Reduction of muscle larvae burden in rats experimentally infected with Trichinella spiralis.

In Wistar rats infected with 500 to 2,500 Trichinella spiralis larvae the muscle larvae intensity (larvae per gram-l.p.g.) was measured from 20 to 180 day post infection (d.p.i). The l.p.g. increased to day 40-50 p.i. and decreased thereafter. The highest reduction took place between 60 and 120 d.p.i. with intermediate inoculum of T. spiralis larvae. The mechanism of the reduction of T. spiralis larvae in muscles is suggested to depend on pericapsular-intercapsular host cells infiltrations attracted by parasite antigens.

Construction and analysis of cDNA library of Necator americanus third stage larvae.

OBJECTIVE: To obtain the genetic information on Necator americanus and to search for the purpose genes. METHODS: mRNA was isolated from the third stage larvae of Necator americanus maintained in hamsters. Double strand cDNA was synthesized and ligated to lambda ZAPII vector to construct the cDNA library. Expressed sequence tages (ESTs) were obtained by single pass sequencing of randomly isolated cDNA clones from the established library. RESULTS: A cDNA library of N. americanus was successfully constructed with high recombinant efficiency. The titer of unamplified library was 1 x 10(7). The i…

Tetracycline inhibits development of the infective-stage larvae of filarial nematodes in vitro.

In recent years, studies have linked tetracycline treatment of filaria-infected animals with reduced adult worm burdens and decreased levels of microfilaremia. These observations are believed to be attributable to clearance of Wolbachia, intracellular rickettsial-like organisms found within filarial tissues. Although maximal worm reductions were observed when treatment was initiated early in infection, it is not known whether tetracycline inhibits development of infective-stage larvae. To address this issue, we studied the effect of tetracycline on three different species of filarial nematodes…

Oral myiasis caused by hypoderma bovis larvae in a child: a case report.

Myiasis is the invasion of living tissue of humans and other mammals by the eggs or larvae of flies of the order of Diptera. It occurs mainly in the tropic, and is associated with inadequate public and personal hygiene. Oral myiasis in humans appears to be rare. This article records a case of oral myiasis caused by larvae of Hypoderma bovis. Two different pathologic soft tissue sockets were observed in the vestibular sulcus at the level of the both deciduous laterals along the deep upper lip tissues.

Spatial distribution and habitat characterization of anopheline mosquito larvae in Western Kenya.

Studies were conducted to characterize larval habitats of anopheline mosquitoes and to analyze spatial heterogeneity of mosquito species in the Suba District of western Kenya. A total of 128 aquatic habitats containing mosquito larvae were sampled, and 2,209 anopheline and 10,538 culicine larvae were collected. The habitats were characterized based on size, pH, distance to the nearest house and to the shore of Lake Victoria, coverage of canopy, surface debris, algae and emergent plants, turbidity, substrate, and habitat types. Microscopic identification of third- and fourth-instar anopheline l…

The morphogenesis of Ascaris suum to the infective third-stage larvae within the egg.

Studies of the morphology of Ascaris suum larvae developing in the egg during embryonation in vitro at room temperature showed that 2 molts take place within the egg. The first larval stage (L1) appeared in the egg after 17-22 days of cultivation, the first molt to the second larval stage (L2) took place from day 22 to day 27, and the second molt to the third larval stage (L3) started on day 27 and continued during the 60-day observation period. Infectivity of the eggs was studied by oral egg inoculation in mice and showed that the L3 are the infective stage for mice. Molting to the L3 stage o…

Water skating in the larvae of dixella aestivalis (Diptera) and hydrobius fuscipes (Coleoptera).

The kinematics of locomotion was investigated in the aquatic larvae of Dixella aestivalis and Hydrobius fuscipes with the aid of high-speed video recordings. Both insects are able to skate on the surface of the water using the dorso-apical tracheal gill as an adhesive organ or 'foot'. Progress relies on the variable adhesion of the foot between 'slide' and 'hold' periods of the locomotory cycle. The flexural body movements underlying skating in D. aestivalis can be derived directly from the figure-of-eight swimming mechanism used in underwater swimming. The latter is shown to be similar to fig…

Method for collecting Hypoderma spp. (Diptera: Oestridae) larvae from cattle.

A new method for collecting 3rd-instar Hypoderma lineatum (Villers) and Hypoderma bovis (L.) is described. A procedure for carrying out chronobiological and other studies involving the life cycle of these flies is included. The larvae are collected from cattle using a dressing made of a circle of cardboard attached to a piece of tulle similar in color to the coat of the animal. These small cloth cages were attached to the animal skin with fast-drying contact glue wherever the swellings produced by 2nd- or 3rd-instar Hypoderma spp. had been previously detected. Each animal was inspected daily t…

Early effect of mosquito larvae extract on mouse cells proliferation in vivo.

It has been demonstrated that mosquito larvae crude extract has an inhibiting effect on the mitotic rate of several mouse cell populations. The sampling period was 16-24 h after treatment, when mitotic peak normally occurs. The present paper reports the effect of mosquito larvae crude extract on the proliferation of hepatocytes, renocytes, Lieberkhün crypt enterocytes, and sialocytes. In this case, the sampling period covered the dark phase of the day, during the first 12 h after treatment. Colchicine-arrested metaphases were controlled at 20/04, 00/08 and 04/12 (Time of Day/Time Post Injectio…

Effect of ivermectin on Caenorhabditis elegans larvae previously exposed to alcoholic immobilization.

First-stage larvae of Caenorhabditis elegans were immersed in 0.15% 1-phenoxy-2-propanol to induce temporary paralysis, including the suppression of pharyngeal pumping. Subsequent addition of ivermectin (to give 50 micrograms/ml) induced coiling and prolonged immobilization of such larvae, as also of control larvae (previously immersed only in water). The results suggest that ingestion of drug by means of pharyngeal pumping is not a prerequisite for the uptake of ivermectin at levels sufficient for antinematodal action.

Immunity to a challenge infection of Strongyloides stercoralis third-stage larvae in the jird.

Jirds support the entire life-cycle of Strongyloides stercoralis. We therefore used this host as a model to define the mechanism of the immune response to a challenge infection, as well as the parasite stage effected by the response. Jirds given a primary infection of S. stercoralis are resistant to re-infection. The use of implanted diffusion chambers containing larvae showed that the immune response killed the third-stage larvae, and this was confirmed by subcutaneous infections. The larvae of a challenge infection are killed within 48 h, a time period too short to allow for the development …

Recovery of Pseudoterranova decipiens (Anisakidae) larvae from codfish of the Antarctic Ocean.

Anisakid larvae were found from the liver and stomach of the codfish (Notothenia neglecta) caught from the Antarctic Ocean, South Pole, where the Polar Research Center of The Korea Ocean Research and Development Institute is operating. Through observations by scanning electron (SEM) and light microscopy (LM), they were identified as the third stage larvae of Pseudoterranova decipiens, a potential agent of human codworm anisakiasis. They measured 25.0-28.5 mm in length, and 0.6-0.8 mm in width. The SEM revealed a prominent boring tooth, three lips, and excretory pore at the anterior end, and a …

Acquisition and expression of humoral reactivity to antigens of infective stages of filarial larvae.

Measurement of anti-larval responses in filaria-exposed populations may shed light on the natural history of exposure to Wuchereria bancrofti. Using serum samples obtained by a cross-sectional survey of 172 individuals from two neighbourhoods in Leogane, Haiti, antibody responses directed against infective stage filarial larvae (L3) were assayed by enzyme-linked immunosorbent assay (ELISA), immunofluorescence (IFA), and immunoblot for the presence of anti-larval antibodies. ELISA results indicated that virtually all members of both neighbourhoods mounted an anti-larval antibody response within…

Studies on the inhibition of rejection of Trichostrongylus colubriformis larvae from immune sheep.

A total of 8 drugs were examined for their ability to suppress the rejection of Trichostrongylus colubriformis infective larvae (L3) from immune sheep. Specific antagonists of leukotrienes (piroxicam), prostaglandin (indomethacin) and the immunosuppressant cyclosporin A were given orally, while injectable preparations of dexamethasone, chlorpheniramine, BN 52051, WEB 2086 (anti-platelet-activating factor) and theophylline were administered as directed. The drugs were given for 5 days prior to and 4 days after challenge with 20,000 L3, when worm counts were done. Corticosteroids inhibited rejec…

Bot fly larvae (Cephenemyia jellisoni) as a cause of neurologic signs in an elk.

In June 1993, a yearling female elk (Cervus elaphus) near John Day, Oregon (USA) was observed twice over a four week period with signs of neurologic disease including weakness, walking in circles with an uncoordinated gait and disorientation. The elk was shot, and the head and neck were examined grossly for parasites and lesions. Thirty-five second and third instar larvae of Cephenemyia jellisoni were recovered from an encapsulated space in the nasopharyngeal area dorsal to the soft palate. Larvae protruded into the caudodorsal end of the ventral nasal meatus, obliterating the opening of the l…

Detection of muscle larvae of Trichinella spiralis by enzyme-linked immunosorbent assay.

ELISA was applied to the detection of Trichinella spiralis larvae in infected rat muscles. Anti-muscle larva serum was prepared by inoculating crude extract of muscle larvae to rabbits. Limb muscles of infected rats were emulsified with carbonate buffer and the supernatant was used for ELISA. ELISA values of positive muscular samples were clearly higher than those of negative ones to prove that ELISA can be applied to the detection of muscle larvae. The present method showed positive values to muscles infected with 10 or more larvae per gram of muscle (LPGM).

Brugia pahangi: production of a monoclonal antibody reactive with the surface of infective larvae.

Monoclonal antibodies against infective third-stage larvae (L3) of Brugia pahangi were generated from mice immunized with L3 antigens. The monoclonal antibodies were L3 stage-specific or stage-nonspecific. A BpG1 monoclonal antibody (IgG1 subclass) showing L3 stage-specificity was examined in detail. BpG1 recognized the surface of B. pahangi L3 and also reacted with the surface of Brugia malayi L3 but not with the surface of filarial worms of other genera, such as Acanthocheilonema viteae and Litomosoides carinii. BpG1 promoted cellular adhesion to the surface of B. pahangi L3. BpG1 bound on l…
