How Children Are Protected From COVID-19? A Historical, Clinical, and Pathophysiological Approach to Address COVID-19 Susceptibility.

The origin and the global spread of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) causing coronavirus disease 2019 (COVID-19) in early 2020 was accompanied by high rates of mortality in regions belonging to the ancient silk road, such as the south of China, Iran, Turkey and the northern parts of Italy. However, children seem to be spared in the epidemic as very small percentage worldwide being ill. The protection of children and neonates suggests the involvement of a specific component of adaptive immunity present at early development. Native immunoglobulin belonging to the clas

Hemophagocytic lymphohistiocytosis: updates and evolving concepts.

PURPOSE OF REVIEW: Hemophagocytic lymphohistiocytosis (HLH) is an immune dysregulatory syndrome that is associated with underlying defects of perforin-dependent cytotoxic function. This review seeks to update readers on new scientific insights and evolving clinical concepts related to this rare but fatal disorder. RECENT FINDINGS: Clinically, HLH is defined by severe inflammation and potentially fatal damage to a variety of organ systems including the bone marrow, liver, or brain. Recent preclinical studies have increasingly defined HLH as a syndrome of abnormal and excessive T-cell activatio

Extracellular cGAMP in health and disease.

The cGAS-STING signaling pathway is a crucial component of the innate immune system that detects aberrant cytosolic DNA, such as that derived from viruses or damaged cells, to activate downstream immune responses. Within this pathway, cyclic guanosine monophosphate-adenosine monophosphate (cGAMP) serves as the essential second messenger linking DNA sensing to immune activation. Upon recognition of cytosolic DNA, cGAS synthesizes cGAMP, whose unique "mixed linkage" structure enables efficient binding to and activation of the STING protein on the endoplasmic reticulum, thereby inducing type I in

[Research progress on the impact and formation mechanism of vascular endothelial cells in cutaneous tuberculosis].

Cutaneous Tuberculosis (CTB) is a dermatological manifestation caused by Mycobacterium tuberculosis (Mtb). Studies have shown that various immune cells are involved in its pathogenesis. Early perspectives suggested that Mtb primarily invades innate immune cells, but recent research has found that adaptive immune cells also play a crucial role during the infection process and significantly influence changes at the inflammation site. As the main component of the vascular endothelium, endothelial cells (ECs) can secrete various inflammatory mediators and chemokines during Mtb infection, recruitin

Role of the cGAS-STING pathway in radiotherapy for non-small cell lung cancer.

One of the most important therapeutic interventions for non-small cell lung cancer is radiotherapy. Ionizing radiation (IR) is classified by traditional radiobiology principles as a direct cytocidal therapeutic agent against cancer, although there is growing recognition of other antitumor immunological responses induced by this modality. The most effective therapeutic combinations to harness radiation-generated antitumor immunity and enhance treatment results for malignancies resistant to existing radiotherapy regimens could be determined by a more sophisticated understanding of the immunologi

Palbociclib and dsRNA sensor co-operate to enhance anti-cancer effects through ER stress and modulation of immune evasion.

Cytoplasmic pattern recognition receptors (PRR) for double-stranded RNA, such as RIG-I/MDA5, are key mediators of anti-viral responses. Here we screen for synergistic drug-virotherapy combinations and find that the reovirus type III Dearing strain (Rt3D)-palbociclib combination augments oncolytic virus-induced stress responses and increases interferon production and signaling. Data from RIG-I agonist and ER stress-inducing agents further confirms the crosstalk between RNA-sensing and ER stress in inducing cancer cell death and interferon production. Combined Rt3D-palbociclib also increases inn

Current concepts in maternal-fetal immunology: Recognition and response to microbial pathogens by decidual stromal cells.

Chorioamnionitis is an acute inflammation of the gestational (extraplacental) membranes, most commonly caused by ascending microbial infection. It is associated with adverse neonatal outcomes including preterm birth, neonatal sepsis, and cerebral palsy. The decidua is the outermost layer of the gestational membranes and is likely an important initial site of contact with microbes during ascending infection. However, little is known about how decidual stromal cells (DSCs) respond to microbial threat. Defining the contributions of individual cell types to the complex medley of inflammatory signa

Woodchuck and deer Hepatitis Delta-like agents show distinct innate immune activation and IFN-resistance compared to human Hepatitis D Virus.

Replication of human hepatitis delta virus (HDV) is sensed by the pattern recognition receptors (PRRs) melanoma differentiation antigen 5 (MDA5) and Laboratory of Genetics and Physiology 2 (LGP2), activating interferon (IFN) responses. Both HDV-induced IFNs and exogenously administered IFN-α and -λ suppress HDV RNA spread via cell division (CDMS) but have only minor effects on replication in resting cells. With the discovery of HDV-like agents (DLAs) in diverse animal species an opportunity arose to comparatively study the interplay between these evolutionarily related replicons with respect t

Cell type-dependent induction of type I interferon and PARP1 activation in astrocytes and neurons during chikungunya virus infection.

Chikungunya virus, a mosquito-borne alphavirus, causes fever, rash, arthritis, and neurological disorders. Its non-structural protein 3 harbors a macrodomain, a key neurovirulence factor that removes adenosine diphosphate ribose from ADP-ribosylated substrates. Notably, chikungunya virus infection results in distinct ADP-ribosylation patterns and non-structural protein 3 macrodomain-mediated replication dynamics in astrocytes and neurons. Understanding the connection between ADP-ribosylation and the activation of innate immunity, particularly interferon release, is key to elucidating how the c

A subgroup of lupus patients with nephritis, innate T cell activation and low vitamin D is identified by the enhancement of circulating MHC class I-related chain A.

The major histocompatibility complex (MHC) class I-related chain A (MICA) is induced upon stress, and labels malfunctioning cells for their recognition by cytotoxic lymphocytes. Alterations in this recognition and also abnormal natural killer (NK) functions have been found in systemic lupus erythematosus (SLE). MICA can be shed from cells, subsequently acting as a soluble decoy receptor (sMICA). Our purpose was to study circulating sMICA levels in relationship with the activation of innate pathways in PBMC in a cohort of lupus patients. NK cells were characterized by flow cytometry. Sequence-structure-function relations of the mosquito leucine-rich repeat immune proteins.

BACKGROUND: The discovery and characterisation of factors governing innate immune responses in insects has driven the elucidation of many immune system components in mammals and other organisms. Focusing on the immune system responses of the malaria mosquito, Anopheles gambiae, has uncovered an array of components and mechanisms involved in defence against pathogen infections. Two of these immune factors are LRIM1 and APL1C, which are leucine-rich repeat (LRR) containing proteins that activate complement-like defence responses against malaria parasites. In addition to their LRR domains, these

A novel immunomodulatory hemocyanin from the limpet Fissurella latimarginata promotes potent anti-tumor activity in melanoma.

Hemocyanins, the huge oxygen-transporting glycoproteins of some mollusks, are used as immunomodulatory proteins with proven anti-cancer properties. The biodiversity of hemocyanins has promoted interest in identifying new anti-cancer candidates with improved immunological properties. Hemocyanins promote Th1 responses without known side effects, which make them ideal for long-term sustained treatment of cancer. In this study, we evaluated a novel hemocyanin from the limpet/gastropod Fissurella latimarginata (FLH). This protein has the typical hollow, cylindrical structure of other known hemocyan

Innate immune signalling, neuroinflammation and network plasticity in temporal lobe epilepsy.

Temporal lobe epilepsy emerges from a cascade of molecular, cellular, and structural disturbances triggered by heterogeneous cerebral insults-including convulsive status epilepticus, viral encephalitis, traumatic brain injury, and blood-brain barrier disruption-that converge on progressive hippocampal reorganization and a chronic predisposition to unprovoked focal seizures. Convergent evidence from chemoconvulsant models, focal intrahippocampal kainate administration, viral encephalitis paradigms, organotypic hippocampal cultures, human iPSC-derived organoids, and resected human tissue shows t

Immunomodulatory Drugs: Immune Checkpoint Agents in Acute Leukemia.

Intrinsic immune responses to acute leukemia are inhibited by a variety of mechanisms, such as aberrant antigen expression by leukemia cells, secretion of immunosuppressive cytokines and expression of inhibitory enzymes in the tumor microenvironment, expansion of immunoregulatory cells, and activation of immune checkpoint pathways, all leading to T cell dysfunction and/or exhaustion. Leukemic cells, similar to other tumor cells, hijack these inhibitory pathways to evade immune recognition and destruction by cytotoxic T lymphocytes. Thus, blockade of immune checkpoints has emerged as a highly p

Trained immunity agonists given to neonatal mice fail to protect against leptospiral infection in adulthood and may cause adverse effects.

Background and ObjectivesTrained immunity (TI) refers to a non-specific, long-lasting protective immune response that occurs following initial stimulation of the immune system and thought to be largely mediated by functional reprogramming of myeloid cells. TI has been demonstrated in BCG-vaccinated infants and can be induced in human cells and adult mice via agonists of pattern recognition receptors (PRR), such as β-glucan or MDP (a muropeptide that activates NOD2). However, its induction in neonates remains poorly understood. Previously, we demonstrated that the synthetic TLR2-NOD2 dual agoni

An update on the microbiology, immunology and genetics of seborrheic dermatitis.

The underlying mechanism of seborrheic dermatitis (SD) is poorly understood but major scientific progress has been made in recent years related to microbiology, immunology and genetics. In light of this, the major goal of this article was to summarize the most recent articles on SD, specifically related to underlying pathophysiology. SD results from Malassezia hydrolysation of free fatty acids with activation of the immune system by the way of pattern recognition receptors, inflammasome, IL-1β and NF-kB. M. restricta and M. globosa are likely the most virulent subspecies, producing large quant

Sequential macrophage DENV and ZIKV infection shows differential expression of CD86, IFN-β, and regulation of TNF-α and IL-1β depending on DENV serotype.

Dengue virus (DENV) is an RNA virus belonging to the Flaviviridae family, comprising four antigenically distinct serotypes. Dengue is the primary arthropod-transmitted virus globally, posing a significant public health challenge, especially in Brazil, where the largest outbreak of Zika virus (ZIKV) was also recorded in 2016. ZIKV shares genomic and structural similarities with DENV, and their co-circulation in Brazil provides evidence of co-infection. The innate immune response against DENV and ZIKV is mediated by pattern recognition receptors that initiate intracellular signaling, leading to

Pattern recognition receptors and divergent disease severity in zoonotic viral infections.

Zoonotic viruses continue to emerge unpredictably, cross species barriers, and inflict major health, economic, and ecological burdens. Recent outbreaks of SARS-CoV-2, H5N1 avian influenza virus (AIV), monkeypox, and Zika virus underscore the unpredictability of viral emergence and the urgent need to understand host/pathogen interactions. This review focuses on how innate immune sensing via pattern recognition receptors (PRRs) shapes disease severity across different species. While PRRs initiate antiviral defenses (interferons, ISGs), inflammatory cytokines, and programmed cell death, host outc

Exploring the proteomic landscape of THP-1 monocytes through two-challenge LPS induction.

Proteome remodelling is central to the regulation of innate immune activation, yet the temporal organisation of protein networks engaged during repeated lipopolysaccharide (LPS) stimulation remains incompletely defined. In the present study, label-free quantitative mass spectrometry-based proteomics was used to characterise protein abundance changes in THP-1 monocytes at early (30 min) and later (2 h) time points following a second LPS challenge. This analysis was complemented by an independent co-immunoprecipitation proteomics experiment designed to identify candidate proteins associated with

RIPK3 Facilitates Host Resistance to Oral Toxoplasma gondii Infection.

Toxoplasma gondii infection activates pattern recognition receptor (PRR) pathways that drive innate inflammatory responses to control infection. Necroptosis is a proinflammatory cell death pathway apart from the innate immune response that has evolved to control pathogenic infection. In this study, we further defined the role of Z-DNA binding protein 1 (ZBP1) as a PRR and assessed its contribution to necroptosis as a host protection mechanism to T. gondii infection. We found that ZBP1 does not induce proinflammatory necroptosis cell death, and ZBP1 null mice have reduced survival after oral T.
