The molecular mechanisms involved in the development of a high level of resistance to a wide range of antimicrobials in Burkholderia pseudomallei and closely related species have not been sufficiently investigated. In the present study, the properties of B. pseudomallei, B. mallei and B. thailandensis mutants with increased resistance to fluoroquinolones and cephalosporins were analysed. Resistance to pefloxacin, ofloxacin and ceftazidime in B. pseudomallei and B. thailandensis was accompanied by an increased resistance to aminoglycosides, beta-lactams, macrolides and chloramphenicol, whereas

Tuberculosis (TB) is a devastating disease caused by Mycobacterium tuberculosis that killed an estimated 4000-5000 person each day during 2005. Although infections with drug sensitive strains can be effectively cured with a 6 to 9 month regimen of multiple antibiotics, the inability to deliver and complete appropriate courses of therapy on a global level has led to the selection of resistant strains over the past 50 years. The selection and spread of multiple drug resistant M. tuberculosis continued for decades leading to two operationally distinct forms of the disease, multiple drug resistant

Acquired drug resistance remains a challenge for treating melanoma. Monotherapies such as vemurafenib and dabrafenib, which target a mutant form of BRAF kinase, become ineffective within six months of treatment. Therapies that combine multiple drugs targeting the same cellular pathway have emerged as a strategy to reduce drug resistance development. To determine whether additional benefits could be achieved by combining anticancer compounds with distinct killing mechanisms, we synthesized a suite of peptide-drug conjugates (PDCs) containing vemurafenib and peptides with either membrane-disrupt

Multidrug-resistant pathogens are the result of cumulative risk constellations and inappropriate antibiotic use. Multidrug-resistant Gram-negative bacteria are of particular relevance, especially in patients with prior antibiotic therapy, recurrent hospitalization, urinary drainage devices or a high density of invasive procedures. A central element of clinical management is the strict distinction between asymptomatic bacteriuria and clinically manifested urinary tract infections. Antimicrobial therapy is justified exclusively in the presence of a clinically manifested infection and, even in th

BACKGROUND: Isolation of individuals colonized or infected with multidrug-resistant organisms (MDROs) is an established strategy to prevent transmission in healthcare settings. Although protective measures such as single-room placement and use of personal protective equipment are designed to safeguard vulnerable patients, growing evidence suggests that community transmission contributes substantially to the spread of certain MDROs, raising questions about the proportionality and necessity of routine isolation. At the same time, isolation has been associated with adverse effects, including redu

Drug-resistant bacteria have become one of the greatest threats to human health in recent decades. With the large-scale abuse of antibiotics, bacteria resistant to traditional antibiotics are becoming increasingly common while clinical options for treating refractory infections are rapidly diminishing. Antimicrobial peptides (AMPs) as a promising alternative to traditional antibiotics, exhibit unique antibacterial mechanisms and potent antimicrobial activity. In recent years, numerous AMPs have been identified and validated in laboratory settings for their efficacy against the 'ESKAPE' pathoge

BACKGROUND: The intestinal microbiome influences health through communication with the brain, the immune system and gut. Travel exposes people to a different microbes and enteric pathogens that can affect health of the traveller and family that continue beyond the journey.
METHODS: PubMed was searched for microbiome changes, travellers' diarrhoea, acquisition of Enterobacteriaceae and multi-drug-resistant (MDR) microbiota during international travel.
RESULTS: When days are shortened during long-haul travel east and diet composition and timing are changed the gut microbiome experiences its fi

Pandoraea species are emerging multidrug-resistant pathogens increasingly associated with respiratory tract infections, particularly in cystic fibrosis patients. Despite their growing clinical relevance, these bacteria are underrepresented in the scientific literature. This review aims to consolidate existing evidence regarding Pandoraea species as emerging multidrug-resistant pathogens, with a focus on their taxonomy, diagnostic methodologies, antimicrobial resistance mechanisms, and treatment challenges. By identifying gaps in current therapeutic strategies and the limited clinical outcome d

The emergence and prevalence of antibiotic-resistant Pseudomonas aeruginosa strains have become a health priority, requiring novel treatment options. Among these, bacteriophages have regained attention in recent years due to their potential for combating bacterial infections and overcoming antibiotic resistance. Here, we present a new Phikmvvirus phage, with an icosahedral structure of 60-62 nm in diameter, which harbors a linear double-stranded DNA genome of 42,679 bp. The genome of Pseudomonas phage vB_Paphi_Mx1 contains 65 coding sequences (CDSs) related to nucleic acid metabolism, morphoge

Genomic alterations and enormous monoclonal immunoglobulin production cause multiple myeloma to heavily depend on proteostasis mechanisms, including protein folding and degradation. These findings support the use of proteasome inhibitors for treating multiple myeloma and mantle cell lymphoma. Myeloma treatment has evolved, especially with the availability of new drugs, such as proteasome inhibitors, into therapeutic strategies for both frontline and relapsed/refractory disease settings. However, proteasome inhibitors are generally not effective enough to cure most patients. Natural resistance

Salmonella is one of the main foodborne pathogens. Irrational antibiotic management has led to an increase in the incidence of multidrug-resistant strains. Bacteriophages may be an alternative method of food biopreservation and contribute to reducing the number of food poisonings requiring pharmacotherapy. This study aimed to isolate a bacteriophage (phage) targeting indigenous multidrug-resistant (MDR) Salmonella strains, followed by their biological, morphological, and genomic characterization. In this study we isolated Salmonella phage KKP_3822, targeting MDR Salmonella Manchester strain KK

Transport proteins are essential for bacterial resistance to antibiotics and toxins, but their mechanisms remain poorly understood in Bacillus subtilis. In the present study, overexpression of yoeA enhanced resistance to various antibiotics, with its expression induced by these antibiotics, especially penicillin and plipastatin. The ΔyoeA strain exhibited significant growth inhibition at 100 μg/mL of plipastatin, while as high as 10,000 μg/mL of iturin/surfactin are required to achieve comparable inhibition, suggesting a higher sensitivity of ΔyoeA to plipastatin. The transcript level of yoeA

Antibiotic and, more broadly, antimicrobial resistance is a naturally occurring biological phenomenon and a major public health problem. Mass emergence of drug-resistant bacterial strains was first observed in the mid-20th century. Since then, cases of resistance have been reported worldwide, and multidrug resistance has been increasingly reported over the past two decades. Overuse of antibacterial agents and their release into the environment contribute to the development of bacterial resistance. Unfortunately, a search and design of new effective antibiotics are declining, while it is necess

Antibiotic-resistant bacteria (ARB) and antibiotic-resistance genes (ARGs) are constantly shed into the aquatic environment, with hospital wastewater potentially acting as an important source for resistance spread into the environment. A systematic review was conducted aiming to investigate the role of hospital wastewater on dissemination of antimicrobial resistance in the aquatic environment. Studies included in the review compared the prevalence of ARB and/or ARGs in hospital versus community wastewater. Data were extracted on ARB and/or ARG prevalence. Data on sampling techniques, microbiol

The continuing rise of infections caused by multi-drug resistant bacteria has led to a renewed interest in bacteriophage therapy. Here we characterize phage vB_AbaM-KARL-1 with lytic activity against multi-drug resistant clinical isolates of Acinetobacter baumannii (AB). Besides genomic and phenotypic phage analysis, the objective of our study was to investigate the antibacterial outcome when the phage acts in concert with distinct antibiotics. KARL-1 belongs to the family of Myoviridae and is able to lyse 8 of 20 (40%) tested clinical isolates. Its double-stranded DNA genome consists of 166,5

- Fosfomycin is a broad-spectrum antibiotic agent used orally for uncomplicated cystitis. The intravenous form of administration has recently been authorised in the Netherlands.- Thanks to its broad spectrum and extensive tissue penetration, fosfomycin offers possibilities for the treatment of infections in different organs.- Infections with multidrug-resistant bacteria pose a significant threat to public health. Many of these multidrug-resistant bacteria are sensitive to fosfomycin, which means fosfomycin may be an option for the treatment of infections with multidrug-resistant bacteria. - Th

Integrons are versatile gene acquisition systems that allow efficient capturing of exogenous genes and ensure their expression. Various classes of integrons possessing a wide variety of gene cassettes are ubiquitously distributed in enteric bacteria worldwide. The epidemiology of integrons associated multidrug resistance in Enterobacteriaceae is rapidly evolving. In the past two decades, the incidence of integrons in enteric bacteria has increased drastically with evolution of multiple gene cassettes, novel gene arrangements and complex chromosomal integrons such as Salmonella genomic islands.
