ZIF-67 and Mn & Zn co-doped ZIF-67 as multifunctional therapeutic platforms: Antibacterial, antioxidant, and anticancer assessment in lung cancer cells in vitro.

The present study is an attempt to delineate the biological efficacy of zinc and manganese doping on ZIF-67 MOF. Pure (ZIF-67) and zinc/manganese-doped samples were synthesised using a one-pot method. Various analytical techniques such as fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), field emission scanning electron microscopy (FESEM), transmission electron microscopy (TEM), dynamic light scattering (DLS) together with elemental mapping analyses through energy dispersive spectroscopy (EDS), support that the MOF formed is a sodalite (SOD) cage type structure. The stud

INPP5K limits PtdIns (4,5)P2 accumulation in CD19 microclusters and restrains CD19 co-receptor membrane proximity and BCR-proximal signaling in B cells.

INTRODUCTION: CD19 is a key B-cell co-receptor that amplifies B-cell receptor (BCR) signaling and tunes B-cell activation thresholds. How the conformation of the CD19 cytoplasmic domain is regulated to restrain basal and BCR-induced signaling remains incompletely understood. We investigated whether the phosphoinositide 5-phosphatase INPP5K controls CD19 conformation and CD19-amplified BCR-proximal signaling through local regulation of phosphatidylinositol 4,5-bisphosphate [PtdIns(4,5)P2]. METHODS: A B-cell-specific Inpp5k knock-down (KD) mouse model was generated using CD21-Cre-mediated recom

Co-delivery of endometrial mesenchymal stem cells and macrophages by an electrospun patch promotes angiogenesis during endometrial injury repair via VEGF related signalling.

BACKGROUND: Intrauterine adhesion (IUA) is a common gynecological disease that contributes to infertility. Decreased endometrial angiogenesis and uterine ischemia are major therapeutic challenges for IUA and cannot be addressed by current treatment strategies. Human endometrial mesenchymal stem cells (H-EMSCs) and macrophages (mø) are both important cell types that reside within the endometrial tissue and participate in its repair and regeneration. However, how to harness the endometrial tissue repair potential of H-EMSCs and mø simultaneously in a co-delivery system and whether there are sign

Safety and efficacy of low-dose, cord blood-derived CD19 CAR-NK cells with 4-1BB co-stimulation in refractory systemic lupus erythematosus.

BACKGROUND: B cell-targeting chimeric antigen receptor (CAR) T cell therapy has shown efficacy in autoimmune diseases but is limited by toxicity, complex manufacturing, and high cost. Umbilical cord blood (UCB)-derived CAR-natural killer (CAR-NK) cells offer an alternative "off-the-shelf" platform with a potentially superior safety profile. We developed a UCB-derived CD19-targeting CAR-NK product incorporating the 4-1BB costimulatory domain (CD19-BBz) and evaluated its safety and efficacy in refractory systemic lupus erythematosus (SLE). This study was registered at ClinicalTrials.gov (Clinica

Engineering a multilayered micro-vascularized soft tissue graft: Integrating polyurethane composite nanofibres with co-cultures of human adipose-derived endothelial and stem cells.

Soft tissue defects and injuries, such as gingival recession, those requiring dermal tissue filling, and other indications, affect millions worldwide, yet autologous grafting remains the standard of care in most instances, despite donor site morbidity and limited tissue availability. Tissue-engineered alternatives have strong potential to overcome these limitations. We evaluated an 8 mm-diameter, 1 mm-thick layered electrospun composite formed from polar/hydrophobic/ionic polyurethane with methacrylated gelatin (FD-PHI) and polycarbonate urethane (PCNU), seeded with a co-culture of human adipo

Loss of PTEN as an Independent Poor Prognosis Indicator in Lung Adenocarcinoma, but Not in Squamous Cell Carcinoma, Is Associated with an Immunosuppressive Tumor Microenvironment and Distinct Co-Mutational Profiles.

Background/Objectives: Non-small cell lung cancer (NSCLC) comprises biologically heterogeneous tumors, primarily lung adenocarcinoma (LUAD) and lung squamous cell carcinoma (LUSC), which differ in genomic landscape and clinical behavior. The tumor suppressor PTEN is a key negative regulator of the PI3K/AKT/mTOR pathway and is frequently inactivated in NSCLC through genetic and non-genetic mechanisms. Although reduced PTEN expression has been associated with poor outcomes in lung cancer, its prognostic relevance across these histological subtypes remains unclear. Methods: Here, we investigated

The impact of EGFR subtype combined with TP53 co-mutation status on survival outcomes with front-line osimertinib in non-small cell lung cancer (NSCLC).

BACKGROUND: Osimertinib is a standard therapy for EGFR-mutant NSCLC. However, markers to better identify those at risk for poor outcomes are needed. This is the largest study to date evaluating the impact of EGFR subtype combined with TP53 co-mutation status on survival endpoints with front-line osimertinib. METHODS: Patients from a U.S. clinical-genomic database with advanced EGFR-mutant NSCLC receiving front-line osimertinib were studied. Real-world progression-free survival (rwPFS) and overall survival (OS) were determined using Kaplan-Meier methods, and multivariable Cox regression compar

Clinical factors and molecular co-alterations impact outcomes in patients receiving first-line osimertinib for EGFR-mutated non-small cell lung cancer.

PURPOSE: Until recently, osimertinib was the preferred first-line therapy for nearly all patients with EGFR-mutated advanced NSCLC. However, with approval of the FLAURA2 and MARIPOSA regimens, identifying molecular or clinical factors that confer higher risk for inferior outcomes to first-line osimertinib may better inform upfront treatment selection for newly-diagnosed patients. METHODS: This real-world, multicenter retrospective study across 4 U.S. academic centers included adult patients with metastatic NSCLC and classic sensitizing EGFR mutations (exon 19 deletion, L858R mutation) receivi

TTF-1 and p40 co-expression defines a distinct subtype of non-small cell lung cancer with frequent TP53 mutations and FGFR pathway dysregulation.

BACKGROUND: Lung cancer remains the leading cause of cancer-related mortality worldwide, with non-small cell lung cancer (NSCLC) accounting for approximately 85% of cases. Accurate differentiation between adenocarcinoma (ADC) and squamous cell carcinoma (SCC) is critical for informing personalized therapies. Thyroid transcription factor-1 (TTF-1) and p40 are traditionally regarded as mutually exclusive markers of ADC and SCC, respectively. However, a subset of tumors exhibits co-expression of TTF-1 and p40, presenting diagnostic challenges and suggesting underlying biological distinctiveness.

Co-Induction of ULK-1 and AHSP mRNAs in Erythroid Precursor Cells Isolated From a Sirolimus-Treated β-Thalassemia Patient: A Case Report Study.

INTRODUCTION: The β-thalassemias are inherited genetic disorders affecting the hematopoietic system and caused by mutations of the adult β-globin gene, leading to low or absent production of adult hemoglobin. In addition, an excess of free α-globin is associated with ineffective erythropoiesis. In fact, the free α-globin molecules are prone to precipitate, causing toxicity to the erythroid cells, and interference with red cell maturation. In order to counteract the detrimental effects of the excess of α-globin, two pathways might be activated in β-thalassemia erythroid cells, i.e. Unc-51-like

Dendritic cell targeting in lymph nodes with modular adapters boosts HAdV5 and HC-HAdV5 tumor vaccination by co-secretion of IL-2v and IL-21.

Adenoviral vectors demonstrate encouraging clinical outcomes for B and T cell vaccines. With such approaches, multiple payloads can be delivered, beyond the antigen itself. Nevertheless, the human adenoviral vector serotype C5 (HAdV5) exhibits limited transduction efficiency to dendritic cells (DCs), therefore necessitating very high viral loads. Targeting antigen-presenting cells (APCs) has remained challenging. To solve this problem, we developed a versatile platform that employs modular retargeting adapters to enhance transduction of specific cell types, including challenging host cells. By

Methadone Requires the Co-Activation of μ-Opioid and Toll-Like-4 Receptors to Produce Extracellular DNA Traps in Bone-Marrow-Derived Mast Cells.

Methadone is an effective and long-lasting analgesic drug that is also used in medication-assisted treatment for people with opioid use disorders. Although there is evidence that methadone activates μ-opioid and Toll-like-4 receptors (TLR-4s), its effects on distinct immune cells, including mast cells (MCs), are not well characterized. MCs express μ-opioid and Toll-like receptors (TLRs) and constitute an important cell lineage involved in allergy and effective innate immunity responses. In the present study, murine bone-marrow-derived mast cells (BMMCs) were treated with methadone to evaluate

Combined Application of Human Amniotic Membrane Mesenchymal Stem Cells and a Modified PGS-co-PCL Film in an Experimental Model of Myocardial Ischemia-Reperfusion Injury.

According to the World Health Organization (WHO), about 3.9 million people die annually of ischemic heart disease (IHD). Several clinical trials have shown that stem cell therapy is a promising therapeutic approach to IHD. Human amniotic membrane mesenchymal stem cells (hAMSCs) positively affect the repair of myocardial ischemia-reperfusion (MI/R) injury by stimulating endogenous repair mechanisms. The differentiated hAMSCs with and without modified PGS-co-PCL film were applied in the myocardium. MI/R injury was induced by ligating the left anterior descending artery in 48 male Wistar rats. Th
