Synergistic inhibitory effects of gastrin and histamine receptor antagonists on Helicobacter-induced gastric cancer.

BACKGROUND & AIMS: Apart from its importance as an acid secretogogue, the role of histamine as a downstream target of gastrin has not been fully explored. Previous studies have shown that the combination of hypergastrinemia and Helicobacter infection resulted in accelerated gastric cancer in mice. We used this model to examine the role of cholecystokinin 2 (CCK2)/gastrin receptor and histamine H2-receptor signaling in the development of gastric atrophy and cancer. METHODS: Male hypergastrinemic mice (INS-GAS mice) were infected with Helicobacter felis and given the CCK2/gastrin receptor antag

Venetoclax Synergizes With Regorafenib for Colorectal Cancer by Targeting BCL-2.

Despite notable advancements in therapeutic modalities, many patients with colorectal cancer (CRC) exhibit inadequate response to regorafenib, largely due to the propensity for drug resistance. Deeper insights into the mechanism of CRC sensitivity to regorafenib therapy are urgently required. The antiapoptotic protein B-cell lymphoma 2 (BCL-2) is closely associated with a variety of malignancies. Therefore, this study investigated the role of BCL-2 in promoting regorafenib resistance in colorectal cancer. Venetoclax, a BCL-2 antagonist, potentiates the antitumor activity of regorafenib. The co

Analysis of the behaviour of selected CCKB/gastrin receptor antagonists in radioligand binding assays performed in mouse and rat cerebral cortex.

1. The previously described complex behaviour of the CCKB/gastrin receptor antagonist, L-365,260, in radioligand binding assays could be explained by a variable population of two binding sites. We have investigated whether other CCKB/gastrin receptor ligands (PD134,308, PD140,376, YM022 and JB93182) can distinguish between these sites. 2. In the mouse cortex assay, Hill slopes were not different from unity and the ligand pKI values did not differ when either [125I]-BH-CCK-8S or [3H]-PD140,376 was used as label as expected for a single site (G2). 3. In the rat cortex, where previous analysis of

Stereotactic body radiotherapy plus lenvatinib and sintilimab with and without transarterial embolization for advanced hepatocellular carcinoma with portal vein tumor thrombus: a dual-center, propensity score-matched retrospective analysis.

INTRODUCTION: Portal vein tumor thrombus (PVTT) in hepatocellular carcinoma (HCC) is associated with poor prognosis and limited efficacy of current first-line therapies. Combining locoregional and systemic therapies may enhance antitumor immunity. However, the safety and efficacy of dual locoregional therapy (LRT) with stereotactic body radiotherapy (SBRT) and transarterial embolization (TAE) along with targeted immunotherapy is unclear. METHODS: In this retrospective real-world study, we analyzed 204 patients with Barcelona Clinic Liver Cancer stage C HCC and PVTT treated with SBRT plus lenv

A case of a patient with advanced HCC with Vp4 on hemodialysis who responded to a multimodality treatment including hepatic arterial infusion, radiation therapy, immunotherapy, and molecular-targeted drugs.

We report a rare case of a 73-year-old woman on chronic hemodialysis who was diagnosed with unresectable advanced hepatocellular carcinoma (HCC) with Vp4 (portal vein tumor thrombosis in the main trunk) at the initiation of dialysis. Initial treatment consisted of hepatic arterial infusion chemotherapy with cisplatin (CDDP) as a one-shot infusion and radiotherapy targeting the main tumor and Vp4, followed by combination therapy with atezolizumab and bevacizumab (Atezo-Bev). Tumor markers decreased but complete response (CR) was not achieved. As second-line therapy, lenvatinib preceded by CDDP

A biomimetic phase-separating hydrogel potentiates local chemo-virotherapy to eradicate recurrent glioblastoma.

Combining local oncolytic virotherapy with chemotherapy offers a logical strategy to prevent glioblastoma (GBM) recurrence, yet physical and biological incompatibilities between viruses and drugs restrict their co-administration. Here, inspired by viral liquid-liquid phase separation (LLPS), we developed a biomimetic phase-separating hydrogel (PSgel) that enables the spatially segregated and sustained co-delivery of oncolytic herpes simplex virus 2 (HSV-2) and the multi-kinase inhibitor sorafenib. Mechanistically, the chemo-viral combination cooperatively depletes the repressive histone mark H

Association between gut microbiota characteristics and therapeutic efficacy of multimodal combination therapy in hepatocellular carcinoma with portal vein tumor thrombus: a secondary analysis of a prospective phase II trial.

BACKGROUND: Hepatocellular carcinoma (HCC) with portal vein tumor thrombus (PVTT) carries a poor prognosis. Although SBRT combined with cadonilimab and lenvatinib has shown encouraging antitumor activity in a prior prospective phase II trial, marked interpatient heterogeneity remains. Gut microbiota may influence antitumor immunity and could help identify patients more or less likely to benefit from this regimen. OBJECTIVE: To characterize baseline and longitudinal gut microbiota features in treatment-naïve HCC patients with PVTT receiving SBRT plus cadonilimab and lenvatinib, and to explore

SOX11 exacerbates ferroptosis to reduce lenvatinib resistance in liver cancer cells by promoting ubiquitination degradation of SREBF1 through upregulating UBE3A.

Lenvatinib is one of the most commonly used first-line drugs for liver cancer. However, lenvatinib resistance occurs in a large proportion of patients, posing a significant challenge. Ferroptosis, an iron-dependent form of cell death, plays a pivotal role in overcoming drug resistance. This study investigates the role of SRY-related HMG-box transcription factor 11 (SOX11) in regulating lenvatinib resistance in liver cancer through its impact on ferroptosis. qRT-PCR, western blot, and immunohistochemistry were performed to examine the expression of key molecules in patient samples and cell line

Protocadherin 17 weakens the lenvatinib resistance of liver cancer through inducing ferroptosis.

Lenvatinib has been employed in the treatment of advanced liver cancer; however, its clinical application is significantly impeded by frequent drug resistance. Recent studies have revealed that lenvatinib treatment triggers ferroptosis in liver cancer cells, providing a novel approach to addressing lenvatinib resistance. In this study, we initially validated the induction of ferroptosis by lenvatinib in liver cancer cells. Remarkably, protocadherin 17 (PCDH17), an adhesion-related protein, was found to be down-regulated in liver cancer, and overexpression of PCDH17 could induce ferroptosis in

The Alpha-7 Nicotinic Receptor Positive Allosteric Modulator PNU120596 Attenuates Lipopolysaccharide-Induced Depressive-Like Behaviors and Cognitive Impairment by Regulating the PPAR-α Signaling Pathway in Mice.

BACKGROUND AND OBJECTIVE: The brain α7 nicotinic acetylcholine receptor (α7 nAChR) has a critical role in the pathophysiology of Major Depressive Disorder (MDD) involving neuroinflammation. The α7 nAChR stimulation has been shown to modulate the anti-inflammatory effects of nuclear peroxisome proliferator-activated receptor-α (PPAR-α) via its endogenous ligands in the brain. The present study determined the effects of α7 nAChR modulator PNU120596 on PPAR-α, an inhibitor of κB (IκB) and nuclear factor-κB (NF-κB) expression and interleukin-1β (IL-1β) level in the hippocampus and prefrontal corte

Deciphering the role of the MALT1-RC3H1 axis in regulating GPX4 protein stability.

Ferroptosis, a unique form of iron-dependent cell death triggered by lipid peroxidation accumulation, holds great promise for cancer therapy. Despite the crucial role of GPX4 in regulating ferroptosis, our understanding of GPX4 protein regulation remains limited. Through FACS-based genome-wide CRISPR screening, we identified MALT1 as a regulator of GPX4 protein. Inhibition of MALT1 expression enhances GPX4 ubiquitination-mediated degradation by up-regulating the E3 ubiquitin ligase RC3H1. Using both rescue assays and functional genetic screening, we demonstrate that pharmacologically targeting
