Whole genome sequencing of SARS-CoV-2: evaluating the genetic diversity of variants circulating in Ebonyi State, Southeastern Nigeria.

BACKGROUND: COVID-19 is primarily considered a viral respiratory and vascular illness as its causative agent, SARS-CoV-2, predominantly targets the respiratory and vascular systems. Since being declared a global pandemic by the WHO in 2020, SARS-CoV-2 has spread to 223 countries with more than 281 million cases, and an estimated 5.4 million deaths reported globally. The cardinal aim of this study was to assess the genetic diversity of the circulating SARS-CoV-2 in Ebonyi State, Southeastern Nigeria. METHODS: Over a span of 24 calendar months (April, 2020 to March, 2022), a total of 20 clinica

From childhood malnutrition to adult mental illness: COVID-19 and the hidden legacy of malnutrition.

INTRODUCTION: The impact of stunting, thinness and obesity on depression and anxiety during challenging times, such as COVID-19, has not been adequately explored. This study aimed to estimate these relationships using longitudinal data from the four countries: Ethiopia, India, Peru and Vietnam. METHODS: We conducted an analysis using data from the Younger Cohort of the Young Lives study, which tracked 8000 children from 2002 to 2021. The first five rounds of the study assessed nutrition (stunting, thinness and obesity). The sixth round evaluated outcomes at three different time points. For th

Graphene foam biosensors for molecular diagnostics of antimicrobial resistance and emerging pathogen threats.

Pandemic preparedness for the surge of antimicrobial resistance (AMR) and newly emerging pathogens creates an urgent need for rapid diagnostic tools which can be mass manufactured and ideally used at the point of care. Rapid adaptation to new diagnostic targets is therefore a key requirement. To meet this need, we developed a sensitive electrochemical biosensor based on disposable graphene foam (GF) electrodes functionalised with peptide nucleic acid (PNA) probes, along with a rapid and generic functionalisation strategy. Pyrene-modified PNA probes were co-immobilised with pyrene-butanol on GF…

A Structure-Based Computational Pipeline for Broad-Spectrum Antiviral Discovery.

The rapid emergence of viruses with pandemic potential continues to pose a threat to public health worldwide. With the typical drug discovery pipeline taking an average of 5-10 years to reach clinical readiness, there is an urgent need for strategies to develop broad-spectrum antivirals that can target multiple viral family members and variants of concern. We present a structure-based computational pipeline designed to identify and evaluate broad-spectrum inhibitors across viral family members for a given target in order to support spectrum breadth assessment and prioritization in lead optimiz

TransFactor-prediction of pro-viral SARS-CoV-2 host factors using a protein language model.

MOTIVATION: Recent pandemics have revealed significant gaps in our understanding of viral pathogenesis, exposing an urgent need for methods to identify and prioritize key host proteins (host factors) as potential targets for antiviral treatments. De novo generation of experimental datasets is limited by their heterogeneity, and for looming future pandemics, may not be feasible due to limitations of experimental approaches. RESULTS: Here, we present TransFactor, a computational framework for predicting and prioritizing candidate host factors using only protein sequence data. It leverages the p

On-site airborne pathogen detection for infection risk mitigation.

Human-infecting pathogens that transmit through the air pose a significant threat to public health. As a prominent instance, the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) that caused the COVID-19 pandemic has affected the world in an unprecedented manner over the past few years. Despite the dissipating pandemic gloom, the lessons we have learned in dealing with pathogen-laden aerosols should be thoroughly reviewed because the airborne transmission risk may have been grossly underestimated. From a bioanalytical chemistry perspective, on-site airborne pathogen detection can be

Influenza A virus replication has a stronger dependency on Raf/MEK/ERK signaling pathway activity than SARS-CoV-2.

The recent COVID-19 pandemic again highlighted the urgent need for broad-spectrum antivirals, both for therapeutic use in acute viral infection and for pandemic preparedness in general. The targeting of host cell factors hijacked by viruses during their replication cycle presents one possible strategy for development of broad-spectrum antivirals. By inhibiting the Raf/MEK/ERK signaling pathway, a central kinase cascade of eukaryotic cells, which is being exploited by numerous viruses of different virus phyla, the small-molecule MEK inhibitor zapnometinib has the potential to address this need.…

Developing a Video Game as an Awareness and Research Tool Based on SARS-CoV-2 Epidemiological Dynamics and Motivational Perspectives.

In mid-2020, the University of Liège (ULiège, Belgium) commissioned the ULiège Video Game Research Laboratory (Liège Game Lab) and the AR/VR Lab of the HEC-Management School of ULiège to create a serious game to raise awareness of preventive measures for its university community. This project has its origins in two objectives of the institutional policy of ULiège in response to the crisis caused by SARS-CoV-2 to raise awareness among community members of various preventive actions that can reduce the spread of the virus and to inform about the emergence and progression of a pandemic. After alm

COVID-19 data reporting systems in Africa reveal insights for future pandemics.

Globally, countries have used diverse methods to report data during the COVID-19 pandemic. Using international guidelines and principles of emergency management, we compare national data reporting systems in African countries in order to determine lessons for future pandemics. We analyse COVID-19 reporting practices across 54 African countries through 2020. Reporting systems were diverse and included summaries, press releases, situation reports and online dashboards. These systems were communicated via social media accounts and websites belonging to ministries of health and public health. Data

End-of-Life Issues in the Era of the COVID-19 Pandemic.

PURPOSE: The coronavirus disease 2019 (COVID-19) pandemic has brought the world to a standstill and has exposed the lack of preparedness of most nations' health care systems. Even in usual times, palliative care has not received its fair share of recognition as an important component of patient care; instead, the emphasis is often placed on aggressive patient management. Now, with the entire medical community and decision-making committees focussed on intensive patient care, end-of-life care has taken a backseat. METHODS: This article is a brief communication. RESULTS: COVID 19 infection has

A consensus-hemagglutinin-based vaccine delivered by an attenuated Salmonella mutant protects chickens against heterologous H7N1 influenza virus.

H7N3 and H7N7 are highly pathogenic avian influenza (HPAI) viruses and have posed a great threat not only for the poultry industry but for the human health as well. H7N9, a low pathogenic avian influenza (LPAI) virus, is also highly pathogenic to humans, and there is a great concern that these H7 subtypes would acquire the ability to spread efficiently between humans, thereby becoming a pandemic threat. A vaccine candidate covering all the three subtypes must, therefore, be an integral part of any pandemic preparedness plan. To address this need, we constructed a consensus hemagglutinin (HA) s…

Media use and communication inequalities in a public health emergency: a case study of 2009-2010 pandemic influenza A virus subtype H1N1.

OBJECTIVES: Studies have shown that differences among individuals and social groups in accessing and using information on health and specific threats have an impact on their knowledge and behaviors. These differences, characterized as communication inequalities, may hamper the strength of a society's response to a public health emergency. Such inequalities not only make vulnerable populations subject to a disproportionate burden of adversity, but also compromise the public health system's efforts to prevent and respond to pandemic influenza outbreaks. We investigated the effect of socioeconomi

Keeping the memory of influenza viruses.

Protection against pathogens is mediated by both humoral responses (neutralizing antibodies) and cellular immunity, both CD4+ and CD8+ cells. In the case of influenza viruses, circulating strains contain both variable and conserved T and B cell epitopes that are challenged after vaccination and/or infection. During infection, the role of T cells is to prevent viral dissemination in the organism by killing the infected cells and helping B cell antibody production to neutralize the virus. The threat of influenza virus increases the preparedness of protective immunity to pandemic and seasonal inf

A Cross-sectional Survey of Public Knowledge and Perspective on Coronavirus Disease, Vaccination, and Related Research in India during the COVID-19 Pandemic.

BACKGROUND AND RATIONALE: The coronavirus disease 2019 (COVID-19) pandemic has left no person unexposed to the wisdom about the need for human preparedness to tackle future pandemics irrespective of individual caste, race, religion, and education status. The extent of this change in knowledge and public perspective is difficult to measure in a populous nation like India subjected to individual freedom and cultural beliefs. Hence, we planned this study with the objective of evaluating the knowledge and perception of the Indian public towards COVID-19 disease, vaccination and research activities
