Reduced proinsecticide activation by cytochrome P450 confers coumaphos resistance in the major bee parasite Varroa destructor.

Varroa destructor is one of the main problems in modern beekeeping. Highly selective acaricides with low toxicity to bees are used internationally to control this mite. One of the key acaricides is the organophosphorus (OP) proinsecticide coumaphos, that becomes toxic after enzymatic activation inside Varroa We show here that mites from the island Andros (AN-CR) exhibit high levels of coumaphos resistance. Resistance is not mediated by decreased coumaphos uptake, target-site resistance, or increased detoxification. Reduced proinsecticide activation by a cytochrome P450 enzyme was the main resi

Two cases of severe falciparum malaria in KMCTH.

Malaria is the most important parasitic disease of man. It is the protozoan infection of RBCs transmitted by bite of blood feeding female anopheline mosquito. Until the 19th century malaria was found throughout Europe, North America and Russia. Since then, it has been eradicated from these areas but in tropics though initial efforts of eradication had been successful, there has been resurgence of disease accompanied by increasing resistance of the anopheline vector to insecticide and of the parasite to antimalarial drugs. We report two cases of falciparum malaria in which there was co-existent

Successes and failures of sixty years of vector control in French Guiana: what is the next step?

Since the 1940s, French Guiana has implemented vector control to contain or eliminate malaria, yellow fever, and, recently, dengue, chikungunya, and Zika. Over time, strategies have evolved depending on the location, efficacy of the methods, development of insecticide resistance, and advances in vector control techniques. This review summarises the history of vector control in French Guiana by reporting the records found in the private archives of the Institute Pasteur in French Guiana and those accessible in libraries worldwide. This publication highlights successes and failures in vector con…

Nematicidal and ovicidal activity of Bacillus thuringiensis against the zoonotic nematode Ancylostoma caninum.

Ancylostoma caninum is a gastrointestinal parasite that affect dogs and humans, considered a worldwide public health problem. The control of these parasitosis is increasingly difficult due to the development of multi-drug resistance. Bacillus thuringiensis is an insecticidal, spore forming bacterium, often species specific. The strain GP526 of B. thuringiensis has toxic effect on the cestode Dipylidium caninum and the trematode Centrocestus formosanus, both of them zoonotic parasites. The high degrees of specificity and environment safe make B. thuringiensis suitable for use against pathogen p

Crystal structures of angiotensin-converting enzyme from Anopheles gambiae in its native form and with a bound inhibitor.

The mosquitoes of the Anopheles and Aedes genus are some of the most deadly insects to humans because of their effectiveness as vectors of malaria and a range of arboviruses, including yellow fever, dengue, chikungunya, West Nile and Zika. The use of insecticides from different chemical classes is a key component of the integrated strategy against An. gambiae and Ae. aegypti, but the problem of insecticide resistance means that new compounds with different modes of action are urgently needed to replace chemicals that fail to control resistant mosquito populations. We have previously shown that…

Factors influencing the selection of Rhipicephalus microplus resistance to acaricides.

Rhipicephalus microplus is a hematophagous ecto-parasite of cattle that has a cosmopolitan distribution in tropical and subtropical regions of the world. This tick species causes economic losses by consuming blood and reducing meat and milk production, as well as facilitating the transmission of pathogens. Different classes of acaricides, including organophosphates, synthetic pyrethroids, insect growth regulators, formamidines, phenylpyrazoles, and macrocyclic lactones, are currently used sequentially or simultaneously to control R. microplus infestations. The frequent use of these acaricides

Acaricide resistance status of deltamethrin and coumaphos in Hyalomma anatolicum ticks collected from different districts of Haryana.

To study the acaricide resistance status and possible mechanisms of action in conferring resistance to commonly used acaricides (deltamethrin and coumaphos), Hyalomma anatolicum ticks were collected from 6 dairy farms of Hisar and Charkhi Dadri districts of Haryana. By using standard larval packet test, H. anatolicum tick larvae of Charkhi Dadri isolates were found to be susceptible (100% mortality) to both the acaricides. Level-I resistance against coumaphos was recorded from four isolates, whereas, level-II was observed in only one isolate, collected from Hisar. One isolates (Kaimri) from Hi

Susceptibility of the invasive malaria vector Anopheles stephensi in Ethiopia to novel chemical insecticides and insect growth regulator.

The presence of invasive Anopheles stephensi has been confirmed from field studies in several urban, peri-urban and rural areas of Ethiopia. Recent studies have shown that these mosquitoes are resistant to common public health insecticides, namely pyrethroids (deltamethrin, permethrin, and alpha-cypermethrin), Carbamates (bendiocarb and propoxur), and organophosphates (pirimiphos-methyl). This study aimed to assess the susceptibility of populations of An. stephensi from Ethiopia to novel chemical insecticides recently recommended for vector control, namely, broflanilide (a meta-diamide targeti

Challenges facing the management of pesticide resistance in weeds, diseases and insect pests in European agriculture and the future of effective IPM implementation.

In recent times, pesticide resistance has been managed reasonably effectively, either proactively or reactively, by monitoring resistance of pest biotypes and the rotation of products with different modes of action (MoAs). However, increased regulation is dramatically limiting the range of MoAs available to farmers, especially in Europe. Innovation and replenishment with new MoAs from industry cannot keep pace with this loss, leaving the need for pragmatic choices in how to manage pests effectively through all methods available. This is crucial for integrated pest management (IPM) adoption to

Beyond Anopheles gambiae sensu lato: exploring the impact of non-dominant Anopheles species on malaria persistence in high-transmission endemic areas of Burkina Faso.

BACKGROUND: Despite widespread implementation of malaria control measures, including insecticide-treated nets (ITNs), indoor residual spraying (IRS), and seasonal malaria chemoprevention (SMC), malaria remains a major public health concern in Burkina Faso. The persistence of transmission is often attributed to increasing insecticide resistance in Anopheles gambiae sensu lato and drug resistance in Plasmodium spp. However, additional factors, such as climatic variability, ecological change, and shifts in vector species composition, may also play a role. This study investigated the geographic di…

First Detection of the Kdr Mutation T929I in Head Lice (Phthiraptera: Pediculidae) in Schoolchildren of the Metropolitan Area of Nuevo Leon and Yucatan, Mexico.

The head louse Pediculus humanus capitis (De Geer) is a hematophagous ectoparasite that inhabits the human scalp. Infestations by this insect are commonly known as pediculosis, which is more common in younger groups. These infestations are asymptomatic; however, skin irritation from scratching occasionally may cause secondary bacterial infections. In recent years, the prevalence of pediculosis has increased in children; this increase has been attributed to louse resistance to the insecticides used as a control measure for infestation. The aim of the present study was to determine the presence

Larvicidal and Adulticidal Activity of Chroman and Chromene Analogues against Susceptible and Permethrin-Resistant Mosquito Strains.

Mosquitoes play a major role as vectors that transmit parasitic and viral diseases worldwide, especially in tropical and subtropical countries. Mosquito borne diseases not only affect humans but they also affect livestock in many parts of the world. They carry diseases that are lethal to dogs and horses. Dog heartworm disease (Dirofilaria immitis) is a parasitic disease spread through mosquitoes. This disease is not limited to dogs, but it can affect other animals and humans as well. Eastern equine encephalitis (EEE) and West Nile virus (WNV) are also mosquito borne diseases that affect the ce

Human pediculosis: a critical health problem and what about nursing policy?

Lice infestation on the human body (also known as pediculosis) is very common. Cases number in the hundreds of millions worldwide. Three distinct presentations of lice infection exist and each is caused by a unique parasite. Head lice (Pediculus humanus capitis) is by far and away the most common infestation and favors no particular socioeconomic group. A genetically close "cousin," Pediculus humanus corporis, is responsible for body lice and is more commonly associated with poverty, overcrowding, and poor hygiene. Pubic lice (crabs) are caused by Pthirus pubis and is transmitted by intimate a

Toxicity and kinetics of methoxyfenozide in greenhouse-selected Spodoptera exigua (Lepidoptera: Noctuidae).

Methoxyfenozide (RH-2485) was found to be 7.5-fold less toxic in terms of LD50 values against last-instar larvae of a greenhouse-selected strain of the beet armyworm Spodoptera exigua (Hübner) that was collected in July 2001 in an experimental greenhouse for resistance at Murcia in southern Spain, in comparison with a laboratory susceptible strain. To date, the compound is the newest member of this new group of moulting hormone accelerating IGRs to reach the marketplace against Lepidoptera. To understand this different potency in the greenhouse-selected S exigua, oxidative metabolism and acety

Permethrin resistance in the head louse Pediculus capitis from Israel.

Head lice, Pediculus capitis, were collected from children aged 3-12 years in Maale Adumin, a town near Jerusalem, after reports of control failure with the pyrethroid insecticide permethrin. A total of 1516 children were examined: living lice and eggs were found on 12.1% of the children; or another 22.8% of the children only nits were found. Twice as many girls as boys (8.1% v 4%) were infested with lice and or nits. Head lice collected from infested children were exposed to permethrin impregnated filter-papers. Log time probit mortality (ltp) regression lines were calculated for mortality da

The diagnosis, management, and prevention of common ectoparasitic infections.

Ectoparasitic diseases share many features in common with emerging infectious diseases, such as Lyme disease, including hyperendemic causative agents afforded selective advantages by changing ecological or socioeconomic conditions; origination as zoonoses or as highly communicable human-to-human diseases; transmission by competent vectors; and introduction into new, susceptible host populations. Many of these ectoparasites are now developing increasing resistances to medical therapies, including the safest insecticides and repellants.

Mortality of Musca domestica (Diptera: Muscidae) in response to exposure to a fungal entomopathogen in combination with eicosanoid biosynthesis inhibitors.

The house fly, Musca domestica L., is a vector of pathogens. When sanitation measures fail to control fly populations, pesticides are often used. Fungal biopesticides have been suggested as another tool in fly control. Our study investigated whether inhibitors of eicosanoid biosynthesis (EBI), and presumably of immunity, combined with the fungal biopesticide BotaniGard ES is a feasible option for a new biopesticide formulation. The EBIs tested were dexamethasone, naproxen, and esculetin. An insecticide-susceptible and an insecticide-resistant house fly strain were tested. Compared to a control
