Multidimensional mechanisms of SFTSV-host interactions: viral protein functions, m6A modification, tick saliva factors, and platelet regulation.

Severe fever with thrombocytopenia syndrome virus (SFTSV), officially designated as Dabie bandavirus by the International Committee on Taxonomy of Viruses (ICTV), is an emerging zoonotic pathogen belonging to the order Bunyavirales and the family Phenuiviridae. First isolated and identified in 2010 from the serum of patients with severe fever with thrombocytopenia syndrome (SFTS) in China, SFTSV has since been reported in multiple provinces across China, as well as in South Korea, Japan, Vietnam, the United Arab Emirates (including Dubai), and Pakistan. With a case fatality rate of up to 30%,

Human serum albumin-based cryogels as a biomimetic platform for 3D co-culture and drug-induced liver injury prediction.

Nonclinical toxicity testing in rodent and nonrodent models often fails to predict human pharmacological responses, and the reliance on poorly characterized biomaterials hinders clinical translation in drug development. To address these limitations, we engineered a human serum albumin methacryloyl (HSAMA) cryogel system incorporating key liver extracellular matrix (ECM) components, collagen I (C) and fibronectin (F), to mimic the liver's architectural complexity. By co-culturing HepG2 and HUVECs within the HSAMA-CF cryogel, we created a three-dimensional, large-pore scaffold that facilitates c

Molecular detection of Brucella species among aborted small ruminants in southeast Iran.

Brucellosis, caused by Brucella bacteria, is a common zoonotic infectious disease with various clinical manifestations in humans and animals. The disease is endemic in human and ruminant populations in Iran, with a particular prevalence in areas where humans have close interactions with livestock. Since domestic animals serve as the primary reservoir for brucellosis, this study aimed to identify the presence of Brucella spp. among aborted small ruminants in southeast Iran. Between 2021 and 2022, aborted fetuses of small ruminants (46 sheep and 4 goats) were collected from Zarand County in the

In vivo phage display to identify new human antibody fragments homing to atherosclerotic endothelial and subendothelial tissues [corrected].

In vivo phage display selection is a powerful strategy for directly identifying agents that target the vasculature of normal or diseased tissues in living animals. We describe here a new in vivo biopanning strategy in which a human phage single-chain antibody (scFv) library was injected into high-fat diet-fed ApoE(-/-) mice. Extracellular and internalized phage scFvs were selectively recovered from atherosclerotic vascular endothelium and subjacent tissues. After three successive biopanning rounds, a panel of six clones with distinct gene sequences was isolated. Four scFvs produced and purifie

Protocols and applications of cellular metabolomics in safety studies using precision-cut tissue slices and carbon 13 NMR.

Numerous xenobiotics are toxic to human and animal cells by interacting with their metabolism, but the precise metabolic step affected and the biochemical mechanism behind such a toxicity often remain unknown. In an attempt to reduce the ignorance in this field, we have developed a new approach called cellular metabolomics. This approach, developed in vitro, provides a panoramic view not only of the pathways involved in the metabolism of physiologic substrates of any normal or pathologic human or animal cell but also of the beneficial and adverse effects of xenobiotics on these metabolic pathw

The endocrine effects of mercury in humans and wildlife.

Mercury (Hg) is well studied and research continues as our knowledge of its health risks increases. One expanding area of research not well emphasized to date is the endocrine effects of Hg. This review summarizes the existing literature on the effects of Hg on the endocrine system and identifies gaps in the knowledge. It focuses on the thyroid, adrenal, and reproductive systems, including the accumulation of Hg in the endocrine system, sex differences that are manifested with Hg exposure, reproductive effects in male and female animals including humans, and Hg effects on the thyroid and adren

Evaluation of quinacrine treatment for prion diseases.

Based on in vitro observations in scrapie-infected neuroblastoma cells, quinacrine has recently been proposed as a treatment for Creutzfeldt-Jakob disease (CJD), including a new variant CJD which is linked to contamination of food by the bovine spongiform encephalopathy (BSE) agent. The present study investigated possible mechanisms of action of quinacrine on prions. The ability of quinacrine to interact with and to reduce the protease resistance of PrP peptide aggregates and PrPres of human and animal origin were analyzed, together with its ability to inhibit the in vitro conversion of the no

Skin sensitization thresholds: determination in predictive models.

For many years, test methods for the prospective identification of skin sensitizing chemicals have been widely available. However, although these techniques have permitted the identification of the great majority of skin sensitizers, their use in assessing the relative potency of a particular chemical as a human contact allergen has not been well described. A primary reason for this is the inherent difficulty of such an exercise. A complex phenomenon involving interactions between the vehicle, the allergen, the skin and its inflammatory responses takes place during the induction and elicitatio

Neuroimmune interactions in the development and chronification of migraine headache.

Migraine is highly prevalent and debilitating. The persistent headaches in this condition are thought to arise from the activation and sensitization of the trigeminovascular pathway. Both clinical and animal model studies have suggested that neuroimmune interactions contribute to the pathophysiology of migraine headache. In this review, we first summarize the findings from human studies implicating the dysregulation of the immune system in migraine, including genetic analyses, measurement of circulatory factors, and neuroimaging data. We next discuss recent advances from rodent studies aimed a

Three-dimensional cell culture models for investigating human viruses.

Three-dimensional (3D) culture models are physiologically relevant, as they provide reproducible results, experimental flexibility and can be adapted for high-throughput experiments. Moreover, these models bridge the gap between traditional two-dimensional (2D) monolayer cultures and animal models. 3D culture systems have significantly advanced basic cell science and tissue engineering, especially in the fields of cell biology and physiology, stem cell research, regenerative medicine, cancer research, drug discovery, and gene and protein expression studies. In addition, 3D models can provide u

Rabies virus assembly and budding.

Rabies virus (RABV) and other negative-strand RNA viruses are the causes of serious diseases in humans and animals worldwide. Assembly and budding are important late events in the replication cycles of these negative-strand RNA viruses that have received much attention in the past decade. Indeed, important insights into the molecular mechanisms by which rhabdoviral proteins usurp and/or interact with host proteins to promote efficient virion assembly and egress has greatly enhanced our understanding of the budding process. Assembly/budding of rhabdoviruses is driven largely by the matrix (M) p

The impact of common micronutrient deficiencies on iodine and thyroid metabolism: the evidence from human studies.

Deficiencies of micronutrients are highly prevalent in low-income countries. Inadequate intake of iodine impairs thyroid function and results in a spectrum of disorders. Other common deficiencies of micronutrients such as iron, selenium, vitamin A, and possibly zinc may interact with iodine nutrition and thyroid function. Randomised controlled intervention trials in iodine- and iron-deficient populations have shown that providing iron along with iodine results in greater improvements in thyroid function and volume than providing iodine alone. Vitamin A supplementation given alone or in combina

Coinfection with Plasmodium falciparum and schistosoma haematobium: protective effect of schistosomiasis on malaria in senegalese children?

Studies with animal models have suggested the possibility of interactions between parasites during concurrent infections and have raised the question of a similar phenomenon in humans. The present survey was undertaken to assess the impact of urinary schistosomiasis on the susceptibility of children to malaria. It was carried out in Senegal between September 2001 and March 2002 among 523 children 3-15 years of age. We tested the association between Plasmodium falciparum densities and the load of Schistosoma haematobium egg excretion using a linear mixed model because data were not independent.

Gastrointestinal parasites of wild carnivores from conservation institutions in the Cerrado of Goiás, Brazil.

Increased interaction between wild and urban environments owing to human population growth, increased anthropization of biomes, and habitat loss for wild animals increases the spread of infectious and parasitic agents. The present study reports on the occurrence of gastrointestinal parasites in carnivorous mammals at two conservation institutions in the state of Goiás, Brazil. Fecal samples from 39 adult carnivores were collected after spontaneous defecation and analyzed by flotation and sedimentation. The structure and management data of each institution were recorded. Parasitism prevalence,

A Review of Pseudorabies Virus Variants: Genomics, Vaccination, Transmission, and Zoonotic Potential.

Pseudorabies virus (PRV), the causative agent of Aujeszky's disease, has a broad host range including most mammals and avian species. In 2011, a PRV variant emerged in many Bartha K61-vaccinated pig herds in China and has attracted more and more attention due to its serious threat to domestic and wild animals, and even human beings. The PRV variant has been spreading in China for more than 10 years, and considerable research progresses about its molecular biology, pathogenesis, transmission, and host-virus interactions have been made. This review is mainly organized into four sections includin

Methods in causal inference. Part 2: Interaction, mediation, and time-varying treatments.

The analysis of 'moderation', 'interaction', 'mediation' and 'longitudinal growth' is widespread in the human sciences, yet subject to confusion. To clarify these concepts, it is essential to state causal estimands, which requires the specification of counterfactual contrasts for a target population on an appropriate scale. Once causal estimands are defined, we must consider their identification. I employ causal directed acyclic graphs and single world intervention graphs to elucidate identification workflows. I show that when multiple treatments exist, common methods for statistical inference

Diet-microbiota crosstalk and immunity to helminth infection.

Helminths are large multicellular parasites responsible for widespread chronic disease in humans and animals. Intestinal helminths live in close proximity with the host gut microbiota and mucosal immune network, resulting in reciprocal interactions that closely influence the course of infections. Diet composition may strongly regulate gut microbiota composition and intestinal immune function and therefore may play a key role in modulating anti-helminth immune responses. Characterizing the multitude of interactions that exist between different dietary components (e.g., dietary fibres), immune c

Estradiol shapes mutualistic behaviour of female cleaner fish (Labroides dimidiatus - Valenciennes, 1839): Potential implications of environmental disturbance.

The presence of endocrine-derived compounds in the environment occurs due to a myriad of human or industrial activity and can disrupt the endocrine system of animals, including fish. One important group of endocrine disruptors are the estrogens, such as 17-β estradiol (E2, estradiol). Estrogens are gonadal steroid hormones, able to be influential even in small concentrations. Here, we demonstrate that E2 is linked to female' decisions made by an important coral reef species, the cleaner fish Labroides dimidiatus, during interactions with other reef fishes (known as clients). E2 treatment in na

Gradual adaptation of animal influenza A viruses to human-type sialic acid receptors.

Influenza A viruses (IAVs) originating from animal reservoirs pose continuous threats to human health as demonstrated by the Spanish flu pandemic. Infection starts by attachment to host receptors, a crucial step that is targeted by immunological, prophylactic, and therapeutic intervention. Fine-tuning of virus hemagglutinin binding to host-specific receptor repertoires needs to remain balanced to receptor-destroying neuraminidase (NA) activity and is a key step in host adaptation. It determines NA-dependent virus motility, enabling IAVs to traverse the mucus layer and to bind to, and migrate o

Malassezia: A Commensal, Pathogen, and Mutualist of Human and Animal Skin.

Identified in the late nineteenth century as a single species residing on human skin, Malassezia is now recognized as a diverse genus comprising 18 species inhabiting not only skin but human gut, hospital environments, and even deep-sea sponges. All cultivated Malassezia species are lipid dependent, having lost genes for lipid synthesis and carbohydrate metabolism. The surging interest in Malassezia results from development of tools to improve sampling, culture, identification, and genetic engineering, which has led to findings implicating it in numerous skin diseases, Crohn disease, and pancr
