Beyond the variant: hereditary cancer awareness in the multi-omics era.

Hereditary cancer awareness has entered a new phase. For decades, awareness has focused on recognizing familial risk, identifying pathogenic germline variants, and helping families access counselling, surveillance and prevention. Yet in 2026, the main challenge is no longer only whether a variant can be detected, but how inherited risk can be interpreted, communicated and translated into action in the biological context of each tissue and tumor. The recent publication landscape of Hereditas illustrates this shift: cancer genetics is now inseparable from RNA regulation, epigenetics, metabolism,

Cellular heterogeneity and multicellular mechanisms in the pathogenesis of late-life depression: insights from single-cell and spatial multi-omics.

Late-life depression (LLD) is a heterogeneous disorder characterised by affective symptoms, cognitive decline, vascular burden, frailty, treatment resistance, and an increased risk of neurodegenerative progression, yet bulk molecular approaches may obscure the cell-type-specific alterations underlying this clinical and biological heterogeneity. This narrative review synthesises single-cell, single-nucleus, spatial, and multi-omic evidence concerning neuronal, glial, vascular, and peripheral immune alterations relevant to LLD, while distinguishing direct findings from clinically defined or olde

Multimodal detection of microplastics in human kidney stones and multi-omics exploration of renal cell metaflammation.

Microplastics (MPs) have emerged as contaminants of increasing concern due to their propensity to accumulate in human tissues and their potential health implications. However, the long-term accumulation behavior of MPs in the kidney and their associated toxic mechanisms remain elusive. Here, we employed advanced multimodal detection techniques to quantify and characterize MPs in kidney stones, and integrated transcriptomic, proteomic, and metabolomic analyses to systematically evaluate the underlying cytotoxic mechanisms. MPs were detected in all kidney stone samples examined, with a median co

Physiology and Multi-Omics Provide Insights into Sperm Activation and Movement in Euryhaline Spotted Seabass (Lateolabrax maculatus).

Sperm activation and movement are pivotal determinants of fertilization success in teleosts, yet the molecular mechanisms in euryhaline species remain largely unresolved. Here, we integrated physiology and multi-omics to elucidate the osmolality, ions, and regulatory networks underlying sperm activation and movement in spotted seabass (Lateolabrax maculatus). Spotted seabass sperm showed strong motility across a broad osmotic range of 500 to 1200 mOsm/kg, with Na+ and K+ promoting motility and Ca2+ exerting inhibitory effects. Integrated transcriptomics and proteomics identified 29 genes and 6

Integration of multi-omics and artificial intelligence for therapeutic insights in Alzheimer's disease: A comprehensive review.

Alzheimer's disease (AD) is a progressively worsening type of brain disorder that damages the nerve cells. It is marked by the buildup of amyloid-β plaques outside the cells, tau neurofibrillary tangles inside the cells, and overall molecular-level dysfunction. The therapies currently available mainly cater to alleviating the symptoms, whereas the newly approved disease-modifying antibodies, such as lecanemab and donanemab, bring out only limited clinical improvements. Being complicated and involving many factors, AD requires sophisticated computer-based methods to combine different biological

Deciphering Sjögren's disease: New insights and perspectives extend from advanced omics approaches and immunology.

Sjögren's disease (SjD), formerly termed Sjögren's syndrome, is a chronic autoimmune exocrinopathy that has seen substantial advances in basic research over the past decade. Classical experimental approaches have revealed immune dysregulation encompassing aberrant activation of the adaptive immune system, activation of humoral autoimmunity against autoantigens, and upstream participation of innate immune mechanisms. In parallel, with the rapid development of sequencing and omics technologies, researchers are able to use these high-throughput methods to gain a more unbiased, sytem-wide view of

Exploring biomarkers for treatment response in psoriatic arthritis: a focus on multi-omics technologies.

Psoriatic arthritis (PsA) is a complex inflammatory disease characterised by a combination of cutaneous and musculoskeletal symptoms, which complicates both diagnosis and management. Despite advances in targeted therapy, the treatment of PsA remains challenging, with up to 40% of patients experiencing inadequate response. Predictive biomarkers may serve as valuable tools to help clinicians select the most appropriate therapy at the optimal time. However, despite substantial research activity, the current evidence remains inconclusive. Moreover, biomarkers that predict treatment response may di

From Proteome to miRNome: A Review of Multi-Omics Ocular Allergy Research Using Human Tears.

Ocular allergy (OA) is a subtype of seasonal allergy that causes symptoms of itchiness, redness, swelling and irritation of the ocular surface and eyelids, often triggering allergy-induced eye rubbing and sustained inflammation for up to six months of the year during peak allergy season. These symptoms, coupled with reduced sleep quality, impaired daily productivity and decreased mood, highlight a significant yet underrepresented disease burden. Recent advances in tear-based multi-omics have enabled detailed characterisation of OA-associated biochemical changes on the ocular surface, highlight

Mosaic integration of spatial multi-omics with SpaMosaic.

With the advent of spatial multi-omics, mosaic integration of diverse datasets with partially overlapping modalities enables the construction of comprehensive multimodal spatial atlases from heterogeneous sources. Here we present SpaMosaic, a tool that uses contrastive learning and graph neural networks to build a modality-agnostic, batch-corrected latent space for spatial domain identification and missing-modality imputation. We systematically benchmarked SpaMosaic against existing integration methods using simulated data and experimentally acquired datasets spanning RNA and protein abundance

Unified representation learning for spatial multi-omics.

MOTIVATION: Integrating spatial multi-omics data is crucial for decoding tissue function, yet existing methods struggle with noise and cross-modal alignment. RESULTS: We introduce SpaAlign, a novel two-stage framework for the robust integration of multiple omics and histological data. At its core, a contrastive learning stage creates a unified semantic space, followed by a self-supervised clustering stage that enforces structural coherence. Our evaluations demonstrate its effectiveness in accurately delineating the capsule-cortex-medulla architecture in human lymph nodes and precisely identif

A nonlinear multi-omics data integration and classification model based on pathway self-attention and graph convolutional networks.

The abundance of omics data has significantly advanced the development of multi-omics data integration techniques. Non-linear embedding approaches for data integration have gradually become the mainstream in multi-omics research, as these approaches can substantially improve cancer analysis by enhancing the quality of the embeddings. However, current multi-omics data integration methods are typically confined to omics measurements, neglecting domain-specific prior knowledge encompassing biological pathways. In this study, we proposed a multi-omics integrated classification model, PathTransGCN,
