Functional anatomy of movement disorders.

Models of basal ganglia function are described which encapsulate the principal pathophysiological mechanisms underlying parkinsonian akinesia on the one hand and abnormal involuntary movement disorders (dyskinesias) on the other. In Parkinson's disease, degeneration of the nigrostriatal dopamine system leads to overactivity of the 'indirect' striatopallidal projection to the lateral (external) segment of the globus pallidus. This causes inhibition of lateral pallidal neurons, which in turn project to the subthalamic nucleus. Disinhibition of the subthalamic nucleus leads to abnormal subthalami

Transmission of the cortical command for human voluntary movement through cervical propriospinal premotoneurons.

In man there is now strong evidence for the existence of a system of cervical propriospinal premotoneurons interposed in the corticospinal pathway to upper limb motoneurons. The relevant neurons are located rostral to upper limb motoneurons. They receive both weak excitation and strong inhibition from low threshold afferents in the upper limb. Ongoing EMG activity recorded in wrist and elbow extensors during voluntary movements of different types (tonic contraction, reaching) is suppressed by a cutaneous volley evoked by stimulation of the superficial radial nerve. There is evidence that this

Changes in retinal arbors in compressed projections to half tecta in goldfish.

In adult goldfish, electrophysiological studies have shown that the retinotectal projection reorganizes, following removal of half of the tectum, to form a complete but compressed projection over the remaining half tectum. As a result, each fiber terminates more rostrally than normal. Electron microscopic studies suggest a competition between retinal fibers for a fixed number of synaptic sites. The current study examines whether retinal arbors in the compressed projection are smaller than normal in extent or branching and whether the fiber paths in the tectum show the rostral movements and the

Connections of the posterior nucleus of the amygdala.

The connections of a relatively homogeneous band of neurons in the caudal amygdala have been examined with anterograde and retrograde axonal tracing methods in the rat. This region, called here the posterior nucleus of the amygdala (PA), corresponds in part to an area that has been referred to as the cortico-amygdaloid transition area, posterior part of the medial nucleus of the amygdala, amygdalo-hippocampal transition area, and posteromedial basal nucleus. Experiments with fluorogold and phaseolus vulgaris leucoagglutinin (PHAL) indicate that the major neuronal input to the PA arises in the

[The parasympathetic direct pathway from the midbrain to the ciliary muscle in cats and monkeys].

The visceral, namely the parasympathetic component of the oculomotor complex which directly innervates ciliary muscle was investigated using horseradish peroxidase (HRP) in three cats and three monkeys. A unilateral total iridectomy to abolish innervation to the iris was made in all animals. After 1 month to allow fiber degeneration in the iris, HRP was injected into the ciliary muscle of ipsilateral eye. Distribution of the labeled cells at the midbrain and at the ciliary ganglion was examined. Labeled cells were present in the ciliary ganglion and midbrain in all three cats and three monkeys

Organization of the zebra finch song control system: II. Functional organization of outputs from nucleus Robustus archistriatalis.

The organization of projections from nucleus robustus archistriatalis (RA) was mapped by a combination of anatomical tracer techniques. After injections of retrograde tracers in the syringeal part of the hypoglossal motor nucleus (nXIIts), labelled cells were seen in ipsilateral RA, in agreement with previous work. However, a shallow band in the dorsal and dorsocaudal part of RA did not contain labelled cells. Cells in this "cap" area were labelled following tracer injections in the dorsomedial nucleus of midbrain ICo (DM), the other known target of RA projections. The topography of outputs to

Olivary branching projections to the flocculus, nodulus and uvula in the rabbit. II. Retrograde double labeling study with fluorescent dyes.

The previous electrophysiological study reported that some of the Purkinje cells in the vestibulocerebellum were innervated by branching climbing fibers. To identify the cells of origin of these branching climbing fibers, a pair of retrograde fluorescent tracers, propidium iodide in combination with 4', 6-diamidino-2-phenylindole or diamidino yellow, were injected into two sites in the flocculus, nodulus and uvula. Proportions of double labeled neurons out of all labeled neurons in each subdivision of the contralateral inferior olive were studied. In cases of injections into the flocculus and

Neuronal pathways to the rat thyroid revealed by retrograde tracing and immunocytochemistry.

The distribution and origin of the nerve fibres innervating the rat thyroid were studied by immunocytochemistry, retrograde tracing and denervation experiments. Immunocytochemistry revealed nerve fibres containing noradrenaline, neuropeptide Y, vasoactive intestinal peptide, peptide histidine-isoleucine, galanin, substance P, neurokinin A and calcitonin gene-related peptide around blood vessels and follicles. Many of these transmitter candidates were found to co-exist with each other in different combinations in different subpopulations of neurons. Sympathectomy eliminated all noradrenaline- a

Cephalic sensory pathways in the central nervous system of larval Manduca sexta (Lepidoptera : Sphingidae).

Central projections of neurons innervating sensory structures on the head of larval Manduca sexta were traced by using methods of anterograde cobalt-diffusion. Regions of the deutocerebrum and tritocerebrum in the brain receive input from the antenna, labrum, maxilla, labial palps, hypopharynx and other unidentified regions of the buccal cavity. Antennal, maxillary and labial inputs project to the larval antennal centre (LAC) of the deutocerebrum. Stemmatal neurons and a few antennal neurons project into the protocerebrum. The suboesophageal ganglion (SEG) receives input from mechanosensory ne

Morphological features of identified trigeminocerebellar projection neurons in the border zone of rat trigeminal nucleus oralis.

The retrograde transport of horseradish peroxidase (HRP) was used to assess the overall morphology of neurons in the dorsal half of the border zone (BZ) of rat trigeminal nucleus oralis (Vo) that project to ipsilateral orofacial portions of four major tactile areas (crura I and II, the paramedian lobule, and the uvula) of the cerebellar cortex. We wished to answer two important questions: Does this group of cells consist of one or more morphologically distinct types, each projecting to different cerebellar tactile areas? Does the overall morphology or morphologies of these BZ neurons resemble

Circadian timekeeping in narcoleptic dogs.

Anatomical and functional aspects of the circadian timekeeping system containing the suprachiasmatic nucleus (SCN) were compared in normal and genetically narcoleptic dogs. The retinohypothalamic tract was delineated by tritiated amino acid autoradiography, the SCN was identified and examined by morphological techniques, and the circadian rhythm of melatonin concentrations in cerebrospinal fluid was measured by radioimmunoassay. Results suggest that the retinal input, cytoarchitecture, and essential timekeeping function of the SCN are intact in narcoleptic dogs.

Distribution in areas 18 and 19 of neurons projecting to the pontine nuclei: a quantitative study in the cat with retrograde transport of HRP-WGA.

Following large injections of horseradish peroxidase - wheat germ agglutinin in the pontine nuclei, corticopontine neurons in areas 18 and 19 were quantitatively mapped and flat maps showing the distribution of retrogradely labeled cells were constructed. The areal borders were defined either cyto- and myeloarchitectonically or from standard retinotopic maps presented in frontal sections (Tusa et al. 1981). Maps of the retinotopic organization in areas 18 and 19 (Tusa et al. 1979) were transferred to the present flat maps. Thus, the number and distribution of pontine projecting cells could be

Topographical projections from the posterior thalamic regions to the striatum in the cat, with reference to possible tecto-thalamo-striatal connections.

Projections from the posterior thalamic regions to the striatum were studied in the cat by the anterograde tracing method after injecting wheat germ agglutinin-horseradish peroxidase conjugate (WGA-HRP) into the caudalmost regions of the lateroposterior thalamic nucleus (caudal LP), suprageniculate nucleus (Sg) and magnocellular division of the medial geniculate nucleus (MGm). The results were further confirmed by the retrograde tracing method after injecting WGA-HRP into the regions of the caudate nucleus (Cd) and putamen (Put) where afferent fibers from the caudal LP, Sg and MGm were distrib

Spastic mutant axolotl: identification of a phenocopy pathway with implications for the control of axolotl swimming by the vestibulocerebellum.

The spastic mutant axolotl shows abnormal swimming behavior, which includes a preponderance of "embryonic" swimming elements (coils) versus mature swimming elements (sinusoids) and a failure to entrain sinusoids into a prolonged swimming sequence. The mutant also shows anatomical disorganization in the area acousticolateralis and cerebellar auricle, but it is unclear (1) to what extent the behavioral abnormalities are traceable to the vestibulocerebellar defect or (2) how the vestibulocerebellar pathway modulates swimming behavior in the normal axolotl. We have performed quantitative cine anal

Identification of cells of origin of tectopontine fibers in the cat superior colliculus: an experimental study with the horseradish peroxidase method.

The pontine projections from the superior colliculus in the cat have been studied by means of retrograde axonal transport of horseradish peroxidase (HRP). Following injections of HRP in the dorsolateral pontine nucleus, where the tectopontine fibers terminate, a fair number of labeled cells are found throughout the rostrocaudal extent of the ipsilateral superior colliculus. Relatively few of the labeled cells are of medium size (25-40 micron in diameter), more than 80% are small (10-25 micron), but no large cells are labeled. The cell bodies giving rise to tectopontine fibers are distributed i

Involvement of the liver-gut peripheral neural axis in nonalcoholic fatty liver disease pathologies via hepatic HTR2A.

Serotonin (5-HT) is one of the key bioamines of nonalcoholic fatty liver disease (NAFLD). Its mechanism of action in autonomic neural signal pathways remains unexplained; hence, we evaluated the involvement of 5-HT and related signaling pathways via autonomic nerves in NAFLD. Diet-induced NAFLD animal models were developed using wild-type and melanocortin 4 receptor (MC4R) knockout (MC4RKO) mice, and the effects of the autonomic neural axis on NAFLD physiology, 5-HT and its receptors (HTRs), and lipid metabolism-related genes were assessed by applying hepatic nerve blockade.
