Metabolic Trade-Off Between Carbon and Phenylpropanoid Pathways in Wheat Under Saline-Alkali Stress Revealed by Multi-Omics Analysis.

Wheat production is severely limited by soil salinity-alkalinity, yet the molecular mechanisms governing seed germination under such stress remain largely unelucidated. This study employed integrated transcriptomics, metabolomics, and WGCNA to decipher the adaptation strategies of germinating wheat seeds to gradient saline-alkali stress. Our results showed that stress induced a functional differentiation in carbon metabolism: fructose and mannose metabolism was enhanced, whereas starch and sucrose metabolism was inhibited. Concurrently, the phenylpropanoid defense network was differentially re

Transcriptome and metabolome analysis of Cycas bifida response to low-temperature stress.

BACKGROUND: Cycas bifida is an endemic species distributed in limited areas of Yunnan and Guangxi, with a notably small population size. Under the context of global climate warming, this species may require introduction to colder habitats. Previous studies have confirmed its weak cold tolerance. To thoroughly investigate the molecular and metabolic mechanisms underlying its response to low temperature, this study employed low-temperature stress treatments combined with transcriptome sequencing and metabolomics technologies for systematic comparative analysis. RESULTS: Through multi-omics anal

The SPL8-YAB3/TCP19 module integrates transcriptional and protein-protein regulatory mechanisms to orchestrate anther development in Arabidopsis.

SQUAMOSA PROMOTER BINDING PROTEIN-LIKE 8 (SPL8) is a plant-specific transcription factor essential for anther development, but its regulatory partners and downstream mechanisms remain largely unknown. We identified 26 SPL8-interacting transcription factors by yeast two-hybrid screening and bimolecular fluorescence complementation (BiFC) assays, with YABBY (YAB) and TEOSINTE BRANCHED 1/CYCLOIDEA/PCF (TCP) family emerging as the predominant interactors. Among these, YAB3 and TCP19 were selected for detailed characterization. Their interactions with SPL8, as well as with each other, were confirme

Damage of brown planthopper (BPH) Nilaparvata lugens and rice leaf folder (LF) Cnaphalocrocis medinalis in parent plants lead to distinct resistance in ratoon rice.

Herbivore-induced defense responses are often specific, whereas plants could induce distinct defense responses corresponding to infestation by different herbivorous insects. Brown plant hopper (BPH) Nilaparvata lugens, a phloem-feeding insect, and rice leaf folder (LF) Cnaphalocrocis medinalis, a chewing insect, are both specialist herbivores on rice. To characterize the distinct resistance primed by prior damage to these two specialist herbivores, we challenged rice plants with two herbivores during vegetative growth of parent plants and assessed plant resistance in subsequent ratoons. Here,

Lipoxygenase (LOX) in Sweet and Hot Pepper (Capsicum annuum L.) Fruits during Ripening and under an Enriched Nitric Oxide (NO) Gas Atmosphere.

Lipoxygenases (LOXs) catalyze the insertion of molecular oxygen into polyunsaturated fatty acids (PUFA) such as linoleic and linolenic acids, being the first step in the biosynthesis of a large group of biologically active fatty acid (FA)-derived metabolites collectively named oxylipins. LOXs are involved in multiple functions such as the biosynthesis of jasmonic acid (JA) and volatile molecules related to the aroma and flavor production of plant tissues, among others. Using sweet pepper (Capsicum annuum L.) plants as a model, LOX activity was assayed by non-denaturing polyacrylamide gel elect

A metal tolerance protein, MTP10, is required for the calcium and magnesium homeostasis in Arabidopsis.

Nutrient antagonism typically refers to the fact that too high a concentration of one nutrient inhibits the absorption of another nutrient. In plants, Ca2+ (Calcium) and Mg2+ (Magnesium) are the two most abundant divalent cations, which are known to have antagonistic interactions. Hence, maintaining their homeostasis is crucial for plant growth and development. In this study, we showed that MTP10 (Metal Tolerance Protein 10) is an important regulator for maintaining homeostasis of Mg and Ca in Arabidopsis. The mtp10 mutant displayed severe growth retardation in the presence of excess Mg2+, to

Human protein paucimannosylation: cues from the eukaryotic kingdoms.

Paucimannosidic proteins (PMPs) are bioactive glycoproteins carrying truncated α- or β-mannosyl-terminating asparagine (N)-linked glycans widely reported across the eukaryotic domain. Our understanding of human PMPs remains limited, despite findings documenting their existence and association with human disease glycobiology. This review comprehensively surveys the structures, biosynthetic routes and functions of PMPs across the eukaryotic kingdoms with the aim of synthesising an improved understanding on the role of protein paucimannosylation in human health and diseases. Convincing biochemica

Heteroexpression of the wheat phytochelatin synthase gene (TaPCS1) in rice enhances cadmium sensitivity.

Phytochelatin synthase (PCS) (EC 2.3.2.15) catalyzes the final step of phytochelatins (PCs) biosynthesis. PCs are a family of cysteine-rich thiol-reactive and heavy metal-binding peptides that play an important role in sequestration and detoxification of heavy metals in plants. Previous studies have indicated that plants that overexpressed PCS displayed contrasting phenotypes, ranging from enhanced cadmium (Cd) tolerance to Cd hypersensitivity in Arabidopsis thaliana. In this study, the wheat phytochelatin synthase gene, TaPCS1, was heteroexpressed in wild-type rice (Oryza sativa L., cv. Zhong

Defects in the rice aconitase-encoding OsACO1 gene alter iron homeostasis.

Rice aconitase gene OsACO1 is involved in the iron deficiency-signaling pathway for the expression of iron deficiency-inducible genes, either thorough enzyme activity or possible specific RNA binding for post-transcriptional regulation. Iron (Fe) is an essential element for virtually all living organisms. When plants are deficient in Fe, Fe acquisition systems are activated to maintain Fe homeostasis, and this regulation is mainly executed at the gene transcription level. Many molecules responsible for Fe uptake, translocation, and storage in plants have been identified and characterized. Howe

Overexpression of SlREV alters the development of the flower pedicel abscission zone and fruit formation in tomato.

Versatile roles of REVOLUTA (REV), a Class III homeodomain-leucine zipper (HD-ZIP III) transcription factor, have been depicted mainly in Arabidopsis and Populus. In this study, we investigated the functions of its tomato homolog, namely SlREV. Overexpression of a microRNA166-resistant version of SlREV (35S::REV(Ris)) not only resulted in vegetative abnormalities such as curly leaves and fasciated stems, but also caused dramatic reproductive alterations including continuous production of flowers at the pedicel abscission zone (AZ) and ectopic fruit formation on receptacles. Molecular dissection of the response of the rice Systemic Acquired Resistance Deficient 1 (SARD1) gene to different types of ionizing radiation.

PURPOSE: Exposure to ionizing radiation induces plant defenses by regulating the expression of response genes. The systemic acquired resistance deficient 1 (SARD1) is a key gene in plant defense response. In this study, the function of Oryza sativa SARD1 (OsSARD1) was investigated after exposure of seeds/plants to ionizing radiation, jasmonic acid (JA) or salicylic acid (SA). MATERIALS AND METHODS: Rice seeds exposed to two types of ionizing radiations (gamma ray [GR] and ion beam [IB]) were analyzed by quantitative reverse transcription PCR (qRT-PCR) to identify the genes that are altered in

Bio-control agents activate plant immune response and prime susceptible tomato against root-knot nematodes.

Beneficial microorganisms are generally known to activate plant defense against biotic challenges. However, the molecular mechanisms by which activated plants react more rapidly and actively to pests remain still largely unclear. Tomato plants pre-treated with a mixture of beneficial bio-control agents (BCAs), as soil-drenches, were less sensitive to infection of the root-knot nematode (RKN) Meloidogyne incognita. To unravel the molecular mechanisms of this induced resistance against RKNs, we used qRT-PCR to monitor the expression, in tomato roots and leaves, of 6 key defense genes. Gene trans

Comparative profiling of gene expression in Camellia sinensis L. cultivar AnJiBaiCha leaves during periodic albinism.

The AnJiBaiCha albino mutant tea cultivar has a reversible albino phenotype at low temperatures. Albino AnJiBaiCha leaves contain high levels of amino acids, which are important components affecting the quality of tea as a beverage. To examine the molecular mechanism of albinism and amino acid enrichment in AnJiBaiCha, we used the amplified fragment length polymorphism (cDNA-AFLP) technique to isolate genes that are differentially expressed during periodic albinism in AnJiBaiCha. A total of 127 transcript-derived fragments (TDFs) were successfully sequenced; among those, 60 TDFs showed high si

Phloem iron remodels root development in response to ammonium as the major nitrogen source.

Plants use nitrate and ammonium as major nitrogen (N) sources, each affecting root development through different mechanisms. However, the exact signaling pathways involved in root development are poorly understood. Here, we show that, in Arabidopsis thaliana, either disruption of the cell wall-localized ferroxidase LPR2 or a decrease in iron supplementation efficiently alleviates the growth inhibition of primary roots in response to NH4+ as the N source. Further study revealed that, compared with nitrate, ammonium led to excess iron accumulation in the apoplast of phloem in an LPR2-dependent m

Expression of Arabidopsis pathogenesis-related genes during nematode infection.

The expression pattern of pathogenesis-related genes PR-1 to PR-5 was examined in the roots and leaves of Arabidopsis thaliana plants on infection with beet-cyst (Heterodera schachtii) and root-knot (Meloidogyne incognita) nematodes. During H. schachtii parasitism of Arabidopsis, the expression of PR-1, PR-2 and PR-5, which are considered to be markers for salicylic acid (SA)-dependent systemic acquired resistance (SAR), was induced in both roots and leaves of infected plants. In addition, the expression of PR-3 and PR-4, which are used as markers for jasmonic acid (JA)-dependent SAR, was not

Harnessing biocompatible bio-based nanoprotectants for phytohormone modulation and heavy metal detoxification in sustainable crop production.

Bio-based nanomaterials (BNMs) have emerged as promising modulators of plant growth hormone (phytohormone) signaling pathways under combined heavy metal (HM) stress conditions. This review investigates how BNMs influence hormone-regulated stress signaling networks to improve plant tolerance to HM stress at biochemical, molecular and physiological levels. Plant extract and microbial-mediated synthesis strategies use biological metabolites as reducing and stabilizing agents, providing environmentally compatible routes for agricultural nanomaterials. BNMs may influence phytohormone-regulated stre

The role of strigolactones in P deficiency induced transcriptional changes in tomato roots.

BACKGROUND: Phosphorus (P) is an essential macronutrient for plant growth and development. Upon P shortage, plant responds with massive reprogramming of transcription, the Phosphate Starvation Response (PSR). In parallel, the production of strigolactones (SLs)-a class of plant hormones that regulates plant development and rhizosphere signaling molecules-increases. It is unclear, however, what the functional link is between these two processes. In this study, using tomato as a model, RNAseq was used to evaluate the time-resolved changes in gene expression in the roots upon P starvation and, usi
