Women with double primary cancers of the colorectum and endometrium: do they have Lynch syndrome?

OBJECTIVE: The aim of this study was to determine the clinical characteristics of women with double primary cancers of the colorectum and endometrium and assess the probability of Lynch syndrome. STUDY DESIGN: We identified 15 women with paraffin-embedded blocks available who were diagnosed, treated and followed for double primary colorectal and endometrial cancers at in a single institution between 1994 and 2014. If there was a family history that met the revised Amsterdam criteria for Lynch syndrome, the woman was considered to have 'clinically defined Lynch syndrome'. If immunohistochemica

From local audit to national learning: revisiting the case for a national colonoscopy clinical quality registry in Australia.

Colonoscopy is central to colorectal cancer prevention and early detection, yet Australia lacks a national mechanism to aggregate and benchmark colonoscopy quality data. The Colonoscopy Clinical Care Standard establishes local monitoring of key quality indicators, which can identify service-level problems, but cannot show whether variation is isolated, systemic, improving over time or concentrated in particular settings. This creates a national blind spot in one of the country's most common cancer-prevention procedures. Current arrangements remain fragmented across health services and do not s

Nanoparticles for colorectal cancer therapy: A review.

Colorectal cancer (CRC) is one of the most common malignancies worldwide and is the second leading cause of cancer-related deaths. Currently, the main therapies for CRC include surgery, chemotherapy and radiotherapy. However, all of them have certain limitations, such as the tendency to trigger cancer recurrence and metastasis, and excessive damage to patients' bodies. To overcome some of these limitations, scholars have developed many nanoparticle-based therapies. This review discusses recent advances in the application of nanoparticles in CRC therapy, presenting the progress of inorganic, or

[Microbiome and Colorectal Cancer].

The number of colorectal cancer patients in Japan continues to rise, placing the country among the highest rates globally, surpassing even Western nations. Furthermore, the increase in colorectal cancer among younger individuals(under 50 years old)is a significant concern. This rapid rise in colorectal cancer patients cannot be explained solely by the adoption of Western lifestyles, suggesting the presence of unknown carcinogenic factors. Colibactin toxin, produced by certain gut bacteria (pks+ Escherichia coli), binds to AT-rich DNA and induces characteristic mutation patterns. An internation

Therapeutic Strategies Targeting the Tumor-Stromal Microenvironment in Colorectal Cancer: A Narrative Review.

Despite advances in colorectal cancer (CRC) treatment, outcomes in advanced disease remain poor. Current therapies mainly target the epithelial compartment of the tumor, yet increasing evidence highlights the tumor microenvironment (TME), particularly the tumor stroma, as a prognostic and potentially predictive factor. This narrative review summarizes current evidence on stromal features as biomarkers and therapeutic targets in CRC. These stromal markers, such as fibroblast-associated protein (FAP), are associated with adverse outcomes, underscoring their clinical relevance. However, therapies

Pangenome analysis reveals both niche-specific "specialists" and microbial "side hustlers" in colorectal cancer microbiomes.

The gut microbiome is reproducibly implicated in colorectal cancer (CRC), yet the inter-study and interpersonal variability of certain species associations suggests that CRC microbiomes may be defined by convergent functional states achievable by phylogenetically diverse organisms. Distinguishing lineage-conserved "specialists" from taxonomically diverse "side hustlers"-organisms whose shared functional traits are dispersed across phylogenetically distant lineages-offers complementary translational insights: "specialists" are primary candidates for lineage-targeted biomarkers and inhibitors, w

Global prevalence and moderating factors of malnutrition in colorectal cancer survivors: A meta-analysis.

PURPOSE: This meta-analysis aims to estimate the global prevalence of severe, moderate, overall malnutrition and moderating factors of malnutrition in colorectal cancer (CRC) survivors. METHODS: A comprehensive search was conducted in Embase, CINAHL, Medline-OVID, PubMed, Scopus, and Web of Science from inception to February 8, 2024, without language, region, or publication date restrictions. A generalized linear mixed model and random-effects model were used to examine the pooled prevalence, and moderator analyses were implemented to investigate variations in the pooled prevalence. RESULTS:

[The Power of the Gut Microbiome: Exploring New Perspectives in Colorectal Cancer Therapy].

The gut microbiome is involved in host physiology, including nutrition, metabolism, and immunity. It was recently known that dysbiosis of the gut microbiome has been implicated in several human diseases such as inflammatory bowel disease. It is altered by environmental factors such as diet, habit and lifestyle and has been directly and indirectly linked to the development and progression of colorectal cancer(CRC). Fusobacterium(F.)nucleatum, which causes periodontal disease, has been shown to play an important role in the initiation and development of CRC, although not as clearly as Helicobact

Obesity-Associated Colorectal Cancer.

Colorectal cancer (CRC) affects approximately 2 million people worldwide. Obesity is the major risk factor for CRC. In addition, obesity contributes to a chronic inflammatory stage that enhances tumor progression through the secretion of proinflammatory cytokines. In addition to an increased inflammatory response, obesity-associated cancer presents accrued molecular factors related to cancer characteristics, such as genome instability, sustained cell proliferation, telomere dysfunctions, angiogenesis, and microbial alteration, among others. Despite the evidence accumulated over the last few ye

National and subnational incidence, mortality and associated factors of colorectal cancer in China: A systematic analysis and modelling study.

BACKGROUND: Due to their known variation by geography and economic development, we aimed to evaluate the incidence and mortality of colorectal cancer (CRC) in China over the past decades and identify factors associated with CRC among the Chinese population to provide targeted information on disease prevention. METHODS: We conducted a systemic review and meta-analysis of epidemiolocal studies on the incidence, mortality, and associated factors of CRC among the Chinese population, extracting and synthesising data from eligible studies retrieved from seven global and Chinese databases. We pooled

Family history of cancer as a potential risk factor for colorectal cancer in EMRO countries: a systematic review and meta-analysis.

The current meta-analysis aims to investigate the existing articles that evaluated the implications of a positive family history of cancer on the risk of colorectal cancer (CRC) within the EMRO countries. We employed PubMed, Scopus, and Web of Science as search databases for this study. To assess the quality of the selected articles, we utilized the Newcastle-Ottawa (NCO) checklist. In comparing the impact of a family history of cancer between the case and control groups, we computed the odds ratio (OR) along with its corresponding 95% confidence interval (CI). Finally, 27 articles were select

Morphology of enteric glia in colorectal carcinoma: A comparative study of tumor site and its proximal normal margin.

OBJECTIVE: Colorectal carcinoma (CRC) is the third most common cancer in the world and fifth most common cancer in India. To understand the extent of perineural invasion (PNI) in CRC it is essential to study the morphology of enteric glial cells (EGCs). The aim of the study was to analyze the numerical density of EGCs and area of myenteric ganglia (MG) in the colonic tissue samples collected from CRC patients. MATERIAL AND METHODS: Fifteen intraoperative tissue specimens were collected from the tumor site and 2cm proximal to the upper extent of tumor. The samples were divided into four groups
