Inhibition of the interaction between the SARS-CoV spike protein and its cellular receptor by anti-histo-blood group antibodies.

Severe acute respiratory syndrome coronavirus (SARS-CoV) is a highly pathogenic emergent virus which replicates in cells that can express ABH histo-blood group antigens. The heavily glycosylated SARS-CoV spike (S) protein binds to angiotensin-converting enzyme 2 which serves as a cellular receptor. Epidemiological analysis of a hospital outbreak in Hong Kong revealed that blood group O was associated with a low risk of infection. In this study, we used a cellular model of adhesion to investigate whether natural antibodies of the ABO system could block the S protein and angiotensin-converting e

Mice are unsuitable for modelling ABO discordance despite strain-specific A cross-reactive natural IgM.

ABO blood group antigens are immunodominant cell surface oligosaccharides. The function of the ABO system is clinically important in blood transfusion and solid organ transplantation but there is no small animal model of ABO discordance. The present study demonstrated A glycoconjugate-reactive IgM in the serum of CBA/Ca mice by enzyme-linked immunosorbent assay but showed with sugar blocking that the specificity of this IgM was different from that of human anti-A IgM. Furthermore, immunisation of CBA/Ca mice with the A antigen did not increase reactive IgM titre. In contrast, knock-out mice fo

Effects of preoperative plasma exchange therapy with albumin replacement fluid on blood coagulation in patients undergoing ABO-incompatible living-donor kidney transplantation using rotational thromboelastometry.

BACKGROUND: ABO-incompatible living-donor kidney transplantation (LDKT) requires immunotherapy and plasma exchange therapy (PEX). PEX with albumin replacement fluid reportedly decreases fibrinogen levels. However, no reports have described the effects of PEX with albumin replacement fluid on blood coagulation parameters and blood loss during the perioperative period. Therefore, we investigated the effects of preoperative PEX on blood coagulation parameters and blood loss during the perioperative period in patients undergoing ABO-incompatible LDKT as measured by rotational thromboelastometry (R

ABO blood groups and salivary gland tumors (Alabama, United States).

Studies of associations between various diseases and the ABO blood groups have shown elevated relative risks for some categories of disease. A strong association between salivary gland tumors and blood group A has been reported, but studies are few and have yielded contradictory results. To investigate this association, we conducted an epidemiologic study of salivary gland tumors diagnosed in Jefferson Country, Alabama (United States), 1968-89. All tumors were histologically confirmed. Blood group information was obtained for 200 individuals with salivary gland tumors, and the distribution of

Association of ABO blood group in Iraqis with hypercholesterolaemia, hypertension and diabetes mellitus.

There is strong evidence to suggest that there is an association between ABO blood group and certain diseases. This study in Baghdad, Iraq investigated the possible association of diabetes mellitus, hypercholesterolaemia and hypertension with ABO type. The data were derived from 920 patients with diabetes mellitus, hypertension and hypercholesterolaemia attending hospitals, clinics and laboratories in Baghdad, and 200 healthy control individuals. Analysing the data by blood group showed that the levels of total cholesterol, glucose and systolic/diastolic blood pressure were all significantly h

Universal red blood cells--enzymatic conversion of blood group A and B antigens.

Accidental transfusion of ABO-incompatible red blood cells (RBCs) is a leading cause of fatal transfusion reactions. To prevent this and to create a universal blood supply, the idea of converting blood group A and B antigens to H using specific exo-glycosidases capable of removing the immunodominant sugar residues was pioneered by Goldstein and colleagues at the New York Blood Center in the early 1980s. Conversion of group B RBCs to O was initially carried out with alpha-galactosidase extracted from coffee beans. These enzyme-converted O (ECO) RBCs appeared to survive normally in all recipient

Carcinoembryonic antigen and blood group-related carbohydrate antigens in glycoproteins in human bile in hepatolithiasis. Japanese Study Group of Hepatolithiasis.

We investigated whether carbohydrate antigens on biliary glycoproteins and carcinoembryonic antigen (CEA) are related to hepatolithiasis. CEA, ABO, and Lewis blood group-related antigens, as well as sialyl-Tn antigen in hepatic bile, were analyzed by Western blotting in samples from 12 patients with hepatolithiasis and 37 with other biliary diseases (choledocholithiasis, 13; cholecystolithiasis, 5; acute cholecystitis, 2; cholangiocarcinoma, 5; common bile duct carcinoma, 4; pancreatic carcinoma, 6; and metastatic carcinoma of liver, 2). CEA was positive on mucinous glycoprotein in six patient

[Immunologic risk analysis of blood transfusion: 1991-1998].

The immunologic risk associated to erythrocyte transfusions is bound to the polymorphism of blood group systems and to the respect of blood transfusion regulations. The results of three studies are presented, which were carried out respectively by the French Society of Blood Transfusion, the National Institute of Blood Transfusion and the National Haemovigilance Network. Two hundred and twenty-seven cases of immunologic accidents are analysed using the Kaplan's interpretation model and the traditional method of process analysis. The results show three critical factors in the occurrence of this

[Yes, we should keep ABO agglutination test within bedside transfusion checks].

ABO incompatible transfusions are still a frequent cause of serious adverse transfusion reactions. Bedside check is intended to detect patient errors and prevent ABO mismatch. France is one of the few countries that includes ABO agglutination test for red blood cells in bedside checks. Evaluation of this ABO agglutination test, performed with a special card, shows that, on the field, despite frequent users' mishandling, it can detect up to 93% of ABO incompatibilities. This is not enough to rely on this sole test for bedside checks. But, linking it with an another test, currently, checks that

Factors affecting the efficacy of human leukocyte antigen-selected platelet provision: A large retrospective study in the United Kingdom.

BACKGROUND: A large, retrospective study was designed to interrogate current NHS Blood and Transplant (NHSBT) HLA matching strategies for the provision of HLA selected platelets (HLA SP) and to determine whether additional factors such as ABO blood group matching, patient diagnosis, patient and/or donor age, sex, ethnicity, age of platelet unit at transfusion and possibly seasonal variation also play a role in transfusion efficacy. MATERIALS AND METHODS: Data for 56 640 HLA SP transfusions over a 3-year period were collected. Transfusions with missing data for any factor under consideration w

Factors related to the outcome of prophylactic platelet transfusions in patients with hematologic malignancies: an observational study.

BACKGROUND: A better knowledge of the connections between platelet concentrate (PC) characteristics and transfusion outcomes in day-to-day practice would help improve the selection process of the most appropriate PC. STUDY DESIGN AND METHODS: In this study of prophylactic platelet transfusions in patients with hematologic malignancies between 2002 and 2012, outcome criteria were corrected count increments (CCIs) and platelet transfusion intervals (TIs, in days). Studied characteristics were ABO matching status, platelet source, dose, storage duration, irradiation, washing, and transfusion seq

Molecular red cell genotyping of rare blood donors in South Africa to enhance rare donor-patient blood matching.

BACKGROUND: Molecular red cell genotyping is devoid of serology limitations such as the scarcity of rare antisera and the possibility of inconclusive results due to biological interferences. Blood incompatibility can result in immune transfusion reactions such as haemolytic transfusion reactions or haemolytic disease of the foetus and newborn. OBJECTIVE: The study aimed to use molecular red cell genotyping to identify rare blood group donors among South African blood donors. METHODS: Red cell genotyping data were extracted retrospectively from the BIDS XT genotyping software in the Immunohae

Fatal acute hemolytic transfusion reaction due to anti-B from a platelet apheresis unit stored in platelet additive solution.

BACKGROUND: Hemolytic transfusion reactions from out-of-group plasma in platelet (PLT) transfusions are uncommon, with most involving passive transfer of anti-A. Only rare reactions have ever been reported due to anti-B. STUDY DESIGN AND METHODS: An apheresis PLT product was donated by a blood group O male, processed using PLT additive solution, and pathogen reduced. Postreaction recipient testing included an antibody screen using gel technology, a direct antiglobulin test (DAT) using immunoglobulin G and C3, and an eluate against group O and B cells. Postreaction donor testing included measu

Determinants of severity of neonatal hyperbilirubinaemia in ABO incompatibility in Nigeria.

The study was designed to determine factors responsible for the severe neonatal hyperbilirubinaemia sometimes found in ABO incompatibility in Nigeria. In 50 jaundiced babies who were ABO-incompatible it was found that the mean serum bilirubin level was significantly higher in outpatients than inpatients; this difference was probably due to the delay in recognizing jaundice among the outpatients and, possibly also to the slightly higher number of G-6-PD deficient babies in the same group, and their greater exposure to icterogenic agents. The results of serological studies suggest that ABO-incom
