Источник пометил публикацию как отозванную

BACKGROUND: In 2008, the first transplantation of a tissue-engineered trachea in a human being was done to replace an end-staged left main bronchus with malacia in a 30-year-old woman. We report 5 year follow-up results. METHODS: The patient was followed up approximately every 3 months with multidetector CT scan and bronchoscopic assessment. We obtained mucosal biopsy samples every 6 months for histological, immunohistochemical, and electron microscopy assessment. We also assessed quality of life, respiratory function, cough reflex test, and production and specificity of recipient antibodies

Methods to reduce the contraction of tissue-engineered buccal mucosa for use in substitution urethroplasty.

BACKGROUND: We previously described the production and clinical outcomes of tissue-engineered buccal mucosa (TEBM) used to treat recurrent urethral strictures. In this study, two patients developed a recurrent stricture and there was also evidence of graft contraction. OBJECTIVE: Assess possible preclinical methods to reduce contraction of TEBM. DESIGN, SETTING AND PARTICIPANTS: Using the model of TEBM in use clinically (ie, oral keratinocytes and fibroblasts cultured on de-epidermised acellular dermal scaffold), three methods of reducing TEBM contraction were investigated in vitro. INTERVE

The delayed repair of sciatic nerve defects with tissue-engineered nerve grafts in rats.

The purpose of this study was to evaluate the feasibility of using tissue-engineered nerve grafts for delayed repair of peripheral nerve defects. A 1-month delayed, 10-mm long sciatic nerve defect was created for rats, which were divided into three grafted groups and a non-grafted group. For bridging the nerve defects, the rats in three grafted groups were subjected to surgical repair with tissue-engineered nerve grafts made of a chitosan/polyglycolic acid (PGA) conduit filled with neural stem cells (NSCs), chitosan/PGA conduits, and autologous nerve grafts, respectively. At 3 months after ner

The repair of large segmental bone defects in the rabbit with vascularized tissue engineered bone.

Management of segmental bone defects is a considerable challenge for orthopedic surgeons. Tissue engineering is a promising method for repairing bone defects, and vascularization is critical to the performance of a tissue engineered bone. We report herein the construction of a vascularized tissue engineered bone with mesenchymal stem cells (MSCs) and MSC-derived endothelial cells (ECs) co-cultured in porous beta-tricalcium phosphate ceramic (beta-TCP) to repair 1.5-cm ulnar defects in the rabbit. Examination by X-ray and single photon emission computed tomography (SPECT), histologic analysis, …

A novel treatment for bile duct injury with a tissue-engineered bioabsorbable polymer patch.

BACKGROUND: With the recent widespread use of laparoscopic cholecystectomy and living-donor liver transplantation, complications involving the biliary system, and stenosis in particular, are encountered frequently. Although various invasive and noninvasive techniques are now available for the treatment of biliary stenosis, recurrence and other problems limit their value and utility. Our group sought to investigate whether a bioabsorbable polymer (BAP) patch could serve as a novel treatment for biliary stenosis. METHODS: In anesthetized hybrid pigs (n = 12), a spindle-shaped portion of the low

Viable osteogenic cells are obligatory for tissue-engineered ectopic bone formation in goats.

In this study we investigated the bone-forming capacity of tissue-engineered (TE) constructs implanted ectopically in goats. As cell survival is questionable in large animal models, we investigated the significance of vitality, and thus whether living cells instead of only the potentially osteoinductive extracellular matrix are required to achieve bone formation. Vital TE constructs of porous hydroxyapatite (HA) covered with differentiated bone marrow stromal cells (BMSCs) within an extracellular matrix (ECM) were compared with identical constructs that were devitalized before implantation. Use of tissue-engineered skin in the dermal atrophy patient with traumatic avulsion injuries.

Tissue-engineered skin has been approved by the Food and Drug Administration for use in certain chronic ulcers (venous stasis ulcers and diabetic foot ulcers). Its use has also been reported in acute (surgical) excisional wounds. The authors report the use of tissue-engineered skin in traumatic avulsion wounds in a series of 10 patients with dermal atrophy. All patients were older than 60 years of age and clinically had thin, atrophic skin. Most were older than 70 years of age and were steroid users. Investigating the impact of microcalcification size and volume on collagenous matrix and tissue mechanics using a tissue-engineered atherosclerotic cap model.

Atherosclerotic plaque rupture can lead to thrombotic cardiovascular events such as stroke and myocardial infarction. Computational models have shown that microcalcifications (calcified particles with a diameter < 50 μm) in the atherosclerotic plaque cap can increase cap tissue stresses and consequently contribute to plaque rupture. Microcalcification characteristics, such as particle size and volume fraction, have been implicated to affect cap stresses. However, the effect of these characteristics on tissue mechanics within a collagenous matrix, has not been investigated experimentally. In th

A physiologically relevant, estradiol-17β [E2]-responsive in vitro tissue-engineered model of the vaginal epithelium for vaginal tissue research.

AIMS: There are many situations where preclinical models of the human vagina would be valuable for in vitro studies into the pathophysiology of vaginally transmitted diseases, microbicide efficacy, irritability testing, and particularly, for assessing materials to be inserted in the vagina for support of the pelvic floor. The aim of this study is to develop a physiologically relevant, low cost, and ethically suitable model of the vagina using sheep vaginal tissue (SVT) to reduce the need for animal testing in gynecological research. METHODS: Tissue-engineered (TE) vaginal models were develope

Acellular dermal matrix and negative pressure wound therapy: a tissue-engineered alternative to free tissue transfer in the compromised host.

Free tissue transfer has revolutionized lower extremity reconstruction; however, its use in elderly patients with multiple medical problems can be associated with elevated rate s of perioperative morbidity and mortality. This study evaluates the use of acellular dermal matrix (ADM) in conjunction with negative pressure wound therapy (NPWT) and delayed skin graft application as an alternative to free tissue transfer in this compromised population. Bilayer, ADM (Integra, Plainsboro, NJ) was used in conjunction with NPWT (Wound V.A.C, Kinetic Concepts Inc., San Antonio, TX) to achieve vascularize

Three-dimensional PCL/collagen electrospun scaffolds with enlarged pores promote cell infiltration for vascular tissue engineering.

Cardiovascular disease remains one of the leading threats to human health. Owing to the limited availability of autologous vessels, the demand for vascular graft substitutes continues to rise. The design of tissue-engineered scaffolds that mimic the composition and architecture of the extracellular matrix (ECM) and facilitate vascular remodeling has become a major research focus in artificial blood vessel development. In this study, we investigated how the surface chemical and physical properties of vascular scaffolds influence cellular behavior. Polycaprolactone/collagen (PCL/Col) composite s

Engineering human multi-organ tissue chip niches for drug absorption, distribution, metabolism, excretion and toxicity prediction.

Predicting human drug responses in vitro remains a central challenge in drug development. Animal models have shown limited accuracy in recapitulating human drug absorption, distribution, metabolism, excretion and toxicity, which arise from coordinated activities across multiple organs. Multi-organ-on-a-chip systems (mOoCs) are a class of promising new approach methodologies that can reconstruct these inter-organ processes. Existing mOoCs have demonstrated proof of concept for inter-organ coupling but translation-ready systems remain out of reach. Two key challenges persist: how to preserve the

A Roadmap to Perfused Skin: Defining the Next Generation of Research Questions in Cutaneous Tissue Engineering.

Cutaneous tissue engineering has advanced from simple coverage substitutes to increasingly complex living constructs, yet the field remains constrained by a decisive problem: timely and durable perfusion. Many engineered skin substitutes can appear vascular in static culture or in small-animal models. However, they still fail when blood flow must be established quickly enough to rescue cells across clinically relevant tissue thickness. Rather than re-catalog platforms already summarized in recent reviews, this critical narrative review reframes the field around perfusion as the master function…

Advances in 3D-printed and biofabricated scaffolds for bone tissue engineering: materials, fabrication and biological integration.

The worldwide incidence of bone disorders is increasing at an alarming rate, especially among the elderly and those with increased obesity and poor physical activity. Therefore, bone tissue engineering, the process of regenerating diseased or damaged bone, is gaining increasing attention from the scientific community. One of the critical components for tissue engineering is the scaffold, an artificial extracellular matrix that promotes bone formation and regeneration activities. Due to the increasing demands for bone repair, bone tissue scaffolds have been extensively studied in state-of-the-a…
