Membrane location of spin-labeled cytochrome c determined by paramagnetic relaxation agents.

The mitochondrial protein horse heart cytochrome c was specifically spin-labeled with succinimidyl-2,2,5, 5-tetramethyl-3-pyrroline-1-oxyl-carboxylate on different lysine residues at positions 86, 87, 72, 8, or 25, respectively. Site-specifically labeled species were separated chromatographically and identified by peptide sequencing of tryptic digests. The monolabeled protein was bound to negatively charged phospholipid membranes composed of dioleoylphosphatidylglycerol, and the accessibility of the spin-labeled lysine residues to lipid-soluble molecular oxygen and to lipid-impermeant chromium

Acidic pH Modulates Headgroup Orientation and Packing in Bis(monoacylglycero)phosphate Bilayers.

Bis-(monoacylglycero)-phosphate (BMP) possesses an atypical headgroup structure, and its naturally occurring 2,2'-isomer rapidly rearranges to the more stable 3,3'-form in aqueous buffers. Because the BMP is a major component of endosomal and lysosomal membranes, understanding its intrinsic physicochemical behavior is broadly relevant to the organization of acidic intracellular interfaces. To quantify how acidity modulates the physicochemical organization of 3,3'-BMP membranes, we characterized model bilayers across a controlled pH range by using electrophoretic mobility, fluorescence spectros

Aggregation and membrane activity of mutant A30P alpha-synuclein on mitochondrial membranes.

The A30P point mutation in the α-synuclein (α-syn) protein is linked to a familial form of Parkinson's disease (PD), the most common motor neurodegenerative disorder. The association of α-syn with lipids in cellular membranes plays a key role in modulating the aggregation behavior of the protein, as well as its toxicity. Here, we studied the impact of the A30P mutation on the interaction of α-syn with mitochondrial membranes, since the interplay between α-syn aggregation and mitochondrial dysfunction is central for the pathogenic process in PD, but still not completely understood. Experiments

A membrane-disruptive action of VBIT-4 challenges its role as a widely used VDAC1 oligomerization inhibitor.

Voltage-dependent anion channel (VDAC) is the most abundant protein of the mitochondrial outer membrane and a key regulator of metabolite ex … scale thermophoresis, and coarse-grained molecular dynamics demonstrate that VBIT-4 partitions into lipid bilayers, increases membrane permeability, and destabilizes membrane structure, without detectable effects on VDAC1 channel properties or assemblies. Consistent with this mechanism, VBIT-4 induces VDAC1-independent cytotoxicity in HeLa cells at concentration …

Loss of STARD7 Impairs Mitochondrial Phospholipid Homeostasis and Contributes to Mitochondrial Myopathy.

Mitochondria are composed of phospholipid bilayers rich in phosphatidylcholine (PC). StAR-related lipid transfer domain-containing protein 7 (STARD7) functions as a lipid transfer protein that plays a crucial role in maintaining mitochondrial PC homeostasis. In this study, we investigated the physiological role of STARD7 in skeletal muscle using muscle-specific knockout (mKO) mice. STARD7 expression was markedly higher in the soleus, a mitochondria-dense slow-twitch muscle, compared with fast-twitch fibers. Although muscle fibers from mKO mice exhibited no apparent structural abnormalities, th…

Pharmaceutical applications of microbial biosurfactants.

Biosurfactants are becoming more and more popular in biomedical and pharmaceutical applications due to their intriguing biological and physi … us. The ring-shaped peptide and hydrophobic chain interact with bacterial membranes, inserting into lipid bilayers and destabilising them, leading to pore formation and membrane leakage. Further, BSs disrupt microbial biofilm formation, making them more permeable and susceptible to antibiotics. The anti-adhesive property is exploited to prevent bacterial colonis …

Polarity dependence of EPR parameters for TOAC and MTSSL spin labels: correlation with DOXYL spin labels for membrane studies.

TOAC (2,2,6,6-tetramethylpiperidine-1-oxyl-4-amino-4-carboxylic acid) is a nitroxyl amino acid that can be incorporated in the backbone of peptides. DOXYL (4,4-dimethyl-oxazolidine-1-oxyl) is a nitroxyl ring that can be attached rigidly at specific C-atom positions in the acyl chains of phospholipids. Isolation and Characterization of Small Extracellular Vesicles Derived from Epicardial Adipose Tissue.

Epicardial adipose tissue (EAT) is a distinct visceral adipose depot located between the myocardium and the visceral layer of the pericardium. Owing to the absence of an anatomical barrier between EAT and the heart, EAT can communicate with the myocardium through paracrine and vasocrine mechanisms. Small extracellular vesicles (sEVs) secreted by EAT, which are lipid bilayer-enclosed nanoparticles that mediate intercellular communication by transferring bioactive cargoes, including proteins, lipids, and nucleic acids, to recipient cells, may serve as key mediators of this crosstalk. However, th

Extracellular Vesicles in Stroke: Drivers of Brain-Body Network Crosstalk.

Mounting evidence reframes stroke not as a single cerebral event but as a dynamic brain-body network disorder. Extracellular vesicles (EVs)-lipid bilayer-enclosed particles released by virtually all cells-serve as intercellular messengers that couple vascular injury, neuroinflammation, and systemic responses. EVs carry nucleic acids, proteins, and lipids reflective of their cellular sources and may reach the circulation and interface with the CNS via context-dependent routes, including blood-brain barrier (BBB) disruption and active transport mechanisms. This review integrates clinical and exp

Nuclear Assembly in Giant Unilamellar Vesicles Encapsulating Xenopus Egg Extract.

The reconstitution of a cell nucleus in a lipid bilayer-enclosed synthetic cell makes great strides in bottom-up synthetic biology. In this study, a method for assembling a nucleus in giant unilamellar vesicles (GUVs) is proposed. To induce reconstitution of the nucleus, the interphase egg extract of African clawed frogs Xenopus laevis is utilized, known as a biochemically controllable cell-free system capable of transforming an added sperm chromatin into a nucleus in vitro. The GUV formation efficiency is enhanced by the inverted emulsion method through incorporating prolonged waiting time an

Exosomes in multiple sclerosis and Alzheimer's disease - Adversary and ally.

Neuroinflammation and the resulting neurodegeneration is a big challenge for the healthcare system, especially with the aging population. Neuroinflammation can result from a variety of insults to the central nervous system leading to an interplay between immune and brain cells that sustains chronic inflammation and injures neural cells. One facilitator of this toxic interplay are exosomes. Exosomes are nano-sized, bilayer lipid vesicles secreted by cells containing proteins, nucleic acids and lipids. Because exosomes can be internalized by other cells, their contents can elicit inflammatory re

Isolation of Extracellular Vesicles for Proteomic Profiling.

Extracellular vesicles (EVs) are nano-sized lipid bilayer surrounded by structures released from most cells, including archaea, bacteria, and eukaryotic cells. EVs play multiple roles in cell-to-cell communication, including immune modulation, angiogenesis, and phenotypic transformation of cells by transferring genetic material and functional proteins. They contain specific subsets of proteins, DNA, RNA, and lipids that represent their cellular origin. Furthermore, EVs are enriched in cell type- or disease-specific proteins, especially plasma membrane proteins, which have pathophysiological fu

Detachment of Membrane Bound Virions by Competitive Ligand Binding Induced Receptor Depletion.

Multivalent receptor-mediated interactions between virions and a lipid membrane can be weakened using competitive nonpathogenic ligand binding. In particular, the subsequent binding of such ligands can induce detachment of bound virions, a phenomenon of crucial relevance for the development of new antiviral drugs. Focusing on the simian virus 40 (SV40) and recombinant cholera toxin B subunit (rCTB), and using (monosialotetrahexosyl)ganglioside (GM1) as their common receptor in a supported lipid bilayer (SLB), we present the first detailed investigation of this phenomenon by employing the quart
