially-produced small molecules. An investigation of one shared environmental niche, the carton material of the Formosan subterranean termite Coptotermes formosanus, identified the participants on one of these molecular exchanges. Molecular characterization of several termite-associated actinobacteria strains identified eleven known antimicrobial metabolites that may aid in protecting the C. formosanus colony from pathogenic fungal infections. One particular ac

Phosphoinositide-mediated oligomerization of a defensin induces cell lysis.

Cationic antimicrobial peptides (CAPs) such as defensins are ubiquitously found innate immune molecules that often exhibit broad activity against microbial pathogens and mammalian tumor cells. Many CAPs act at the plasma membrane of cells leading to membrane destabilization and permeabilization. In this study, we describe a novel cell lysis mechanism for fungal and tumor cells by the plant defensin NaD1 that acts via direct binding to the plasma membrane phospholipid phosphatidylinositol 4,5-bisphosphate (PIP2). We determined the crystal structure of a NaD1:PIP2 complex, revealing a striking o

Common bile duct obstruction due to candidiasis.

Biliary obstruction with its wide range of potential causes (e.g. neoplastic lesions, gallstones and inflammatory processes) is a common disease in gastroenterology. Although infections with Candida and other fungal species have increasingly been recognized in patients with certain predispositions, fungal involvement of the biliary tract is extremely rare. We report the case of a male patient with a past history of long-time mechanical ventilation and who was referred to our department with cholangitis. Endoscopic retrograde cholangio-pancreatography (ERCP) of the septic patient revealed a hig

Architectural confinement and seasonal forcing shape cross-domain pathogen-associated assemblages in complex built environments.

Airborne microbial pathogens in built environments (BEs) may pose health threats, yet the ecological mechanisms governing their assembly and persistence across interconnected architectural spaces are less studied. Here, we conducted a year-long, multi-spatial investigation across a university building complex, sampling the exhaust outlets of three indoor environments and adjacent outdoor inlets. By integrating the 16S rRNA gene and ITS sequencing from 437 paired bacterial-fungal samples, we characterized the spatiotemporal dynamics of airborne opportunistic pathogen-containing genera. Identification of regions in which positive selection may operate in S-RNase of Rosaceae: implication for S-allele-specific recognition sites in S-RNase.

A stylar S-RNase is associated with gametophytic self-incompatibility in the Rosaceae, Solanaceae, and Scrophulariaceae. This S-RNase is responsible for S-allele-specific recognition in the self-incompatible reaction, but how it functions in specific discrimination is not clear. Window analysis of the numbers of synonymous (dS) and non-synonymous (dN) substitutions in rosaceous S-RNases detected four regions with an excess of dN over dS in which positive selection may operate (PS regions). The topology of the secondary structure of the S-RNases predicted by the PHD method is very similar to th

Designing a chimeric multi-epitope vaccine against Candida auris using reverse vaccinology approach targeting the agglutinin-like protein N-terminal domain.

Candida auris is an emerging multidrug resistant fungal pathogen associated with high mortality rates, rapid global dissemination and resistance to conventional antifungal therapies. It's remarkable ability to evade host immune responses and persist in health care setting demands the development of effective immunotherapeutic strategies. In this study, a reverse vaccinology and immunoinformatics based approach was employed to design a novel chimeric multi-epitope vaccine targeting surface expose N-terminal domain of the agglutinin like protein involved in host pathogen interactions. The Trisubstituted Isoxazole MMV688766 Exerts Broad-Spectrum Activity against Drug-Resistant Fungal Pathogens through Inhibition of Lipid Homeostasis.

Candida species are among the most prevalent causes of systemic fungal infection, posing a growing threat to public health. While Candida albicans is the most common etiological agent of systemic candidiasis, the frequency of infections caused by non-albicans Candida species is rising. Among these is Candida auris, which has emerged as a particular concern. Since its initial discovery in 2009, it has been identified worldwide and exhibits resistance to all three principal antifungal classes. Here, we endeavored to identify compounds with novel bioactivity against C. auris from the Medicines fo

Two forms and two faces, multiple states and multiple uses: Properties and applications of the self-assembling fungal hydrophobins.

The fungal hydrophobins are small proteins that are able to spontaneously self-assemble into amphipathic monolayers at hydrophobic:hydrophilic interfaces. These protein monolayers can reverse the wettability of a surface, making them suitable for increasing the biocompatibility of many hydrophobic materials. The self-assembling properties and amphipathic nature of hydrophobins make them attractive candidates for biotechnological applications. Recently, there have been significant advances in the understanding of the structure and assembly of these remarkable proteins. This opens up the way for

Formulations Based on Natural Ingredients for the Treatment of Nail Diseases.

Nail is a strong and resistant structure, characterized by a low permeability to foreign molecules. Nails can be subjected to many diseases, among which fungal infections (e.g. onchomycosis) are the most common and responsible for nail structure alteration. Many formulations have been produced for the delivery of active ingredients to treat nail disorders, based on newly synthesized active molecules or containing chemical enhancers or chemically-modified polymers able to improve the drug transungual penetration. To avoid permanent alterations of the nail structure due to the use of chemical co

A role for LHC1 in higher order structure and complement binding of the Cryptococcus neoformans capsule.

Polysaccharide capsules are important virulence factors for many microbial pathogens including the opportunistic fungus Cryptococcus neoformans. In the present study, we demonstrate an unusual role for a secreted lactonohydrolase of C. neoformans, LHC1 in capsular higher order structure. Analysis of extracted capsular polysaccharide from wild-type and lhc1Δ strains by dynamic and static light scattering suggested a role for the LHC1 locus in altering the capsular polysaccharide, both reducing dimensions and altering its branching, density and solvation. These changes in the capsular structure

Rim Pathway-Mediated Alterations in the Fungal Cell Wall Influence Immune Recognition and Inflammation.

UNLABELLED: Compared to other fungal pathogens, Cryptococcus neoformans is particularly adept at avoiding detection by innate immune cells. To explore fungal cellular features involved in immune avoidance, we characterized cell surface changes of the C. neoformans rim101Δ mutant, a strain that fails to organize and shield immunogenic epitopes from host detection. These cell surface changes are associated with an exaggerated, detrimental inflammatory response in mouse models of infection. We determined that the disorganized strain rim101Δ cell wall increases macrophage detection in a contact-de
