Relationship of nervous tissue transketolase to the neuropathy in chronic uremia.

Patients with chronic uremia develop neurologic defects which are similar to the demyelinating lesions seen in thiamine deficiency. The present study describes inhibitory effects of uremic material on nervous tissue transketolase, a thiamine-dependent enzyme of the pentose phosphate pathway which has been reported to have functional importance in the metabolism of myelinated nervous structures. Transketolase activity (TKA) of normal human brain and spinal cord was measured by the conversion of ribose-5-phosphate (R5P) to sedoheptulose-7-phosphate (S7P). TKA was significantly inhibited by plasm

Role of oxidant stress in lawsone-induced hemolytic anemia.

Lawsone (2-hydroxy-1,4-naphthoquinone) is the active ingredient of henna (Lawsonia alba), the crushed leaves of which are used as a cosmetic dye. Application of henna can induce a severe hemolytic anemia, and lawsone is thought to be the causative agent. Administration of lawsone to rats has been shown to induce a hemolytic response that is associated with oxidative damage to erythrocytes. However, direct exposure of isolated erythrocytes to lawsone did not provoke oxidative damage, suggesting that lawsone must undergo extra-erythrocytic bioactivation in vivo. In the present study, the surviva

Krüppel-like factor 1 is a core cardiomyogenic trigger in zebrafish.

Cardiac regeneration requires dedifferentiation and proliferation of mature cardiomyocytes, but the mechanisms underlying this plasticity remain unclear. Here, we identify a potent cardiomyogenic role for Krüppel-like factor 1 (Klf1/Eklf), which is induced in adult zebrafish myocardium upon injury. Myocardial inhibition of Klf1 function does not affect heart development, but it severely impairs regeneration. Transient Klf1 activation is sufficient to expand mature myocardium in uninjured hearts. Klf1 directs epigenetic reprogramming of the cardiac transcription factor network, permitting coord

Malate-aspartate shuttle inactivation drives pulmonary hypertension through glucose carbon flux rewiring and α-ketoglutarate-dependent epigenetic remodeling.

Pulmonary hypertension (PH) is characterized by progressive pulmonary vascular remodeling, primarily driven by the hyperproliferation of pulmonary artery smooth muscle cells (PASMCs) and their resistance to apoptosis. Metabolic reprogramming, particularly enhanced glycolysis, has been proposed to be involved in PASMCs activation. However, the implicated mechanisms remain incompletely understood. Here, using in-vitro cultured PASMCs and in-vivo PH models, we showed that malate-aspartate shuttle (MAS), a critical system connecting glycolysis to oxidative phosphorylation, was significantly inhibi

Microcella aerolata GA224 exhibits preventive potential against Streptococcus pneumoniae infection via the gut-lung axis.

Antibiotics and vaccines have long been major key interventions against Streptococcus pneumoniae (Spn) infection. However, alternative therapies are urgently needed with the original therapies becoming suboptimal efficacy. A beneficial bacterium, Microcella aerolata strain GA224, with protective potential against Spn infection was isolated in previous study. Here, this protective effect was investigated at the bacterial, cellular and animal levels, exploring the mechanisms from the perspective of the gut-lung axis. Cellular and animal models of Spn infection were established and GA224 was admi

A response to iron involving carbon metabolism in the opportunistic fungal pathogen Candida albicans.

UNLABELLED: Iron (Fe) is an essential micronutrient, and during infection, the host attempts to starve pathogens of this vital element through a process known as nutritional immunity. Successful pathogens have evolved means to evade this attack, an example being Candida albicans, the most prevalent human fungal pathogen. When Fe-starved, C. albicans induces multiple pathways for Fe uptake using the SEF1 trans-regulator, and we now describe a previously unrecognized effect of Fe on C. albicans metabolism that occurs independent of SEF1. Specifically, Fe limitation leads to inhibition of pyruvat

Multi-omics dissection of CO2-induced metabolic reprogramming in broccoli: key regulators of growth and nutritional quality.

The rising global CO2 level influences plant growth, productivity, and nutritional quality, and elevated CO2 (ECO2) is commonly utilized in greenhouses to enhance vegetable crop yields. However, the molecular mechanism underlying plant responses to ECO2 remains unclear. This study examined the impacts of ECO2 on the nutritional quality and growth characteristics of two widely cultivated broccoli varieties, 'Yanxiu' and 'Qiuli'. A pot experiment was carried out in environmentally auto-controlled growth chambers over a period of 100 days, under both ambient CO2 (ACO2, 420/470 ppm, daytime/nightt

Differential metabolic pathways underlie THC- and CBD-mediated inhibition of B-cell activation in both young and aged mice.

OBJECTIVE: B lymphocytes play a crucial role in immunity but also contribute to the pathogenesis of various diseases. Cannabis plants produce numerous biologically active compounds, including cannabinoids. The two most studied phytocannabinoids are Δ9-tetrahydrocannabinol (THC) and cannabidiol (CBD). These cannabinoids exert diverse and potent biological effects primarily through the endocannabinoid system (ECS), which also plays a key role in mature B-cell function. Both the immune system and the ECS undergo age-related changes that lead to a clinically significant decline in function. METHO

Dependence of Mycoplasma bovis on a novel nucleoside transporter for survival in association with host cells.

UNLABELLED: Transport systems play a vital role in metabolism in mycoplasmas by facilitating the uptake of essential nutrients from host cells, on which mycoplasmas are highly dependent. Co-culture of Mycoplasma bovis with Madin-Darby bovine kidney cells has shown that the gene MBOVPG45_0748 is essential for the survival of M. bovis in association with host cells. To characterize the function of this protein, intracellular and extracellular metabolomic profiles of a mutant strain with a transposon inserted into the MBOVPG45_0748 gene (∆MBOVPG45_0748) were compared to those of the parent strain

Stimbiotic and wheat bran inclusion in maize or wheat-based diets for broiler chickens; effects on jejunal histomorphology, digesta oligosaccharides and caecal short-chain fatty-acids profiles.

1. A stimbiotic (STB) is any feed additive that stimulates caeca fibre fermentation, although the additive itself contributes little to the caeca short-chain fatty acid (SCFA) production. A 42 d experiment investigated the interactive effects of STB and wheat bran (WB) in broiler chickens receiving maize or wheat-based diets.2. The treatments were arranged in a 2 × 2 × 2 factorial (eight replicates each), the dietary factors being diet (maize-SBM or wheat-SBM), STB (with or without) and WB (0 or 50 g/kg). Jejunal tissue, gizzard, jejunal and ileal digesta and caecal contents were collected on

Changes in composition and concentration of differential metabolites in root exudates are associated with aluminum-tolerance of Ricinus communis under a high CO2 environment.

Root exudates are the most direct performance for plants responding to adverse environments, and are also important media for materials exchange, energy transmission and information communication between the roots and rhizosphere. However, how plant roots and exudates respond to aluminum (Al) stress under elevated CO2 concentration (eCO2) is still unclear. Ricinus communis is a famous oilseed crop throughout the world, which has strong tolerance to metal contaminated soil. In the present study, root physiological changes and the exudates of this species under aluminum stress and eCO2 based on

Evaluating the phytotoxicological effects of bulk and nano forms of zinc oxide on cellular respiration-related indices and differential gene expression in Hordeum vulgare L.

The increasing use of nanoparticles is driving the growth of research on their effects on living organisms. However, studies on the effects of nanoparticles on cellular respiration are still limited. The remodeling of cellular-respiration-related indices in plants induced by zinc oxide nanoparticles (nnZnO) and its bulk form (blZnO) was investigated for the first time. For this purpose, barley (Hordeum vulgare L.) seedlings were grown hydroponically for one week with the addition of test compounds at concentrations of 0, 0.3, 2, and 10 mg mL-1. The results showed that a low concentration (0.3

PPM1K-regulated impaired catabolism of branched-chain amino acids orchestrates polycystic ovary syndrome.

BACKGROUND: Polycystic ovary syndrome (PCOS) is one of the most common diseases with the coexistence of reproductive malfunction and metabolic disorders. Previous studies have found increased branched chain amino acid (BCAA) levels in women with PCOS. However, it remains unclear whether BCAA metabolism is causally associated with the risk of PCOS. METHODS: The changes of BCAA levels in the plasma and follicular fluids of PCOS women were detected. Mendelian randomization (MR) approaches were used to explore the potential causal association between BCAA levels and the risk of PCOS. The function

Hemolysis and Metabolic Lesion of G6PD Deficient RBCs in Response to Dapsone Hydroxylamine in a Humanized Mouse Model.

Glucose 6-phosphate dehydrogenase (G6PD) deficiency is the most common enzymopathy in humans (∼5% of all individuals). G6PD deficiency (G6PDd) is caused by an unstable enzyme and manifests most strongly in red blood cells (RBCs) that cannot synthesize new protein. G6PDd RBCs have decreased ability to mitigate oxidative stress due to lower levels of NADPH, as a result of a defective pentose phosphate pathway. Accordingly, oxidative drugs can result in hemolysis and potentially life-threatening anemia in G6PDd patients. Dapsone is a highly useful drug for treating a variety of pathologies but or
