A novel RGB-trichrome staining method for routine histological analysis of musculoskeletal tissues.

Morphometry and histology are essential approaches for investigation and diagnosis of musculo-skeletal disorders. Despite the advent of revolutionary methods of image analysis and high resolution three-dimensional imaging technology, basic conventional light microscopy still provides an incisive overview of the structure and tissue dynamics of the musculoskeletal system. This is crucial to both preclinical and clinical research, since several clinically relevant processes, such as bone repair, osteoarthritis, and metabolic bone diseases, display distinct, if not pathognomonic, histological fea…

Reliability of the Multidimensional Pain Inventory and stability of the MPI classification system in chronic back pain.

BACKGROUND: This cross validation study examined the reliability of the Multidimensional Pain Inventory (MPI) and the stability of the Multidimensional Pain Inventory Classification System of the empirically derived subgroup classification obtained by cluster analysis in chronic musculoskeletal pain. Reliability of the German Multidimensional Pain Inventory was only examined once in the past in a small sample. Previous international studies mainly involving fibromyalgia patients showed that retest resulted in 33-38% of patients being assigned to a different Multidimensional Pain Inventory subg

Musculoskeletal adverse effects associated with inhaled levofloxacin in a patient with cystic fibrosis: A case report.

Inhaled levofloxacin is widely used in cystic fibrosis (CF) patients with chronic Pseudomonas aeruginosa infection and is generally well tolerated. The most commonly reported adverse effects are bronchospasm and dysgeusia, with limited evidence of systemic toxicity. We report a case of a 26-year-old woman with CF who developed recurrent diffuse musculoskeletal pain temporally associated with inhaled levofloxacin therapy. Symptoms occurred exclusively during treatment cycles, resolved upon discontinuation, and have not recurred since cessation. This case highlights a potential association betwe

Prospective associations between 24-h device-measured occupational and leisure-time physical activity and register-based musculoskeletal-related primary healthcare utilization among Danish workers.

Musculoskeletal pain is a leading cause of seeking care and imposes an immense economic burden to the healthcare systems. The guidelines recommend physical activity for managing musculoskeletal pain but do not differentiate between occupational and leisure-time physical activity. Research indicates that occupational physical activity may not have similar health benefits as leisure-time physical activity. Thus, we investigated the association between occupational and leisure-time physical activity and musculoskeletal-related primary healthcare utilization. At baseline, physical activity data (s

Validation of Claims-Based Algorithms for Identifying Congenital Urinary Tract, Genital, Gastrointestinal, and Musculoskeletal Malformations.

AIM: Congenital malformations are important outcomes when evaluating medication safety in pregnancy. However, the accuracy of claims-based algorithms for identifying organ-specific malformations remains understudied. We validated algorithms for four malformation groups: urinary tract, genital, gastrointestinal, and musculoskeletal. MATERIALS AND METHODS: Using the Mass General Brigham (MGB, 2007-2020) and Stanford Medicine (2016-2023) databases, we identified infants with potential malformations of interest based on diagnosis and procedure codes within 90 days of birth. A total of 150 cases w

On Lingering Pelvic Floor Musculoskeletal Injuries in the Postpartum Athlete: Are We Adequately Addressing Them?

Although vaginal birth has the highest risk of musculoskeletal injury per hour of exposure of any natural human activity, the resulting injuries often go unrecognized - sometimes for decades. The types of tissues injured during a difficult vaginal birth are similar to those involved in other musculoskeletal injuries. It is not widely appreciated that multiple structures can be injured, including pubic bone stress fractures, injury to the nerve innervating the right or left levator ani (LA) muscle, stretch injuries of the LA muscles themselves, and/or passive tissue stretch injuries to the peri

Lipid-Driven Immunometabolism in Mesenchymal Stromal Cells: A New Axis for Musculoskeletal Regeneration.

The immunosuppressive and anti-inflammatory potential of mesenchymal stromal cells (MSCs) underpins their therapeutic value in musculoskeletal disorders. However, the underlying mechanisms remain ill-defined. Traditionally associated with immune cells, immunometabolism (the cellular metabolism-immune system interplay) is now recognized as central in a broader range of processes, including tissue homeostasis, repair, and chronic inflammation. Depending on the context and cell type, distinct metabolic pathways (e.g., fatty acid oxidation, lipid mediator biosynthesis) can drive pro-inflammatory/p

Deciphering the toxicological mechanisms of per- and polyfluoroalkyl substances (PFAS) in musculoskeletal disorders via integrated network toxicology and molecular docking.

Per- and polyfluoroalkyl substances (PFAS) have been extensively used in everyday products; however, their effects on human health remain largely unknown. This study aimed to investigate the potential mechanisms by which PFAS may contribute to functional disorders in the human musculoskeletal system. Four representative musculoskeletal disorders were selected, which included intervertebral disc degeneration, myositis, osteoarthritis, and osteoporosis. Advanced network toxicology approaches integrating data from the Comparative Toxicogenomics Database (CTD) and GeneCards identified potential PF…

Rheumatic manifestations of sleep disorders.

PURPOSE OF REVIEW: We have long assumed that rheumatic pain causes sleep problems, fatigue, and functional disability. This paper reviews the accumulating evidence from human and animal experimental research studies that show a bidirectional relationship of disordered sleep to pain and fatigue. RECENT FINDINGS: The studies demonstrate that both disturbances of sleep and sleep restriction result in increased sensitivity to noxious stimuli and musculoskeletal pain symptoms. The notion of central nervous system hypersensitivity affecting widespread pain in patients with fibromyalgia syndrome is

[Hand-arm vibration syndrome and upper limbs diseases in the forest workers of Italia meridionale].

Vibration exposure of the hand-arm system is associated with an increased risk of upper-limb vascular, neurological and musculoskeletal lesions, or hand-arm vibration syndrome (HAVS). The prevalence of occupational HAVS and upper-limb disorders was studied among 278 Forestry Service workers in Sicily and Calabria. Subjects who used chain-saws (18 weeks/year) had a greater prevalence of peripheral sensory-neural disturbances (28%), upper-limb musculoskeletal disorders (33%) and carpal tunnel syndrome (19%) compared with 260 manual workers from the same Corps not exposed to hand-transmitted vibr

Prenatal sonographic diagnosis of focal musculoskeletal anomalies.

Focal musculoskeletal anomalies vary, and can manifest as part of a syndrome or be accompanied by numerous other conditions such as genetic disorders, karyotype abnormalities, central nervous system anomalies and other skeletal anomalies. Isolated focal musculoskeletal anomaly does, however, also occur; its early prenatal diagnosis is important in deciding prenatal care, and also helps in counseling parents about the postnatal effects of numerous possible associated anomalies. We have encountered 50 cases involving focal musculoskeletal anomalies, including focal limb dysplasia [radial ray abn

Quantitative MRI for Evaluation of Musculoskeletal Disease: Cartilage and Muscle Composition, Joint Inflammation, and Biomechanics in Osteoarthritis.

Magnetic resonance imaging (MRI) is a valuable tool for evaluating musculoskeletal disease as it offers a range of image contrasts that are sensitive to underlying tissue biochemical composition and microstructure. Although MRI has the ability to provide high-resolution, information-rich images suitable for musculoskeletal applications, most MRI utilization remains in qualitative evaluation. Quantitative MRI (qMRI) provides additional value beyond qualitative assessment via objective metrics that can support disease characterization, disease progression monitoring, or therapy response.
