From Signaling Pathways to Combination Therapy: Bioactive Compounds against Cancer Stem Cells.

Cancer Stem Cells (CSCs) represent a heterogeneous group of tumor cells that possess the innate ability to self-renew and differentiate, which also contributes to their resistance to first-line therapies. What sets CSCs apart from others is their crucial role in the recurrence of cancer, metastasis, and varied clinical responses against anti-cancer drugs, which makes them challenging to target. In recent years, there has been growing evidence that therapies capable of eliminating CSC niches or specifically targeting their core survival mechanisms are a potential means of providing a sustainabl

Multifaceted Attack Networks of Artemisinin in Reversing Chemoresistance in Colorectal Cancer.

Chemotherapy resistance in colorectal cancer (CRC) represents a critical clinical challenge leading to treatment failure and poor patient prognosis. Artemisinin is a natural product isolated from Artemisia annua, and its clinically relevant derivatives include dihydroartemisinin (DHA) and artesunate. Beyond their established antimalarial efficacy, both artemisinin and its derivatives-collectively referred to as artemisinin-derived compounds (ADs)-have been increasingly recognized for their unique potential to reverse multidrug resistance in cancer. Unlike previous reviews focusing on isolated

Unlocking ferroptosis to overcome cancer stem cells-mediated treatment failure and immune evasion.

Cancer stem cells (CSCs), a small subset of tumor cells with stem cell - like properties, are central to driving tumorigenesis and metastasis. Growing evidence highlights the importance of ferroptosis in maintaining CSCs, positioning ferroptosis as a promising therapeutic strategy. This review explores the interplay between ferroptosis and CSCs, which drive tumor initiation, therapy resistance, and recurrence. It summarizes the mechanisms of ferroptosis involving iron metabolism, lipid peroxidation, and antioxidant defense. The article highlights how non-coding RNAs, metabolic reprogramming, r

Arboviruses in Cancer therapy: pros and cons.

Arboviruses, arthropod-borne RNA viruses, are increasingly being recognized not only as causative agents of infectious diseases but also as promising tools in cancer therapy. Recent studies have demonstrated that certain arboviruses possess inherent oncolytic properties and can stimulate potent antitumor immune responses. This review critically examines the therapeutic potential of arboviruses in oncology alongside the key biosafety and translational challenges they present. Several members of the Flaviviridae, Reoviridae, Togaviridae, and Bunyaviridae families, such as Zika virus (ZIKV), Deng

The role of quantum dots in enhancing the therapeutic targeting of cancer stem cells.

In recent years, cancer stem cells have emerged as an interesting field in oncology due to their metastatic and resistance potential to chemotherapy and radiation therapy, thus resulting in the resurfacing of cancer even after multiple treatment attempts. The interest in these cells aims to address the key challenges associated with cancer treatments and to offer insights that may aid in better understanding the biology of cancer, with the possibility of introducing advanced or novel treatment methods. Conventional treatments often fail to eradicate the cancer stem cells, which then results in

Glioma-derived small extracellular vesicles induce pericyte-phenotype transition of glioma stem cells under hypoxic conditions.

BACKGROUND: Glioblastoma (GBM) is the most common and lethal primary brain tumor characterized by extensive vascularization. Anti-angiogenic therapy for this cancer offers the possibility of universal efficacy. However, preclinical and clinical studies suggest that anti-VEGF drugs, such as Bevacizumab, actively promote tumor invasion, which ultimately leads to a therapy-resistant and recurrent phenotype of GBMs. Whether Bevacizumab can improve survival over chemotherapy alone remains debated. Herein, we emphasize the importance of small extracellular vesicles (sEVs) internalization by glioma s

Heterogeneity of cancer stem cell-related marker expression is associated with three-dimensional structures in malignant pleural effusion produced by lung adenocarcinoma.

INTRODUCTION: Cancer stem cells have been described in lung adenocarcinoma-associated malignant pleural effusion. They show clinically important features, including the ability to initiate new tumours and resistance to treatments. However, their correlation with the three-dimensional tumour structures in the effusion is not well understood. METHODS: Cell blocks produced from lung adenocarcinoma patients' pleural effusion were examined for cancer stem cell-related markers Nanog and CD133 using immunocytochemistry. The three-dimensional cancer cell structures and CD133 expression patterns were

mTOR inhibition suppresses salinomycin-induced ferroptosis in breast cancer stem cells by ironing out mitochondrial dysfunctions.

Ferroptosis constitutes a promising therapeutic strategy against cancer by efficiently targeting the highly tumorigenic and treatment-resistant cancer stem cells (CSCs). We previously showed that the lysosomal iron-targeting drug Salinomycin (Sal) was able to eliminate CSCs by triggering ferroptosis. Here, in a well-established breast CSCs model (human mammary epithelial HMLER CD24low/CD44high), we identified that pharmacological inhibition of the mechanistic target of rapamycin (mTOR), suppresses Sal-induced ferroptosis. Mechanistically, mTOR inhibition modulates iron cellular flux and thereb

A Novel Type of Monocytic Leukemia Stem Cell Revealed by the Clinical Use of Venetoclax-Based Therapy.

UNLABELLED: The BCL2 inhibitor venetoclax has recently emerged as an important component of acute myeloid leukemia (AML) therapy. Notably, use of this agent has revealed a previously unrecognized form of pathogenesis characterized by monocytic disease progression. We demonstrate that this form of disease arises from a fundamentally different type of leukemia stem cell (LSC), which we designate as monocytic LSC (m-LSC), that is developmentally and clinically distinct from the more well-described primitive LSC (p-LSC). The m-LSC is distinguished by a unique immunophenotype (CD34-, CD4+, CD11b-,

Curcumin-based-fluorescent probes targeting ALDH1A3 as a promising tool for glioblastoma precision surgery and early diagnosis.

Glioblastoma (GBM) is the most aggressive primary brain tumour for which both effective treatments and efficient tools for an early-stage diagnosis are lacking. Herein, we present curcumin-based fluorescent probes that are able to bind to aldehyde dehydrogenase 1A3 (ALDH1A3), an enzyme overexpressed in glioma stem cells (GSCs) and associated with stemness and invasiveness of GBM. Two compounds are selective versus ALDH1A3, without showing any appreciable interaction with other ALDH1A isoenzymes. Indeed, their fluorescent signal is detectable only in our positive controls in vitro and absent in

Survival of salivary gland cancer stem cells requires mTOR signaling.

Advanced salivary gland mucoepidermoid carcinoma (MEC) is a relentless cancer that exhibits resistance to conventional chemotherapy. As such, treatment for patients with advanced MEC is tipically radical surgery and radiotherapy. Facial disfigurement and poor quality of life are frequent treatment challenges, and many patients succumb to loco-regional recurrence and/or metastasis. We know that cancer stem-like cells (CSC) drive MEC tumorigenesis. The current study tests the hypothesis that MEC CSC are sensitive to therapeutic inhibition of mTOR. Here, we report a correlation between the long-t

Adipose-derived stem cells in ovarian cancer progression, metastasis, and chemoresistance.

Ovarian cancer is the deadliest gynecological malignancy due to its symptomless early stage, metastasis, and high recurrence rate. The tumor microenvironment contributes to the ovarian cancer progression, metastasis, and chemoresistance. Adipose-derived stem cell in the tumor microenvironment of ovarian cancer, as a key player, interacts with ovarian cancer cells to form the cancer-associated fibroblasts and cancer-associated adipocytes, and secretes soluble factors to activate tumor cell signaling, which can promote ovarian cancer metastasis and chemoresistance. We summarize in this review th

Salinomycin Suppresses Tumorigenicity of Liver Cancer Stem Cells and Wnt/Beta-catenin Signaling.

BACKGROUND: Accumulating evidence has revealed the important role of Cancer Stem Cells (CSCs) in driving tumor initiation and tumor relapse or metastasis. Therapeutic strategies that selectively target CSCs may be effective approaches to eliminate cancer. Salinomycin, an antitumor agent, was identified as a selective inhibitor of several types of CSCs. We previously reported that salinomycin inhibits the migration and invasiveness of Liver Cancer Stem Cells (LCSCs). OBJECTIVE: This study was conducted to explore the role of salinomycin in suppressing the stemness properties of LCSCs and the m

Targeting Cancer Stem Cells to Overcome Therapy Resistance in Ovarian Cancer.

Ovarian cancer is the most lethal gynecological malignancy due to its late detection and high recurrence rate. Resistance to conventional platinum-based therapies and metastasis are attributed to a population of cells within tumors called cancer stem cells, which possess stem-like features and are able to recapitulate new tumors. Recent studies have deepened the understanding of the biology of ovarian cancer stem cells and their special properties and have identified multiple markers and signaling pathways responsible for their self-renewal abilities. Targeting cancer stem cells represents the

Cancer Stem Cells and Immunosuppressive Microenvironment in Glioma.

Glioma is one of the most common malignant tumors of the central nervous system and is characterized by extensive infiltrative growth, neovascularization, and resistance to various combined therapies. In addition to heterogenous populations of tumor cells, the glioma stem cells (GSCs) and other nontumor cells present in the glioma microenvironment serve as critical regulators of tumor progression and recurrence. In this review, we discuss the role of several resident or peripheral factors with distinct tumor-promoting features and their dynamic interactions in the development of glioma. Locali

CD114: A New Member of the Neural Crest-Derived Cancer Stem Cell Marker Family.

The neural crest is a population of cells in the vertebrate embryo that gives rise to a wide range of tissues and cell types, including components of the peripheral nervous system and the craniofacial skeleton as well as melanocytes and the adrenal medulla. Aberrations in neural crest development can lead to numerous diseases, including cancers such as melanoma and neuroblastoma. Cancer stem cells (CSCs) have been identified in these neural crest-derived tumors, and these CSCs demonstrate resistance to treatment and are likely key contributors to disease relapse. Patients with neural crest-der

ADARs Edit MicroRNAs to Promote Leukemic Stem Cell Activity.

Leukemic stem cells (LSCs) drive progression of chronic myeloid leukemia (CML) and tyrosine kinase inhibitor resistance through poorly understood mechanisms. Now in Cell Stem Cell, Zipeto et al. (2016) show targeting the RNA editing enzyme ADAR1 restores expression of let-7 and efficiently kills LSCs, providing an innovative therapeutic target in CML.

Dysplastic eosinophils in three patients with acute promyelocytic leukaemia.

Three cases of acute promyelocytic leukaemia (M3) with dysplastic eosinophils are reported. Promyelocytes from these cases showed classical features of hypergranular acute promyelocytic leukaemia. All the cases had minimal organomegaly and disturbances of coagulation were minimal. All the cases were associated with dysplastic eosinophils in the marrow but very few of these eosinophils were found in peripheral smear. One of the three patients presented with a mass in the vertebral column, a feature not uncommonly seen in a variant form of AML-M2 with eosinophilia. Dysplastic eosinophils appear
