Different distribution of phosphorylated tau protein isoforms in Alzheimer's and Pick's diseases.

Tau proteins aggregate into different neuronal inclusions in several neurodegenerative disorders. In Alzheimer's disease (AD), hyperphosphorylated Tau from paired helical filaments (PHF) of neurofibrillary tangles, named PHF-Tau, have an electrophoretic profile with four main bands (Tau 55, 64, 69, 74 kDa). In Pick's disease, phosphorylated Tau from Pick bodies are made of two major components (Tau 55, 64 kDa) and a minor 69 kDa. Here we show, using specific antibodies against translated exon 2, 3 or 10 of Tau isoforms, that the set of Tau isoforms engaged in the most insoluble part of PHF in

On the regulation of the expressed L-type calcium channel by cAMP-dependent phosphorylation.

The Ca2+ channel subunits alpha 1C-a and alpha 1C-b were stably expressed in Chinese hamster ovary (CHO) and human embryonic kidney (HEK) 29 … X. These results indicate that the L-type Ca2+ channel expressed is not modulated by cAMP-dependent phosphorylation to the same extent as in native cardiac myocytes.

Site-specific modulation of c-Myc cotransformation by residues phosphorylated in vivo.

c-Myc is a nuclear phosphoprotein which binds DNA as a heterodimer with Max. We have identified two in vivo phosphorylation sites, Thr58 and Ser62, within a domain highly conserved among all Myc family members. Thr58 is mutated in several viral forms of the protein and constitutes a mutational hot-spot in Burkitt's lymphoma. Members of the GSK-3 and MAP kinase families, but not CKII, specifically phosphorylated these sites in vitro. The effect of these phosphorylation sites on Myc function was assessed by cotransformation of primary rat embryo fibroblasts with Ras. Mutagenesis of Thr58 to alan…

Dephosphorylation of Alzheimer's disease abnormally phosphorylated tau by protein phosphatase-2A.

Microtubule-associated protein tau is abnormally hyperphosphorylated in the brain of patients with Alzheimer's disease, and is the major protein subunit of paired helical filaments. There is also a significant pool of non-paired helical filament abnormally phosphorylated tau in Alzheimer's disease brain. In the present study, the site-specific dephosphorylation of this Alzheimer's disease abnormally phosphorylated tau by protein phosphatase-2A was studied and compared with that by protein phosphatase-2B. The dephosphorylation was detected by its interaction with several phosphorylation-depende…

[Influence of phosphorylation on the functional properties of the sodium channel reconstructed in an artificial membrane].

Phosphorylation of the reconstructed TTX-sensitive cytosolic protein of the bovine brain has been studied. Some properties of the protein are similar to those of the membrane potential-dependent sodium channel. It is shown that the influence of phosphorylation by protein kinase A on the reconstructed channel greatly depends on the mode of reconstruction. Phosphorylation fo reconstructed channels in the open state leads to their closing. Preliminary phosphorylation of channel-forming protein results in a considerable increase of the activation effect of veratrine and scorpion toxin.

Structure and antigenicity of the phosphorylated lipopolysaccharide antigens from the leprosy and tubercle bacilli.

A family of major arabinose- and mannose-containing phosphorylated lipopolysaccharides was isolated from Mycobacterium leprae and Mycobacterium tuberculosis. The only antigenic member of the family, lipoarabinomannan (LAM)-B, was purified by anion exchange and gel filtration chromatography in detergent and recovered in large quantities (15 mg/g of bacteria). It yielded a broad diffuse band on polyacrylamide gel electrophoresis but appeared homogeneous by this criterion and gel filtration. Besides arabinose and mannose, it contained glycerol and a polyol phosphate and was acylated by lactate, s

Tau proteins-enhanced Ca2+/calmodulin (CaM)-dependent phosphorylation by the brain supernatant of diisopropyl phosphorofluoridate (DFP)-treated hen: tau mutants indicate phosphorylation of more amino acids in tau by CaM kinase II.

Diisopropyl phosphorofluoridate (DFP) produces organophosphorus ester-induced delayed neurotoxicity (OPIDN) in hen, human, and other sensitive species. A single dose of DFP (1.7 mg/kg, s.c.) produces mild ataxia in 7-14 days in hens, followed by progression to severe ataxia or paralysis. We studied the effect of DFP administration on Ca2+/calmodulin-dependent phosphorylation of tau proteins by the brain supernatants of control and DFP-treated hens. Brain supernatants from DFP-treated hens showed enhanced in vitro phosphorylation of htau40 and its various mutants, but no change in the two-dimen…

WOX1 is essential for tumor necrosis factor-, UV light-, staurosporine-, and p53-mediated cell death, and its tyrosine 33-phosphorylated form binds and stabilizes serine 46-phosphorylated p53.

WW domain-containing oxidoreductase WOX1, also named WWOX or FOR, undergoes Tyr33 phosphorylation at its first N-terminal WW domain and subsequent nuclear translocation in response to sex steroid hormones and stress stimuli. The activated WOX1 binds tumor suppressor p53, and both proteins may induce apoptosis synergistically. Functional suppression of WOX1 by antisense mRNA or a dominant negative abolishes p53-mediated apoptosis. Here, we determined that UV light, anisomycin, etoposide, and hypoxic stress rapidly induced phosphorylation of p53 at Ser46 and WOX1 at Tyr33 (phospho-WOX1) and thei…

Antiviral innate immunity induces alpha synuclein phosphorylation at serine129 in neurons independent of aggregation.

Alpha synuclein (asyn) is expressed in neurons and is associated with the pathogenesis of synucleinopathies like Parkinson's Disease. Asyn aggregates are associated with phosphorylation at serine position 129 (pS129), which can be induced by environmental triggers, including viral infection, and the risk increases with aging. We show that brain tissue from West Nile Virus (WNV) infected patients exhibit increased expression of pS129 asyn in neurons of brain tissue. We found that WNV infection, Poly I:C treatment, and type 1 interferon (T1 IFN) treatment elicit the acute induction, followed by …

Prochlorperazine inhibits the proliferation of hepatocellular carcinoma cells by promoting phosphorylation of β-catenin.

Hepatocellular carcinoma (HCC) is the most prevalent form of primary liver cancer and ranks as the third leading cause of cancer-related fat … use model. Subsequent studies have revealed that PCZ can directly bind to β-catenin and enhance the phosphorylation level of β-catenin. This, in turn, inhibits the Wnt signaling pathway, thereby exerting its anticancer effects. This novel finding sheds light on the potential molecular mechanisms of PCZ as a therapeutic strategy for HCC. In summary, our study prov …

Proteoform-Resolved Cross-Linking Reveals Environment-Dependent Structural Effects of α-Synuclein S129 Phosphorylation.

Intrinsically disordered proteins (IDPs) drive many neurodegenerative disorders, but their structures remain difficult to define because they populate dynamic ensembles that change with the chemical environment. This problem is central for α-synuclein (aSyn), a Parkinson's disease-linked IDP in which S129 phosphorylation is highly enriched in disease-associated aggregates. Although S129 phosphorylation stabilizes a more compact aSyn ensemble in dilute solution, whether this structural effect persists across other biochemical environments has remained unknown. Here, we developed a quantitative …

Structural and molecular basis of O-GlcNAcylation-phosphorylation crosstalk in protein Kinase-B (Akt2).

Post Translational Modifications significantly influence conformational preferences and activity of proteins. In Protein Kinase-B (Akt2), numerous residues undergo phosphorylation and O-GlcNAcylation, in same region. This results in a crosstalk mechanism modulating Akt2 activity, and is dysregulated in metabolic diseases such as type-II diabetes. However, atomic level details of how such modifications influence Akt2 dynamics and activity is poorly understood. In this work, we employed in silico approaches to explore the conformational preferences and functional consequences of Akt2 in O-GlcNAc…

Site-specific phosphorylation modulates p16/CDK4 binding dynamics and energetics: Insights from molecular simulations.

Phosphorylation of the tumor suppressor p16INK4a is known to influence its inhibitory interaction with cyclin dependent kinase 4 (CDK4), yet the structural and energetic consequences of site specific modifications remain unclear. Here, we employed all-atom molecular dynamics simulations to systematically investigate Wild-type and single-site phosphorylated p16 variants in complex with CDK4, aiming to decipher phosphorylation driven changes in conformational dynamics, interfacial stability, and binding energetics. Region wise analyses revealed distinct alterations in flexibility and stabilizati…

CMV-GLA: contrastive multi-view graph layer attention for predicting phosphorylation site-disease associations.

Protein phosphorylation is a critical posttranslational modification involved in cell signaling and metabolic regulation, and phosphorylation dysregulation is closely associated with various diseases. However, studies on phosphorylation site-disease associations remain scarce, and existing methods often fail to fully leverage complementary information across heterogeneous data sources, resulting in suboptimal predictive performance. We present CMV-GLA (Contrastive Multi-View Graph Layer Attention Network), a novel framework integrating protein sequences, disease semantics, and known associatio…
