Cordyceps sinensis and Sonodynamic Therapy in Non-Small Cell Lung Cancer: Pharmacological Basis, Therapeutic Potential, and Future Directions.

Non-small cell lung cancer (NSCLC) accounts for the highest cancer-related mortality worldwide. Chemotherapy, the backbone of treatment for advanced disease, is constrained by drug resistance and off-target toxicity. Extracts of Cordyceps sinensis (CORS), a medicinal fungus, contain cordycepin, cordyceps polysaccharides (CSP), and various sterols that suppress NSCLC cell proliferation and induce apoptosis in preclinical models. Translating these findings to the clinic, however, faces three problems. First, the active compounds are poorly absorbed orally. Second, water-based decoction inevitabl

A roadmap for focused ultrasound applications in psychiatry: Proceedings of the 2024 symposium on focused ultrasound in psychiatry (FUS-PULSE).

Transcranial focused ultrasound (FUS) is an emerging neuromodulation modality that enables incisionless, spatially precise targeting of deep brain structures implicated in neuropsychiatric conditions. Growing clinical applications in FUS psychiatry encompass both transient and permanent bioeffects, including focal lesioning, neurostimulation, and targeted drug delivery. In response to rapid advances in the field, an in-person multidisciplinary symposium, FUS-PULSE, was held in Toronto, Canada from June 5-7 2024. The meeting convened over 70 international experts across neurosurgery, psychiatry

Gaseous Microemboli and Postoperative Delirium in Coronary Artery Bypass Grafting.

Background: Postoperative delirium (POD) is a neurocognitive syndrome affecting patients undergoing surgery. It is a frequent complication of coronary artery bypass grafting (CABG) and is associated with higher morbidity, mortality and treatment costs. This study aimed to investigate the relationship between gaseous microemboli (GME) load during cardiopulmonary bypass (CPB) and subsequent POD in patients undergoing CABG. Methods: In total, 102 patients undergoing elective on-pump CABG were evaluated in this observational study. An ultrasonic microbubble counter, with probes placed on the arter

Ultrasound-activated miR-195-5p/shikonin nanobubbles remodel immunosuppressive microenvironment via immunogenic cell death to potentiate PD-1/PD-L1 blockade in hepatocellular carcinoma.

Despite the transformative clinical impact of programmed cell death ligand 1/programmed death 1 (PD-1/PD-L1) blockade in hepatocellular carcinoma (HCC), therapeutic efficacy remains limited by the tumor immunosuppressive microenvironment (TIME), with objective response rates persistently below 20 %. To address this critical clinical challenge, we engineered ultrasound (US)-responsive lipid nanobubbles (NBs) co-encapsulating microRNA (miR)-195-5p and shikonin (SK) (designated miR-195-5p/SK-NBs), a dual-functional platform designed to synergize PD-L1 suppression with immunogenic cell death (ICD)

Novel use of contrast enhanced ultrasonography with urodynamics in children: A comparison study to fluoroscopy.

INTRODUCTION: Video urodynamics (UDS) has classically been performed using fluoroscopy (fluoro). Contrast enhanced voiding ultrasonography (ceVUS) has rarely been reported for use with UDS. This is the first study to compare the imaging characteristics of ceVUS versus fluoro UDS. METHODS: Children were enrolled for ceVUS UDS who previously underwent fluoro UDS. Demographics, imaging data for ceVUS and fluoro UDS, time between studies, and clinical data between studies were recorded. Changes in clinical status included implantation/cessation of catheterization or anticholinergic medications, l

BACKGROUND: KCNE2 is a promiscuous auxiliary subunit of voltage-gated cation channels. A recent work demonstrated that KCNE2 regulates L-type Ca2+ channels. Given the important roles of altered Ca2+ signaling in structural and functional remodeling in diseased hearts, this study investigated whether KCNE2 participates in the development of pathological hypertrophy. METHODS AND RESULTS: We found that cardiac KCNE2 expression was significantly decreased in phenylephrine-induced cardiomyocyte hypertrophy in neonatal rat ventricular myocytes and in transverse aortic constriction-induced cardiac h

Cerebral Gaseous Microemboli are Detectable During Continuous Venovenous Hemodialysis in Critically Ill Patients: An Observational Pilot Study.

BACKGROUND: Continuous venovenous hemodialysis (CVVHD) may generate microemboli that cross the pulmonary circulation and reach the brain. The aim of the present study was to quantify (load per time interval) and qualify (gaseous vs. solid) cerebral microemboli (CME), detected as high-intensity transient signals, using transcranial Doppler ultrasound. MATERIALS AND METHODS: Twenty intensive care unit (ICU group) patients requiring CVVHD were examined. CME were recorded in both middle cerebral arteries for 30 minutes during CVVHD and a CVVHD-free interval. Twenty additional patients, hospitaliz

Nonthermal ablation in the rat brain using focused ultrasound and an ultrasound contrast agent: long-term effects.

OBJECTIVE Thermal ablation with transcranial MRI-guided focused ultrasound (FUS) is currently under investigation as a less invasive alternative to radiosurgery and resection. A major limitation of the method is that its use is currently restricted to centrally located brain targets. The combination of FUS and a microbubble-based ultrasound contrast agent greatly reduces the ultrasound exposure level needed to ablate brain tissue and could be an effective means to increase the "treatment envelope" for FUS in the brain. This method, however, ablates tissue through a different mechanism: destruc

The bubble study: ultrasound confirmation of central venous catheter placement.

STUDY OBJECTIVE: The objective was to determine if ultrasound (US) can more rapidly confirm central venous catheter (CVC) position in comparison to chest radiography (CXR) in the emergency department. METHODS: The study included a convenience sample of emergency department patients with supradiaphragmatic CVCs and a CXR for confirmation. Ultrasound was used for CVC confirmation by visualizing microbubble artifact in the right atrium after injection of saline through the distal port. To evaluate for pneumothorax (PTX), "sliding sign" of the pleura was noted on US of the anterior chest. Blinded

Assessment of myocardial perfusion with real-time myocardial contrast echocardiography: methodology and clinical applications.

Real-time myocardial contrast perfusion imaging (RTMCI) with echocardiography is a promising technique for evaluation of patients with known or suspected coronary artery disease. The technique is based on the utilization of small (<10 mum) microbubbles, which are capable of crossing the pulmonary circulation after intravenous injection. Unlike radioactive isotopes, which are taken actively or diffuse passively in the myocytes, myocardial contrast agents remain extracellularly in the capillaries and present a measure of the myocardial capillary blood volume and microvascular integrity. RTMCI ha…

Advances in acoustic tweezers for biological detection: from mechanisms to applications.

Acoustic tweezers use acoustic radiation force and acoustic streaming generated by ultrasound fields to achieve contactless and relatively low-damage manipulation of biological targets, including cells, bacteria, extracellular vesicles, and other micro-/nanoscale objects. In biosensing workflows, they mainly function as modules for sample preparation, target enrichment, sorting, reaction organization, and mass-transfer enhancement before downstream signal readout. This review summarizes the fundamental mechanisms, representative device configurations, and integration strategies of acoustic twe

Nod-like receptor protein 3 (NLRP3) inflammasome activation and podocyte injury via thioredoxin-interacting protein (TXNIP) during hyperhomocysteinemia.

NADPH oxidase-derived reactive oxygen species (ROS) have been reported to activate NLRP3 inflammasomes resulting in podocyte and glomerular injury during hyperhomocysteinemia (hHcys). However, the mechanism by which the inflammasome senses ROS is still unknown in podocytes upon hHcys stimulation. The current study explored whether thioredoxin-interacting protein (TXNIP), an endogenous inhibitor of the antioxidant thioredoxin and ROS sensor, mediates hHcys-induced NLRP3 inflammasome activation and consequent glomerular injury. In cultured podocytes, size exclusion chromatography and confocal mi
