Dynamics of leukocyte populations, immune-regulatory cytokines, and biochemical parameters in wild boar and domestic pigs experimentally infected with a virulent African swine fever virus genotype II strain.

African Swine Fever Virus (ASFV) is the causative agent of devastating disease affecting domestic and wild pigs globally. A previous study of the intranasal inoculation of domestic pigs (DP) and wild boar (WB) with the ASFV genotype II strain "Armenia 2007" demonstrated distinct disease outcomes. This study aims to compare the leukocyte, cytokine and biochemical profiles in experimentally infected DP and WB. Blood and serum samples were collected before infection (day 0), from animals euthanized in groups of six (comprising 3 DP and 3 WB) on days 1, 2, 3 and 5 post infection (pi) and from anim

Characterization of α, β, and muscarinic receptor distribution in porcine ureters: a translational model for pharmacological ureteral dilation.

Intraoperative ureteral dilation through smooth muscle relaxation is of interest for improving upper urinary tract access during retrograde intrarenal surgery. Porcine ureters are widely used translational models due to their anatomical similarities to humans; however, the distribution of receptors essential for pharmacological feasibility studies has not been fully characterized. This ex vivo study defines the regional expression of adrenergic and muscarinic receptors throughout the porcine ureter. Ureters from juvenile female Yorkshire swine (n = 4) were segmented into renal pelvis, proximal

Immunopathogenesis and pathological features of NADC34-like PRRSV infection in pregnant sows during late gestation.

The recently emerged NADC34-like PRRSV-2 strain (Lineage 1A) has been increasingly linked to severe reproductive failures in sow herds, yet its immunopathogenic mechanisms remain poorly understood, especially in pregnant sow models. In this study, we compared the pathogenic and immunological characteristics of a NADC34-like isolate (JBNU-22-N01) with a prevalent NADC30-like strain (PJ73, Lineage 1C) in pregnant sows during late gestation. JBNU-22-N01 infection resulted in earlier peak viremia, more severe maternal pathology, and greater fetal growth restriction compared with PJ73, despite comp

Influence of Chlamydia suis on reproductive outcome in farms with and without tetracycline treatment of sows.

Porcine genital chlamydiosis is considered a potential cause of reproductive failure, with Chlamydia (C.) suis being the most prevalent chlamydial species. To assess its pathogenic relevance, 50 Austrian piglet-producing farms were investigated: 40 farms with reproductive disorders (20 with tetracycline-treatment (tet), 20 without (no-tet)) and 10 clinically unaffected control farms. Samples from 770 sows (tet: n = 333, no-tet: n = 337, control: n =100) including blood, cervical and rectal swabs, dust samples and one pooled nursery fecal sample per farm were analyzed. Thirty-seven reproductive

Genetic Diversity and Selection Signatures of Three Indonesian Indigenous Pig Populations From Eastern Indonesia Using Whole Genome Sequencing Data.

Indonesia is widely recognized as a megadiversity country, harboring a rich diversity of indigenous pig species that play significant roles in the cultural and religious practices of local communities. However, the limited availability of genomic and demographic data of these indigenous pigs has greatly impeded our understanding of their evolutionary history and population structure. To date, the genetic characteristics of indigenous pigs from Maluku, Southeast Sulawesi (Sultra), and East Nusa Tenggara (NTT) remain unknown. Here, we aimed to investigate the genetic diversity, population struct

Post-outbreak dynamics and persistence of Actinobacillus pleuropneumoniae serotype 15 in finisher pigs in Iowa.

In 2021, Actinobacillus pleuropneumoniae serotype 15 (App15) mediated a high mortality respiratory outbreak in finisher hogs, affecting multiple companies within a 30-km radius of Iowa, USA. The atypical regional spread raised concerns for the strain's unusual environmental persistence and survivability. This prospective longitudinal study aimed to determine the duration of App15 persistence in convalescent pigs at a naturally infected 1200 head site. Sixty-seven pigs were sampled individually for 6 weeks using nasal swabs (NS), tonsil scrapings (TS), and serum samples (SS); pen-based oral flu

Optimizing Oral Vaccine Distribution Strategies for Wild Boars Through Bias-Corrected Habitat Modeling: A Case Study of Classical Swine Fever Control in Japan.

Control of infectious diseases in wildlife is often considered challenging due to the limited availability of information. Some infectious diseases in wildlife can also affect livestock, posing significant problems for the animal farming industry. In Japan, classical swine fever (CSF) reemerged in September 2018. Given the availability of commercial vaccines, control measures mainly involve the vaccination of domestic pigs and the distribution of oral vaccines to wild boars. Despite these efforts, the disease continues to spread, primarily due to wild boars. This transmission is further exacer

Strategic risk factor selection in disease risk mapping applied to the African swine fever outbreak in Sweden.

Risk mapping in epidemiology is a strategic tool that identifies high-risk areas for disease outbreaks, guiding preventive, surveillance, and control measures. In this study, we investigated how the selection of risk factors in ASF risk mapping affected the output of risk maps using the outbreak in Fagersta, Sweden as a study area. We identified ASFV risk factors by considering the sequence of introduction, release, and spread. Introduction was linked to pathways through which the virus could enter new areas, while release was associated with human activities and infrastructure that may lead t

Changes in mucin expression in the lungs of infant piglets following cardiopulmonary bypass with deep hypothermic circulatory arrest.

Atelectasis and mucus plugging are common complications following pediatric cardiac surgery with cardiopulmonary bypass (CPB). Mucins increase in airway lavage after pediatric CPB; however, tissue expression and regional variation remain to be evaluated in a translational model. The objective of this study was to analyze mucins in the lungs of infant pigs undergoing CPB with deep hypothermic circulatory arrest (DHCA). Piglets underwent CPB with DHCA followed by survival for 4 h (n = 5). Anesthesia controls (n = 7) were mechanically ventilated for 7 h without CPB. Gene expression of Muc4, Muc5A

Pathogenicity and pathobiological characterization of a recombinant genotype I/II African swine fever virus in pigs.

African swine fever virus (ASFV) is a highly contagious pathogen affecting domestic pigs and wild boar, causing substantial economic losses. Recently, a highly virulent recombinant ASFV strain combining genotypes I and II (rASFV I/II) emerged in Asia. Genotype II ASFV vaccine candidates have failed to effectively protect against rASFV I/II infection, presenting a critical challenge. Here, we investigated the pathobiological characteristics of rASFV I/II in domestic pigs. Ten healthy 7-week-old female pigs were intramuscularly inoculated with an rASFV I/II strain. Clinical signs, rectal tempera

Infection of domestic pigs with a genotype II potent strain of ASFV causes cytokine storm and lymphocyte mass reduction.

The whole-genome sequence of an African swine fever virus (ASFV) strain (HuB/HH/2019) isolated from Hubei, China, was highly similar to that of the Georgia 2007/1 strain ASFV. After infection with strong strains, domestic pigs show typical symptoms of infection, including fever, depression, reddening of the skin, hemorrhagic swelling of various tissues, and dysfunction. The earliest detoxification occurred in pharyngeal swabs at 4 days post-infection. The viral load in the blood was extremely high, and ASFV was detected in multiple tissues, with the highest viral loads in the spleen and lungs.

Structure of the recombinant RNA polymerase from African Swine Fever Virus.

African Swine Fever Virus is a Nucleo-Cytoplasmic Large DNA Virus that causes an incurable haemorrhagic fever in pigs with a high impact on global food security. ASFV replicates in the cytoplasm of the infected cell and encodes its own transcription machinery that is independent of cellular factors, however, not much is known about how this system works at a molecular level. Here, we present methods to produce recombinant ASFV RNA polymerase, functional assays to screen for inhibitors, and high-resolution cryo-electron microscopy structures of the ASFV RNAP in different conformational states.

Identifying Effective Biosecurity Measures for Preventing the Introduction of Classical Swine Fever in Pig Farms in Japan: Under the Condition of Absence/Presence of Observable Infected Wild Boar.

The outbreak of infectious diseases in swine, such as classical swine fever (CSF), has become a significant concern in the pig-farming industry. In Japan, after the re-emergence of CSF in 2018, farms are now exposed to the risk of transmission from infected wild boar and CSF-contaminated farms. This study aimed to identify biosecurity measures that were effective for the prevention of CSF introduction into farms during the period from the beginning of the CSF epidemic to the implementation of a vaccination campaign for domestic pigs at risk. The probability of virus introduction was assumed to

African Swine Fever Vaccine Candidate ASFV-G-ΔI177L Produced in the Swine Macrophage-Derived Cell Line IPKM Remains Genetically Stable and Protective against Homologous Virulent Challenge.

ASFV vaccine candidate ASFV-G-ΔI177L has been shown to be highly efficacious in inducing protection against challenges with the parental virus, the Georgia 2010 isolate, as well as against field strains isolated from Vietnam. ASFV-G-ΔI177L has been shown to produce protection even when used at low doses (102 HAD50) and shows no residual virulence even when administered at high doses (106 HAD50) or evaluated for a relatively long period of time (6 months). ASFV-G-ΔI177L stocks can only be massively produced in primary cell macrophages. Alternatively, its modified version (ASFV-G-ΔI177L/ΔLVR) gr
