Butyrate improves the metabolism of proximal tubular cells and attenuates tissue damage in oxalate-induced acute kidney injury.

Metabolic reprogramming of renal cells is closely linked to kidney injury and repair. In proximal tubular cells (PTCs), impaired mitochondrial and peroxisomal fatty acid oxidation leads to a compensatory increase in glycolysis to meet energy demands. Short-chain fatty acids such as butyrate modulate cell metabolism and can attenuate inflammation through activation of the G protein-coupled receptors. We investigated whether butyrate influences PTC metabolism in calcium oxalate (CaOx) crystal-induced acute kidney injury (AKI). In vitro, primary murine PTCs were pretreated with sodium butyrate (N

Cuproptosis-associated genes DLD, ULK2, CCL8, and CCS link metabolic and immunological dysfunction in Parkinson's disease: a multi-omics study.

Parkinson's disease (PD) is a progressive neurodegenerative disorder whose pathogenesis involves complex interactions between metabolic dysfunction and neuroinflammation. The role of cuproptosis, a newly defined copper-dependent cell death pathway that is triggered during mitochondrial aerobic respiration, in PD remains incompletely understood. In this study, we employed integrated bioinformatics on bulk and single-cell RNA sequencing datasets to identify four hub cuproptosis-associated genes (DLD, ULK2, CCL8, and CCS) strongly linked to PD. Functional enrichment and immune infiltration analys

[Comparative assessment of circulatory and respiratory maintenance by hemoglobin dissolved in plasma and contained in erythrocytes].

Basic values of blood circulation and respiration were compared in two groups of cats. The blood of group I animals was replaced by a 8% solution of human Hb completely free of stroma and procoaglulant activity. In group II animals the blood was diluted with polyglucine to the Hb level of 8 g%. The solution is well saturated in the lungs and releases O2 to tissues, but much less than Hb contained in red cells. The dissolved Hb did not completely meet the requirements of the organism in oxygen with the resultant hypoxia. Moderate anemic hypoxia caused by blood dilution with polyglucine was easi

Urinary multi-omics reveal non-invasive diagnostic biomarkers in clear cell renal cell carcinoma.

Clear cell renal cell carcinoma (ccRCC) is the most common kidney malignancy. Yet, no rapid, non-invasive biomarkers are available for diagnosis or screening. Urine represents an ideal analyte matrix due to its accessibility, low invasiveness, longitudinal sampling, and the kidney's central role in filtration. Here, we integrated proteomic, lipidomic, and metabolomic analyses of urine from ccRCC patients and controls to identify diagnostic biomarkers. Multi-omics profiling revealed urogenital metabolic dysregulation in ccRCC, including increased lipid metabolism, altered mitochondrial respirat

Psoriasis-like inflammation induces structural and functional changes in mitochondria.

Mitochondrial structural and functional changes accompany psoriasis, yet the mitochondrial response to psoriatic inflammation in keratinocytes and fibroblasts remains unexplored. In this study, we investigated the effect of psoriasis-like inflammation (PLI) induced by a cytokine cocktail (interleukin (IL)-17A, IL-22 and tumour necrosis factor (TNF)-α) on mitochondrial network morphology and function in cultured keratinocytes (HaCaT) and fibroblasts (BJ-5ta). In both cell types, PLI triggered the expression of psoriasis-related Elafin and high amounts of cytokines (IL-1, IL-6), interferons (IFN

Oxidative stress enhances the therapeutic action of a respiratory inhibitor in MYC-driven lymphoma.

MYC is a key oncogenic driver in multiple tumor types, but concomitantly endows cancer cells with a series of vulnerabilities that provide opportunities for targeted pharmacological intervention. For example, drugs that suppress mitochondrial respiration selectively kill MYC-overexpressing cells. Here, we unravel the mechanistic basis for this synthetic lethal interaction and exploit it to improve the anticancer effects of the respiratory complex I inhibitor IACS-010759. In a B-lymphoid cell line, ectopic MYC activity and treatment with IACS-010759 added up to induce oxidative stress, with con

The emerging role of cranial nerves in shaping craniofacial development.

Organs and structures of the vertebrate head perform a plethora of tasks including visualization, digestion, vocalization/communication, auditory functions, and respiration in response to neuronal input. This input is primarily derived from afferent and efferent fibers of the cranial nerves (sensory and motor respectively) and efferent fibers of the cervical sympathetic trunk. Despite their essential contribution to the function and integration of processes necessary for survival, how organ innervation is established remains poorly understood. Furthermore, while it has been appreciated for som

OPA1-dependent cristae modulation is essential for cellular adaptation to metabolic demand.

Cristae, the organized invaginations of the mitochondrial inner membrane, respond structurally to the energetic demands of the cell. The mechanism by which these dynamic changes are regulated and the consequences thereof are largely unknown. Optic atrophy 1 (OPA1) is the mitochondrial GTPase responsible for inner membrane fusion and maintenance of cristae structure. Here, we report that OPA1 responds dynamically to changes in energetic conditions to regulate cristae structure. This cristae regulation is independent of OPA1's role in mitochondrial fusion, since an OPA1 mutant that can still oli

Effects of sodium picosulfate and other laxatives in cultured Chang cells.

Cytotoxic effects were studied in Chang liver cells exposed to 4,4'-dihydroxy-diphenyl-(2-pyridyl)-methane-bis(hydrogen sulfate) (ether)-disodium salt (sodium picosulfate) and the results were compared with the effects of other laxatives. Sodium picosulfate caused a decrease in cell growth parallel to a decrease in glucose consumption and lactate production at 2000 and 4000 microgram/ml. On the contrary, 4,4'-dihydroxy-diphenyl-(2-pyridyl)-methane (bis-acodyl) decreased cell growth at 25 and 50 microgram/ml, while glucose consumption, lactate production and the molar ratio of glucose converted

Gramine sensitizes Klebsiella pneumoniae to tigecycline killing.

BACKGROUND: The presence of plasmid-mediated resistance-nodulation-division (RND) efflux pump gene cluster tmexCD1-toprJ1 and its related variants has been associated with heightened resistance to tigecycline, thus diminishing its effectiveness. In this study, we explored the potential of gramine, a naturally occurring indole alkaloid, as an innovative adjuvant to enhance the treatment of infections caused by K. pneumoniae carrying tmexCD-toprJ-like gene clusters. METHODS: The synergistic potential of gramine in combination with antibiotics against both planktonic and drug-tolerant multidrug-

Single-plate Runs of Mito Stress And Glycolysis Stress Tests using Adherent or Non-Adherent Cells.

Real-time metabolic analysis provides label-free measurements of mitochondrial respiration and glycolysis, allowing researchers to link cellular energy metabolism with disease mechanisms and therapeutic responses across diverse fields. When comparing energy metabolism across different mice, other animals, humans, or cells under varying treatments, it is crucial to perform measurements within a single run to ensure accurate and reliable results. Moreover, experimental design must consider the cell type being analyzed, for example, whether they are adherent, like bone marrow-derived macrophages

Periderm invasion contributes to epithelial formation in the teleost pharynx.

The gnathostome pharyngeal cavity functions in food transport and respiration. In amniotes the mouth and nares are the only channels allowing direct contact between internal and external epithelia. In teleost fish, gill slits arise through opening of endodermal pouches and connect the pharynx to the exterior. Using transgenic zebrafish lines, cell tracing, live imaging and different markers, we investigated if pharyngeal openings enable epithelial invasion and how this modifies the pharyngeal epithelium. We conclude that in zebrafish the pharyngeal endoderm becomes overlain by cells with a per

Oxygen sensing in airway chemoreceptors.

Pulmonary neuroepithelial bodies, composed of innervated clusters of amine- and peptide-containing cells, are widely distributed throughout the airway mucosa of human and animal lungs. Structurally, neuroepithelial bodies resemble chemoreceptors (such as carotid body, taste buds) and are thought to function as hypoxia sensitive airway sensors. Evidence for this is indirect, however, and the mechanism of oxygen sensing by these cells is unknown. Here we culture neuroepithelial bodies isolated from rabbit fetal lungs and identify voltage-activated potassium, calcium and sodium currents using the

ATP depletion rather than mitochondrial depolarization mediates hepatocyte killing after metabolic inhibition.

The importance of ATP depletion and mitochondrial depolarization in the toxicity of cyanide, oligomycin, and carbonyl cyanide m-cholorophenylhydrazone (CCCP), an uncoupler, was evaluated in rat hepatocytes. Oligomycin, an inhibitor of the reversible mitochondrial ATP synthase (F1F0-adenosinetriphosphatase), caused dose-dependent cell killing with 0.1 microgram/ml being the minimum concentration causing the maximum cell killing. Oligomycin also caused rapid ATP depletion without causing mitochondrial depolarization. Fructose (20 mM), a potent glycolytic substrate in liver, protected completely

Carbon Monoxide: A Pleiotropic Redox Regulator of Life and Death.

Despite recent technological progress, carbon monoxide poisoning is still one of the leading causes of domestic and industrial morbidity and mortality. The brain is particularly vulnerable to CO toxicity, and thus the majority of survivors develop delayed movement and cognitive complications. CO binds to haemoglobin in erythrocytes, preventing oxygen delivery to tissues, and additionally inhibits mitochondrial respiration. This renders the effect of CO to be closely related to hypoxia reperfusion injury. Oxygen deprivation, as well as CO poisoning and re-oxygenation, are shown to be able to ac

Antituberculosis Activity of the Antimalaria Cytochrome bcc Oxidase Inhibitor SCR0911.

The ability to respire and generate adenosine triphosphate (ATP) is essential for the physiology, persistence, and pathogenicity of Mycobacterium tuberculosis, which causes tuberculosis. By employing a lead repurposing strategy, the malarial cytochrome bc1 inhibitor SCR0911 was tested against mycobacteria. Docking studies were carried out to reveal potential binding and to understand the binding interactions with the target, cytochrome bcc. Whole-cell-based and in vitro assays demonstrated the potency of SCR0911 by inhibiting cell growth and ATP synthesis in both the fast- and slow-growing M.

Pyruvate decarboxylase deficiency in a patient with Leigh's encephalomyelopathy.

A 23-month-old boy with progressive muscular hypotonia and mental and motor deterioration was described. Four days after the admission he had a respiratory arrest and required the care in a respirator thereafter. Laboratory examinations showed metabolic acidosis and high levels of pyruvate, lactate and alanine in blood and cerebrospinal fluid. Oral administration of thiamine-HCl and lipoic acid was noneffective and he died 7 months after the admission. Autopsy findings were compatible with subacute necrotizing encephalomyelopathy (Leigh's disease). The activity of pyruvate decarboxylase in aut

A study of the action of a bisquaternary ammonium salt, an amine oxide and an alkoxy phenylcarbamic acid ester on some metabolic functions in Staphylococcus aureus.

Aspects of the mode of action of three antibacterial agents, N,N'-bis (dodecyldimethyl)-1,2-ethane diammonium dibromide (BDED), 2-piperidinoethyl-4-heptyloxyphenylcarbamate hydrochloride (XXI) and 1-dodecylpiperidine N-oxide (DPNO) against Staphylococcus aureus have been examined. Their ability to modify respiration, proton translocation, potassium leakage and ATP synthesis are reported. In all cases there was a striking correspondence between the minimum inhibitory concentration and the dose level which completely prevented ATP synthesis. It is suggested that for these three chemically distin

O2-sensing and HIF regulation: from oxygen homeostasis to tumor-associated immune responses.

Oxygen is needed to generate adenosine triphosphate (ATP) via aerobic respiration in mitochondria. Hypoxia-when the tissue's oxygen demand exceeds its supply-characterizes not only sites of acute inflammation but also many tumors and their microenvironment. But even then, (immune) cells must ensure sufficient ATP synthesis. Under hypoxic conditions, hypoxia-inducible factors (HIFs) are capable of adapting cellular metabolism by induction of hundreds of target genes that, among others, regulate glycolysis, cell survival, and angiogenesis. This may hold true for immune cells under homeostatic co

Mitochondria as target to inhibit proliferation and induce apoptosis of cancer cells: the effects of doxycycline and gemcitabine.

Doxycycline has anti-tumour effects in a range of tumour systems. The aims of this study were to define the role mitochondria play in this process and examine the potential of doxycycline in combination with gemcitabine. We studied the adenocarcinoma cell line A549, its mitochondrial DNA-less derivative A549 ρ° and cultured fibroblasts. Treatment with doxycycline for 5 days resulted in a decrease of mitochondrial-encoded proteins, respiration and membrane potential, and an increase of reactive oxygen species in A549 cells and fibroblasts, but fibroblasts were less affected. Doxycycline slowed
