Expression, purification and crystallization of CLK1 kinase - A potential target for antiviral therapy.

Cdc-like kinase 1 (CLK1) is a dual-specificity kinase capable of autophosphorylation on tyrosine residues and Ser/Thr phosphorylation of its substrates. CLK1 belongs to the CLK kinase family that regulates alternative splicing through phosphorylation of serine-arginine rich (SR) proteins. Recent studies have demonstrated that CLK1 has an important role in the replication of influenza A and chikungunya viruses. Furthermore, CLK1 was found to be relevant for the replication of HIV-1 and the West Nile virus, making CLK1 an interesting cellular candidate for the development of a host-directed anti

Dissection of the Effects of JAK and BTK Inhibitors on the Functionality of Healthy and Malignant Lymphocytes.

Despite the emergence of small molecule inhibitors, current treatment strategies for chronic lymphocytic leukemia (CLL) are not curative, and the search for new therapeutic modalities continues. Prosurvival signaling derived from the microenvironment is often mediated via JAK signaling. However, whether JAK inhibitors are useful in CLL therapy has not been studied extensively. JAK inhibitors are valuable therapeutic agents in myelofibrosis and show promising results in graft-versus-host-disease. However, JAK inhibition is associated with an increased infection risk, presumably because of the e

Treatment with Entinostat Heals Experimental Cholera by Affecting Physical and Chemical Barrier Functions of Intestinal Epithelia.

We have shown previously that oral treatment with sodium butyrate or phenylbutyrate in an experimental model of shigellosis improves clinical outcomes and induces the expression of the antimicrobial peptide CAP-18 in the large intestinal epithelia. In a subsequent study, we found that entinostat, an aroylated phenylenediamine compound, has similar therapeutic potential against shigellosis. In this study, we aimed to evaluate entinostat as a potential candidate for host-directed therapy against cholera in an experimental model. Vibrio cholerae-infected rabbits were treated with two different do…

A novel C5a-derived immunobiotic peptide reduces Streptococcus agalactiae colonization through targeted bacterial killing.

Streptococcus agalactiae (group B Streptococcus [GBS]) is a Gram-positive bacterium that colonizes the cervicovaginal tract in approximately 25% of healthy women. Although colonization is asymptomatic, GBS can be vertically transmitted to newborns peripartum, causing severe disease such as pneumonia and meningitis. Current prophylaxis, consisting of late gestation screening and intrapartum antibiotics, has failed to completely prevent transmission, and GBS remains a leading cause of neonatal sepsis and meningitis in the United States. Lack of an effective vaccine and emerging antibiotic resist

Pharmacotherapies to manage bone loss-associated diseases: a quest for the perfect benefit-to-risk ratio.

In this review, benefits and side-effects of current and emerging therapies to treat and prevent pathological bone loss are described. Bisphosphonates are the antiresorptive compounds most widely used in the treatment of bone-loss associated diseases. They are generally well-tolerated although have recently been associated with osteonecrosis of the jaw and other complications. Therapies modulating estrogen receptor activation are indicated in the prevention and treatment of either breast cancer or osteoporosis in postmenopausal women. Thus, hormone replacement therapy is effective in preventio

Activation of lytic Epstein-Barr virus (EBV) infection by radiation and sodium butyrate in vitro and in vivo: a potential method for treating EBV-positive malignancies.

The consistent presence of the EBV genome in certain tumors offers the potential for novel EBV-directed therapies. Switching the latent form of EBV infection present in most EBV-positive tumor cells into the cytolytic form may be clinically useful because lytic EBV infection leads to host cell destruction, and very few normal cells contain the EBV genome. It would also be therapeutically advantageous to induce expression of EBV-encoded lytic proteins that convert the nucleoside analogues ganciclovir (GCV) and 3'-azido-3'deoxythymidine (AZT) into their active, cytotoxic forms. The interaction of infection and the adult respiratory distress syndrome.

Infection, particularly that involving the respiratory tract, is commonly seen in the patient with ARDS. It can be either an etiologic factor leading to the syndrome or a complicating factor leading to a high likelihood of mortality. Pneumonia develops in up to 70% of individuals with ARDS, and when present, converts the syndrome to its most severe and mortal form. In addition, when systemic injury coexists with any type of infection, ARDS will develop with an increased frequency. Nosocomial pneumonia results when upper and lower respiratory tract defenses fail and these sites are overwhelmed

From Microbiota Correction to Host Protection: A New Therapeutic Target for the Prevention and Treatment of Postoperative Complications.

Background/Objectives. The intestinal microbiota is a key contributor to postoperative complications, yet direct interventions targeting dysbiosis-antibiotics, probiotics, and synbiotics-have produced inconsistent results. This paradox indicates a fundamental gap in understanding host-microbiota interactions under surgical stress. We aimed to re-examine the causal role of dysbiosis in postoperative pathogenesis and propose a revised therapeutic paradigm centered on host barrier protection. Methods. A narrative literature review was conducted, searching PubMed/MEDLINE, Scopus, and Web of Scienc

Context-dependent roles of the NLRP3 inflammasome in osteomyelitis and strategies for precision modulation.

Osteomyelitis is an inflammatory bone disease caused by pathogen infection, characterized by persistent infection, bone destruction, and impaired repair. Its refractoriness stems from the interplay between pathogen persistence and dysregulated host immune responses within the unique bone microenvironment. As a critical sensor of innate immunity, the NLRP3 inflammasome integrates pathogen-associated molecular patterns (PAMPs) and damage-associated molecular patterns (DAMPs), playing a pivotal regulatory role in the initiation and chronicity of osteomyelitis through caspase-1 activation, maturat

Adjunctive Therapies in Periodontitis: Current Concepts and the Future.

Periodontitis is a chronic, host-mediated inflammatory disease in which microbial dysbiosis and dysregulated immune responses drive the destruction of tooth-supporting tissues. Although conventional therapy remains centered on mechanical biofilm control, persistent inflammation and alveolar bone loss in susceptible individuals underscore the need for adjunctive strategies. Translating preclinical discoveries into predictable clinical outcomes, however, remains a major challenge in periodontal research. This narrative review integrates two interrelated themes, translational research methodology

COVID-19 and metabolic health: Lessons from a global pandemic.

The COVID-19 pandemic has highlighted the critical role of metabolic health in shaping susceptibility to infection, disease severity, therapeutic response, and vaccine-induced immunity. Accumulating clinical and experimental evidence has shown a bidirectional relationship between SARS-CoV-2 infection and metabolic dysfunction, whereby pre-existing cardiometabolic disorders exacerbate clinical outcomes, while COVID-19 itself induces acute and persistent metabolic derangements. This Special Issue explores COVID-19 through an immune-metabolic lens, integrating mechanistic insights, clinical persp

Extracellular Vesicles from Lactic Acid Bacteria: Emerging Postbiotic Effectors in Host-Microbe Crosstalk.

This article addresses extracellular vesicles derived from lactic acid bacteria (LAB-EVs), which have become fundamental mediators of host-microbe communication, yielding new insights about the molecular mechanisms underlying probiotic and postbiotic activity. The review integrates current evidence on the biogenesis, structural characteristics, and functional roles of LAB-EVs, stressing their capacity to transport bioactive cargos - including proteins, nucleic acids, and metabolites - to host cells. We highlight how LAB-EVs interact with the intestinal epithelial barrier, modulate innate and a

Current understanding and advances regarding the adipose-immune-metabolic axis in disease tolerance during sepsis.

Sepsis remains a critical global health challenge characterized by high mortality and morbidity, primarily due to the limitations of current pathogen-centric therapies and a poor understanding of host-defense mechanisms. This review synthesizes the pivotal role of the adipose-immune-metabolic axis as a central regulator of disease tolerance-a host defense strategy that limits tissue damage without directly reducing pathogen load. We delineate how adipose tissue is reprogrammed from a passive energy reservoir into an active immunometabolic hub during sepsis. This functional shift is governed by

Bifidobacteria and Clostridioides difficile infection: review of emerging therapeutic perspectives.

Clostridioides difficile infection (CDI) remains a major cause of antibiotic-associated diarrhea and colitis, with a high rate of recurrence. Current CDI treatments rely largely on antibiotics that perpetuate dysbiosis or on microbiota-based interventions with limited accessibility, emphasizing the need to explore novel, targeted microbiota-derived therapies. Probiotics have emerged as a potential approach for the prevention and management of CDI, owing to their capacity to modulate the gut microbiota and host immune responses. Among them, bifidobacteria are prominent commensal anaerobes of th

Research advances in early diagnosis and treatment strategies for peri-implantitis: from microecology to regenerative therapy.

Peri-implantitis, a plaque-associated inflammatory disease characterized by progressive bone loss around functional implants, poses a significant threat to the long-term success of oral rehabilitation. The understanding of its etiology has evolved from a traditional infection model to a dysbiosis-based host-microbe interaction disorder model. This paradigm shift underscores the critical need for early detection and novel therapeutic strategies. This review comprehensively summarizes recent research advances within a logical framework: microbial dysbiosis, early diagnosis, treatment evolution,

Cellular and molecular basis of PfCoronin function in artemisinin resistance in Plasmodium falciparum.

Artemisinin combination therapies are central to malaria control, but their efficacy is threatened by the emergence of resistant Plasmodium falciparum strains. While the role of Pfkelch13 mutations is well established, mutations in Pfcoronin have also been implicated in treatment failure, revealing layers of resistance complexity. Here, we define the cellular mechanism of Pfcoronin‑mediated artemisinin resistance. PfCoronin interacts with PfActin and localizes to the parasite plasma membrane, the digestive vacuole membrane, and small vesicles containing host cell material. Mutations in PfCoron

The Multifaceted Role of T-Helper Responses in Host Defense against Aspergillus fumigatus.

The ubiquitous opportunistic fungal pathogen Aspergillus fumigatus rarely causes infections in immunocompetent individuals. A healthy functional innate immune system plays a crucial role in preventing Aspergillus-infection. This pivotal role for the innate immune system makes it a main research focus in studying the pathogenesis of aspergillosis. Although sometimes overshadowed by the innate immune response, the adaptive immune response, and in particular T-helper responses, also represents a key player in host defense against Aspergillus. Virtually all T-helper subsets have been described to
