Impact of high-inoculum Staphylococcus aureus on the activities of nafcillin, vancomycin, linezolid, and daptomycin, alone and in combination with gentamicin, in an in vitro pharmacodynamic model.

We evaluated the impact of high (9.5 log10 CFU/g) and moderate (5.5 log10 CFU/g) inocula of methicillin-susceptible and -resistant Staphylococcus aureus (MSSA and MRSA, respectively) on the activities of nafcillin, linezolid, vancomycin, and daptomycin, alone and in combination with gentamicin in an in vitro pharmacodynamic model with simulated endocardial vegetations over 72 h. Human therapeutic dosing regimens for nafcillin, daptomycin, vancomycin, linezolid, and gentamicin were simulated. At a moderate inoculum, nafcillin (MSSA only), vancomycin, and daptomycin demonstrated equivalent and s

The prevalence of faecal carriage of ampicillin-resistant and high-level gentamicin-resistant enterococci among inpatients at 10 major Norwegian hospitals.

From March to October 1999, 854 patients hospitalized at 10 major Norwegian hospitals were screened for rectal carriage of ampicillin-resistant enterococci (ARE) and high-level gentamicin-resistant enterococci (HLGRE). A total of 59 ARE carriers (prevalence 6.9%, range 0-22% among hospitals) and 28 HLGRE carriers (prevalence 3.3%, range 1-11%) were detected. All ARE or HLGRE strains were susceptible to vancomycin, whereas 77% of the ARE isolates were resistant to ciprofloxacin. All the ARE strains were identified as Enterococcus faecium, and 48% of these were genomically closely related as sho

Protective effects of KW-3902, a novel adenosine A1-receptor antagonist, against gentamicin-induced acute renal failure in rats.

We investigated the possible renal protective effects of KW-3902 (8-(noradamantan-3-yl)-1,3-dipropylxanthine), an adenosine A1-receptor antagonist, against gentamicin (GM)-induced acute renal failure (ARF) in rats. ARF was induced by subcutaneous injection of GM at 80 mg/kg/day for 12 days. KW-3902 (0.001-0.1 mg/kg, p.o., twice daily) attenuated the increases of serum creatinine and urea nitrogen and the decrease of creatinine clearance in rats treated with GM. In contrast, furosemide and trichlormethiazide aggravated the GM-induced nephrotoxicity. These results suggest that KW-3902 can amelio

Combined Exposure to Polystyrene Nanoplastics and Gentamicin Promotes Liver Toxicity Through Oxidative Stress, Inflammation and Apoptotic Pathways: In Vivo and In Silico Studies.

Human exposure to nanoplastics and antibiotics is increasingly recognized as a major public health concern; however, the potential impacts of their combined exposure on human health remain poorly understood. In this study, we evaluated the hepatotoxic effects of polystyrene nanoplastics (PsNPs) and gentamicin (GEN), administered alone or in combination, and investigated the underlying molecular mechanisms using both in vivo and in silico approaches. Our findings demonstrated that co-exposure to PsNPs and GEN induces combined treatment response in liver by triggering oxidative stress, inflammat

β-Sitosterol attenuates gentamicin-induced nephrotoxicity via ADAM-17/ACE2/Ang 1-7/MasR Axis modulation in rats.

BACKGROUND: Gentamicin (Genta) is a widely used aminoglycoside antibiotic, but its clinical value is limited by nephrotoxicity involving oxidative stress, inflammation, apoptosis, and dysregulation of the ADAM-17/ACE2/Ang (1-7)/MasR axis. β-Sitosterol (BSST) is a phytosterol with reported antioxidant and anti-inflammatory properties, yet its nephroprotective activity and mechanistic link to this signaling pathway have not been explored. This study investigated whether BSST mitigates Genta-induced renal injury in association with modulation of this axis together with autophagy and apoptotic pat

Rosmarinic acid activates the Nrf2/HO-1 axis and suppresses NF-κB to protect against gentamicin-induced acute kidney injury.

BACKGROUND: Rosmarinic acid (RA), a polyphenolic ester abundant in Melissa officinalis, exhibits potent antioxidant properties. While the traditional use of M. officinalis for detoxification is well documented, the molecular mechanisms by which RA protects against drug-induced acute kidney injury (AKI), particularly whether it actively induces endogenous cytoprotective pathways, remain incompletely defined. PURPOSE: This study investigated whether RA activates the Nrf2/HO-1 antioxidant pathway to confer nephroprotection in a rat model of gentamicin-induced AKI, and explored additional anti-in

EDTA potentiates gentamicin and ofloxacin antibacterial activity against biofilms of methicillin-resistant Staphylococcus pseudintermedius derived from canine superficial pyoderma.

Canine pyoderma often results from infections caused by methicillin-resistant Staphylococcus pseudintermedius (MRSP), which frequently exhibits multidrug resistance and potent biofilm-forming capabilities. Therefore, veterinary clinicians have limited effective antibiotic options. This study aimed to investigate the potential of ethylenediaminetetraacetic acid (EDTA) combined with antibiotics commonly used to treat canine pyoderma (gentamicin and ofloxacin) on MRSP biofilms in vitro. We performed in vitro biofilm survival assays to determine whether EDTA enhances the efficacy of gentamicin and

Comprehensive biological evaluation of infuzide as a potent antimicrobial, alone and in combination with gentamicin, linezolid, and minocycline targeting MDR Staphylococcus aureus and Enterococcus sp.

Antimicrobial resistance is the greatest threat to modern healthcare systems. The unrelentingly increasing drug resistance manifests in ESKAPE pathogens, thus necessitating urgent novel drug discovery. Whole cell-screening assays were used to identify Infuzide, expressing potent antimicrobial activity against gram-positive pathogens Staphylococcus aureus and Enterococcus. To probe if Infuzide's inactivity against gram-negative pathogens was due to entry issues, its activity was determined in the presence of Polymyxin B nonapeptide, a known membrane permeabilizer. Furthermore, its activity agai

Differential Pre- and Post-Treatment Effects of Low-Dose Antidyslipidemic Drugs on Gentamicin-Induced Acute Nephrotoxicity in the Rat: A Histopathological and Biochemical Study.

Aminoglycoside-induced acute nephrotoxicity is a worldwide concern, while no effective preventive therapy exists. This study examined pre- and post-treatment effects of low doses of antidyslipidemic drugs such as fenofibrate (30 mg/kg/day, p.o.) and rosuvastatin (2 mg/kg/day, p.o.) in gentamicin (100 mg/kg/day, i.p.)-induced experimental acute nephrotoxicity in rats. Gentamicin-evoked nephrotoxicity was assessed via biochemical, molecular, and renal histopathological studies. Compared with normal rats, gentamicin-treated rats presented marked renal functional abnormalities with significant inc

Effect of mitochondrial coenzyme-Q10 precursor solanesol in gentamicin-induced experimental nephrotoxicity: Evidence from restoration of ETC-complexes and histopathological alterations.

Nephrotoxicity occurs when the body is exposed to certain drugs or toxins. When kidney damage occurs, the kidney fails to eliminate excess urine and waste. Solanesol (C45H74O) is a tri-sesquiterpenoid alcohol first isolated from tobacco, and it is widely distributed in plants of the Solanaceae family. Solanesol (SNL) is an intermediate in the synthesis of coenzyme Q10 (CoQ10), an antioxidant which protects nerve cells. This study investigated the protective effect of SNL at doses of 30 and 60 mg/kg in gentamicin-induced nephrotoxicity in Wistar albino rats. Animals were distributed into six gr
