Functional characterization of an IRF7 homologue in large yellow croaker (Larimichthys crocea).

Interferon regulatory factor 7 (IRF7) functions as a central orchestrator of type I interferon (IFN) responses in mammals, yet the functional mechanisms of its homologue in teleost remains incompletely understood. Here, we characterized an IRF7 homologue (LcIRF7) in large yellow croaker (Larimichthys crocea). The expression of LcIRF7 was strongly induced by poly (I:C), lipopolysaccharide (LPS), and recombinant type I IFN (rLcIFNi). These inductions were dependent on the JAK-STAT signaling pathway, suggesting that LcIRF7 is an interferon-stimulated gene (ISG). Overexpression of LcIRF7 significa

The ubiquitin E3 ligase TRIM10 promotes STING aggregation and activation in the Golgi apparatus.

STING is an endoplasmic reticulum-resident protein regulating innate immunity. After binding with cyclic guanosine monophosphate-AMP (cGAMP), STING translocates from the endoplasmic reticulum (ER) to the Golgi apparatus to stimulate TBK1 and IRF3 activation, leading to expression of type I interferon. However, the exact mechanism concerning STING activation remains largely enigmatic. Here, we identify tripartite motif 10 (TRIM10) as a positive regulator of STING signaling. TRIM10-deficient macrophages exhibit reduced type I interferon production upon double-stranded DNA (dsDNA) or cGAMP stimul

Psoriasis triggered by toll-like receptor 7 agonist imiquimod in the presence of dermal plasmacytoid dendritic cell precursors.

BACKGROUND: It has been proposed that the innate immune system plays a central role in driving the autoimmune T-cell cascade leading to psoriasis; however, there is no direct evidence for this. OBSERVATIONS: We observed aggravation and spreading of a psoriatic plaque when treated topically with the toll-like receptor (TLR) 7 agonist imiquimod. The exacerbation of psoriasis was accompanied by a massive induction of lesional type I interferon activity, detected by MxA expression after imiquimod therapy. Since imiquimod induces large amounts of type I interferon production from TLR7-expressing p

CFTR modulators partially restore the epithelial interferome in Aspergillus infection to improve clinical outcome.

BACKGROUND: The impact of CFTR modulator therapy on host immunity and outcomes in people with Cystic Fibrosis (CF)-related Aspergillus lung disease is poorly defined. We aimed to characterise fungal-relevant clinical outcomes post-CFTR modulators and assess effects on the Aspergillus-dependent Type I/III interferome. METHODS: Biomarkers of Aspergillus-related lung disease (Aspergillus-specific IgE/IgG), anti-fungal and corticosteroid therapy were analysed in a retrospective cohort of people with CF pre and post Elexacaftor/Tezacaftor/Ivacaftor (ETI) modulator therapy. Homozygous F508del (CF)

Specific binding of interferon-gamma and -alpha 2a to tumor cells and their antitumor activities.

Binding activities of a recombinant human interferon-gamma (IFN-gamma) preparation with 5 human tumor cell lines, including lung large cell carcinoma PC-13, lung small cell carcinoma QG-90, amelanotic melanoma HMV-1, renal carcinoma ACHN and Burkitt lymphoma Daudi, were compared with those of IFN-alpha 2a. Three out of five human tumor cell lines, ACHN, HMV-1, and QG-90, were highly susceptible to IFN-gamma: the concentrations required to inhibit 50% of the growth of cells (IC50) of ACHN, HMV-1, and QG-90 were 26, 11 and 7 pM, respectively. IFN-alpha 2a also markedly inhibited the growth of AC

IFNAR2 Deficiency Causing Dysregulation of NK Cell Functions and Presenting With Hemophagocytic Lymphohistiocytosis.

We describe a 2 year old boy with two previously undescribed frameshift mutations in the interferon (IFN)α/β receptor 2 (IFNAR2) gene presenting with hemophagocytic lymphohistiocytosis (HLH) following measles-mumps-rubella vaccination. Functional analyses show the absence of response to type I IFN in the patient's cells, as revealed by the lack of phosphorylation of STAT1 and the lack of induction of interferon-stimulated genes upon ex vivo stimulation with IFNα. HLH has been reported in patients with inborn errors of type I IFN-mediated immune responses following vaccination with live-attenua

Phase I study of recombinant beta-interferon given by four-hour infusion.

Twelve patients with advanced malignant disease were entered onto a Phase I study of escalating doses of beta-interferon serine given by 4-h i.v. infusion twice a wk. Three patients each were entered at starting doses of 0.01, 1, 10, and 30 million units (MU)/m2. Doses escalation within individual patients was allowed to a maximum dose of 400 MU/m2. Fever, chills, fatigue, and acral cyanosis were commonly seen and increased in frequency at higher doses. Myalgia, nausea, diarrhea, headache, and confusion were seen at lesser frequencies. Mild leukopenia, paresthesia, infusion site erythema, and

[Interferon alfacon-1 (Advaferon): a novel synthetic interferon for the treatment of hepatitis C, its pharmacological and clinical profile].

Interferon alfacon-1 (Advaferon, also referred to as consensus interferon) is a novel synthetic recombinant type-I interferon (IFN) developed by comparing the amino acid sequences of several natural IFN-alpha subtypes and assigning the most frequently observed amino acid in each corresponding position to generate a consensus molecule. Interferon alfacon-1 binds with high affinity to type-I IFN receptors and has greater biological activity than naturally occurring IFN-alpha as assessed by its increased antiviral, anti-proliferative, and natural killer cell activities, as well as its stronger IF

The Ro60 autoantigen binds endogenous retroelements and regulates inflammatory gene expression.

Autoantibodies target the RNA binding protein Ro60 in systemic lupus erythematosus (SLE) and Sjögren's syndrome. However, it is unclear whether Ro60 and its associated RNAs contribute to disease pathogenesis. We catalogued the Ro60-associated RNAs in human cell lines and found that among other RNAs, Ro60 bound an RNA motif derived from endogenous Alu retroelements. Alu transcripts were induced by type I interferon and stimulated proinflammatory cytokine secretion by human peripheral blood cells. Ro60 deletion resulted in enhanced expression of Alu RNAs and interferon-regulated genes. Anti-Ro60

Lichenoid graft-versus-host disease shows a high interferon score, with IFNAR1 inhibition preventing skin inflammation.

BACKGROUND: Chronic cutaneous graft-versus-host disease (ccGVHD) is a debilitating complication of allogeneic haematopoietic stem cell transplantation, manifesting as either sclerotic or lichenoid eruptions (lGVHD). Although frequent, the pathogenesis of lGVHD remains mainly unknown and represents a therapeutic challenge. OBJECTIVES: This study aims to decipher the immunological mechanisms underlying lichenoid GVHD and to evaluate the effect of blocking the Type I interferons (IFN-I) pathway in a mice model of ccGVHD on local inflammation. METHODS: First, we performed single-cell gene expres

Cleavage of the adaptor protein TRIF by enterovirus 71 3C inhibits antiviral responses mediated by Toll-like receptor 3.

Enterovirus 71 (EV71) causes hand-foot-and-mouth disease and neurological complications in young children. Although the underlying mechanisms remain obscure, impaired or aberrant immunity is thought to play a role. In infected cells, EV71 suppresses type I interferon responses mediated by retinoid acid-inducible gene I (RIG-I). This involves the EV71 3C protein, which disrupts the formation of a functional RIG-I complex. In the present study, we report that EV71 inhibits the induction of innate immunity by Toll-like receptor 3 (TLR3) via a distinct mechanism. In HeLa cells stimulated with poly

Long-term HIV-1 infection induces an antiviral state in primary macrophages.

HIV-1 infection is thought to impair type I interferon (IFN-I) production in macrophages, a cell type that is also relatively resistant to HIV-1 cytotoxic effects. Here, we show that monocyte differentiation into macrophages by M-CSF led to cell proliferation and susceptibility to HIV-1 infection that induced cell cycle arrest and increased cell death. Established HIV-1 infection of monocyte-derived macrophages induced the upregulation of the pattern recognition receptors MDA5 and Rig-I that serve as virus sensors; production of interferon-β, and transcription of interferon-stimulated genes in

The role of vitamin A in natural killer cell cytotoxicity, number and activation in the rat.

The cause of low natural killer (NK) cell activity in vitamin-A-deficient rats was investigated in the current study. Cytotoxicity correlated with the percentage of NK cells (3.2.3-positive cells) in peripheral blood mononuclear cells (PBMC; r = 0.85) and spleen (r = 0.70). Lytic activity per NK cell did not differ between vitamin-A-deficient rats and controls in PBMC, whereas it did in spleen. Vitamin A deficiency did not impair the ability to produce interferon (IFN), increase lytic efficiency and expand the NK cell pool upon poly(I:C) stimulation in vivo. Nor was the in vitro activation by

Nonsegmented Negative-Sense RNA Viruses Utilize N6-Methyladenosine (m6A) as a Common Strategy To Evade Host Innate Immunity.

N6-Methyladenosine (m6A) is the most abundant internal RNA modification catalyzed by host RNA methyltransferases. As obligate intracellular parasites, many viruses acquire m6A methylation in their RNAs. However, the biological functions of viral m6A methylation are poorly understood. Here, we found that viral m6A methylation serves as a molecular marker for host innate immunity to discriminate self from nonself RNA and that this novel biological function of viral m6A methylation is universally conserved in several families in nonsegmented negative-sense (NNS) RNA viruses. Using m6A methyltrans

TANK-binding kinase 1 as a novel therapeutic target for viral diseases.

TANK-binding kinase 1 (TBK1) is vital for the induction of antiviral innate immune responses. Both RNA and DNA viral infection induces TBK1 activation, triggers phosphorylation of interferon regulatory factor (IRF) 3 and subsequent expression of type I interferons (IFNs; IFN-α/β). Type I IFNs can induce the expression of numerous antiviral genes called interferon-stimulated genes (ISGs) to build a remarkable antiviral state and limit viral replication. Thus, optimal TBK1 activity is crucial for IRF3-induced type I IFNs expression and ISGs-mediated viral elimination. Areas covered: This review

Comparative analysis of the impact of interferon regulatory factors on basal and induced interferon and interferon stimulated gene expression in human and canine keratinocytes.

INTRODUCTION: Keratinocytes are key barrier cells able to mount a robust interferon (IFN) antiviral response to defend against infection in the skin. Similar to humans, dogs spontaneously develop skin disorders associated with dysregulation of IFN immunity and can be used as a large animal model to investigate these diseases. One of the critical factors driving IFN regulation are interferon regulatory factors (IRFs). IRFs are crucial in upregulating the antiviral type I or type III IFNs, which then subsequently upregulate hundreds of antiviral effector proteins called interferon stimulated gen

Parasympathetic neural activity and the reciprocal regulation of innate antiviral and inflammatory genes in the human immune system.

The vagus nerve mediates parasympathetic nervous system control of peripheral physiological processes including cardiovascular activity and immune response. In mice, tonic vagal activation down-regulates inflammation via nicotinic acetylcholine receptor-mediated inhibition of the pro-inflammatory transcription factor NF-κB in monocyte/macrophages. Because Type I interferon and pro-inflammatory genes are regulated reciprocally at the level of transcription factor activation and cell differentiation, we hypothesized that vagal activity would up-regulate Type I interferon response genes concurren

Caspase-3 cleaves CYLD to restrict interferon signaling during mitochondrial apoptosis.

Mitochondrial outer membrane permeabilization is a pivotal event in programmed cell death by apoptosis, leading to the activation of the cysteine protease caspase-3 (CASP3) and the release of mitochondrial nucleic acids. This release triggers the activation of the Interferon Regulatory Factor 3 (IRF3) transcription factor and the subsequent IRF3-mediated type I interferon production and cell death. CASP3 ensures apoptosis remains immunologically silent, though the mechanisms are unclear. We report that CASP3 cleaves CYLD, a deubiquitinating enzyme crucial for cell fate and inflammatory signali

Expression of interferon-stimulated genes, but not polymorphisms in the interferon α/β receptor 2 gene, is associated with coronavirus disease 2019 mortality.

Excessive inflammatory response is a hallmark of severe COVID-19. This study investigated the associations between interferon-stimulated genes (ISGs) expression, genetic variation in the interferon α/β receptor 2 (IFNAR2) gene, and COVID-19 mortality. We investigated 67 patients with moderate-to-severe COVID-19. Of them, 22 patients (32.8 %) died because of COVID-19. We examined the expression of ISGs in total RNA of peripheral whole blood. We observed a significant increase in the expression of all ISGs examined in non-surviving patients, indicating a heightened interferon type I signaling ac…
