Noninvasive assessment of hypoxia with 3-[18F]-fluoro-1-(2-nitro-1-imidazolyl)-2-propanol ([18F]-FMISO): a PET study in two experimental models of human glioma.

Despite multiple advances in cancer therapies, patients with glioblastoma (GBM) still have a poor prognosis. Numerous glioma models are used not only for the development of innovative therapies but also to optimize conventional ones. Given the significance of hypoxia in drug and radiation resistance and that hypoxia is widely observed among GBM, the establishment of a reliable method to map hypoxia in preclinical human models may contribute to the discovery and translation of future and more targeted therapies. The aim of this study was to compare the hypoxic status of two commonly used human

Improved methodology to obtain large quantities of correctly folded recombinant N-terminal extracellular domain of the human muscle acetylcholine receptor for inducing experimental autoimmune myasthenia gravis in rats.

BACKGROUND: Adolescents possess distinct physiological characteristics, including immature skeletal and neuromuscular development, which necessitate training modalities different from those designed for adults. Conventional training methods often present limitations in both efficiency and safety for this population. OBJECTIVES: This study aimed to quantify the effects of sprint interval training (SIT) on the physical fitness of adolescents, thereby providing evidence to support the optimization of training regimens for this demographic. METHODS: PubMed, Web of Science, MEDLINE, and the Cochr

[Further research on the role of prostanoids in controlling renal function in humans in normal potassium balance and acute experimental potassium depletion. I: Studies of normal potassium balance. Effects of indomethacin].

The renal function was studied by clearance (cl.) method during hypotonic polyuria (oral water load followed by 5% dextrose solution infusion) and successive relative antidiuresis induced by lysine-8-vasopressin (LVP) administration (5 microU in bolo followed by continuous infusion at a rate of 0.04 microU/min). Four 15 min and two 60 min clearance (cl.) periods were performed during hypotonic polyuria and antidiuresis, respectively. Glomerular filtration rate was estimated by creatinine cl.; the osmotic cl. (Cosm, CH2O), the absolute and fractional excretions of water, sodium, potassium and c

Experimental and field studies on the viability of eggs, and the infectivity of third-stage larvae of the human hookworm Necator americanus recovered from the creek waters of the Niger Delta, Nigeria, and their epidemiological significance.

In a combined field and laboratory study, the eggs and 3rd-stage infective larvae of the human hookworm Necator americanus were recovered from the creek waters of the Niger Delta, Nigeria. They were tested for viability and infectivity. Eggs recovered from fresh water creeks showed a viability or hatching index of between 17.5 and 23.7%. Eggs recovered from brackish water showed a viability index of 18.8-21.4% in 'normal' cultures, and a maximum index of 6.3% when cultured in brackish water. The differences in the viability indices of these eggs were not significant (P greater than 0.05). Infe

Martin Salgo and the Dawn of Patient Autonomy.

On January 8, 1954, Martin Salgo underwent a translumbar aortogram for a presumptive diagnosis of mid-aortic occlusion. Postprocedure paraplegia occurred, and Mr. Salgo filed a medical malpractice lawsuit claiming, among other causes of action, negligence, breach of a duty to warn, and unauthorized human experimentation. The trial jury initially awarded the plaintiff $250,000. On appeal, Judge Absalom F. Bray reversed the verdict, noting errors of law, specifically, an incorrect theory of negligence. The judge's opinion highlighted two other important elements of patient autonomy and most famo…

Development and characterization of novel T-cell receptor-like antibodies targeting human papillomavirus type 16 oncoprotein peptides for early cervical cancer detection using experimental and computational approaches.

Cervical cancer remains one of the most prevalent malignancies among women worldwide, with persistent infection by high-risk human papillomavirus (HPV), particularly types 16 and 18, as the principal etiological factor. The viral oncoproteins E6 and E7 are well-established drivers of cellular transformation and the maintenance of malignant phenotypes. This study aimed to generate T-cell receptor-like antibodies (TCR-like antibodies) capable of recognizing HPV16-derived E6 and E7 peptides presented by the major histocompatibility complex class I molecule HLA-A2402, with the goal of developing a

Human Cord Blood Derived Unrestricted Somatic Stem Cells Restore Aquaporin Channel Expression, Reduce Inflammation and Inhibit the Development of Hydrocephalus After Experimentally Induced Perinatal Intraventricular Hemorrhage.

Intraventricular hemorrhage (IVH) is a severe complication of preterm birth associated with cerebral palsy, intellectual disability, and commonly, accumulation of cerebrospinal fluid (CSF). Histologically, IVH leads to subependymal gliosis, fibrosis, and disruption of the ependymal wall. Importantly, expression of aquaporin channels 1 and 4 (AQP1 and AQP4) regulating respectively, secretion and absorption of cerebrospinal fluids is altered with IVH and are associated with development of post hemorrhagic hydrocephalus. Human cord blood derived unrestricted somatic stem cells (USSCs), which we p

Genetically modified NT2N human neuronal cells mediate long-term gene expression as CNS grafts in vivo and improve functional cognitive outcome following experimental traumatic brain injury.

Human Ntera-2 (NT2) cells can be differentiated in vitro into well-characterized populations of NT2N neurons that engraft and mature when transplanted into the adult CNS of rodents and humans. They have shown promise as treatments for neurologic disease, trauma, and ischemic stroke. Although these features suggest that NT2N neurons would be an excellent platform for ex vivo gene therapy in the CNS, stable gene expression has been surprisingly difficult to achieve in these cells. In this report we demonstrate stable, efficient, and nontoxic gene transfer into undifferentiated NT2 cells using a

A toxicokinetic model for predicting the tissue distribution and elimination of organic and inorganic mercury following exposure to methyl mercury in animals and humans. I. Development and validation of the model using experimental data in rats.

The objective of this study was to develop a biologically based dynamic model for predicting the distribution and elimination of methyl mercury and its metabolite, inorganic mercury, under a variety of exposure scenarios in rats. A model is proposed based on a multicompartment approach; each compartment represents an organ or a group of organs or an excreta. The model translates into a set of coupled differential equations taking into account interorgan rates of exchanges and excretion together with the biotransformation process. The free parameters of the model are determined from statistical

In Vitro Gene Therapy Using Human iPS-Derived Mesoangioblast-Like Cells (HIDEMs) Combined with Microdystrophin (μDys) Expression as the New Strategy for Duchenne Muscular Dystrophy (DMD) Experimental Treatment.

Duchenne Muscular Dystrophy (DMD) is a genetic disorder characterized by disruptions in the dystrophin gene. This study aims to investigate potential a therapeutic approach using genetically modified human iPS-derived mesoangioblast-like cells (HIDEMs) in mdx mouse model. This study utilizes patient-specific myoblasts reprogrammed to human induced pluripotent stem cells (iPSCs) and then differentiated into HIDEMs. Lentiviral vectors carrying microdystrophin sequences have been employed to deliver the genetic construct to express a shortened, functional dystrophin protein in HIDEMs. The study i

A New Hemodynamic Ex Vivo Model for Medical Devices Assessment.

BACKGROUND: In-stent restenosis (ISR) remains a major public health concern associated with an increased morbidity, mortality, and health-related costs. Drug-eluting stents (DES) have reduced ISR, but generate healing-related issues or hypersensitivity reactions, leading to an increased risk of late acute stent thrombosis. Assessments of new DES are based on animal models or in vitro release systems, which have several limitations. The role of flow and shear stress on endothelial cell and ISR has also been emphasized. The aim of this work was to design and first evaluate an original bioreactor

Isolation of live human δ cells for genetic and functional analysis.

Investigations of human pancreatic islets have been empowered by strategies to isolate and study live islet cell subsets, like β cells and α cells. To advance experimentation with human islet δ cells, which remain relatively understudied, we generated combinatorial cell sorting approaches to separate human δ cells from β cells, yielding highly-enriched human δ cells. We used molecular analysis, immunohistology, and electroporation-based targeting to demonstrate the quality of δ cell purification. We also demonstrated the feasibility of prospectively studying human δ cell function in pseudoisle

Audiogenic startle reflex of man and its relationship to startle syndromes. A review.

The normal human audiogenic startle reflex is defined from a review of the literature, substantiated by a current investigation of normal subjects, and clarified pathophysiologically by a discussion of animal experimentation. The basic information is used to evaluate critically a variety of syndromes loosely termed 'startle' in the past. A new classification of startle syndromes is proposed.

Long-term blood-free rearing of Anopheles mosquitoes with no effect on fitness, Plasmodium infectivity nor microbiota composition.

Mosquito-borne diseases kill millions of people each year. Therefore, many innovative research and population control strategies are being implemented but, most of them require large-scale production of mosquitoes. Mosquito rearing depends on fresh blood from human donors, experimentation animals or slaughterhouses, which constitutes a strong drawback since high blood quantities are needed, raising ethical and financial constraints. To eliminate blood dependency and the use of experimentation animals, we previously developed BLOODless, a patented diet that represents an important advance towar

The response of human nasal and bronchial organotypic tissue cultures to repeated whole cigarette smoke exposure.

Exposure to cigarette smoke (CS) is linked to the development of respiratory diseases, and there is a need to understand the mechanisms whereby CS causes damage. Although animal models have provided valuable insights into smoking-related respiratory tract damage, modern toxicity testing calls for reliable in vitro models as alternatives for animal experimentation. We report on a repeated whole mainstream CS exposure of nasal and bronchial organotypic tissue cultures that mimic the morphological, physiological, and molecular attributes of the human respiratory tract. Despite the similar cellula

Evaluation of Gas Exchange and Hemocompatibility of an Experimental Oxygenator at Anticipated Human Fetal Flow Rates in Fetal Lambs Using a Dual Oxygenator Platform in the EXTra-Uterine Environment for Neonatal Development (EXTEND) System.

BACKGROUND: We previously reported the EXTEND (EXTra-uterine Environment for Neonatal Development) system provides physiologic support of fetal lambs (105-111 days gestational age; GA) via a pumpless arteriovenous circuit using a commercially available oxygenator (Maquet-Ox). However, for anticipated human subjects at 23-28 weeks of GA, a smaller oxygenator is required. To meet this requirement, a proprietary oxygenator (Experimental Oxygenator: Exp-Ox) was developed. METHODS: Fourteen lambs (mean GA 99 days, mean weight at cannulation 1.36 kg) were placed on the EXTEND system. Thereafter, Ex

Network Pharmacology Combined with Molecular Docking, Molecular Dynamics, and In Vitro Experimental Validation Reveals the Therapeutic Potential of Thymus vulgaris L. Essential Oil (Thyme Oil) against Human Breast Cancer.

Breast cancer is a major global health issue for women. Thyme oil, extracted from Thymus vulgaris L., has shown promising anticancer effects. In the present study, we investigated how Thyme oil can influence breast cancer treatment using a multimethod approach. We used network pharmacology to identify the active compounds of Thyme oil, their molecular targets, and the pathways involved in breast cancer. We found that Thyme oil can modulate several key proteins (EGFR, AKT1, ESR1, HSP90AA1, STAT-3, SRC, IL-6, HIF1A, JUN, and BCL2) and pathways (EGFR tyrosine kinase inhibitor resistance, prolacti
