Effect of a High-Fat Meal on the Pharmacokinetics of an Immediate Release Atogepant Tablet.

Atogepant, an oral calcitonin gene-related peptide receptor antagonist, is approved for the preventive treatment of migraine. A phase 1, open-label, single-dose, 2-period crossover study evaluated the effect of a high-fat meal on the pharmacokinetics and safety of atogepant in 20 healthy adults. Administration of atogepant 60 mg immediate-release (IR) tablets under fed conditions reduced the area under the plasma concentration-time curve (AUC) from 0 to time t and from 0 to time infinity by approximately 18% and reduced the maximum plasma concentration (Cmax) by 22%. The 90% confidence interva

Intermittent fasting and exercise therapy abates STZ-induced diabetotoxicity in rats through modulation of adipocytokines hormone, oxidative glucose metabolic, and glycolytic pathway.

Diabetes is a global, costly, and growing public health issue. Intermittent fasting (IF) and exercise therapy have been shown to improve insulin sensitivity (IS) in large studies, although the underlying processes are still unknown. The goal of this study, which included both nondiabetic and diabetic rats, was to look at the mechanisms of intermittent fasting and exercise in the management of diabetotoxicity. The effects of starvation and honey on the oral glucose tolerance test, insulin tolerance test, adipocytokines, oxidative glucose metabolic enzymes, glycolytic enzymes, food intake, and b

Neuropeptide B in Nile tilapia Oreochromis niloticus: molecular cloning and its effects on the regulation of food intake and mRNA expression of growth hormone and prolactin.

Neuropeptide B (NPB) regulates food intake, energy homeostasis and hormone secretion in mammals via two G-protein coupled receptors, termed as GPR 7 and GPR 8. However, there is no study that reports the function of NPB in teleosts. In this study, the full-length cDNA of prepro-NPB with the size of 663bp was cloned from the hypothalamus of Nile tilapia. The CDS of the prepro-NPB is 387bp which encodes a precursor protein with the size of 128a.a. This precursor contains a mature peptide with the size of 29a.a, and it was named as NPB29. Tissue distribution study showed that this gene was mainly

Glucose regulates the release of orexin-a from the endocrine pancreas.

Orexins (hypocretins) are novel neuropeptides that appear to play a role in the regulation of energy balances. Orexin-A (OXA) increases food intake in rodents, and fasting activates OXA neurons in both the lateral hypothalamic area and gut. OXA is also found in the endocrine pancreas; however, little is known about its release or functional significance. In this study, we show that depolarizing stimuli evoke the release of OXA from rat pancreatic islets in a calcium-dependent manner. Moreover, OXA release is stimulated by low glucose (2.8 mmol/l), similar to glucagon secretion, and inhibited b

Recent plant photosynthate as a driver of ecological and evolutionary processes.

Plants rapidly allocate carbon from photosynthesis belowground via root exudates and root-associated microorganisms. Here we synthesise recent evidence of our understanding of the consequences of this 'fast' pathway of recent photosynthate flux for major ecological and evolutionary processes. This rapid input of plant-derived carbon to soils shapes food webs and drives processes including soil CO2 efflux, nutrient cycling, plant community dynamics, stability of symbioses and evolution of contrasting modes of nutrition, such as parasitism and mycoheterotrophy. We also highlight how seasonality

Time-restricted eating: Watching the clock to treat obesity.

Time-restricted eating (TRE) has become a popular strategy to treat obesity. TRE involves confining the eating window to 4-10 h per day and fasting for the remaining hours (14-20 h fast). During the eating window, individuals are not required to monitor food intake. The sudden rise in popularity of TRE is most likely due to its simplicity and the fact that it does not require individuals to count calories to lose weight. This feature of TRE may appeal to certain individuals with obesity, and this could help produce lasting metabolic health improvements. The purpose of this review is to summari

[Enteral forced feeding using blind fixed stomach probes: two practical methods].

Whereas fasting is well tolerated by the healthy organism, protein-energy malnutrition in the sick animal has a negative impact on organ functions, immunocompetence, wound healing, and reconvalescence. Sick animals, physically unable to eat or held off food for medical reasons for more than 5 days, consequently need some sort of nutritional support. Force feeding through a gastrostomy tube has several advantages compared to other forms of enteral nutrition. Gastrostomy tube feeding is relatively physiological, is quite comfortable for the animals and their owners, can replace oral feeding for

Effects of isolated single fibers, fiber mixtures, and fiber-rich whole foods on glucose homeostasis in individuals with overweight and obesity: A systematic review and meta-analysis.

BACKGROUND & AIMS: Increasing evidence from epidemiological studies indicates that dietary fibers are protective against the development of type 2 diabetes mellitus. Supplementation of dietary fiber may therefore be a strategy to improve metabolic health in individuals with overweight and obesity at high risk to develop type 2 diabetes mellitus (T2DM). However, results from intervention studies are inconsistent, which may be related to the form of supplementation. In this systematic review and meta-analysis, we evaluated randomized controlled trials (RCTs) investigating the effects of dietary

Food intake and food choice: the role of the endogenous opioid peptides in the marsupial Sminthopsis crassicaudata.

Endogenous opioid peptides activate food seeking behaviour and influence macronutrient choice in a number of animal species and previous studies have suggested that the palatability of food is strongly modulated by the opioid feeding system. The effect of opioid peptides on appetite and food choice in marsupials has not been evaluated. The aim of these studies was to determine the effect of mu, delta and K opioid receptors on food intake and food choice in the marsupial Sminthopsis crassicaudata. When offered a choice of mealworms or laboratory diet after 24 h food deprivation, S. crassicaudat

Food poisoning due to a TSST1-producing Staphylococcus aureus.

Toxic Shock Syndrome (TSS) is a very rare and severe complication of Staphylococcus aureus infections. However, bacteremia is very uncommon in this disease. We present here the case of a healthy 15-year old boy who presented septic shock and diffuse exanthema four hours after eating in a fast food restaurant. Blood cultures were positive for a TSST-1 producing Staphylococcus aureus. The patient was treated with antibiotics and fully recovered.

[Research advances in liver diseases associated with parenteral nutrition].

For patients with gastrointestinal dysfunction or failure, although parenteral nutrition can maintain nutrition and save their lives, lack of food stimulus during the fasting period can cause abnormal secretion of gastrointestinal fluid, bile, and pancreatic juice, which may result in the abnormal secretion of a series of enzymes and lead to the development of various liver diseases. Recent research has focused on the prevention and treatment of such diseases, and this article reviews the research advances in recent years.

Are patients with Hirschsprung's disease prediabetic?

Hirschsprung's disease (HD) is considered a focal disease usually confined to the distal colon and rectum. However, autonomic dysfunction and dysmotility in the upper gastrointestinal tract have been reported, suggesting that this disease is not only confined to the distal gastrointestinal tract. This study examines the fasting and postprandial levels of glucose and insulin in adult patients with HD to elucidate whether there might also be an endocrine involvement in this disease. Sixteen patients with surgically treated HD during early childhood and 17 healthy subjects were studied. All subje

Mitochondria-targeted NIR fluorescent probe for detection of hydrogen sulfide and its application in monitoring food freshness and bioimaging.

BACKGROUND: The hydrogen sulfide (H2S) in spoilage of raw meat poses significant food safety risks to human health. Meanwhile, as a signaling molecule, H2S is crucial for maintaining human physiological homeostasis. Thus, the establishment of an efficient method for H2S detection is essential for safeguarding human health. RESULTS: In this work, a NIR fluorescent probe TPAW (emission at 690 nm) based on ICT effect for detection of H2S was constructed. TPAW exhibited fast response (<10 s), high selectivity, high sensitivity, good biocompatibility, and mitochondrial targeting ability. The testi

A fluorogenic capped mesoporous aptasensor for gluten detection.

Celiac disease is a complex and autoimmune disorder caused by the ingestion of gluten affecting almost 1% of global population. Nowadays an effective treatment does not exist, and the only way to manage the disease is the removal of gluten from the diet. Owing the key role played by gluten, clear and regulated labelling of foodstuff and smart methods for gluten detection are needed to fight frauds on food industry and to avoid the involuntary ingestion of this protein by celiac patients. On that scope, the development of a novel detection system of gluten is here presented. The sensor consists
